ROS Scavenger, Ebselen, Has No Preventive Effect in New Hearing Loss Model Using a Cholesterol-Chelating Agent

  • Background and Objectives: The antioxidant ebselen will be able to limit or prevent the ototoxicity arising from 2-hydroxypropyl-β-cyclodextrin (HPβCD). Niemann-Pick Type C (NPC) disease is a disorder of lysosomal storage manifested in sphingolipidosis. Recently, it was noted that experimental use of HPβCD could partially resolve the symptoms in both animals and human patients. Despite its desirable effect, HPβCD can induce hearing loss, which is the only major side effect noted to date. Understanding of the pathophysiology of hearing impairment after administration of HPβCD and further development of preventive methods are essential to reduce the ototoxic side effect. The mechanisms of HPβCD-induced ototoxicity remain unknown, but the resulting pathology bears some resemblance to other ototoxic agents, which involves oxidative stress pathways. To indirectly determine the involvement of oxidative stress in HPβCD-induced ototoxicity, we tested the efficacy of an antioxidant reagent, ebselen, on the extent of inner ear side effects caused by HPβCD. Materials and Methods: Ebselen was applied prior to administration of HPβCD in mice. Auditory brainstem response thresholds and otopathology were assessed one week later. Bilateral effects of the drug treatments also were examined. Results: HPβCD-alone resulted in bilateral, severe, and selective loss of outer hair cells from base to apex with an abrupt transition between lesions and intact areas. Ebselen co-treatment did not ameliorate HPβCD-induced hearing loss or alter the resulting histopathology. Conclusions: The results indirectly suggest that cochlear damage by HPβCD is unrelated to reactive oxygen species formation. However, further research into the mechanism(s) of HPβCD otopathology is necessary.

ROSA/LSTF test and RELAP5 code analyses on PWR 1% vessel upper head small-break LOCA with accident management measure based on core exit temperature

  • An experiment was performed using the large-scale test facility (LSTF), which simulated a 1% vessel upper head small-break loss-of-coolant accident with an accident management (AM) measure under an assumption of total-failure of high-pressure injection (HPI) system in a pressurized water reactor (PWR). In the LSTF test, liquid level in the upper head affected break flow rate. Coolant was manually injected from the HPI system into cold legs as the AM measure when the maximum core exit temperature reached 623 K. The cladding surface temperature largely increased due to late and slow response of the core exit thermocouples. The AM measure was confirmed to be effective for the core cooling. The RELAP5/MOD3.3 code indicated insufficient prediction of primary coolant distribution. The author conducted uncertainty analysis for the LSTF test employing created phenomena identification and ranking table for each component. The author clarified that peak cladding temperature was largely dependent on the combination of multiple uncertain parameters within the defined uncertain ranges.

공기실을 사용한 압력수두의 완화효과에 대한 분석 (An Analysis of Attenuation Effect of Pressure Head Using an Air Chamber)

  • 공기실(air chamber)은 관망에 있어 압력수두가 설계치를 초과하지 않고 또한 최저 압력수두가 발생하지 않도록 하기 위해 설계된다. 따라서 공기실은 갑작스런 펌프의 중단에 따르는 순간적인 최고 및 최저 압력수두의 발생을 방지하기 위해 사용되어 진다. 본 연구에서는 가상적인 관망에 공기실을 적용하여 공기실의 공기부피, 위치, 면적, 오리피스에서의 손실계수 그리고 polytropic지수의 변화에 따른 압력수두의 완화효과를 분석하였다. 분석결과 공기의 부피가 증가할수록 최고압력수두는 감소하고 최저 압력수두는 증가함을 보여주었으며, 공기실의 위치나 단면적의 변화는 완화효과의 변화가 별로 없었다. 또한 오리피스 손실계수를 증가할수록 최고 압력수두가 감소함을 보이고, 공기실의 공기가 등온변화과정을 따를때가 단열변화과정을 따를 때보다 최고 압력수두가 낮음을 보여주었다.

