Breeding of a New Cultivar, 'Sanggang' with Upright Stipe and Improved Shelf life in Agrocybe aegerita (직립형이고 저장성이 개선된 버들송이 신품종 '상강' 육성)

  • 'Sanggang', a new cultivar of Agrocybe aegerita, was bred with mating between monokaryotic strains isolated from 'GMAG45109' and 'GMAG45107' in Mushroom Research Station, Gyonggi Province A.R.E.S. in 2009. The optimum temperature for the mycelial growth of 'Sanggang' was 26 to 28$^{\circ}C$ on PDA(potato dextrose agar) medium. The optimum temperature for the primordia formation and fruiting body development of 'Sanggang', was 18 to 20$^{\circ}C$. In the bottle cultivation of 'Sanggang', the period of spawn running was around 38 days at 22 to 23$^{\circ}C$ and the period from scratching of inoculum to harvest was 12 days. These characteristics of 'Sanggang' were not different from those of 'Mihwang' (control). 'Sanggang' had brown-colored pileus, whereas 'Mihwang' had yellowish brown-colored one. In bottle cultivation, 'Sanggang' hada little smaller but stronger stipe and pilus than 'Mihwang', and 'Sanggang', as well as 'Mihwang', had upright stipe. And 'Sanggang' was shown to have lower veil opening ratio than that of 'Mihwang' at harvest time.The yield of fruiting bodies of 'Sanggang' was 134 g/850ml bottle, which was similar to that of 'Mihwang'. Resistance of'Sanggang' against Trichoderma spp., as well as 'Mihwang', was weak. Freshness of 'Sanggang' was maintained for 13 days under storage temperature 4$^{\circ}C$, while that of 'Mihwang' was maintained for 10 days.

Starch Properties of Daehak Waxy Corn with Different Harvest Times (수확시기에 따른 대학찰옥수수의 전분특성)

  • This study investigated starch properties of 'Daehak waxy corn (DWC)' with different harvest times. The DWCs were harvested at 4 days before suitable time (BST), suitable time (ST) and 4 days after suitable time (AST). As harvest time was delayed, starch yield and amylopectin content of DWC starch increased from 43.21 to 52.73%, and from 90.79 to 92.83% based on dry weight, respectively. As harvest time was postponed, enzymatic digestibility and water solubility of DWC starch decreased from 81.43 to 80.58%, and from 10.23 to 9.23%. However, water binding capacity and swelling power of DWC starch increased from 227.94 to 244.88%, and from 24.75 to 29.74%, respectively. Retrogradation viscosity of starch was the lowest in DWC harvested at AST. There was a high correlation coefficient among starch properties of DWC, such as starch yield, enzymatic digestibility, water binding capacity, swelling power, water solubility and retrogradation viscosity (p<0.01). As harvest time was deferred, enzymatic digestibility, water solubility and retrogradation viscosity decreased; however, amylopectin content increased, and water binding capacity and swelling power significantly increased with increasing amylopectin content.

Comparison of Mineral Contents of Three Rice Varieties under Different Cultural Practices (재배조건(이앙시기, 수확시기, 질소시비)별 쌀의 무기질 조성 비교)

  • The objectives of this study were to analyze mineral contents of three rice varieties (Josangheugchal, Heugjinjubyeo and Dongjinchalbyeo) under three different cultural practices: five transplanting time (May 15, May 25, June 4, June 14, and June 24), five harvest times (30, 40, 50, 60, and 70 days) and N-fertilization levels (0.0, 4.5, 9.0, 13.5, and 18.0 kg/10a). Mineral contents of Josangheugchal were increased by late transplanting time with increased nitrogen level; however, it was not significantly different by harvest time. Heugjinjubyeo did not show significant difference in three cultivations. Mineral contents of Dongjinchalbyeo increased at the transplanting time of May 25 and June 14 and according to late harvest time. Also mineral contents of Dongjinchalbyeo that did not show significant difference except for calcium according to different nitrogen levels.

The difference of occurring pattern of Hericium erinaceus by pinheading induction methods (노루궁뎅이버섯(Hericium erinaceus) 봉지재배시 발이유도 처리방법에 따른 자실체 발생양상)

  • In recent years, Hericium erinaceus became famous for comsumer because it contains some materials to assist in human health. This study was carried out to find the difference of occurring pattern of H. erinaceus by pinheading induction methods. Harvest period of T1, T2 treatments were longer than T3~T6. All treatments fresh weights were not significantly different, but T3, T6 treatments's dry weight in 2nd flush were lower than others. T5 treatment's 2nd flush's regeneration rate was the highest than others and looks best quality. Therefore, it is suggested that pinheading induction method would be changed not T1, T2 treatment which were used some farm as Pluerotus ostreatus cultivation, but to scratch surface method which is T5 treatment for cultivation of high quality H. erinaceus.

