Virucidal Efficacy against Avian Influenza Virus of a Disinfectant Spray Containing Grapefruit Seed Extracts, Citric Acid, Malic Acid and Benzalkonium Chloride (자몽종자추출물, 구연산, 사과산 그리고 염화벤잘코늄을 주성분으로 하는 스프레이형 소독제의 조류인플루엔자바이러스에 대한 살바이러스 효과)

  • Objectives: This study evaluated the virucidal efficacy against avian influenza virus (AIV) of a disinfectant spray containing 0.25% grapefruit seed extract, 0.2% citric acid, 0.0625% malic acid and 0.0125% benzalkonium chloride. Methods: The disinfectant spray was diluted several times with hard water (HW) and organic matter (OM). Two point five mL of each diluent was added into each test tube, and 2.5 mL of AIV suspension was inserted into each test tube. After 30 minutes of virus-disinfectant contact reaction at $4^{\circ}C$, 2.5 mL of 10% inactivated fetal bovine serum was added into each test tube to neutralize the sanitizer efficacy. The neutralized solutions were serial 10-fold dilutions with phosphate buffer solution, and 0.2 mL of the diluents was injected into the allantoic cavity of five ten-day-old-chickens per dilution time. After incubation of the embryos for five days, the viability of the AIV was examined by hemagglutination titer. The valid dilution of the disinfectant spray was estimated according to the dilution time that the virus titer was inactivated more than $10^4$ 50% egg-infective dose (EID50)/mL compared with pathogen control. Results: In HW and OM conditions, the valid dilutions of the disinfectant spray against AIV were seven- and three-fold dilutions, respectively. The AIV titer of the pathogen control was more than 6.1 log10EID50/mL, and there was no embryonic toxicity. Conclusion: The present study showed that this disinfectant spray has effective virucidal activity against AIV.

Comparison of Efficacy of Newcastle Disease Virus $B_1$ Vaccine by Different Administration .Methods in Commercial Layer Chickens (백신접종 방법에 따른 뉴캣슬병 $B_1$생독백신의 효능비교)

  • The immune responses of commercial layer chickens against Newcastle disease (ND) were compared among different administration methods and times of vaccination during 4 weeks of age. A total of 372 day-old chickens were devided into 4 groups of 93 birds each. Each of 3 groups was received a commercially available B$_1$ live vaccine via drinking water, eye instillation or spray method at one, 14 and 28 days of age. One group was used as an unvaccinated control. At two and 4 weeks after each time of vaccination, 15 birds from each group were challenged with virulent ND virus at the dose of 10$^{5}$ EID$_{50}$ per bird to examine the pretection rate. Ten to 15 birds from each group were bled at two weeks intervals from day old to 8 weeks of age to determine hemagglutination inhibition antibody titer. The protection rate was generally low regardless of the times of vaccination although two or more times vaccination gave higher protection than once vaccination. The low protection was considered due to low titer of the vaccine used since the vaccine titer was less than 10$^{2.5}$ EID$_{50}$ per bird. Spray method gave better protection compared to eye instillation of drinking water method which resulted in lowest response. When birds were challenged majority showed clinical signs on ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Major clinical signs observed were depression (96%), drowsy(90%), anorexia (84%), diarrhoea (29%), difficult breath (15%) and torticollis (10%). Hemorrhagic lesions on post mortem were seen in duodenum (51%), trachea(36%), illeum (13%), ceacal tonsil (11%), proventriculus (10%) and some other organs. When birds were challenged majority showed clinical signs on ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Etiologic Survey on the Acute Diarrhea of Puppies in Seoul (서울지역에서 문제되고 있는 강아지의 급성설사증에 관한 병인학적 연구)

