Isolation and Culture Characteristics of a Bacterial Symbiont from Entomopathogenic Nematode Steinernema galseri (Steinernema glaseri 곤충병원선충으로부터 공생박테리아의 분리 및 배양특성)

  • Asymbiotic bacterium with highly effective toxins was isolated from entomopathogenic nematode Steinernema glaseri which has been widely used against various soil-inhabiting pests. The symbiont of S. glaseri was identified as Xenorhabdus nematophilus sp. by using several biochemical and physiological tests. When this strain was released into the hemolymph of insect larva, it produced highly toxic substances and killed the larva within 2 days. Two colony forms that differed n some biochemical characteristics were observed when cultures in vitro. Phase l colonies were mucid and difficult to be dispersed in liquid. Phase II was not mucoid and was easily dispersed in liquid. It did not adsorb neutral red or bromothymol blue. Rod-shaped cell size was highly variable between two phases, ranging 2-10 ${\mu}{\textrm}{m}$. It was also found that only infective-stage nematodes can carry only primary-phase Xenorhabdus in their intestine.

  • Ventral nephrocytes in the larva of the Lucilia illustris comprise ellipsoid cells situated onto the salivary glands. The cells are $60{\sim}100{\mu}m$ in diameter. Junctional complex beneath the basement membrane hold the plasma membrane in a even contour. Intracellular channels from the juntion complex are well developed at the cortex part of the cell. Coated vesicles pinched off from the channels seems to be connected with the ${\alpha}$-vacuoles via the tubular elements, which is regared as selective absorption system from the hemolymph. Two nuclei are sometimes observed in the medulla part of the cell. Ventral nephrocytes contain well-developed rough endoplasmic reticulum and Golgi complex, and numerous mitochondria. These cellular organelles synthesize lysosome. The lysosome not only digest some cell organells but also seems to be related with the ${\beta}$-vacuoles.

Recent Advances in the Innate Immunity of Invertebrate Animals

  • Invertebrate animals, which lack adaptive immune systems, have developed other systems of biological host defense, so called innate immunity, that respond to common antigens on the cell surfaces of potential pathogens. During the past two decades, the molecular structures and functions of various defense components that participated in innate immune systems have been established in Arthropoda, such as, insects, the horseshoe crab, freshwater crayfish, and the protochordata ascidian. These defense molecules include phenoloxidases, clotting factors, complement factors, lectins, protease inhibitors, antimicrobial peptides, Toll receptors, and other humoral factors found mainly in hemolymph plasma and hemocytes. These components, which together compose the innate immune system, defend invertebrate from invading bacterial, fungal, and viral pathogens. This review describes the present status of our knowledge concerning such defensive molecules in invertebrates.

Genetic Relationships of Silkworm Stocks in Korea Inferred from Isozyme Analyses (동위효소 다형특성에 의한 누에 품종의 유연관계)

  • Isozyme was used to characterize general protein patterns of genetic relationships among 303 silkworm stocks preserved in National Sericultural and Entomology Research Institute, RDA. Six isozymes (esterase, acid phosphatase, alkaline phosphatase, amylase, glucose-6-phosphate dehydrogenase and sucrase) from hemolymph, midgut, and digestive juice were employed to construct dendograms(UPGMA method) using a polycrylamide gel electrophoresis. A cluster analysis revealed four major group, which were divided into several subgroups within each group, contained assemglages of Japanese and Chinese races. Especially, genetic differentiation in the first and second group was greatest rather than within Japanese and Chinese races repectively and was concordant with the hypothesis of phyletic sorting of initial variability in China many years ago. Hypothesized recent introgression between groups was also plausible, but the eviednce suggested bidirectional gene flow between the Chinese and the Japnaese lineages. Interpreting the results in light of evidence from the current study, the genetic diversity and relationship showed in Korean silkworm race, Hansammyun reflected early and independent evolution from the Chinese ancestor, limited addition of new variability and phyletic sorting within Korean peninsula more than 4,000 years.

Identification and Purification of Juvenile Hormone Binding Protein from nyphantria cunea Drurv (미국흰불나방(Hyphuntrio cuneo D.)의 유충호르몬 결합단백질의 확인 및 정제)

  • We identified juvenile hormone binding protein (JHBP) from last instar larval hemollvnph of Hvphantria cunea using gel filtration and non-SDS PAGE. Two kinds of JHBP in hemollnnph were found at two peaks by gel filtration (Sephadex G-100) and also at Rm values of 0.13 and 0.57 by non-SDS PAGE. JHBP was partially purified using anion exchange chromatosraphv, preparative gel filteration, and preparative PAGE. Dextrin coated charcoal (DCC) binding assay was employed to monitor the location of JHBP in chromatographic profile during the purification process. Purity of JHBP was checked by silver staining of 1091 SDS-Polyacrvlamide.

