C57BL/6 마우스에서 나노밀 가공된 초미세분말(상황버섯)의 식이 안전성 연구 (The Food Safety of Superfine Powder (Phellinus linteus) Processed by Nanomill in C57BL/6 Mice)

  • 상황버섯(Phellinus linteus)은 건강 증진 및 질병의 치료를 목적으로 널리 사용되는 약용버섯으로서 항산화효과와 항암효과를 비롯한 다양한 생물학적 활성을 갖는다. 상황버섯은 주로 추출물이나 분말의 형태로 복용되고 있으며, 최근 분쇄기법의 발달로 초미세분말 형태의 제품이 상용화되면서 식이 안전성에 대한 연구의 필요성이 제기되었다. 본 연구는 상황버섯 초미세분말과 일반 미세분말을 동물에게 먹인 후 입자의 크기에 따른 식이안전성을 평가하기 위해 실시되었다. 부피평균에 의한 입도분석 결과, 상황버섯 초미세분말(SPL)과 미세분말(FPL) 입자의 평균지름은 각각 11.8 ${\mu}m$와 216.1 ${\mu}m$였고, d (0.5)는 각각 5.5 ${\mu}m$와 147.9 ${\mu}m$였다. 동물의 체중, 사료 섭취량 및 각 기관의 무게를 관찰한 결과 SPL군은 정상군(N)이나 FPL군과 비교하여 통계학적 차이를 나타내지 않았다. 혈액학 및 생화학적 검사에서는 ALT(N vs. FPL, P<0.001)와 BUN(N vs. FPL, P<0.01; N vs. SPL, P<0.01)이 정상군과 비교하여 차이를 보였으나 FPL군과 SPL군 사이의 유의성은 발견되지 않았으며, 그 밖의 다른 검사항목들은 모두 정상범위 내에 포함되었다. 또한 조직학적 검사 결과에서도 비정상적인 소견은 관찰되지 않았다. 이 연구 결과는 초미세 가공된 상황버섯을 경구투여할 경우 혈액학, 생화학, 조직학적인 측면에서 동물에게 유해하지 않다는 것을 말해준다. 그러나 다량의 초미세입자가 소화기와 장기간 접촉할 경우 일어날 수 있는 조직학적 유해성에 대한 심도 깊은 연구는 필요할 것으로 판단된다.

복합나트륨염의 Salmonella gallinarum 감염 육계에 대한 치료효과 (Therapeutic Effect of a Sodium Salt Mixture Against Salmonella gallinarum Infection in Broiler)

  • The objectives in the present study were to evaluate the therapeutic effect of a sodium salts mixture against Salmonella gallinarum infection in broiler. Of this study, sixty broilers at two weeks of age (body weight, $450{\pm}35$ g) were used to estimate the efficacy of a sodium salts mixture (3.25 mg sodium azide, 2.45 mg sodium cyanide, 0.8 g sodium chlorate) against Salmonella gallinarum infection in broiler with drinking water. Broilers challenged with S. gallinarum were administered with ${\times}$ 1(group I) and ${\times}$ 2(group II) sodium salt mixture for seven days, and cecal content samples were collected at the gate of treatment and on 1st, 3rd, 5th, and 7th day after administration. Changes in body weight and cecal shedding of S. gallinarum were monitored during the experimental period. All groups treated with the sodium salt mixture slightly increased body weight compared to control group but there is no significant difference. At 7th day after administration, the number of S. gallinarum in group I and II was significantly decreased compared to control group (p < 0.001). In the hematological and blood biochemical analysis, values of parameters were not significantly different between the treated groups and control group. From results of the present study, the sodium salt mixture had therapeutic effect on S. gallinarum infection in broilers.

