Genetic Analysis of Carcass Traits in Hanwoo with Different Slaughter End-points (세가지 도축 종료 시점을 공변량으로 하는 한우 도체형질에 대한 유전능력 분석모형)

  • Data from Hanwoo steers and bull calves were analyzed to see the phenotypic and genetic relationships between carcass traits from four different covariance models. Four models fit test station and test period as fixed effect of contemporary group and sire as random effect assuming paternal half-sib relationships among animals. Each model fits one of linear covariate (s) of different slaughter end points-age at slaughter in the first order, age at slaughter in the first and second order, slaughter weight or back fat thickness at 12-13th rib of cold carcass. Age at slaughter in its second order was not significant. Age at slaughter accounted for signifi- cant amount of genetic variances and covariances of carcass traits. Heritability estimates of back fat thickness, rib eye area, carcass weight, marbling score and dressing percentage were 0.34, 0.22, 0.24, 0.42 and 0.18, respectively at constant age basis. The genetic correlation between carcass weight and the other variables were all positive and low to high in magnitude. Genetic correlations between back fat thickness and rib eye area and between marbling score and dressing percentage were low but negative. Variance and covariance structure between these traits were shifted to a great extent when these variables were regressed on slaughter weight or on back fat thickness. These two covariates counteracted to each other but they adjusted each carcass variable or their interrelationship according to differential growth of body components, bone, muscle and fat. Slaughter weight tended to decrease genetic variances and covariances of carcass weight and between component traits and back fat thickness tended to increase those of rib eye area and between rib eye area and carcass weight.

Molecular Characterization of Hanwoo Glucose Transporter 4 Gene (한우 Glucose Transporter 4 유전자의 분자생물학적 해석)

  • The uptake of glucose for metabolism and growth is essential to most animal cells and is mediated by glucose transport protein. In the glucose transport protein family, GLUT4 plays a key role in cellular glucose uptake stimulated by insulin in skeletal muscles and adipose tissue in rodents and human. In this studies, we reported the identification, characterization, and expression of Hanwoo GLUT4 gene. The Hanwoo GLUT4 cDNA includes a 1527 bp open reading frame encoding a protein of 509 amino acids. The GLUT4 amino acid sequences of the Hanwoo show strong conservation with the corresponding sequences reported in other species. The highest mRNA expression of GLUT4 was detected in heart and lower expression was detected in rib meat, sirloin, and colon. We confirmed the expression of GLUT4 in the subcutaneous and small intestinal adipose tissue using RT-PCR. To investigate the expression of GLUT4 in the bovine intramuscular adipose differentiation, fibroblast-like cells were isolated from the sirloin of Hanwoo bull aged 12 months by collagenase digestion of minced tissue and cultured with activators of PPAR gamma. We identified that GLUT4 mRNA expression decreased during differentiation of preadipocytes into adipocyte in Korean cattle. These results indicated that function of GLUT4 in bovine adipose tissue was different from that of mouse and human.

A Comparative Analysis of the Illumina Truseq Synthetic Long-read Haplotyping Sequencing Platform versus the 10X Genomics Chromium Genome Sequencing Platform for Haplotype Phasing and the Identification of Single-nucleotide variants (SNVs) in Hanwoo (Korean Native Cattle) (일루미나에서 제작된 TSLRH (Truseq Synthetic Long-Read Haplotyping)와 10X Genomics에서 제작된 The Chromium Genome 시퀀싱 플랫폼을 이용하여 생산된 한우(한국 재래 소)의 반수체형 페이징 및 단일염기서열변이 비교 분석)

