Effects of water temperature changes on oxygen consumption and hematological factors in olive flounder Paralichthys olivaceus

  • Water temperature (WT) is a major environmental factor of metabolic rate in fish; it directly affects food intake, ammonia excretion, oxygen consumption (OC), growth, and survival. Thus, the purpose of this study was to investigate changes in the OC and the hematological response of olive flounder Paralichthys olivaceus because of WT changes. In Exp. I, WT was increased from 20℃ to 29℃ within 18 h and maintained at 29℃ for 96 h. Then, WT was decreased from 29℃ to 20℃ within 18 h and maintained at 20℃ for 24 h. In Exp. II, WT was decreased from 20℃ to 11℃ within 18 h and maintained at 11℃ for 96 h. Then, WT was increased from 11℃ to 20℃ within 18 h and maintained at 20℃ for 24 h. The Exp. III maintained that the Exp. I and II was consecutively. In Exp. I, the OC increased from 116.7 mg O2 kg-1 hr-1 to 317.5 mg O2 kg-1 hr-1 with increasing WT. In Exp. II, the OC decreased from 96.5 mg O2 kg-1 hr-1 to 71.3 mg O2 kg-1 hr-1 with decreasing WT. In Exp. III, OC tended to increase or decrease in inverse proportion to temperature. In Exp. I, cortisol, glucose, and aspartate aminotransferase (AST) values increased with increasing WT. In Exp. II, Cl-, osmolality, AST, and alanine aminotransferase (ALT) values significantly changed during the experimental period: glucose values increased, whereas cortisol values decreased with decreasing WT. Exp. III was shown to be a more stressful environment to olive flounder than Exp. I and Exp. II. The results of our study will be useful for evaluating current aquaculture procedures of olive flounder and developing techniques to minimize stress in aquaculture farms.

Hematological values of Korean indigenous neonatal goats (한국재래산양 신생자축의 혈액학치 변화에 관한 연구)

  • The present study was undertaken to clarify the changes in hematological values of Korean indigenous goats according to age. Blood samples were collected from 16 goats (5 females and 11 males) from birth up to the age of 6 weeks and the levels of various hematological values were analyzed. The body weight was increased from 2.54 kg at birth to 6.41 kg at 6 weeks age. The erythrocytes (RBC) counts and hemoglobin (Hb) values of goats were decreased at 2 weeks after birth, and increased gradually from 4 weeks after birth. The hematocrit (Ht) values were decreased until 4 weeks after birth, and increased at 6 weeks after birth. The mean cell volume (MCV) and mean cell hemoglobin (MCH) were decreased until 6 weeks after birth. The mean cell hemoglobin concentration (MCHC) was increased slightly from 2 weeks after birth. The red cell distribution width (RDW) was increased significantly at 2 weeks after birth, and decreased from 4 weeks after birth. The platelet (PLT) counts were increased significantly from 2 weeks after birth. The mean platelet volume (MPV) was decreased at 2 weeks after birth, and increased gradually from 4 weeks after birth. The total white blood cell (WBC) counts and the mean absolute numbers of neutrophil, lymphocyte, monocyte and eosinophil were increased from 2 weeks after birth. In conclusion, the data obtained from this study may be valuable as a standard for interpreting results of hematological analyses in Korean indigenous goats.

Effects of Unsaturated Fatty Acid Diets Feeding PEriods on the Antithrombosis the Hematological Changes in the Blood and Fatty Acid Compositions of Platelets in Rats (불포화 지방산의 종류와 사육기간이 흰쥐의 항혈전 작용, 혈액구성 및 혈소판의 지방산 조성 변화에 미치는 영향)

  • This study was performed to investigate the effect on polyunsaturate fatty acid diets and feeding periods on the antithrombosis. the hematological changes in the blood and fatty acid compositions of platelets in rats. Each group of rats was fed a diet containing 20%(W/W) corn oil beef tallow sardine oil and the general stock diet for 10, 20. 40 and 80 days. Rats fed sardine oil diet showed significantly longer bleeding time than any other diet groups after 20 days feeding The whole blood clotting time of sardine oil group fed for 80 days was increased significantly. The number of platelet and the concentration of hemoglobin showed no significant difference among all groups. The number of white blood cell was decreased continously in sardine oil group after 10 days feeding. The level of malondialdehyde generation during thrombin-induced aggregation of platelets was decreased continously in sardine oil grou after 20 days feeding. With regard to the composition of platelet fatty acid the ratio of eicosapentaenoic acid(EPA 20: 5 $\omega$-3) to arachidonic acid(AA 20:4 $\omega$-6) was increased in sardine oil group but decreased in corn oil groups and beef tallow groups with days. In conclusion the rats fed sardine oil diet for more than 20 days showed the fact that EPA induced the antithrombosis. the changes in number of white blood cell and the fatty acid composition of platelets.

