  • This study conducted to investigate the effects of dietary Korean medical herb residue on egg quality and serum cholesterol of laying hens under heat stress. One hundred forty four, 41 weeks old ISA brown commercial layer, were used in a 28 d growth assay with a 7d adjustment period. Dietary treatments Included 1) Control (basal diet), 2) KMHR1.0 (basal diet+1.0% korean medical herb residue), 2) KMHR2.0 (basal diet+2.0% Korean medical herb residue). For overall Period, laying hens fed KMHR1.0 diet were higher hen-day egg production than laying hens fed Control and KMHR2.0 diets with significant difference (quadratic effect, p<0.01). Egg weight, egg shell breaking strength and egg shell thickness were not influenced by Korean medical herb residue supplementation (P>0.05). As adding level of Korean medical herb residue increased in the diets, yolk color tended to increase, however, the differences failed to reach significance (P>0.05). Also, egg yolk index was not influenced by korean medical herb residue supplementation (P>0.05). Total-, HDL- LDL+VLDL-cholesterol and triglyceride concentrations in serum tended to increase as the concentration of Korean medical herb residue in the diets was increased without significant difference (P>0.05). In conclusion, supplementing 1.0% Korean medical herb residue to a corn SBM meal diet for laying hens increased hen-day egg production.

  • This study was conducted to develop a new synthetic breed of Korean native chicken. The combining ability and reciprocal effects for production traits were estimated on 1,157 hens from a $5{\times}5$ diallel cross-mating design using grand parent stock (GPS) lines of Korean native chicken. Body weight, viability, age at first egg laying, egg weight, and hen-day egg production were measured and analyzed. The results showed that the general combining ability (GCA) of the survival rate during laying periods was -9.6 to 11.1, with the highest value obtained in the W strain. Additionally, the GCA of the body weight at 12 weeks was -209.7 to 162.2, with the highest value obtained in the F strain. The GCA for age at fist egg laying was estimated to be -2.8 to 3.7, while the GCA of egg weight was -0.91 to 0.96, and the GCA of hen-day egg production was -4.9 to 6.0. In the estimation of specific combining ability, the YW combination showed the highest survival rate, FW showed the highest body weight at 12 weeks, and GW showed the highest hen-day egg production. The reciprocal effects were significantly different among crosses for almost all productivity traits. In identical breeding combinations, differences in ability were observed when the maternal or paternal breeds were switched. The mean value based on combining ability was higher in WY, WF, and GW combinations for survival rate; GF, HG, and HF combinations for body weight at 12 weeks; and GW, YW, and FW combinations for hen-day egg production. It is concluded that the GF and HF combinations, which have excellent growth performance and moderate survival rate, are the most desirable paternal parent stock (PS) strains, and the GW and FW combinations, which have great laying performance and moderate body weight, are the most desirable maternal PS strains.

  • The aim of the study was to evaluate the effect of sodium nitrate consumption on egg quality and quantity, and some blood parameters of native breeder hens of West Azerbaijan province. One hundred native hens were used from wk 25 to 32 of age. These birds were divided into two groups. One group was fed the control diet (CD) but the other fed the same diet supplemented with 4.2 g/kg sodium nitrate (ND). After 2 wks of adaptation, eggs were collected daily and egg mass and egg production were measured weekly for five weeks. To assess the egg quality parameters, two eggs from each replicate pen were collected for three consecutive days each week. At the end of experimental period (wk 32 of age), blood samples of 5 birds per replicate were collected from the wing vein into anticoagulant tubes. Dietary sodium nitrate didn't affect the egg production, shell stiffness, shell thickness and Haugh unit (p>0.05) but it decreased the both egg production and egg mass during the last three weeks (wks 30, 31 and 32) (p<0.05). Furthermore, a treatment effect was observed for yolk colour (p<0.05). Both the egg production and egg mass were increased over time (p<0.05). No significant treatment${\times}$time interaction was observed for egg weight, egg production and egg mass (p>0.05). No effect of time or treatment${\times}$time were observed for shell stiffness (p>0.05). Over time, shell thickness was decreased while Haugh unit increased (p<0.05). None of the blood TP and TG or the activity of ALT, AST and LDH enzymes were affected by dietary consumption of sodium nitrate at wk 32 of age (p>0.05). Sodium nitrite decreased both the TAC and TC at wk 32 of age (p<0.001). It was concluded that the lower body antioxidant capacity of nitrate fed birds resulted in the lower performance (egg weight, egg production and egg mass).

  • The eggshell, which is a complex and highly ordered structure, is very important factor for food safety and egg marketing. This study investigated the changes in eggshell structure and shell components in relationship to hen age. For this study, we examined the histological change of the endometrium of the 30-, 60-, and 72-wk-old commercial layers, and analyzed the ultrastructure and ionic composition of their eggshells. The results showed that histological deformation, fibrosis, atrophy and elimination of micro-villi in the uterus endometrium were found through microscopic observation that was associated with increasing hen age. Concentration of blood-ion components such as $Ca^{2+}$, $Na^+$, $K^+$, and $Cl^-$ ions did not change with age. Along with the results from the ultrastructure analysis of the eggshell, the palisade layer ratio and the density of mammillary knobs were significantly decreased in older hens. In addition, the type B mammillary knobs were frequently observed with increasing hen age. In the mineral element assay from the eggshell, $Ca^{2+}$, $S^{2-}$, and $Co^{2+}$ significantly decreased with increasing hen age, whereas $Na^+$, $K^+$, and $V^{2+}$ significantly increased. Therefore, the damages of endometrial tissue inhibit the processes of ion transmission and the crystallization of eggshell formation, resulting in a large and non-uniform mammillary knob formation. This means the conditions of endometrial cells affect the formation of the eggshell structure. In conclusion, hen aging causes the weakness of the eggshell and degrades the eggshell quality.