  • In Hanwoo cattle (Korean native cattle), there is a scarcity of comparative analysis papers using highdepth sequencing and haplotype phasing, particularly a comparative analysis of the Truseq Synthetic Long-Read Haplotyping sequencing platform serviced by Illumina (TSLRH) versus the Chromium Genome Sequencing platform serviced by 10X Genomics (10XG). DNA was extracted from the sperm of a Hanwoo breeding bull (ID: TN1505D2184/27214) provided by Hanwoo research canter and used for the generation of sequence data from both the sequencing platforms. We then identified SNVs using an appropriate analysis pipeline tailored for each platform. The TSLRH and 10XG platforms generated a total of 355,208,304 and 1,632,772,004 reads, respectively, corresponding to a Q30 (%) of 89.04% and 88.60%, respectively, of which 351,992,768(99.09%) and 1,526,641,824(93.50%) were successfully mapped. For the TSLRH and 10XG platforms, the mean depth of the sequencing was 13.04X and 74.3X, the longest phase block was 1,982,706 bp and 1,480,081 bp, the N50 phase block was 57,637 bp and 114,394 bp, the total number of SNVs identified was 4,534,989 and 8,496,813, and the total phased rate was 72.29% and 87.67%, respectively. Moreover, for each chromosome, we identified unique and common SNVs using both sequencing platforms. The number of SNVs was directly proportional to the length of the chromosome. Based on our results, we recommend the use of the 10XG platform for haplotype phasing and SNV identification, as it generated a longer N50 phase block, in addition to a higher mean depth, total number of reads, total number of SNVs, and phase rate, than the TSLRH platform.

Development of nuclear Transfer Embryos using Somatic Cell Nuclei from Korean Native Cattle (Hanwoo) with High Genetic Value (고능력 한우 종모우 체세포를 이용한 핵이식 배아의 발달에 관한 연구)

  • This study was conducted to determine developmental ability of reconstructed embryos by nuclear transfer using somatic cell of Korean bull with high genetic value. Fibroblast cells obtained from ear biopsy of the bull were cultured in Dulbecco's Modified Eagle's medium (DMEM) at 37$^{\circ}C$ in air containing 5% $CO_2$. The cummulus-oocyte complexes were collected from slaughterhouse and were matured in vitro for 20 h in TCM 199 culture medium and the oocytes were then enucleated in modified phosphate buffered saline with cytochalasin B. Matured bovine oocytes were enucleated by aspirating the first polar body and metaphase chromatin using a beveled pipette in modified phosphate buffered saline. The ear fibroblast cells were fused into enucleated oocyte by electrical stimulation. The reconstructed oocytes were activated with ionomycine and 6-dimethylaminopurine, and then cultured in CR1aa medium for 7.5 days. Out of 524 bovine eggs reconstructed by nuclear transfer 65.6%(277/422) embryos were cleaved, and 30.7% (85/277) cleaved embryos were developed to the morula to blastocysts. There was no difference of developmental ability in vitro of reconstructed embryos regardless of donor cell passages. In order to determine fate of foreign mitochondria of donor nucleus, the Mito Tracker stained cells were fused into enucleated oocytes. The donor mitochondria were detected early stage of embryos, but disappeared rapidly. The developmental ability of reconstructed embryos was not impaired by Mito Tracker treatments. The results indicate that viable reconstructed embryos can be producted by nuclear transfer using somatic cell of Korean bulls.bulls.

Expression of brain-derived neurotrophic factor and neurotrophic tyrosine receptor kinase-2 in bovine testes

