Effects of Changes in Composition of Herb Extract Product on Growth of Spoilage Yeast, Zygosaccharomyces sp. (생약복방제의 조성 변화가 부패성 효모 Zygosaccharomyces sp. 의 성장에 미치는 영향)

  • The aim of the present study was to investigate effects of food preservative addition and changes in composition of herb extract product on the growth of spoilage yeast, Zygosaccharomyces sp. Herbs such as Panax ginseng, Cinnamomum cassia, Lycium chinense, Zyzyphus juiuba and Jingiber officinale were altogether put into water and essence was extracted at 80oC, and then the extract was concentrated at 75oC. The herb extract product was made by adding vitamins, amino acids and honey to the concentrated herb extract. The amount of gas produced from the herb extract product was increased as inoculated cell number increased but decreased as Brix concentration increased. Gases were produced in small amount when incubation was made at 4oC but large amounts of gases were produced at 25 or 40oC of incubation. The gas production and growth of Zygosaccharomyces sp. were measured after browning reaction was induced by heating at 85oC for 12 hours. It appeared that heating treatment did not induce any significant change in the gas production and growth of the cell. The effects of addition of various sugar to the herb extract produce were also invesigated. Amounts of gas production were in the order of glucose>sucrose>oligosaccharide>stevioside. The viable cell count was measured as 6.0$\times$107 CFU/g when glucose was added to the herb extract product. The viable cell counts were 5.0$\times$106, 3.0$\times$103, and 3.0$\times$102 CFU/g in sucrose, oligosaccharide and stevioside added herb extract product, respectively. The amount of gas production from the herb extract product was remarkably reduced by addition of such food preservatives as sodium benzoate and DF 100. TLC(thin layer chromatography) chromatogram of the herb extract showed stability of the herb extract in the above treatments.

Effect of Water Extract from Hordeum vulgare L. with Medicinal Herb on Plasma Lipid Status and Glucose in Rats Fed High Fat Diet (맥아와 한약재추출물이 고지방 식이를 섭취시킨 흰쥐의 혈장지질 및 혈당농도에 미치는 영향)

  • Objectives : This study was designed to investigate the effect of water extract from Paeonia Lactiflora Pasll. with medicinal herb on plasma lipid and glucose in rats fed high fat diet. Methods : Experimental groups were divided into control diet group(C), high fat diet group(HF), high fat diet and 5% water extracts from Paeonia Lactiflora Pasll. with medicinal herb group(HFE I), high fat diet and 10% water extract from Paeonia Lactiflora Pasll. with medicinal herb group(HFE II). Weight gain, food intake, food efficiency, plasma lipid concentration and glucose were determined for 10 weeks during high fat diet. Results : In this experiment, body weight and food efficiency were significantly increase in high fat diet. High fat diet group showed elevated levels of total cholesterol, LDL-cholesterol and free fatty acid. Consumption of 10% water extract Paeonia Lactiflora Pasll. with medicinal herb lowers level of total cholesterol, LDL-cholesterol and free fatty acid. But levels of plasma triglyceride, HDL-cholesterol and glucose did not affect by high fat diet and water extract Paeonia Lactiflora Pasll. with medicinal herb Conclusions : In these results, water extract Paeonia Lactiflora Pasll. with medicinal herb in high fat diet group decreased plasma total cholesterol, LDL-cholesterol and free fatty acid. Thus the Paeonia Lactiflora Pasll. with medicinal herb might be useful in the treatment of hyperlipemia.

Administration Duration Dependent Effects of Epimedium Herb Extract Solution on the Reproductive Capacities in the Mice (음양곽(淫羊藿)의 수컷 생쥐 생식능력(生殖能力)에 대한 투약기간별(投藥期間別) 효과(效果))

  • Purpose : This study was undertaken to evaluate the effects of the different administration duration of Epimedium Herb extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase. Methods : We used the 2-month-old mice and administered the extract solution of Epimedium Herb 0.3 ml/g/day for 30, 60, 90 and 120 days. The control group was administered the normal saline as the same way. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase. Also we observed changes of isolated testis before and after administration of Epimedium Herb extract solutions in the mice. And we compared the testicular tissue especially seminiferous tubules with the control and treated group by histochemical methods. Results : The significant differences were observed in the concentration of total sperm and normality of spermatozoa of the Epimedium Herb extract solution administered groups compared to the control group in 60, 90 and 120 days groups. The significant differences were observed the motility of the Epimedium Herb extract solution administered groups compared to the control group in 60 days group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Epimedium Herb extract solution administered groups compared to the control group, respectively. And the activity of hyaluronidase was significantly increased in the Epimedium Herb extract solution administered groups compared to the control group. Conclusion : This study shows that Epimedium Herb has the beneficial effect on the concentration, morphology and motility of sperm, the testicular tissues and the activities of sperm hyaluronidse in 60 days administration group. We can suggest that Epimedium Herb extract solution be useful for the treatment of male sexual dysfunction and infertility.

