Development of Clubroot Resistant Doubled-Haploid Inbred Lines in Kimchi Cabbage (Chinese Cabbage) (Brassica rapa L.)

  • Kimchi cabbage (Chinese cabbage), radish and Cabbage are major Brassicaceae vegetables in Korea. Especially, we can easily develop whole plant from one microspore in Kimchi cabbage. To develop clubroot resistant doubled-haploid (DH) inbred lines, we pollinated a clubroot resistant turnip of 'IT 033820' with a Kimchi cabbage (Chinese cabbage) inbred of 'BP 079'. More than 85 DH inbred lines were developed from this combination. We screened about 400 materials including these DH inbred lines, commercial cultivars and breeding materials during 3 years using hydroponic system after inoculating single spore isolation race 4(SSI-04) inoculate. One inbred line derived from this combination selected as clubroot resistant and registered as 'Wonkyo20036ho'. We inoculated 26 DH inbred lines derived from 'Zoong-baek 2ho' using SSI-4, the percent of resistant plants varied from 0 to 83%. However the horticultural traits of highly resistant DH inbred line was poor. Thus we selected one DH line showing 77% resistant with yellow inner leaf and maid good head, was registered as 'Wonkyo20034ho'. Another DH inbred line derived from Korean variety of 'Wol-dong' showing 86% resistant was registered as 'Wonkyo20037ho'. Other DH inbred lines were derived from Chinese cultivar of 'Choon-hi-go-hang-wang' and 'Hwang-shim-zo48' showed 80 and 71% resistant, respectively, was also selected for registration. Even though DH inbred lines derived from turnip showed highly resistant to SSI-04 and provincial inoculate, they showed poor characteristics in horticultural traits. However, commercial seed companies showed interesting for adapting these DH inbred lines in commercial breeding.

  • We investigated the possible association of interleukin-10 (IL-10) single nucleotide polymorphisms (SNPs) and susceptibility to acute myeloid leukemia (AML) in 115 patients and 137 healthy controls. Genetic analysis of IL-10 SNPs at -819 and -592 was carried out with the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) approach. The IL-10 mRNA expression of AML patients and controls with different genotype was detected by real-time quantitative polymerase chain reaction (RT-PCR). Genetic analysis of IL-10 revealed that the -819AA genotype frequencies and the -819A allele frequencies in the AML group were higher than in the controls (59.1% vs 40.9%; 75.6% vs 63.9%, respectively); there were remarkable differences in -819T/C and -592A/C gene distribution (P<0.05) and the TA haploid frequencies were higher in the AML group (75.6% vs 63.9%, P<0.05). IL-10 mRNA expression in incipient AML patients was obvious higher than the non-tumor group and the remission group ($7.78{\times}10^{-3}$ vs $2.43{\times}10^{-3}$, $3.64{\times}10^{-3}$, P<0.05).The study suggested that the haploid TA and genotype TA/TA may be associated with AML in Han people in Hunan province.The IL-10 SNPs at -819 and -592 sites were associated with AML and may affect IL-10 mRNA expression in AML patients.

  • Accurate analysis of nuclear status is needed when biopsied-blastomeres are used for embryo sexing. In this study, the nuclear status of blastomeres derived from 8- to 16-cell stage IVF bovine embryos was analyzed to evaluate the representative of single blastomere for embryo sexing. When 55 embryos were analyzed by PCR following biopsy, the coincident rate of sex determination between biopsied-single blastomere and matched blastocyst by PCR was 80 %. Karyotyping of biastomeres in 8- 16-cell stage bovine embryos was conducted to assess chromosome status of IVF embryos. To establish karyotyping of blastomeres, concentrations of vinblastine sulfate and duration of exposure time for metaphase plate induction with 8- to 16-cell stage bovine embryos were tested. The most effective condition for induction of metaphase plate (>45%) was 1.0 ug/ml vinblastine sulfate treatment for 15 h. In 22 embryos under the condition, only 8 embryos out of ten that had a normal diploid chromosome complement showed a sex-chromosomal composition of XX or XY (36.4%) and 2 diploid embryos showed mosaicism of the opposite sex of XX and XY in blastomeres of embryo (9.1%). One haploid embryo contained only one X-chromosome (4.5%). Four out of the other 11 embryos having a mixoploid chromosomal complement contained haploid blastomere with wrong sex chromosome (18.2%). These results suggested that morphologically normal bovine embryos derived from IVF had considerable proportion of mixoploid and sex-chromosomal mosaicism which could be the cause of discrepancies of the sex between biopsied-single blastomere and matched blastocyst by PCR analysis.