  • In momentum exchange theory the loss models for the circulatory flow is critically important. But because of lack of loss model on the circulatory flow, analysis model on regenerative turbomachines is not available in the open literature. In the present study circulatory loss is evaluated by combining bend's losses. Through the comparison with the previous experimental data on linear pressure gradient, a combination factor is suggested in terms of the aspect ratio of a channel. Applying this factor to two kinds of regenerative blowers the predicted results are found to be in good agreement with the experimental data of the overall performance and the head distribution along the rotational direction. Especially, the comparison with the head distribution demonstrates the accuracy of hydraulic model and loss model suggested in the present study. And the comparison with the overall performance confirms the validness of physical models as well as loss models suggested in the present study.

  • The amount of grain loss incurred during harvesting operation through the chaff-outlet of combine may not be negligible. To minimize this grain loss and optimize condition of combining, it may be necessary that the amount of chaff-outlet loss dependent on varying crop condition is to be estimated as exactly as possible. This study was thus intended to develop the monitor that could indicate the amount of grain loss occurred through the chaff-outlet of combine during harvesting operation. The function of this monitor is to measure at the sounding board the impact sound of paddy kernels which could be distinguished from those of other threshing products through chaff-outlet, and from vibration or noise created by the combine engine and other moving parts. To develop such monitor, the frequency distributions of each sound generated by the impact of grain and chaff, the sound generated by the impact of the mixture of grain and chaff, and vibration or noise created by the combine engine and other moving parts were investigated experimentally. From the results of frequency analyses, the trainsducer adequate for the monitering system was selected and sounding board was constructed. The grain loss monitor thus obtained was tested by attaching the sounding board to the chaff-outlet of combine.

효율적인 Source Head 개발에 관한 연구 (A Study on Development of Efficient Source Head)

  • 본 연구는 이온주입(Ion Implanter)장비의 성능향상과 재현성 있는 Source Head를 개발하기 위한 방법이다. Source Head의 Life time이 부품 간 사용되는 Life time과 틀리기 때문에 보통 20일 정도 사용하고, 한번 Source Head를 재생하기 위해 교체되는 부품들도 고가일 뿐 만 아니라 50% 이상이 일회성으로 사용되고 있다. 본 개발에서는 원자의 유입방식을 공중 분산방식으로 적용함으로써 열전자의 손실로 발생하는 부분을 억제하는 효과와 Arc Chamber의 압력을 낮게 가지고 갈 수 있고 Chamber의 오염을 억제하는 효과를 얻을 수 있었다.

돌발성난청에서 홍삼추출물의 추가적인 치료효과 (Beneficial Effect of Ginseng Extract on Sudden Sensorineural Hearing Loss)

  • Sudden idiopathic sensorineural hearing loss is a disease that develops within several hours to several days. Its etiology has not yet been verified, but the disturbance of the circulation of blood in the inner ear, inner-ear hydrops, and viral infection are considered possible causes of the disease. This study was conducted to evaluate the effect of Panax ginseng extract, which is known to have a vasodilatory effect, on sudden sensorineural hearing loss. Sixty-nine patients suffering from sudden sensorineural hearing loss were admitted to Korea University Anam Hospital from March to December 2008. They were divided into the experimental (30 ears) and control (39 ears) groups. Ginseng extract (2700 mg/day, 4 weeks) was added to the therapeutic regimen in the experimental group. The effect of ginseng extract therapy was analyzed according to the factors relating to the prognosis. A considerable hearing improvement was documented in both groups (32.2 dB in the experimental group and 25.8 dB in the control group). However, there was little beneficial effect of ginseng extract on additional hearing improvement compared with control. The total recovery rate of the experimental group (80.0%) was better than that of the control group (58.9%), and the experimental group's high-tone hearing gain at 3 kHz (29.7 dB) was better than that of the control group (21.7 dB). The results of the study suggest that the effects of ginseng therapy tend to be superior to those of the conventional therapy, but the difference between the two is not statistically significant. The hearing gains tend to be in the higher frequencies and may be due to the promotion of cellular differentiation from the supporting cells.