  • Background: Brain-derived neurotrophic factor (BDNF) and its receptor, neurotrophic tyrosine receptor kinase-2 (NTRK2), are well known for their roles in the central nervous and animal reproductive systems. Several studies have observed the extensive expression of BDNF and NTRK2 in non-neuronal tissues, especially reproductive organs. However, most of these studies focused on ovarian development and regulation; thus, scientific research on BDNF and NTRK2 in males is required to determine their roles in the male reproductive system. Therefore, this study aimed to investigate BDNF and NTRK2 expression in bovine testes. Methods: Testes were collected from six Hanwoo bulls (6-8 months old). Reverse transcription-polymerase chain reaction (RT-PCR) analysis was performed to investigate the mRNA expression of BDNF and NTRK2 in the testes. Western blot analysis was performed to verify the cross-reactivity of BDNF and NTRK2 antibodies with bovine testicular tissues. Immunohistochemistry was conducted to determine BDNF and NTRK2 protein expression in the testes. Results: RT-PCR analysis revealed BDNF and NTRK2 mRNA expression in bovine testes. In Western blotting, BDNF and NTRK2 protein bands were observed at 32 and 45 kDa, respectively. Immunofluorescence demonstrated BDNF expression in the nuclei of spermatogonia and Sertoli cells as well as in the cytoplasm of Leydig cells. NTRK2 was exclusively expressed in Sertoli cells. These results suggest that BDNF plays a potential role in spermatogenesis via BDNF and NTRK2 signaling in bovine testes, a finding supported by previous results in different animal species. Conclusions: The expression patterns of BDNF and NTRK2 indicate their functional importance in the bovine reproductive system.

Feed Intake Evaluation of Korean Cattle (Hanwoo) Fed Diets Containing Different Levels of Compound Fattening Periods (한우의 육성 및 비육기간중 배합사료 급여 수준에 따른 사료섭취량 조사)

  • A study was conducted to evaluate feed intake of Hanwoo bulls and steers fed diets of compound feed and rice straw. Twenty bull calves and sixty steers at 5 to 7 months old were used. The experimental period was divided into three feeding stages which are growing period (〈300 kg body weight (BW)), early fattening period (300-450 kg BW) and late fattening period (〉450 kg BW). The animals were given the diets containing 14.1% crude protein (CP) and 70.0% total digestible nutrients (TDN) in the growing period, 12.1% CP and 70.6% TDN in the early fattening period, and 11.2% CP and 71.9% TDN in the late fattening period, respectively. Experiment 1 was designed to compare feed intake (as-fed basis) between Hanwoo bulls and steers fed the experimental diets ad libitum. In Experiment 2, Hanwoo steers were allocated in one of three compound feed feeding treatments to investigate feed intake (as-fed basis). The treatment groups were ① feeding level 1 group fed compound feed ad libitum through the whole periods; ② feeding level 2 group fed 1.0% compound feed per kg BW in the growing period, 1.5% compound feed per kg BW in the early fattening period and compound feed ad libitum in the late fattening period; and ③ feeding level 3 group fed 1.5% compound feed per kg BW in the growing period, 2.0% compound feed per kg BW in the early fattening period and compound feed ad libitum in the late fattening period. In Experiment 1, the average daily feed intake of bulls increased linearly through the whole experimental period while the feed intake of steers increased until their body weight was reached upto 521 kg, afterward reduced. Average daily feed intake was about 3.5% per kg BW of both bulls and steers at the beginning (150 kg BW) of Experiment 1 while bulls and steers at 600 kg BW consumed the diets of 2.0 and 1.5% per kg BW, respectively. In Experiment 2, the average daily feed intake of steers in the feeding level 1 group gradually increased through the growing and early fattening periods and then steadily reduced over the late fattening period. The average daily feed intake in the feeding level 2 group linearly increased through the whole period while the feed intake in the feeding level 3 group showed a relatively rapid increase and reached a peak at 455 kg BW, and then sharply dropped. The average daily feed intake of steers in the feeding level 1 at the beginning (150 kg BW) of Experiment 2 was about 3.5% per kg BW but there was a reduction (1.5% per kg BW) at 600 kg BW. Besides, The feed intake of steers in the feeding level 2 and 3 in which compound feed was given with limitation increased to 2.0-3.0% per kg BW in the growing period and then reduced to 1.5-2.0% per kg BW. The limited compound feed feeding of steers in Experiment 2 resulted in higher rice straw intakes up to two to three folds and two folds in the growing and early fattening periods, respectively, than the ad libitum feeding.

