Histological Indicator Change of the Abalone, Haliotis discus hannai Chronic Exposed to Zinc Chloride (ZnCl2) (염화아연 (ZnCl2) 만성노출에 따른 북방전복, Haliotis discus hannai의 조직학적 지표 변화)

  • This study was conducted to find out concentration of trace metal, survival rate, falling rate and structural changes of abalone, Haliotis discus hannai exposed to zinc chloride. Experimental groups were composed of one control condition and four exposure conditions (0.3, 0.8, 1.3, $1.8\;ZnCl_2\;mg/L$). The accumulation of trace metal (zinc) in abalone was significantly increased all exposure group. Though the exposure groups had lower survival rate than the control group, higher falling rate than the control group. Histopathological changes in the foot, gill and hepatopancreas of abalone revealed obvious with exposure group than control group.

  • A filamentous form of mollicute-like bacterium was detected during a routine health survey of wild adult kuruma shrimp Penaeus japonicus. The kuruma shrimp were native to Japan and were imported to Korea. The histology showed no pathological changes. Transmission electron microscopy revealed extensive infections of hepatopancreatic epithelial cell by a pleomorphic, filamentous intracellular bacterium. The filamentous bacterium was of about 60 nm in diameter and 300 nm to more than $1{\mu}m$ in length. Teh morphology of bacteria were filamentous and branched with terminal blebs, or knobs, on the branches. They lacked the cell wall, and were bounded by the plasma membrane. They contained typical prokaryotic ribosomes and fibrillar DNA-like strands. No additional internal structure has been observed. They are considered to be mollicutes, based upon the morphological appearance and upon the cellular location.

Optimum Feeding Rate in Growing Olive Flounder Paralichthys olivaceus Fed Practical Extruded Pellet at Optimum Water Temperature (21℃) (적수온(21℃)에서 사육한 성장기 넙치 (Paralichthys olivaceus)의 배합사료 적정 공급률)

  • This study was conducted to investigate the effects of feeding rate on growth performance, blood components and histology of growing olive flounder. The experiment was carried out to determine the optimum feeding rate of the fish (initial fish mean weight of $97{\pm}3.0$ g) at the optimum water temperature. Two replicated groups of fish were fed commercial diet at the feeding rates of 0, 1.0, 1.65 and 2.3% body weight (BW)/day and satiation. Feeding trial was conducted under a flow-through system with ten 1.2 metric ton aquaria receiving filtered seawater at $21{\pm}2^{\circ}C$ for three weeks. Weight gain and specific growth rate (SGR) for fish fed to satiation were significantly higher than those for the unfed fish and for fish fed at 1.0 and 1.65% BW/day. There were no significant differences in values of such parameters between fish fed at 1.65% and those fed at 2.3% BW/day and between fish fed at 2.3% and those fed to satiation. Weight gain and SGR for the unfed fish were significantly lower than those for fish in the other treatments. All the tissues (hepatopancreas, kidney and anterior intestine) were in good condition in fish fed the experimental diet at different feeding rates. This result indicates that the optimum feeding rates of olive flounder (97-160 g) was approximately 2.52% BW/day at the optimum water temperature.

Four Members of Heat Shock Protein 70 Family in Korean Rose Bitterling (Rhodeus uyekii)

  • Heat shock protein (HSP) 70, the highly conserved stress protein families, plays important roles in protecting cells against heat and other stresses in most animal species. In the present study, we identified and characterized four Hsp70 (RuHSP4, RuHSC70, RuHSP12A, RuGRP78) family proteins based on the expressed sequence tag (EST) analysis of the Korean rose bitterling R. uyekii cDNA library. The deduced RuHSP70 family has high amino acid identities of 72-99% with those of other species. Phylogenetic analysis revealed that RuHsp70 family clustered with fish groups (HSP4, HSC70, HSP12A, GRP78) proteins. Quantitative RT-PCR analysis showed the specific expression patterns of RuHsp70 family members in the early developmental stages and several tissues in Korean rose bitterling. The expression of 4 groups of Hsp70 family was detected in all tested tissue. Particularly, Hsp70 family of Korean rose bitterling is highly expressed in hepatopancreas and sexual gonad (testis and ovary). The expression of Hsp70 family was differentially regulated in accordance with early development stage of Rhodeus uyekii.

  • The histopathologieal effects of his(tri-n-butyltin)oxide (TBTO) on the flounder, Paralichthys otivaceus were examined by means of histological methods. The experimental fishes were exposed to 0.17, 0.36, 0.60, 3.20, 6.30, 12.50 ${\mu}g \;L^{-1}$TBTO concentrations for 42 days. Histopathological change of the fish exposed to TBTO is dependent on the exposure duration and concentration. In the lower concentrations early histological changes included activated mucous cells and chloride cells, capillary hyperemia and epithelial hyperplasia in the gill; hepatocyte activation, degeneration of bile duct and pancyeatic zymogen reduction in the hepatopancreas; and capillary hyperemia, appearance of eosinophilic cell and melano-macrophagocytes in the kidney. At the higher concentrations histological changes of dysfunctionality included epithelial lifting and deformation of the lamellae in the gill; pycnosis and cytoplasmic degeneration of hepatocyte; pycnosis of haemopoietic cell and deformation of renal tubules and glomerulus in the kidney. It is indicated that TBTO induced histopathological changes in the fish as other aquatic pollutants.