Hematological and histological changes of black porgy Acanthopagrus schlegeli in ozonated recirculating systems

  • This study evaluated hemato-histological changes of black porgy in recirculating aquaculture systems (RAS) with three different ozone doses (no ozone, 20 g, and $40g\;ozone/kg\;feed\;day^{-1}$). During the 44-day study, black porgy did not show significant behavior changes or mortalities in both the ozonated systems displaying average total residual oxidants concentrations of 0.12 and 0.25 mg/L. There were no differences in growth and blood parameters among the systems. However, histological alterations on gills and livers were observed in both the treatment systems. In the higher ozone dose, signs of cellular damage were more apparent. Although the ozone doses did not manifest a serious adverse effect on growth and hematological observations in this short-term study, an ozone dose should not exceed $20g\;ozone/kg\;feed\;day^{-1}$ for black porgy based on the histological result. In order to use ozone in a seawater RAS, further studies will be needed to evaluate long-term effects of total residual oxidants.

Devocalization of Dogs by Fixation of Vocal Cords and Vocal Processes (개에서 성대 및 성대돌기의 고정에 의한 무성술)

  • This study was performed to evaluate the new surgical technique for debarking. Of the 8 mongrel dogs which underwent devocalization, 4 heads (group 1) received complete surgical removal of the vocal cords and another 4 heads (group 2) received fixation of vocal cords and vocal processes onto the thyroid cartilage. Volume of each dogs' voices was measured by a sound level meter(Function A) over 7 months at intervals of 10 days. All experimental animals'hematological values were measured before operation and postoperative at 2i 5 and 10 days. The changes of volume of dogs'voices and hematological values were analyzed by paired t-test. The volume in the dogs'voices in each group after operation was significantly lower than that before operation. At this results we detected that devocalizing effect was the same in each group. Postoperative volume of dogs'voices in group 1 had a tendency to increaser but group 2 had a tendency to decrease. The number of erythrocytes at 2 days after operation was significantly fewer than those before operation in group 1, but the group 2 did not have significant changes. The number of leukocytes at 2 days after operation significantly increased from those before operation in group 11 but the group 2 did not have significant changes. It was concluded that the new surgical technique, fixation of vocal cords and vocal processes onto the thyroid cartilages could be available for the devocalization of dogs and for the prevention of postoperative inflammation and blood loss.

Single and Four-week Intravenous Toxicity Studies of DA-3585, a Recombinant Human Erythropoietin, in Rats (재조합 사람 적혈구 조혈인자 DA-3585의 랫드에 대한 단회 및 4주반복 정맥투여 독성시험)

  • DA-3585 is a recombinant human erythropoietin produced by Dong-A pharmaceutical Co. Ltd. using recombinant DNA technique. Recently, recombinant human erythropoietin (rHu-EPO) has been used to treat various types of anemia. In this study, we examined acute and subacute toxicity of DA-3585 in rats. DA-3585 was intravenously administered to rats at dose levels of 0, 6,250, 12,500 and 25,000 lU/kg for single dose toxicity study and at dose levels of 0,100,500 and 2,500IU/kg daily for 4 week-repeated dose toxicity study. In the single dose toxicity study, there were no death, clinical signs and changes in body weight gain related to the treatment. Necropsy revealed no evidence of toxicity related to DA-3585, In the repeated dose toxicity study, all the rats survived throughout the study. There were no treatment-related changes in clinical signs, food and water intake, and body weight. Hematological examination showed increases in the number of erythrocytes, hemoglobin concentration, hematocrit value and mean corpuscular volume, and decrease in the number of platelet in 500 and 2,500 lU/kg dosed groups. Extramedullary hematopoiesis in the spleen and erythroid hyperplasia in the bone marrow were noted as treatment-related histological changes. Toxicologically significant changes were not observed in blood biochemistry, urinalysis, organ weights and in any other examinations. The treatment-related changes observed in this study were hematological or histological changes associated with pharmacological effects of DA-3585. On the basis of the results of this study, LD5n value of DA-3585 was above 25,000 lU/kg and the no-observed-adverse-effect-level was estimated to be 100 lU/kg.