The Effects of Fig Fermented Product Supplementation on Animal Performance, Serum Profile and Meat Quality in Hanwoo Bulls (무화과 발효물의 급여가 한우의 생산성, 혈액성상 및 육질에 미치는 영향)

  • We investigated the effect of fig fermented product(FFP) supplementation on growth performance, serum profile, carcass performance, meat performance and meat quality in 10 bulls of Korean cattle. Concentrates diet was supplemented with substrate fermented from fig fruit and leaves at 10% of the diet. The feed intake of FFP were slightly higher than the control, but the final weight showed no sifnificant difference between the two. Daily weight gain and feed intake were increased in FFP. The serum profile had no significant difference in the treatment. In carcass performance, the meat quantity grade of the treatment had no significant difference, but in meat quality grade the marbling score of FFP was significantly(P<0.05) increased therefore it showed a positive effect on meat quality grade. Also there was no significant(P<0.05) difference of meat cut performance in the treatment. Due to the proximate characteristics of longissimuss muscles the crude fat content of the FFP was significantly(P<0.05) increased. There was no significant difference(P<0.05) in physical characteristics ; pH level, meat color and heat loss of the treatment, but the shear force value and the cholesterol content of FFP significantly(P<0.05) decreased. Crude fat was increased(P<0.05) and cooking loss, shear force and cholesterol concentration were decreased. In fatty acids composition of the FFP, the linoleic acid from the longissimus increased significantly(P<0.05). In subcutaneous fat of longissimus of the FFP, C16:0(palmitic acid) significantly(P<0.05) decreased, but C18:1 significantly (P<0.05) increased. Therefore in FFP, the concentration of saturated fatty acid significantly decreased (P<0.05), but on the other hand the concentration of unsaturated fatty acids significantly (P<0.05) increased. In sensory evaluation of the FFP, the evaluation of odor increased slightly in a positive manner, also the appearance and the taste increased significantly(P<0.05). In conclusion when annexing additional fig fermented product to Hanwoo bulls, the carcass grade improves and the livestock production increases. Also the shear force, lower cholesterol, improved appearance and taste will open the doors to high quality meat production.

Estimation of Genetic Parameters for Growth and Carcass Traits in Hanwoo Bull Populations (비거세 한우 집단에 있어 성장형질과 도체형질에 대한 유전모수의 추정)

  • Genetic parameters for growth and carcass traits were estimated on the basis of the data from 3,415 heads of Hanwoo bulls tested at Livestock Improvement Main Center. The data were divided into two groups : the first period group consisted of 1,592 heads of bulls tested in individual pens from 1986 to 1990 and the second period group consisted of 1,823 heads of bulls tested in group pens from 1991 to 1996. The testing period was from 6 to 18 months of age in the first period group and 10 to 22 months of age in the second period group. The average daily gains during 6 to 12 months and 12 to 18 months were 1.01kg and 0.77kg, respectively, in the first period group and 0.80kg and 0.95kg in the second period group, and the compensated growth appeared to have occurred in the second period group. The heritability of body weight at 12 months estimated was 0.37 during the first period and 0.47 during the second period. The heritability of body weight at 18 months was 0.29 during the first period and 0.33 during the second period. The heritability of average daily gain during 6${\sim}$12 months estimated was 0.26 for the first period and 0.33 for the second period. The heritability of average daily gain during 12${\sim}$18 months estimated was 0.11 for the first period and 0.22 for the second period. The heritabilities of body weights and average daily gains estimated tended to be higher in the second period than in the first period. The heritability of body measurements at 12 months during the first period estimated was 0.30 for withers height, 0.25 for rump height, 0.11 for thurls width and 0.13 for chest girth. The heritability of body measurments at 12 months during the second period was 0.53 for withers height, 0.44 for rump height, 0.36 for thurls width and 0.56 for chest girth. The heritabilites of withers height, rump height, thurls width and chest girth at 18 months of age were 0.40, 0.22, 0.12 and 0.21 during the first period and 0.45, 0.42, 0.42 and 0.35 during the second period, respectively. The heritability of carcass traits estimated during the first period was 0.11 for carcass weight, 0.24 for dressing percent, 0.25 for eye muscle area, 0.18 for backfat thickness and 0.21 for carcass length. The heritabilities of carcass weight, dressing percent, eye muscle area, backfat thickness, carcass length and marbling score during the second period estimated were 0.32, 0.52, 0.33, 0.51, 0.58 and 0.31, respectively. The genetic correlations between the first and second periods estimated were higher than 0.8 for chest girth at 12 months, rump height at 18 months, carcass weight and dressing percent, but were lower for other traits.