Development of Gonad and External Genitalia in Exopalaemon carinicauda (Decapoda: Palaemonidne) (밀새우 (Exopalaemon carinicauda)의 생식소와 교접기의 발달)

  • The morphological and histological changes of gonad in ridgetail prawn (Expalaemon carinicauda) were examined with the development of the external genitalia. The samples were collected monthly in Hampyung Bay on the western coast of Korea, from January to December 2003. The ovaries lie on the dorsal side of the female prawns thorax. The ovary cavity is located below the heart and above the hepatopancreas. The anterior part of the ovary is round and fused, and the posterior part has a pair of tubular structures. Testes are bilaterally symmetrical; the anterior part shows fused feature, and the posterior part consists of a pair of tubules. Based on histological examination, ripe eggs and spermatozoa were observed when mature females and males reach a length of about 14.9 and 11.5 mm CL, respectively. The immature female are almost same shape and structure with matured one in the endopod of first pleopod and appendix interna of the second pleopod, except the number of setae, and appear to be structurally complete after females reach a length of about 14.9 mm CL. The appendix masculina reaching beyond the distal end of the appendix interna of the second pleopod after males reach a length of more than 5.5 mm CL and a fully developed appendix masculina and appendix interna of the second pleopod appear up to a length of 11.5 mm CL.

EST-based Survey of Gene Expression in Seven Tissue Types from the Abalone Haliotis discus hannai

  • The analysis of expressed sequence tags (ESTs) is an efficient approach for gene discovery, expression profiling, and the development of resources for functional genomics. To analyze the transcriptome of the abalone Haliotis discus hannai, we conducted EST analysis using seven cDNA libraries made from gill, gut, hepatopancreas, skin, muscle, testis, and ovary. Redundant ESTs were assembled into overlapping contiguous sequences using the assembly program ICAtools. We found that the total 1,393 ESTs formed 135 clusters and 951 singletons, indicating that the overall redundancy of the library was 22%. Of the 1,393 clones, BLAST identified 1,278 clones (91.7%) as known genes; 115 clones (8.3%) did not match any previously described gene. Based on the major functions of their encoded proteins, the identified clones were classified into 16 broad categories. Sequence analysis revealed the presence of micro satellite-containing genes that may be valuable for further gene mapping studies. This study contributes to the identification of numerous EST clones that can be applied to further clarifying the genetics and developmental biology of abalone.

Studies on Effect of $CO_2$ Concentration in Air and Pb Concentration in Soil on Pillbug Growth and Bio-accumulation (대기 중 $CO_2$ 및 토양 중 Pb 농도 증가가 공벌레의 성장과 공벌레 체내 Pb 축적에 미치는 영향에 관한 연구)

  • In nature, the overall effect of heavy metals on the biota can be influenced by a number of environmental factors like soil characteristics and air pollution by elevated $CO_2$. Pillbugs(Isopoda, Armadillium vulgare) take up heavy metals with their food and store them mainly in the vesicles of hepatopancreas. They accumulate certain metals, occuring in relatively large numbers, are easily collected and identified. Therefore, it has been suggested that total body concentration of metals in pillbugs could be positively correlated to the levels of environmental exposure and that pillbugs could be used as biological indicators of metal pollution and global change by $CO_2$. The aim of the study is to determine effects of heavy metal concentrations in soil and elevated $CO_2$ on pillbugs'body accumulation of heavy metal and growth rate. In this study, pillbugs were collected at five sites (N=287) May 2006. Cu and Zn concentrations in pillbugs were higher than in soils (1.39-41.70 times) than in control. The high bioconcentration of lead in Sangam may be partly associated with reclaimed land uses. Pillbugs in low $CO_2$ and Pb condition showed higher growth rate than in elevated $CO_2$ and Pb condition.

The samples were collected monthly around the Ochongdo area on the western coast of Korea, from April 1997 to December 1998. Thelycum appear structurally complete after females reach a length of about 12.0 mm CL. Petasma rarely Join after males reach a length of 10.6 mm CL and a fully developed petasma shows up to a length of 12.0 mm CL. The ovaries lie on the dorsal side of the female shrimps thorax. The ovary cavity is located below the heart and above the hepatopancreas. The anterior part of the ovary is round and fused, and the posterior part shows a pair of tubule-like structures. Testes are bilaterally symmetrical, the anterior part shows a convoluted tubule, and the posterior part consists of a pair of tubules. In histological observations, ripe eggs and spermatozoon show when mature females and males reach a length of about 12.0 mm CL.

Experimental groups were composed of one control condition and five nickel chloride exposure conditions (5.9, 8.8, 13.3, 20.0, 30.0 mg/L). The accumulation of nickel in abalone was significantly increased all exposure group. Though the exposure groups had lower survival rate than the control group, higher falling rate than the control group. Histopathological changes in the foot, gill and hepatopancreas of abalone revealed obvious with exposure group than control group.