편백 정유의 마우스에 대한 급성경구독성 (Acute Oral Toxicity Test of Chamaecyparis obtusa Essential Oil on ICR Mice)

  • 본 연구는 세균과 진균에 대한 항균작용, 살충작용, 항아토피 활성, 항염증 효과, 혈압강하 및 스트레스 완화 효과 등이 있는 것으로 알려진 편백 정유를 이용하여, 마우스에서의 급성경구독성시험을 수행하였다. 편백 정유를 마우스에 0, 125, 250, 500, 1,000, 2,000 mg/kg body weight의 농도로 각각 1회 경구투여한 결과, 마우스 암 수 모두 2,000 mg/kg에서 모두 생존하였으며, 모든 투여군의 체중 및 모든 혈액학적 혈액생화학적 지표값들이 대조군과 비교하여 통계적으로 유의적인 차이를 보이지 않았다. 따라서 편백 정유의 마우스에서의 $LD_{50}$은 2,000 mg/kg body weight 이상으로 확인되었다.

Effect of a mixture of Galla rhois and Cinnamomum cassia extracts on susceptibility to the colonization of Campylobacter jejuni in broiler chickens

  • The present study evaluated the effects of a mixture of Galla rhois and Cinnamomum cassia extracts (GCE) (1 : 1, w/w) on susceptibility to the colonization of Campylobacter (C.) jejuni in broilers. Eighty two-week-old broilers (n = 20 per group) were used to estimate the efficacy of GCE against C. jejuni infection via drinking water. Antibacterial activity testing revealed that the minimum bactericidal concentration of GCE against C. jejuni was 2.5 mg/mL. Broilers challenged with C. jejuni were administered 0.0 (Non-GCE), 2.5 (GCE-2.5), 5.0 (GCE-5.0) and 10.0 g/L (GCE-10) GCE for 7 days, and the cecal contents were collected from five broilers per group on the 1st, 3rd, 5th, and 7th day post-treatment. On day 3 post-administration, the number of C. jejuni in GCE-5.0 (p < 0.05) and GCE-10 (p < 0.01) was significantly decreased relative to Non-GCE, while on day 7 those in all GCE-treated groups were significantly decreased compared to the Non-GCE group (p < 0.001). Hematological and blood biochemical analysis revealed no significant differences in parameters between the Non-GCE and GCE-treated groups. Based on the results of the present study, GCE was identified as a safe and alternative candidate to suppress C. jejuni colonization in broilers.

메틸사이클로헥산의 독성과 신경에 미치는 영향 연구 (Toxicity of Methylcyclohexane and the Effects on Nervous System)

  • Methylcyclohexane is frequently used in industrial sites (2,592tons/year) as rubber adhesives, ink, paint thinners, organic solvents, and so on. However, there are limited data on the toxic evaluation of methylcyclohexane. This study aims to predict the hazards and neurological effects of methylcyclohexane using SD rats in order to prevent health disorders of workers. The OECD Guideline for Testing of Chemicals (OECD, 2001) was used as a reference during the tests. For 13 weeks (once a day, five days per week) 0, 10, 100 and 1,000mg/kg/day of methylcyclohexane was injected to SD rats to observe any changes in the body or organ weight, hematology, histopathology, mobility, blood pressure, and neurotransmitter. As a result, some male and female SD rats injected with 1,000mg/kg/day of methylcyclohexane died. On the other hand, surviving rats showed significant changes such as hematological changes involving the decrease in the number of red blood corpuscles, and the decrease or increase in the weight of the lungs, kidneys, spleens, and livers (p< 0.05, p<0.01). Also histopathological lesions were observed in the hearts and kidneys. In the test for the effect on the nervous system, SD rats injected with 100mg/kg/day of methylcyclohexane had higher blood pressure levels compared to the control group. However, no abnormal effects was observed in the mobility, serotonin, neurotransmitter, and the biopsy of the brain and coronary arteries. The study results revealed that the livers, hearts, and kidneys were affected by methylcyclohexane. The absolute toxic dose of methylcyclohexane is 1,000mg/kg/day, NOAEL is 100 mg/kg/day, and it is not a toxic substance to the nervous system.