Plant Regeneration from Leaf Segments Culture of Several Jeju Native Lilies (잎절편 배양에 의한 제주 자생나리의 재분화)

  • This study was conducted to examine the effects of plant growth regulators on the plant regeneration from leaf segments of Lilium callosum, L. concolor var. partheneion, and L. formosanum. Leaf segments were sectioned about 5 mm long and cultured on the basal medium (MS medium with $3\%$ sucrose and $0.8\%$ agar) under dark condition, The most effective plant regulators on harvesting more shoots from leaf culture of L. callosum were $0.2\;mg{\cdot}L^{-1}\;BA$ and $0.5\;mg{\cdot}L^{-1}\;NAA$. Culturing in the basal medium with $0.2\;mg{\cdot}L^{-1}\;BA$ and $2.0\;mg{\cdot}L^{-1}\;NAA$ was effective for leaf culture of L. concolor var. partheneion. The treatment of $1.0\;mg{\cdot}L^{-1}\;BA$ and $1.0\;mg{\cdot}L^{-1}\;NAA$ was the most effective condition for shoot harvest at the leaf culture of L. formosanum.

Study on Wine Quality of Domestic Grape Cultivar 'Cheonghyang' classified by Ripening Stage (국내 육성 품종 '청향' 포도의 숙도별 양조 적성 연구)

  • In this study, Red pearl (4x) and MBA (2x) were crossed Gangwon-do Agricultural Research and Extension Services in 2000 to compare the brewing characteristics of Cheonghyang, a seedless kind of grape registered in 2009, by maturity. Fruit characteristics of Cheonghyang were examined after the first racking and 300 days after fermentation during aging in manufacturing wine based on maturity categories of being immature, mature, and overmature after harvest between August 25 and 27, 2014. The soluble solid matter content did not differ between the first racking following manufacturing wine and during ripening, and the alcohol content following the first racking was 12.9% for immature ones, 13.0% for mature ones, and 12.7% for overmature ones., The alcohol content decreased by appropriately 1.3% to 1.8% during ripening. The total acidity content was 0.9% for immature ones, 0.8% for mature ones, and 0.5% for overmature ones following the first racking and showed insignificant variations during ripening of 0.8%, 0.7%, and 0.5%, respectively, with wine made from mature ones showing proper acidity. The volatile acid content following the first racking during ripening, showing insignificant variation by maturity; however, it decreased after ripening. The sensory evaluation detected differences in acidity of wine between mature and overmature grapes. There was no difference in sour taste between immature and mature grapes; however, overmature ones had low levels of sour taste. Our results showed that mature wine had the best overall preference.

Changes in Marketability of Strawberry 'Maehyang' for Export as Affected by Harvest Time of the Day and Temperature of Precooling and Storage (수출 딸기 '매향'의 일중 수확시기와 예냉 및 저장 온도에 따른 상품성 변화)

  • This study was conducted out to investigate the effect of harvest time of the day, precooling or not, and temperature of precooling and storage on the marketability in strawberries 'Maehyang' for export in May. Strawberry colored with 60±5% of the skin was harvested at 07:00 am or 15:00 pm, respectively. After harvesting, some strawberries were precooled to 0, 2, 4℃ for 3 hours in the cold store, respectively, and the others were kept at room temperature. And then, strawberries were stored at low temperature in the cold store set at 4, 8 or 10℃ storage temperatures. The weight loss rate, firmness, soluble solids content, color, incidence of gray mold of strawberries were measured at two days intervals during storage for 14 days. Both 07:00 am and 15:00 pm harvest, fruits as the storage periods lapses increased weight loss rate compared to the weight at harvest time of the day, and the difference in the weight loss rate of fruits depending on the treatment was greater at 15:00 pm harvest than at 07:00 am. Firmness tended to increase again after 8th day at 07:00 am or 15:00 pm harvest, respectively. In the afternoon harvest, 10℃ storage without precooling showed the lowest fruit firmness on the 2nd day after storage. The soluble solids content at 07:00 am or 15:00 pm harvest tended to be maintained at high value with precooled and stored at low temperature as storage days elapse. The color values were significantly higher at 'L' indicating brightness and lower at 'a', indicating redness at low storage temperature regardless of harvesting time of the day and whether it was precooling or not. The incidence of gray mold was higher at 15:00 pm harvest than at 07:00 am harvest, and it was significantly higher in the treatments stored at 10℃ after precooling similarly. In this study, strawberry 'Maehyang' for export harvested at 07:00 am and stored at 4℃ after precooling at 0℃ maintained the best shelf life, therefore, it is judged that desirable to harvest in the morning with low temperature and to precool and store at low temperature.