Effect of Parathion on Hematological Parameters in the Serum of a Male Bagrid Catfish (Pseudobagrus fulvidraco )

  • To evaluate the impact of parathion on aquatic organisms, a freshwater bagrid catfish (Pseudobagrus fulvidraco) was exposed to sublethal concentrations $(63,\;95,\;190\;and\;380\;{\mu}g/L)$ of parathion, organophosphorus pesticide for 30 days. Glucose level in the serum of the bagrid catfish was significantly increased than that of control groups in the $190\;{\mu}g/L$ concentration at 30 days and in the $380\;{\mu}g/L$ concentration after 10 days. Bilirubin level was significantly increased in the $190\;{\mu}g/L$ concentration at the end of the experiment. After 10 days, a significant differences of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) activity increased in the ${\geq}190\;{\mu}g/L$ and $380\;{\mu}g/L$ groups. Though cholesterol concentration was stable, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities in serum were significantly reduced in the $190\;{\mu}g/L$ at 10 days and in the $380\;{\mu}g/L$ concentration at the end of the experiment. In conclusion, if P. fulvidraco was chronically exposed to ${\geq}190\;{\mu}g/L$ concentration of parathion, the hematological changes may be induced due to the stress response.

Evaluation of clinical usefulness of reference interval of some selected hematological parameters in canine blood

  • To estimate the source of variance components for some hematological parameters and assess the utility of the conventional population-based reference interval, this study computed index of individuality for blood samples, which were from 13 dogs drawn once weekly for 4 consecutive weeks. Results were subjected to nested analysis of variance. For all parameters measured between-dog variations were greater than within-dog variation. Except for the parameters RBC and MCHC the index of individuality was <1.4. The low reliability coefficient and high index of individuality of ${\leq}0.8$ were found for the majority of hematological parameters. In practical term, the present study indicated that use of hemogram profiles alone in the evaluating clinical state of a single patient should be avoided because of their physiological or natural random variations, and that comparing a single measurement on the blood analytes from an individual dog to the conventional population-based reference range may be too insensitive to detect any significant changes in the blood components of that particular dog. A single measurement may not characterize an individual's average concentration of the parameters even shortterm period.

The Acute Toxicity of 1,2,4-Trichlorobenzene in Sprague-Dawley Rats Depleted of Glutathione by Treatment with Buthionine Sulfoximine (BSO 유도 글루타치온 저감 흰쥐에서 1,2,4-trichlorobenzene의 급성독성)

  • 1,2,4-trichlorobenzene (1,2,4-TCB) is used as a dye carrier, an intermediate in the syn[hesis of herbicides, aflame retardant, and for other purpose. After a single oral administration of 1,2,4-TCB (200 mg/kg, 400 mg/kg) in rats, toxic effects were studied by means of serum biochemical and hematological analysis, and liver calcium concentration. Administration of 1,2,4-TCB resulted in dose-dependent manner liver and kidney damage being suggested by increased serum alanine aminbtransferase (ALT) activities, liver calcium concentration and blood urea nitrogen (BUN). Pretreatment with DL-buthionine sulfoximine (BSO, 2 mmol/kg, i.p.) considerably decreased liver glatathione concentration, which was accompanied by markedly elevated serum ALT activites. It is well-known that toxicity of halogenated benzene such as bromobenzene, 1,4-dichlorobenzene is increased by pretreatment of phenobarbital, and protected by pretreatment of cytochrorn P450 inhibitor including metyrapone. However, there were no obvious alterations in toxicity of 1,2,4-TCB by pretreatment of phenobarbital or metyrapone. In comparison with control group, treatment groups exhibited significant changes in some parameters of hematological analysis but all hematological values remained within normal ranges.

Changing Salinity Affects Hematological and Histological Response in Hybrids and Hybrid Triploids between River Puffer, Takifugu obscurus and Tiger Puffer, T. rubripes

  • We investigated the physiological responses of the river puffer, Takifugu obscurus, the tiger puffer, T. rubripes, a hybrids of the two (river puffer${\times}$tiger puffer) and a hybrid triploids to acute changes of salinity from 30 psu to 0 psu and from 0 psu to 30 psu. The blood and plasma factors of each species were elevated for 48, 72, or 96 hrs, and thereafter decreased due to hyper-osmoregulation and hypo-osmoregulation. In hyper-osmoregulation and hypo-osmoregulation, the cortisol concentration of river puffer, hybrids, hybrid triploids and tiger puffer increased for 12 or 48 hrs, and decreased thereafter. Chloride cells in the gill filaments of each species increased with increasing salinity, and melano-macrophages in the kidney tissue of each species increased with decreasing salinity. In conclusion, the hematological and stress responses of the hybrids were between those of the river puffer and tiger puffer, and the hematological responses of the hybrid triploids were higher than those of the other groups. The stress response of the hybrids was more sensitive than that of the hybrid triploids. In all groups, the histological responses of kidney in hyper-osmoregulation were more sensitive than those in hypo-osmoregulation.