1,4-Dichlorobutane의 랫드 2주 반복경구투여독성시험 (2-Week repeated oral dose toxicity study of 1,4-dichlorobutane in rats)

  • Objectives: The present study investigated the potential subacute toxicity of 1,4-dichlorobutane (1,4-DCB) by a 2-week repeated oral dose in male Sprague-Dawley rats. Materials and Methods: The test chemical was administered once daily by gavage to male rats at dose levels of 0, 74, 222, 667, and 2000 mg/kg/day for 2 weeks. All rats were sacrificed at the end of treatment period. During the test period, clinical signs, mortality, body weights, food and water consumption, urinalysis, hematology, serum biochemistry, gross findings, and organ weights were examined. Results: At 2000 mg/kg/day, treatment-related clinical signs, as evidenced by hypothermia, decreased locomotor activity, piloerection, lying on side, and prone position were observed. All the rats were found dead on test day 2. At 667 mg/kg/day, polyuria, suppressed body weight gain, food consumption, and spleen and thymus weights, and increased adrenal gland and liver weights were observed.Hematological and serum biochemical investigations revealed increases in the alanine aminotransferase, alkaline phosphataseand total bilirubinand decreases in the serum $Na^+$ level, white blood cell count and lymphocyte ratio. There were no treatment-related adverse effects in the 74 and 222 mg/kg/day groups. Conclusions: In the present experimental conditions, target organs were determined to be spleen, thymus,and liver. The no-observed-adverse-effect level was considered to be 222 mg/kg/day in male rats.

이산화규소 나노입자의 마우스 복강 내 주입에 의한 급성독성 (In vivo Acute Toxicity of Silicon Dioxide Nanoparticle to Mice after Intraperitonial Injection)

  • 본 연구는 이산화규소 나노입자의 급성독성을 확인하기 위해 여러 농도로 마우스의 복강에 주사한 다음 24시간 후에, 혈액학적, 혈액생화학적, 그리고 병리조직학적인 검사를 수행하였다. 혈액학적 검사에서, group II (100 mg/kg 이산화규소 나노입자)에서는 lymphocyte와 monocyte의 수치가 대조군과 비교하여 통계적으로 유의한 차이를 나타내었으며(p < 0.05), group III (200 mg/kg 이산화규소 나노입자)에서는 lymphocyte, monocyte, 그리고 hemoglobin의 수치가 대조군과 비교하여 통계적으로 유의한 차이를 나타내었다(p < 0.05). 혈액생화학적 검사에서, group II의 경우에는 ALT가, group III의 경우에는 AST, ALT, BUN, 그리고 creatinine이 대조군과 비교하여 통계적으로 유의한 차이를 나타내었다(p < 0.05). 병리조직학적 관찰에서는, group III의 간과 신장 조직에서 미약한 독성작용이 관찰되었다. 향후, 이산화규소 나노입자에 대한 장기적인 연구를 통해 독성영향 및 작용기전을 규명할 필요가 있는 것으로 사료된다.

체세포 복제 자견의 성장 및 혈액학적 특성 분석 (Analysis of Growth and Hematologic Characteristics of Cloned Puppies)

  • The objective of this study was to monitor health conditions of four genetically identical somatic cells cloned Labrador retriever puppies by estimation of body weight and analysis of hematologic and serologic characteristics. Naturally ovulated oocytes and donor cells were used for somatic cell nuclear transfer (SCNT). Donor cells and enucleated oocytes were followed by electric fusion, chemical activation and surgical embryo transfer into the oviducts of surrogate females. Two recipients became pregnant; two maintained pregnancy to term, and four live puppies were delivered by Caesarean section. The cloned Labrador retrievers were genetically identical to the nuclear donor dog. The body weight of clone-1, -2, -3, and -4 was increased from 0.66, 0.40, 0.39, and 0.37 kg at birth to 6.2, 6.6, 6.2, and 6.0 kg at 8 weeks of age, respectively. Although clone-4 had lower numbers of RBC than reference range, the most of RBC and WBC related heamatologic results of cloned puppies were not different when compared to reference range. In serological analysis, Glucose, ALP and inorganic phosphate level of four cloned puppies was significantly higher than the reference ranges. However, there was no significant difference among four cloned dogs. This study suggests that cloned puppies derived from SCNT did not have remarkable health problems, at least in the growth pattern and hematological and serological parameters.