  • The life cycle of 17 species of Trichoderma was elucidated to seize the proper stage for observing the nuclear behavior and chromosome count. The most convenient stage for the purpose in their life cycle was the stage just before producing the asexual spore. Of the 17 species in the genus Trichoderma the haploid chromosome numbers were counted 5,6,7 and 10. Six chromosomes were most frequently observed. It is believed that the basic chromosome number is placed between 4 and 10, and that the number might be 6, referring to the related papers. It appears necessary to reclassify the single genus of Trichoderma into at least two or three genera.

  • Bulbosum method, which haploids are developed by pollinating with H. bulbosum have been productive to be recommended as an effective method for the production of genetically stable normal barley hybrids. The purpose of this study is to investigate several characters of H. bulbosum, seed set and embryo development rate pollinated with H. bulbosum in order to establish a method for improvement of embryo development and to find conditions favoring plant development from the embryos cultured in vitro. Three lines of H. bulbosum used as pollinators: GBC(2${\times}$), Spenish diploid(2${\times}$) and var. Jaaska (4${\times}$) were morphologically similar, having characteristically narrow leaves, narrow and long culms, long spikes and anthesis duration in comparison with H. vulgare. H. bulbosum, var. Jaaska(4${\times}$) on being pollinated to barley cultivars, increased embryo formation by 8% and plant development in vitro by three times compared to the other diploid lines. the plants developed were not normal barley hybrids but had some H. bulbosum chromosomes uneliminated, indicating that the line was unstable as a pollinator to induce barley haploids.

  • This experiment was conducted to know the effects of mutagen treatments on callus induction, plant regeneration and their ploidy in the anther culture of wheat. The winter wheat cultivars, 'apos;and 'apos;Wonkwang'apos;, were treated at the mid or late-uninucreate stage under 4 different doses (100, 200, 500 and 1,000 rad.) of X-ray and 3 different levels(0.1, 0.2 and 0.3 mole) of Ethyl Methane Sulphonate. The anthers treated were set on the C$\_$17/medium for callus induction, and callus induced was transfered to 1/2 MS medium for plant regeneration. The mutagen treatments inhibited the callus induction but increased the plant regeneration in the callus which were induced from the anther set on the medium for the long time of 60 to 80 days. Also, the chromosome number to the regenerated plant varied largely by increasing of haploid plants(n=3x=21) and by occurring of aneuploidy having n=20 and n=22 of chromosome number.aried largely by increasing of haploid plants(n=3x=21) and by occurring of aneuploidy having n=20 and n=22 of chromosome number.

  • The present study was conducted to compare the relative efficiency of two different haploid breeding methods in tobacco varietal development. A single FI hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L., Bright Yellow4(BY4) and NC95, was used to develop the 30 anther derived dihaploid (ADH) lines and maternally-derived dihaploid (MDH) lines utilizing Nicotiana africana, respectively. As compared with mid-parent, mean of ADH lines showed increase in number of leaves, delay in days to flower and narrow in width of leaf. However, no significant differences in the other investigated characters were recognized. MDH lines also showed narrow width of leaf, while no significant differences in the other characters were observed. The variations of the investigated characters were generally greater in ADH than MDH lines. MDH lines had higher height of plant and shorter days to flower than ADH lines, while the other characters did not show remarkable differences. The degree of heritability for each of the characters observed between ADH and MDH was almost the same. The characters showing high heritability value were height of plant, number of leaves, days to flower, and yield, while those showing relatively low value were length of leaf, width of leaf, and total alkaloid content. Predicted gains from selection for increased yield were calculated for both populations(ADH, MDH) and correlated responses associated with selection for yield were estimated. Height of plant, width of leaf, days to flower, and concentration of reduceing sugar would be expected to improve with selection for yield much faster in the MDH population than in the ADH.
