Shifts in Protein Metabolism in Hemolymph and Fat Body of the Silkworm, Bombyx mori L. in Response to Fluoride Toxicity

  • Changes in protein metabolism were studied in hemolymph and fat body on days 1, 3, 5 and 7 of the fifth-instar silkworm, Bombyx mori, exposed to lethal, sublethal doses and prevailing levels of fluoride in groundwater in Karnataka and Andhra Pradesh States of India. The total protein content indicated a depletion followed by a concomitant increase in accumulation of free amino acids. Concurrently, the activity of protease in both of the tissues was also increased. A steady enhancement in the activities of alanine aminotransferase and aspartate aminotransferase paralleled the elevation of glutamate dehydrogenase activity in the tissues studied. It is presumed, on the basis of these results, that the fluoride toxicity causes major changes in protein metabolism of the silkworms.

  • The fine structure of the hemocytes in Wolf spider, Pardosa astrigera, is discribed and compared with that of similar cells in other spider species and insects. Five hemocyte types are identified in the hemolymph: prohemocyte, plasmatocyte, granulocyte, spherulocyte and adipohemocyte. Prohemocytes are small with a relatively undifferentiated cytoplasm. The nucleus is comparatively large and has a perinuclear space. Plasmatocytes and granulocytes are pinocytotic function in the hemolymph of the body. The plasmatocytes have some coated pits on the plasma membrane and well developed Golgi complex, The granulocytes appear sequence of events in the formation of coated vesicle from a coated pit on its plasma membrane. Golgi complex become well expressed and give rise to small secretory vesicles which fuse to large bodies. The spherulocytes are larger in cell size than other hemocytes. Their cytoplasm is filled with spherules. The spherules contain the floccurent materials and the helical structured materials, which are 220nm in length and 80nm in width. The adipohemocytes are oval shaped and have a number of lipid droplets.

  • Since 1975, the studies on in vitro rearing Trichogramma spp., Anastatus japonicus Ashmead, Telenomus dendrolimusi Chu, Dibrachys cavus Walker, Habrobracon hebetor(Say), Bracon greeni Ashmead have been conducted successfully in China. The simulated “host-eggs" are made of polyethylene or polypropylene semispherical capsules, containing artificial diets, in which insectan pupal hemolymph is the main component. Manufacture of simulated “host larvae" are made of small rectangular parafilm of cotton-paper bags, containing artificial diets with insectan pupal hemolymph as the main component. Mass production of in vitro reared Trichogramma spp. and Anastatus japonicus and its utilization in the fields showed good effectiveness in controlling cotton bollworm, pine caterpillars, sugarcane borers and litchi stink bug.

  • We have purified and characterized cecropin A-like antibacterial peptide from the hemolymph of immunized Galleria mellonella larvae. Acid extraction, gel filtration, preparative acid urea PAGE, and reversed-phase HPLC were used for purification of peptide. The molecular mass of the purified peptide was estimated to be 4160.68 Da by MALDI-TOF mass spectrometry. (omitted)

Phenotypic Characteristics of New Mutant, Non-molting f(nm-f) of Bombyx mori (신돌연변이 불면잠 nm-f의 유전형질)

  • Studies were carried out to investigate the phenotypic characteristics of the non-molting mutant (nm-f) which was mapped on the 2nd linkage group. The results obtained were as follows : The nm-f mutant was distinguishable in the 3rd day of hatching. About 80 percentage of the non-molting mutant larvae died at the first instar within 10 days of hatching. The remaining larvae survived to the 2nd ad the 3rd instar but did not live to the final instar. There was no difference in non-molting nutant manifestation between hibernating and artificial hatching eggs. As a result of hemolymph protein analysis, the protein content on nm-f mutant was less than the normal larvae's. Therefore, we conclude that the characterization of nm-f is similar to the already known strains of non-molting mutant and the shortage of hemolymph protein is closely related to the non-molting characteristic in nm-f.

  • Two molecular species of apolipophorin-III (spoLp-III) were purified from the last instar larval hemolymph of Galleria mellonella by gel permeation chromatography (Sephadex G-100), ion exchange chromatography (DE-52), heat treatment (90C for 30 min) and Mono S FPLC, and were named apoLp-III-a and apoLp-lll-b, respectively. They were indistinguishable by SDS-PAGE but could be separated by native PAGE. The molecular mass of apoLp-III determined by SDS-PAGE was approximately 18 kDa. The N-terminal amino acid sequence of apoLp-III-b revealed high similarities with the apoLp-III from Manduca sexta.

  • Hemolymph SHBP (hJHBP) was partially purified from last instar larvae of Bombyx zori by gel filtration and their optimal reaction conditions of dextrin coated charcoal binding assay were determined. Dissociation constant (KD) of hJHBP for JH III was calculated to be 1.45 $\times$ 10-7 M at $4^{\circ}C.$ The molecular weight of hJHBP was estimated to be 30 kDa by gel filtration on a calibrated Sephadex G-100 column and 33 kDa by SDS-PAGE. These results indicate that hSHBP consists of a single polvpeptide chain. Isoelectric point of hJHBP was found to be pH 5.1 and 19 of the first 20 amino acid residues were determined from N-terminus of purified hJHBP.