Cancers of the Young Population in Brunei Darussalam

  • Background: Globally, the overall incidence of cancer is increasing as a result of ageing populations and changing lifestyles. Cancer is one of the leading causes of death, especially in the developed nations. Cancers affecting the young population are generally considered uncommon. This study assessed the demography and trends of cancers of the young in Brunei Darussalam, a small and developing Southeast Asia nation. Materials and Methods: All patients diagnosed with cancers between 2000 and 2012 were identified from the cancer registry maintained by the State Histopathology Laboratory. Cancers of the young was defined as any cancers diagnosed under the age of 40 years. Demographic data and the type of cancers were collected and analysed using SPSS Statistics 17.0. Results: Among the 6,460 patients diagnosed with cancer over the study period, 18.7% (n=1,205) were categorized as young with an overall decline in the proportion from 26.6% in 2000 to 18.8% in 2012 (p<0.001 for trend). Among all cancers of the young, the most common systems affected were gynecological (24.1%), hematological/lymphatic (15.8%), subcutaneous/dermatological/ musculoskeletal (10.5%), breast (10.5%) and gastrointestinal (9.9%). Overall, among the different systems, neurological (54.9%) had the highest proportion of cancers of the young followed by gynecological/reproductive (30.6%), hematological/lymphatic (39.9%), endocrine (38.7%), subcutaneous/dermatological/ musculoskeletal (22.3%) and the head and neck region (20.1%). There was a female predominance (66.9%) and the incidence was significantly higher among the Malays (20.1%) and expatriates (25.1%) groups compared to the Chinese (10.7%) and indigenous (16.8%) groups (p<0.001 for trend). Conclusions: Cancers of the young (<40 years) accounted for almost a fifth of all cancers in Brunei Darussalam with certain organ systems more strongly affected. There was a female preponderance in all racial groups. Over the years, there has been a decline in the overall proportion of cancers of the young. Selective screening programs should nevertheless be considered.

부산·경남 지역 성인의 담낭용종 위험인자 및 초음파 영상의 형태학적 분석 (Analysis of Risk factors & Morphological Ultrasound Image for Gallbladder Polyp in Adults Living in Busan and Gyeongnam Provinces)

  • 본 연구는 부산 경남지역에서 담낭용종의 위험인자 및 초음파영상의 형태학적 분포를 알아보고자 하였다. 실험 대상은 2016년 1월~5월까지 부산 P병원 내원환자의 복부초음파 영상을 대상으로 하였다. 그 중 복부초음파와 혈청학적 검사를 동시에 실시한 399명을 대상으로 위험인자를 분석하였다. 담낭용종 위험인자들의 통계분석은 독립표본 t검정(independent t-test)과 카이제곱 검정(chi-square test)을 시행하였다. 차이검정 결과를 고려하여 독립변수에 대한 상대 위험비(odds ratio, OR) 산출을 위해 다중 로지스틱 회귀분석(multiple logistic regression analysis)을 시행하여 변수들로부터 예측모형을 산정하여 타당성을 검정하였다. 그 결과 담낭용종 위험인자로 남성, HBsAg 양성, 중성지방이 관련이 있음을 알 수 있었다. 담낭용종의 위험인자로 확인된 남성, HBsAg 양성, 중성지방으로 예측모형 및 예측 확률값을 산정하였다. 예측확률의 민감도 61.0%, 특이도 76.8%를 보였으며, ROC 곡선의 AUC 결과는 0.735를 보여 예측모형의 타당성을 확인할 수 있었다. 복부 초음파검사 상 관찰되는 담낭용종의 형태학적 분석 결과는 고 에코, 유경, 균질한 형태가 가장 많은 분포(27.5%)를 나타내었으며, 용종 개수는 2개(38%), 크기는 5~10 mm (53%)로 가장 많았다. 담낭용종과 관련된 간질환으로는 mild fatty liver (23%), diffuse hepatopathy (21%)로 나타났다.