  • In Seoul area, there are so many kennel clubs, veterinary hospital, pet establishment and breeding confers that various problems have occurred. They are crowed pet houses, poor sanitation, stress to puppies, sudden environmental changes to puppies and unvaccination against parvovirus, canine distemper virus, parainfluenza virus, infectious hepatitis caused by Adenovirus type I and Leptospira. Several studies were made to survey the infectious agents involved in acute diarrhea of poppies in Seoul are, such as history taking, physical examination, complete blood count and serum chemistry, histopathological finding, bacterial isolation and identification, and hemagglutination test in feces and hemagglutination inhibition test in serum against parvovirus, respectively. The results obtained are summarized as followed. 1. The percent of PCV (30.5$\pm$5.6) and concentration of Total protein(5.0$\pm$0.8) resulted from statistical analysis are significantly lower than normal values (p<0.05), respectively. In addition, fibrinogen (505$\pm$326) was significantly higher an normal value (p<0.001), Band neutrophil (22.9$\pm$12.7) showed signifiant difference (P<0.01). decreased monocyte (3.2$\pm$2.1) and eosinophil (0.7$\pm$0.8) values appeared statistically significant (p<0.001), lymphocyte (16.7$\pm$9), as well. 2. The concentrations of calcium .(8.0$\pm$2.8), glucose (40.1$\pm$31.4), and albumin (2.0$\pm$0.39) were lower than normal values (p<0.01, p<0.01, p<0.001), respectively. Also Inorganic phosphate (7.1$\pm$2.4), pH (p<7.9f:0.2), and Blood Urea Nitrogen (40.41=37.1) were significantly higher than normal values (p<0.001, p(0.001, p<0.05), respectively. 3. Simple and mixed infections occupied 18% and 82% in the distribution of causes in puppies with acute diarrhea, respectively. 4. As puppies got older, incidence of acute diarrhea caused by Staphylococcus aureus was decreased to 13% and infection of canine distemper virus was increased to 53%, but E coli and canine parvovirus always showed high frequency of outbreak in the body weight ranged from 35g to 7.8Kg. 5. As showed in table 5, infections of E coli and Canine Parvovirus showed high outbreak regardless of the age which is classified into three stages, 35~50 day, 60 day and 75~day to 1 year, canine distemper virus appeared increased, but in case of Staphylococcus aureus, visa versa. 6. In comparison wi methods for the laboratory diagnosis diagnosis parvovirus, Hemagglutination test showed positive reaction in 25% and mean serum antibody titer measured by Hemagglutination inhibition test showed 2779 (n=20). In addition, positive reaction was 90% (18/20). 7. In histopathological studies, enteritic and pneumonic lesions were indicated in 53.7% and 39.5% of samples, respectively. 8. Etiologic diagnosis based on the history taking, clinical signs and histopathological findings in puppies with acute diarrhea and vomiting indicated that 50% of puppies were infected by canine parvovirus and distemper virus. 9. In the parasitological examination made by simple flotation with saturated zinc sulfate tour parasites found were Isospora canis, Toxocara canis, Ancylostoma spp and Toxocara spp. Isospora canis and Toxocara canis were more frequently found among those parasites of diarrhoeic causes in puppies ranged from 35 days to 1 year. Their infestation rates were 15% and 13% respectively.

Effect of n-6, n-3 Fatty Acids and Vitamin A Levels on Humoral Immunity in Rats (n-6, n-3 지방산과 비타민 A 수준이 흰쥐의 체액면역에 미치는 영향)

  • This study was carried out to observe the effects of dietaryn-6, n-3 fatty acids and vitamin A levels on humoral immunity in rat. Sixty eight male Sprague-Dawley rats were fed 6 different experimental diets for 6 weeks. The diets were composed of 10% of either corn oil or fish oil with various levels of vitamin A ; deficient(12450 IU/kg diet) adequate(4000IU/kg diet) and excess(400,000 IU/kg diet) The weight of spleen from the excess vitamin A-fish oil group showed the lowest value of all the groups when spleen weight was expressed/100g body weight. The number of PFC to SRBC was not affected by dietary at type and vitamin A levels. Hemagglutination titers were significantly lower in fish oil groups compared to corn oil groups and the values of vitamin A deficient groups were lower than the ones of adequate and excess vitamin A groups. IgM contents is serum were significantly lower in fish oil groups than in corn oil groups. IgG contents were higher in fish oil groups than in corn oil groups and the highest levels was recorded in excess vitamin A-fish oil group which showed the smallest speen size. Light microscopical examination showed that spleen tissues of fish oil groups were well developed than those of the corn oil groups and vitamin A deficient and excessive groups showed poor development than the adequate groups. Therefore it is suggested that adequate amounts of vitamin A consumption is necessary for healthy individuals and fish oil intake along with excess vitamin A should be avoided in order to maintain immune function properly.