  • Many studies on crystal growth mechanisms of the hen egg-white lysozyme protein crystals have mainly performed by optical microscopy and atomic force microscopy (AFM). As results, two types of growth mechanisms, which are a two-dimensional nucleation mechanism and a spiral growth mechanism, were identified. However, there was no direct evidence of grown-in screw dislocations at the spiral sites. We first observed the screw dislocations in tetragonal lysozyme crystals using synchrotron X-ray topography. In addition, to confirm the characteristics of dislocations, we have observed some elastic constants in lysozyme crystals in terms of the sound velocity measurement by pulse echo methods. Tetragonal hen egg-white lysozyme crystals were grown by the concentration gradient method. The crystals were grown in test tubes, with an inner diameter of 8 ㎜ and 80 ㎜ in length, held vertically. The test tubes were kept at 23C for 2 weeks. The maximum size of crystals were 3×3×4 ㎟. The high quality crystals were examined by Laue topography with a water filter using synchrotron radiation. Figure is a X-ray topograph. Several straight screw dislocations were observed. We also determined Burgers vector to be a [110] direction. The measurement of sound velocity was performed by the digital signal processing method. the crystals were placed in stainless steel vessel, which was filled with lysozyme solution used for crystal growth. We observed the longitudinal sound velocity along the [110] direction in the tetragonal is obtained to be 1817 ㎧. Therefore, Young modulus and shear modulus were evaluated to be 2.70 Gpa and 1.02 Gpa, respectively, if we assumed Poisson ratio is 0.33. These results will be discussed at the meeting.

  • The effects of several variables on the refolding of hen egg white lysozyme have been studied, Lysozyme was denatured in both urea, and guanidine hydrochloride(GuHCl), and batch refolded by dilution (100 to 1000 fold) into 0.1 M Tris-HCI, pH 8.2 mM EDTA 3 mM reduced glutathione and 0.3 mM oxidised glutathions. Refolding was found to be sensitive to temperature, with the highest refolding yield obtained at 50$\^{C}$. The apparent activation energy for lysozyme re-folding wasf ound to be 56kJ/mol, Refolding by dilution results in low concentrations of both de-naturant and reducing agent species. It was found that the residual concentrations obtained dur-ing dilution(100-fold dilution:[GuHCI]=0.06 mM, [DTT]=0.15 mM) were significant and could inhibit lysozyme refolding. This study has also shown that the initial protein concentration (1-10mg/mL) that is refolded is an important parameter. In the presence of residual GuHCl and DTT higher refolding yields were obtained when starting from higher initial lysozyme concentra-tions. This trend was reversed when residual denaturant components were removed from the re-folding buffer.

  • An experiment was conducted to investigate the effects of feeding fermented food waste on the egg production and egg qualities in laying hens. A lot of 30 commercial layer(ISA Brown) at the age 58 weeks were placed in individual of 80% commercial feed and 20% fermented food waste(C80%+F20%), a mixture of 50% commercial feed and 50% fermented food waste (C50%+F50%), and a mixture of 20% commercial feed and 80% fermented food waste (C20%+F80%). Daily measurements were made on feed intake, egg production rate, egg weight, yolk color, Haugh unit, shell color during the experimental period. At the end of the experiment, body weight change and egg cholesterol contents were determined. The results indicate that up to 50% of basal diet could be supplied by fermented food waste with little depression in feed intake and efficiency in egg production (p<0.01). Egg weight, egg shell thickness and Haugh units were not significant different between the treatments and the control. Egg color quality improved with increasing the proportion of the fermented food waste in the diet.

  • The objective of this study was to develop a user-friendly computer model for economic analysis on the commercial egg production that could help the egg farmer make managerial diagnosis and rational decision in the changing environment. To raise the adequacy of the model, the program was run for every sample and adjusted to fit the data. The model, programmed with Microsoft QuickBASIC, was a user-friendly computer program in supporting the Korean language. The basic analytical tool used in the study was an engineering-type computerized simulation model which incorporates a cost-benefit analysis of a full-time egg farmer. The computer model developed in this study may be the powerful analytical tool used to evaluate both a managerial decision whether to alter the production system and its impact on production, costs, revenue, and profits. Ultimately, the program is expected to enable the egg farmer to make managerial planning and diagnosis. The program can also calculate the values of economic variables at user-chosen incremental values of market eggs and feed prices. It provides the information on the profit and cost. This may lead the egg farmer, by allowing to establish the best managerial strategy, to increase the profit aor to lessen the cost. The results of this study could be utilized in the evaluation and improvement of the management. It also may be utilized for the researchers and guiding farmers in collecting and analyzing the data on the laying hen. In particular, such a program would be potentially useful to researchers who wish to quickly estimate profits associated with various laying hen treatments. The program could also benefit the egg farmer interested in making managerial decisions based on either current or predicted market conditions. The model would make the egg farmer respond actively to the information-oriented society by promoting to use personal computer.