  • This study was conducted to study the environment effects on live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score, which are the carcass traits of Hanwoo, based on the estimates and all the possible regression for the selection of variable and significance test for 231,382 heads that underwent the carcass measurements. The average and standard deviation for the live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score were 654.79${\pm}$91.61 kg, 362.30${\pm}$67.15 kg, 59.52${\pm}$0.03%, 81.79${\pm}$12.21 $cm^2$, 11.39${\pm}$5.40 mm, 4.38${\pm}$2.29, respectively. The live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score for cow were 532.79${\pm}$78.38 kg, 313.40${\pm}$44.90 kg, 56.50${\pm}$0.03%, 75.24${\pm}$10.69 $cm^2$, 11.82${\pm}$5.10 mm, 4.30${\pm}$2.06, respectively, while for bull were 619.74${\pm}$93.27 kg, 376.89${\pm}$48.62 kg, 58.61${\pm}$0.02%, 85.61${\pm}$10.46 $cm^2$, 5.64${\pm}$2.71 mm, 1.41${\pm}$0.83, respectively, and for steer were 681.78${\pm}$70.72 kg, 415.23${\pm}$49.43 kg, 60.19${\pm}$0.02%, 88.29${\pm}$10.27 $cm^2$, 12.71${\pm}$5.23 mm, 5.42${\pm}$1.99, respectively. In the environmental variables selection based on the variables selection method, the examination by carcass traits suggested that the most appropriate model could be determined when five variables were selected for the live weight, carcass weight, dressing percentage, eye muscle area, and four variables for backfat thickness, and marbling score. When they were considered at a time altogether based on multiple traits, it was deemed to be desirable to insert all five variables into the variables for analysis. In addition, high significance was found by carcass traits.

Analysis of Carcass Quality Grade Components and ChemicoPhysical and Sensory Traits of M. longissimus dorsi in Hanwoo (한우 도체 육질등급 요인 분석과 육질등급에 따른 이화학 및 관능 특성)

  • The current study was conducted to analysis the effects of sex and live weight on carcass characteristics using total 773 Hanwoo, and to assess the effects of quality grade on chemico-physical and sensory characteristics in longissimus muscle. Results show that both steer and cow received superior quality grade to bull within the same weight c1ass(P < 0.05). Marbling score, texture, maturity, meat color and fat color were significantly( P< 0.01) affected by sex, whilst only marbling score and maturity were significantly affected by live weight. The ultimate quality grade showed significant relationships with marbling score(r = - $0.81^{**}) meat color (r= 0.21^{**}) fat color(r = 0.10^{**}) and with texture(r = 0.41^{**})$ but that had no such a relationship with maturity. WB-shear force and cooking loss were decreased when carcass grade was increased(P < 0.01), but water-holding capacity was identical between the quality grades. Objective meat color dimensions in lightness, redness and yellowness were increased for higher quality grade(P< 0.01). Meat flavor, juiciness and tenderness received higher scores for higher quality grade(P< 0.01). Intramuscular fat content had significantly relationships with WB-shear force(r = 0.$56^*), water-holding capacity(r = 0.18^{**}), juiciness(r = 0.46^{**}), tenderness(r= 0.49^{**})$, and flavor intensity(r = 0.$34^*$). In addition, a higher WB-shear force was related to a lower flavor, tenderness and juiciness scores.
