ACUTE TOXICITY STUDY OF HEPACCINE-B(HEPATITIS B VACCINE)

  • Acute toxicity study was conducted on a Hepatitis B vaccine (Hepaccine-B-inj.) with mice, guinea pigs, and rabbits, in accordance with the norms suggested by the F.D.A. in U.S.A. Dose ranges were 2 doses/mouse, 5 doses/guinea pig, 10 doses/rabbit. They received the vaccine subcutaneously and intraperitoneally. Thereafter, all animals injected were observed of general signsdaily, and of body weight for two weeks. At the end of the observation period (or at the time of death), all animals received the highest dose group were autopsied and gross observation was made on various organs and tissues. No significant toxicity was noted.

  • To investigate the development of a subunit vaccine against theileriosis in cattle, the DNA fragments encoding piroplasm surface protein (p33) of Theileria sergenti of a Korean isolate were expressed in baculoviruses. The expressed p33 was characterized by indirect fluorescent antibody (IFA) and western blotting analysis. The expression of p33 was mainly detected on the surface of infected Sf21 cells by IFA. The immunoblotting analysis revealed the presence of a same molecular weight protein band of p33. The antigenicity of expressed polypeptide was further examined through the inoculation of a guinea pig. The sera of guinea pigs immunized with p33 expressed cell Iysate showed similar fluorescent antibody patterns and reacted with the same molecular weight protein of T. sergenti in immunoblotting analysis, thus indicating that this protein can be a promising candidate for a subunit vaccine in the future.

  • This experiment was performed in order to investigate the effect of radioiodine upon the uterus and the ovary of guinea pigs. The animals were injected single dose of 4. 5 mCi of radioiodine per kg of body weight. They were sacrificed on various time intervals; 1, 7, 14, 28, 42 and 56 days after the injection. The results were as follows: 1. In the ovary, the follicle cells and the ova were degenerated with lapse of time after the injection. 2. In the uterus, hyperplasia of the lamina propria of the endometrium and atrophy of the myometrium were observed in accordance with time after the injection.

  • Due to the fact that an inactivated anthrax vaccine may show no or lower immunogenicity and stability, a number of spore vaccines were exclusively used worldwide. In these studies non or less allergic strain of anthrax bacillus was selected and made a capsulated vegetative organisms. Anthrax organisms of a virulent strain were cultivated on sodium bicarbonate medium with or without adding I-alanine in which B. anthracis grew luxuriantly without forming spores. Inactivation of the organisims was carried out at $37^{\circ}C$ water bath for 3 days after the bacterial culture was mixed with formalin in a final concentration of two per cent. Aluminum hydroxide gel was added to the mixture of anthrax and blackleg bacterin. Guinea pigs were injected with the vaccine via subcutaneous or intramuscular route and challenged after three weeks, and the possibilities of protection was tested. Throughout the studies, the vaccines possibly protected the vaccinated guinea pigs more than 80 per cent compared to that of the controls. This experimental results strongly suggest that the vaccine may possibly applicable to the prevention of bovine anthrax and blackleg.

  • Antigenic potential of a recombinant human erythropoietin (rhEPO) produced by Dong-A charm. Co. Ltd. was examined by active systemic anaphylaxis (ASA) test in guinea pigs, mouse-rat passive cutaneous anaphylaxis (PCA) reaction and passive hemagglutination (PHA) test. In ASA test, rhEPO induced the signs of restlessness, rubbing or licking nose, sneezing and coughing in the animals immunized with rhEPO 1000 lU/kg alone or rhEPO 1000 lU/kg incorporated into Freund\\\\`s complete adjuvant. In the mouse-rat PCA test, only one of six sera from the animals immunized with rhEPO 1000 lUng incorporated into Alum showed positive result. In the PHA test, rhEPO revealed negative results in all of the rhEPO-immunized groups. From these results, rhEPO was considered to produce IgE in guinea pigs and mice, but not IgG and/or IsM in mice. The results of this study were similar to those of the other recombinant human erythropoietin and these positive results were thought to be caused due to the fact that rhEPO were heterogeneous proteins to guinea pigs and mice. Considering the fact that rhErO has an identical structure with indigenous human erythropoietin, rhEPO is not thought to cause immunological problems in clinical use.