  • Objectives : The effects of cotreatment of adriamycin and ethanol extract of herb (Palgin-tang hab Hwajuck-hwan a traditional medicine for cancer treatment in oriental medicine) on the induction of apoptotic cell death were investigated in human liver origin cell lines, Chang. Methods : Chang(ATCC) liver cells were cultured in RPMI-1640(Gibco SRL Co, Gaithersburg, MD) badge including 10% fetal bovine serum. Chang liver cells were treated with various concentrations(from 10 to $0.16{\mu}l$) of adriamycin and herb extract(from 500 to $31.25{\mu}l$) After 48h later, the cells were tested for viability by Crystal violet staining assay. Adriamycin and Herb extract induced ladder pattern of DNA fragmentation in Chang cells. Genomic DNA was isolated and separated on 1.5% agarose gels. The DNA was stained with ethidium bromide and visualized under UV light. Results : The death of Chang cells was synergistically induced by the cotreatment of adriamycin and ethanol extract of herb. In addition, the cotreatment-induced cell death of Chang cells was mediated by apoptotic death signal processes. The phosphotransferase activity of JNK1 remained in a basal level in Chang cells which was treated individually with the adriamycin and ethanol extract of herb. However, it was markedly increased in Chang cells which was cotreated with adriamycin and ethanol extract of herb. In addition, the expression of Fas and FasL was markedly induced by the cotreatment of adriamycin and herb extract. For a while, the expression of Sax was a eminently increased by the ethanol extract of herb. However, Scl2 expression was not affected by the individual or cotreatment of adriamycin and herb extract. Conclusions : our results suggest that the cotreatment of adriamycin aM ethanol extract of herb induces synergistic apoptotis of human liver origin Chang cells via the upregulation of JNK, Fas, FasL and Bax.

Studies on the Safety Assessment of Red-ginseng Radix Extract Solution for Herb-acupuncture (III) (약침용 홍삼추출액의 면역독성, 발열성 및 용혈성시험에 관한 연구)

  • According to the Medical Product Safety Administration Guidelines for safety assessment guinea pigs, mice and rats were used for antigenicity test, rabbits for pyrogenic test and hymolytic test. The results were summarized as follows; 1. The antigenicity test of Red-ginseng radix extract solution for herb-acupuncture did not show any anaphylatic shock but produced IgE in high dose group. 2. The result of the pyrogen test of Red-ginseng radix extract solution for herb-acupuncture was negative. 3. The result of the hemolytic test of Red-ginseng radix extract solution for herb-acupuncture were negative in low and middle dose group but was positive in high dose group.

Quality Characteristics of Teriyaki Sauces containing Medicinal Herb Extracts

  • This study determined the effects of addition of medicinal herb extracts (Zanthoxylum piperitum extract(TZ), Acanthopanax sessiliflorus(TA), Panax ginseng(TP), Cinnamomum lureitri Nees(TC), Angelica gigantis radix(TAG) and Crataegii fructus extract (TCF) to teriyaki sauce with regard to taste and health benefit. After adding the medicinal herb extracts to teriyaki sauce, moisture content was found to be inversely proportional to the viscosity. The sauce with Panax ginseng extract(TP) had lowest sweetness, but highest salinity. The results showed that the sauce with Korean herb extracts had lower salinity content. In terms of colour value, the sauce with Crataegii fructus extract(TCF) had much higher lightness, redness, and yellowness than sauce prepared with other herb extracts. It had the strongest acidic taste and lowest pH. Sensory evaluation test revealed that the sauce with Anthoxylum piperitum extract(TZ) was the most preferred sauce among all the sauces tested. The degree of brown color of teriyaki sauces was correlated positively with taste preference(r=.570, p<0.01) and overall preference(r=.505, p<0.01).