  • Male-specific protein (MSP) is a soluble protein which is accumulated in high amounts In the hemolymph and other organs of adult male wax moth. The MSP was purified from adult male wax moth by gel filtration and reversed Phase column chromatography, and its amino acid sequence was determined. Three internal amino acid sequences of MSP were obtained by the in-gel digestion method using trypsin because of its blocked N-terminus. (omitted)

  • In order to understand effect of polycyclic aromatic hydrocarbon (PAH) on shell repair of the Pacific oyster, Crassostrea gigas, shell regeneration experiments were carried out using oysters drilled a hole on the right valve. The change of pH and hemocytic characteristics in both extrapallial fluid and hemolymph were observed during the shell repair. The thickness of mantle tissue was apparently decreased, while necrosis in epithelium and periostracal gland was increased in response to PAH exposure. Our finding suggested that PAH could adversely influence on shell repair.

  • The insecticidal compound priduced by Streptomyces SS-4993 strain was indicated to be a relatively stable small compound by pancreatin treatment. The active compound was adsorbed on HP20 and eluted with 70% acetone. Then, the eluate was extracted with ethyl acetate. The content of the extract was determined by thin layer chromatography (Silica gel 60 F254, hexane-ethyl ether-acetic acid,80 : 20 :1). Three active bands (Rf 0.08, 0.07, 0.88) were identified by dermal application using Bombyx mori larvae. Electrophoretic analysis revealed that the injection of strain SS-4993 culture broth into B. mori larvae induce the reduction of the hemolymph proteins level. Especially, the level of apolipophorin-I was decreased drastically.

Cucurbitacin B Suppresses Hyperglycemia Associated with a High Sugar Diet and Promotes Sleep in Drosophila melanogaster

  • Secondary metabolites enable plants to protect themselves from herbivorous insects. Among these, cucurbitacin B (cuc-B) is a bitter-tasting compound with promising pharmacological potential. Dietary exposure to cuc-B lowered the hemolymph glucose levels of Drosophila melanogaster fed with a high carbohydrate diet, which is homologous to high blood glucose in humans, and its effect was comparable to that of metformin, a well-known glucose-lowering drug. Furthermore, cuc-B reduced tissue sugar levels and glycogen levels, as well as triacylglycerol levels. Our results thus highlight the potential applicability of this compound to treat chronic metabolic diseases such as diabetes and obesity. Additionally, we analyzed sleep quality and taste-associative memory enhancement after cuc-B and metformin treatment. Both supplements increased nighttime bout length and metformin increased memory consolidation. Therefore, discarded shell of Cucurbitaceae could be processed into health supplements.

Studies on the Effects of various Levels of Protein in the Artificial Diet on Nutritional Physiology of the Silkworm, Bombyx mori (인공사료의 단백질수준이 가잠의 영양생리에 미치는 영향에 관한 연구)

  • Larvae of the silkworm (Bombyx mori L.) were reared during the 5th instar on the four kinds of artificial diets on the basis of the different amounts of soybean meal used as the protein source. In this experiment it was shown that the various levels of protein in the diet affected not only the growth and silk production but the digestibility of the diet. haemolymph protein and uric acid excretion. The results obtained are summarized as follows; 1. By an increase of the level of protein in the diet the apparant digestibility was increased. but the protein digestibility was comparatively decreased. 2. Larval body weight increment was not observed by the 3rd day of the 5th instar, but was increased from the 4th day as the level of protein was increased in the diet. 3. After the 3rd day of the 5th instar, protein content in the hemolymph was increased steeply by an increase of the protein content in the diet. However, the percentage of hemolymph protein to the ingested protein was decreased from the 2nd day of the 5th instar and increased more or less after the 4th day. 4. An increase of the uric acid excretion was observed as the content of protein in the diet was increased but the pattern of the uric acid excretion was different between high and low-protein diet. However, the percentage of the uric acid excretion to the ingested protein and to the hemolymph protein were both decreased steeply after the 2nd day of the 5th instar. 5. It was also evident that the high-protein diet increased the cocoon productivity. 6. It showed that the feed efficiency for body weight increment and silk formation was high by an increase of the level of protein in the diet, but the protein efficiency was not.