Characteristics of Newly Bred Lentinula edodes Cultivar 'Hwadam' for Sawdust Cultivation (표고 톱밥재배용 신품종 '화담' 육성 및 특성)

  • A new oak mushroom cultivar 'Hwadam' was bred from monokaryotic strains of 'GMLE36062' and 'LE15401'. The optimum temperature for mycelial growth of 'Hwadam' on potato dextrose agar was 25℃. The cultivation period of 'Hwadam' lasted for 129 days until the first harvest of the fruiting bodies, including 35 days for spawn run, 84 days for browning, and 10 days for development of fruiting bodies. The fruiting bodies of 'Hwadam' had a hemispherical, brighter pileus and thicker stipe than the control cultivar (Sanjo 701ho). The three-cycle yield of 'Hwadam' was 554 g, which was higher than that of control cultivar (405 g). In addition, 'Hwadam' contained 79.4 mg/g of beta-glucan and 0.93 mg GAEs/g of phenolic compounds, which was similar to that of the control cultivar but higher than that of 'L808' and 'Chujae 2ho'.

Effects of reflective film treatments on fruit quality attributes and peel color variables in 'Fuji' apple fruit

  • This study evaluated the effects of agricultural reflective film and HANSPEK film on the fruit quality attributes and peel color variables of 'Fuji' apples. The reflective films were applied under the tree canopies at 20 and 30 days before harvest (DBH), and a tree with no films was used as the control. Depending on the measurement locations, the light intensity was 7.4 - 14.6% higher in the reflective film treatments than that of the control. However, no difference in the light intensity was observed between the reflective and HASPEK films. In addition, the light intensity was not also different between the 20 and 30 DBH. At harvest, the flesh firmness, fruit weight, soluble solids content, titratable acidity, and internal ethylene concentration were not different regardless of the type of reflective film and treatment time. In addition, the peel color variables were not affected in the equatorial region. However, in the calyx region, the L* and b* values were decreased, and the a* value was increased by the reflective and HANSPEK films compared with the control. Hence, the hue angle was decreased to near 0o (red), and the peel color was improved. Therefore, this study suggests that the application of the HANSPEK film under a tree canopy could improve the fruit peel color of 'Fuji' apples.

Effect of preharvest application of chitosan on the growth and quality of peach fruit (Prunus persica L.)

  • Chitosan with a natural antimicrobial property has been introduced to protect horticultural crops from diseases as an environmentally friendly method. The purpose of this study was to investigate the effects of the pre-harvest application of chitosan on growth and quality during the late stage of fruit development and on the simulated marketing of the peach fruit (Prunus persica L.). The application of chitosan with calcium chloride ($100mg{\cdot}L^{-1}$) three times at one week intervals 4 weeks before the harvest significantly increased the fruit weight, changed the fruit shape, and reduced the fruit length/diameter ratio giving the peach fruits a round oblate shape. The calcium treatment contributed to enhancing or maintaining the storage potential by increasing the flesh firmness. However, at higher concentrations of $CaCl_2$, i.e., > $600mg{\cdot}L^{-1}$, the positive effects of the chitosan application were offset, and fruit growth was not affected by calcium alone. The application of the chitosan/calcium mixture delayed fruit softening; however, this effect was shortened when the storage temperature was $20^{\circ}C$ rather than $15^{\circ}C$. The internal quality of the fruit was profoundly affected by the concentration of calcium added to the chitosan, and delayed fruit maturation was observed at a higher concentration of calcium. The pre-harvest application of chitosan with calcium contributes to the enhancement of food safety by inhibiting the occurrence of diseases during postharvest handling. Considering the above results, chitosan has the potential to improve both the yield of peach fruits and their storability. Because chitosan can enhance the freshness and shelf-life of fresh produce, it is necessary to examine its effects on other horticultural crops.