  • The components of Red-ginseng radix extract solution for herb- acupuncture were analyzed by HPLC. According to the Medical Product Safety Administration Guidelines for safety assessment, mice and rats were used for acute toxicity test. The results were summarized as follows; 1. In the Saponin contents(%) of Red-ginseng radix extract, Ginsenoside $Rb_1$ Saponin was 0.27% in raw material, 1.67% in extract powder and Ginsenoside Rc Saponin was 0.16% in raw material, 1.12% in extract powder and Ginsenoside Rd Saponin was 0.08% in raw material, 0.54% in extract powder. 2. There were no abnormal findings in acute toxicity test treated with Red-ginseng radix extract solution for herb-acupuncture and $LD_{50}$ could not be measured.

Anti-oxidant and Hepatoprotective Effect of White Ginsengs in H2O2-Treated HepG2 Cells

  • The antioxidant activity of white ginseng was not recorded in Korea Functional Food Code, while its activity of red ginsengs was recorded. The aim of this study was to evaluate the antioxidant and hepato protective effect of different ginsengs in H2O2-treated HepG2 cells. White and red ginseng were prepared from longitudinal section of the same fresh ginseng (4-year old). The whole parts of white and red ginsengs were separately extracted with 70% ethanol and distilled water respectively, at 70 ℃ to obtain therapeutic ginseng extracts namely, WDH (distilled water extract of white ginseng), WEH (70% ethanol extract of white ginseng), RDH (distilled water extract of red ginseng) and REH (70% ethanol extract of red ginseng). In this work, we have investigated the DPPH, hydroxyl radical, Fe2+-chelating activity, intracellular ROS scavenging capacity and lipid peroxidation of different ginsengs. All these extracts showed a dose dependent free-radical scavenging capacity and a ROS generation as well as lipid peroxidation was significantly reduced by treatment with bioactive extracts of white ginsengs (WDH) than red ginsengs. Additionally, white ginseng extracts (WDH) has dramatically increased intracellular antioxidant enzyme activities like superoxide dismutase and catalase in H2O2-treated HepG2 cells. All these results explain that administration of white ginseng is useful as herbal medicine than red ginseng for chemoprevention of liver damage.

The Amelioration Effect of the Ethanolic Extract of Cnidium officinale in Mice with Imiquimod-induced Psoriasis-like Skin Lesion

  • Psoriasis is an auto-immune skin disease, which is characterized by the excessive generation of plaques on the skin with typically a long-lasting red, itchy and scaly symptoms. Imiquimod, which has been used for the treatment of external genital warts, actinic keratosis, and superficial basal cell carcinoma, induced of psoriasis-like skin disorders with skin erythema and thickness in mice. In the present study, we tried to find the bioactive herbal extract against imiquimod-induced psoriasis-like skin disorder in mice. During the searching of the herbal extract with anti-psoriatic effect, the ethanolic extract of Cnidium officinale ameliorated imiquimod-induced psoriasis-like skin disorder in mice. The morphological evaluation, H&E staining and Psoriasis Area and Severity Index (PASI) score showed that ear and back thickness, and erythema induced by imiquimod were significantly reversed after the treatment of the cream of the ethanolic extract of C. officinale. The overexpressed myeloperoxidase (MPO) and keratin 6A levels were decreased by the treatment of C. officinale cream. Also, $IFN-{\gamma}$, c-fos and $I{\kappa}B-{\alpha}$ mRNA levels, which are related to the progression of psoriasis, were reduced by C. officinale cream. Thus, the ethanolic extract of C. officinale ameliorated psoriasis-like skin disorder induced by imiquimod and might be the therapeutic agent for psoriasis.

Effects of Antioxidant and Quality Characteristics of Herb Soy Sauce (허브간장의 항산화성 및 품질 특성에 미치는 영향)

  • Herb soy sauce was prepared by extracting rosemary, lemon balm, spearmint and peppermint at a low temperature in fresh soy sauce. The antioxidant and physicochemical properties of herb soy sauces were examined. Herb soy sauces were prepared by adding 2.5% (w/v) of herbs to fresh soy sauce at 60℃. The total polyphenol, DPPH%, ABTS% increased with extracting time but five minutes extraction with sufficient antioxidant herb soy sauce could be prepared. The antioxidant powers of herb soy sauces were higher in lemon balm extract, and the rosemary extract showed the lowest. Soy sauces added with herbs had lower pH and salinity (p<0.05) and higher sugar content than without herbs. The colorness according to the type of herbs showed significant difference.