Immunization of broiler chicks deprived food and water with live Newcastle disease vaccine(LaSota strain) by drinking water

  • To confirm the effect of food and water deprivation prior to Newcastle disease(ND) virus vaccination, three hundred chicks were divided into five groups with three replications. ND vaccine were sprayed to at 1 -day old chicks at commercial hatchery. Secondary and third vaccination was conducted at 2-week old and 24-day old chicks by LaSota strain. Control was conventionally vaccinated without withdrawing the food and water before or after vaccination. In group 2(G2) and 3(G3), LaSota strain was vaccinated to chicks before and after fasting the food and water for 3 and 2 hours, respectively. Group 4(G4) has the same fasting time of group 2, but supplemented the skim milk in vaccin dilution water. In group 5(G5), skim milk was added into group 3. Weight gain, feed intake and feed conversion were weekly measured for 5 weeks. Blood was collected from wing vein at 24 and 35 days of age. Each serum antibody level were measured by hemagglutination inhibition(HI) test. The average weight gain, feed intake, feed conversion of all group were not significantly different. Weight gain of each groups was 1910.30(control), 1875.28(G2), 1952.12(G4) and 1896.05(G5), respectively. Feed intake of all group was recorded at 3160.67(control), 3167.07(G2), 3189.48(G3), 3157.85(G4) and 3178.16(G5), respectively. The feed conversion of each groups was 1.655(control), 1.688(G2), 1.633(G3), 1.699(G4) and 1.676(G5), respectively. The HI titer of G4 was $ 5.50{\Pm}$1.40 and significantly higher than the other groups (p<0.05)(control : $4.36{\Pm}$1.87 , G2 : $5.18{\Pm}$2.14, G3 : $4.51{\Pm}$2.19, G : $5.28{\Pm}$1.58 at 35 days old. The results of this experiment indicated that two or three hours of fasting time before or after vaccination would be able to show the higher antibody level against ND virus.

Evaluation of immunogenicity of the 2008-2009 seasonal influenza vaccines by microneutralization test

  • Purpose: For evaluating the immunogenicity of an influenza vaccine, the microneutralization (MN) test has a higher sensitivity and specificity as compared to the hemagglutination inhibition (HI) test. However, the MN test is more time consuming and is difficult to standardize. We performed the MN test to determine its usefulness as an alternative or complementary test to the HI test for evaluating the immunogenicity of influenza vaccines. Methods: We compared the MN test with the HI test using 50 paired samples taken from a previous clinical study (2008-2009) in Korean children under 18 years of age. Results: The linear correlation coefficients of the 2 tests for H3N2, H1N1, and influenza B were 0.69, 0.70, and 0.66, respectively. We identified a high index of coincidence between the 2 tests. For an influenza vaccine, the postvaccination seroprotection rates and seroconversion rates determined by the MN test were 78.0% and 96.0%, 90% and 42.0%, and 42.0% and 48.0% for H3N2, H1N1, and influenza B, respectively. Geometric mean titer fold increases of H3N2, H1N1, and influenza B were 2.89, 5.04, and 4.29, respectively, and were 2.5-fold higher. We obtained good results in the evaluation of the immunogenicity of the 2008-2009 seasonal influenza vaccines. Conclusion: We found that the MN test was as effective as the HI test. Therefore, we suggest that the MN test can be used as an alternative or complementary test to the HI test for evaluating the immunogenicity of influenza vaccines.

Antiviral Effects of Ethyl Acetate Fraction of Distylium racemosum Leaf Extract on Adenovirus 36 (조록나무 잎 에틸 아세테이트 분획물의 아데노바이러스 36에 대한 항바이러스 효과)

  • Distylium racemosum is an evergreen tree growing wild on Jeju Island, which has been reported to exert biological activity. Obesity is induced by genetic, metabolic, environmental, and other factors. Among these, certain bacterial and viral infections have been shown to cause obesity, which is known as infectobesity. Human adenovirus (HAdV)-36 is one of the viruses that are known to cause infectobesity in humans. Unlike extensive research on preventing obesity and developing anti-obesity drugs, little research has been conducted specifically on the prevention and treatment of infectobesity. Therefore, this study aimed to evaluate the effects of phytochemicals from D. racemosum on the replication of HAdV-36. A549 cells infected with HAdV-36 were treated with an ethyl acetate fraction of a D. racemosum leaf extract (DRE), and the virus titer was calculated based on the hemagglutination (HA) titer. The results showed a concentration-dependent inhibitory effect of DRE treatment on HA titers. DRE treatment was also found to inhibit the cytopathic effects of the virus and the expression of viral genes. Quercitrin was identified as the constituent of DRE exerting an inhibitory effect on HAdV-36 replication. This study shows that DRE can be used as a candidate substance for the development of treatment for HAdV-36 infections. In addition, this study provides a basis to further investigate DRE for the development of anti-infectobesity medication.