매끄러운 관수로 내 난류흐름특성을 반영한 수리학적 매개변수에 의한 마찰속도의 산정 (The Estimation of Friction Velocity by Hydraulic Parameters Reflecting Turbulent Flow Characteristics in a Smooth Pipe Line)

  • 스마트워터그리드의 중요한 요소기술로써 그리드(관망)의 설계는 압력, 마찰계수, 마찰속도, 수두손실, 그리고 에너지 경사와 같은 수리학적 매개변수를 추정하는 것이 필수적이다. 특히, 그리드의 마찰속도는 에너지 경사, 마찰계수, 압력, 수두손실 등의 결합에 있어 매우 중요한 인자이다. 그러나 마찰 수두손실, 마찰속도 등 마찰인자를 정확히 산정하는 것은 매우 어려우며, 경험적 마찰 인자는 여전히 약 100년 전에 개발된 공식과 이론을 사용함으로써 산정된다. 따라서, 본 논문에서는 최대유속과 마찰속도 사이의 새로운 공식을 Darcy-Weisbach의 마찰수두 손실공식과 Schlichting 공식 사이의 적분관계 및 회귀분석을 통하여 개발하였다. 개발된 공식을 증명하기 위하여 매끄러운 관 자료가 사용되었으며 제안한 공식은 관측 자료와 비교하여 높은 정확성을 보여준다. 이 연구의 결과는 안정성 향상과 그리드 설계에 사용이 가능할 것으로 판단된다.

지하저수지(ASR) 유입수 전처리기법 개발: 물리적 폐색 저하 및 수량공급원활을 위한 2 단계 급속여과지 (Development of pre-treatment for the injection water into the aquifer storage and recovery(ASR) in Korea: The two-step rapid filter to reduce physical clogging and secure the volume of the injected water)

  • Two step rapid filter system as a pre-treatment for the injected water into aquifer storage and recovery (ASR) in Korea was developed to reduce physical blockage and secure the volume of the injected water. First, single rapid sand filters with three different media sizes (0.4~0.7, 0.7~1.0 and 1.0~1.4 mm) were tested. Only two sizes (0.4~0.7 and 0.7~1.0 mm) satisfied target turbidity, below 1.0 NTU. However, they showed the fast head loss. To prevent the fast head loss and secure the volume of the injected water, a rapid anthracite filter with roughing media size (2.0~3.4 mm) were installed before a single rapid sand filter. As results, both the target turbidity and reduction of head loss were achieved. It was determined that the media size for a rapid sand filter in two step rapid filter system (i.e. a rapid anthracite filter before a rapid sand filter) was 0.7~1.0 mm. In addition, the effects of coagulant doses on the removal of natural organic matter (NOM), which might cause a biological clogging, were preliminarily evaluated, and the values of $UV_{254}$, dissolved organic carbon (DOC) and SUVA were interpreted.

Comparison of the Effects of Intratympanic Steroid Injection at Different Intervals in Sudden Sensorineural Hearing Loss

  • Background and Objectives: Systemic steroid therapy (SST) and intratympanic steroid injection (ISI) have been the treatment of choice for sudden sensorineural hearing loss (SSNHL). We studied the effect of ISI administered at different intervals on hearing outcomes in patients with SSNHL. Subjects and Methods: We performed a retrospective study of 427 patients diagnosed with SSNHL at Bundang Jesaeng Hospital, of whom 51 patients with SSNHL who received SST and four ISIs were included in this study. Patients were treated with four ISIs either every day for 4 days (group 1) or at intervals (mean duration of interval: 2.21 days) (group 2). Hearing outcomes were evaluated using the pure-tone test before the injection and 14 days, 1 month, and 3 months after the final injection. Recovery rates were classified based on Siegel's criteria. Results: The amount of improvement was 27.67 dB (±20.45) in group 1 and 32.79 dB (±21.42) in group 2. However, there were no significant differences between the two groups (p= 0.714). The recovery rates based on Siegel's criteria were 18/27 (66.7%) and 16/24 (66.7%) in groups 1 and 2, respectively, with no significant difference (p=1.000). Considering only complete recoveries in hearing recovery, the recovery rates were 15/27 (55.6%) and 14/24 (58.3%) in groups 1 and 2, respectively, with no significant difference (p=0.842). Conclusions: There were no significant differences in hearing outcomes or recovery rates after ISI administration every day or at intervals of 2-3 days.