Effects of Walgookwhanhab-Bojoongikgitang administration on immune-function in Balb/c mice stressed by electric footshock (월국환합보중익기탕(越鞠丸合補中益氣湯)이 전기자극(電氣刺戟)스트레스를 받은 mouse의 면역기능(免疫機能)에 미치는 영향(影響))

  • The present experiments were designed to study the influence of WalgookwhanhabBojoongikgitang on immune functions of Balb/c mice under stress condition. WalgookwhanhabBojoongikgitang was orally administered to the mice for 15 days. On the 10th day the mice were immunized with sheep red blood cells (SRBC) and then subjected to electric footshock for 5 days (2 sessions a day, 11 footshocks a 31 min-session). The immune responses to SRBC were determined by means of hemagglutination and BIT cell populations in the spleen were studied by FACS analysis on the 16th day. The results were as follows. 1. After electric footshock, IDlce became sluggish and crowded to one side of the cage. Increased antibody titer for SRBC, increased B cell population, and decreased T cell population in the spleen were also observed. These results confirm that electric footshock caused stress inducing immunological and behavioral changes in Balblc mice. 2. Walgookwhanhab-Bojoongikgitang administration significantly antagonized the effect of electric footshock on the antibody titer for SRBC. As a result, antibody titers for SRBC in the mice treated with Walgookwhanga-Bojoongikgitang were maintained at the similar levels as those of control group mice even after the electric foots hock. 3. Walgookwhanhab-Bojoongikgitang administration also antagonized the effect of electric footshock on the BIT populations in spleen. As a result, Band T populations in the mice treated with Walgookwhanhab-Bojoongikgitang were maintained at the similar levels as in the control group mice even after the electric footshock. Taken together, Walgookwhanhab-Bojoongikgitang seem to help Balb/c mIce to maintain their humoral immune response and immune cell populations at a normal range under the stress conditions, suggesting its possible therapeutic use as a immune function modulator.

The Analysis of the Value of the Thyroid Autoantibody Measured by Radioimmunoassay (방사면역측정법에 의한 갑상선 자가항체 측정의 기본적 및 임상적 검토)

  • To evaluate the values of the thyroid autoantibody measured by radioimmunoassay (RIA) and compare it with hemagglutination method (HA) in the normal and the thyroid disease, data were obtained from total 618 persons; 236 healthy persons, 217 patients with Graves' disease (including 113 patients with undertreated Graves' disease), 100 Hashimoto's disease, 31 thyroid nodule, and 34 simple goiter. RSR kit made in England was used and could be detected to at least 3 U/ml. The positive rates of normal group were antimicrosomal antibody (AMA) 31.8%, antithyroglobulin antibody (ATA) 44.5% by RIA and there was no considerable change in sex and age distribution. In Graves' disease, the positive rates of AMA and ATA were 90.4, 76.9% by RIA, 85, 39% by HA. In Hashimoto's disease, 94,91 % by RIA, and 87,48% by HA, respectively. The autoantibody titer by RIA in thyroid autoimmune disease as well as in normal group was more senisitive than that by HA, especially in ATA. There were linear relationships between the titer of RIA and that of HA in AMA of Graves' disease and AMA and ATA of Hashimoto's disease. There was no relationship among thyroid autoantibody, free $T_4$ index, TBII, and TSH. The titers of AMA and ATA were found to decrease in patients with Graves' disease during the course of antithyroid drug therapy. Of the 236 normal subjects, thirty-seven (15.7%) had concentrations of above 7.5 U/ml in AMA, forty. four (18.6%) above 9 U/ml in ATA. These values were considered as the upper limit for the normal range. In Graves' disease, 82.7, 53.8% were above 7.5, 9 U/ml, respectively; In Hashimoto's disease, 82, 79% were positive. We conclude that RIA was more sensitve than HA in measuring the thyoird autoantibody, but we will study further more for determining the normal range and its interpretation.

  • A total of 3 hybridoma clones producting menoclonal antibody (MCA) against Newcastle disease virus(NDV) was raised by cell fusion method. The MCAs did not cross react against other avian or mammalian viruses tested. However, these antibodies reacted with all strains of velogenic and lentogenic NDVs tested indicating that they are unable to discriminate the possible antigenic differences among NDVs. All. the MCAs were classified as IgG type and did not show neutralizing and hemagglutination inhibition (HAI) activity except one clone which has low HAI activity. One of these MCA raised in mouse ascites revealed the titer of $10^6$ by indirect immunofluorescent antibody (IFA) test Using the MCA, virulent NDV could easily be detected from tracheal and conjunctival smears made 2 to 3 days after experimental infection.