Inhibitory effects of 2,6-di-tert-butyl-4-hydroxymethylphenol on asthmatic responses to ovalbumin challenge in conscious guinea pigs

  • This study evaluated the anti-asthmatic activities of 2,6-di-tert-butyl-4-hydroxymethylphenol (DBHP) that is a potent phenolic antioxidant in edible vegetable oil. The effects of DBHP on bronchial asthma were evaluated by determining the specific airway resistance (sRaw) and tidal volume (TV) during the immediate asthmatic response (IAR) and the late-phase asthmatic response (LAR) in guinea pigs with aerosolized ovalbumin-induced asthma. Recruitment of leukocytes and the levels of biochemical inflammatory mediators were determined in the bronchoalveolar lavage fluids (BALFs), and histopathological surveys performed in lung tissues. DBHP significantly inhibited the increased sRaw and improved the decreased TV on IAR and LAR, and also inhibited recruitment of eosinophils and neutrophils into the lung, and release of biochemical inflammatory mediators such as histamine and phospholipase $A_2$ from these infiltrated leukocytes, and improved pathological changes. However, anti-asthmatic activities of DBHP at oral doses of 12.5 to 50 mg/kg was less than those of dexamethasone (5 mg/kg, p.o.) and cromoglycate (10 mg/kg, p.o.), but more potent or similar to that of salbutamol (5 mg/kg, p.o.). These results in the present study suggest that anti-asthmatic effects of DBHP in the guinea pigs model of OVA-induced asthmatic responses principally are mediated by inhibiting the recruitments of the leukocytes and the release of biochemical inflammatory mediators from these infiltrated leukocytes.

Effect of Korean Red Ginseng on Clinical chemical Parameters in Male Guinea Pigs Exposed Acutely to 2, 3, 7, 8-Tetrachlorodibenzo-p-dioxin (2, 3, 7, 8-Tetrachlorodibenzo-p-dioxin 투여로 급성독성을 유도한 웅성 기니픽에 있어 임상화학지수에 미치는 홍삼의 효과)

  • This study was carried out to investigate the protective effect of Korean red ginseng water extract (KRG WE) on clinical chemical parameters in male guinea pigs acutely exposed to 2,3,7,8 tetrachlorodibenzo p dioxin(TCDD). Forty male guinea pigs(200 $\pm$20g) were divided into 4 groups. Normal controls(group 1) received vehicle and saline; group 2(single TCDD treated) received TCDD(5 g/kg, single dose) intraperitoneally; group 3 received KRG WE(200mg/kg, i.p.) for 2 weeks from 1 week before TCDD exposure; group 4 received KRG WE for 1 week since the day of TCDD exposure. Increase in body weight was retarded greatly by TCDD exposure. Body weight of animals in group 2 was significantly decreased starting 2 days after TCDD exposure. However, body weight of animals in group 3 increased throughout the experimental period, although the increasing rate was slower than that of group 1. Decrease in body weight was not observed during the experimental period in group 4. Increases in blood glucose, amylase, lipase, total cholesterol, triglyceride, GOT, GPT, and LDH levels by TCDD intoxication were significantly attenuated by the KRG WE treatment(p<0.05). These results provide a strong evidence that Korean red ginseng might be a useful protective agent against TCDD, an endocrine disruptor.

Specificity of cell-mediated immunity in guinea pigs sensitized with Mycobacterium spp using the leukocyte adherence inhibition test (Leukocyte adherence inhibition test를 이용한 Mycobacterium 속균(屬菌) 감작(感作)기니픽의 세포면역반응(細胞免疫反應)의 특이성(特異性))

  • In order to measure in vitro cell mediated immunity in the guinea pigs sensitized with the killed bacilli of Mycobacterium bovis ($AN_5$), M avium (serotype 2), M tuberculosis and M intracellulare (serotype 8), leukocyte adherence inhibition (LAI) test was established using the antigens of purified protein derivatives (PPD) tuberculin. By using LAI test, specificity of cell-mediated immune responses of the guinea pigs inoculated with various Mycobacterium spp was investigated, and comparison between values of LAI and skin test was also made to evaluate the specificity of the newly designed test. The results obtained throughout the experiments were summarized as follows; 1. The optimal concentration of PPD antigens for LAI test was 1 to 2mg per ml of medium. 2. When the leukocytes of guinea pigs sensitized with both M bovis($AN_5$) and M avium (serotype 2) for 2 to 8 weeks were incubated with homologous or heterologous PPD antigens, mean values of LAI test were $61.2{\pm}11.2$ and $65.6{\pm}5.1%$ in homologous PPD antigens respectively, while $30.0{\pm}3.7$ and $32.8{\pm}5.7%$ in heteNlogous PPD antigens, showing the prominently high value of LAI in the homologous syst,em (p<0.01). 3. When the leukocytes of guinea pigs sensitized with both M tuberculosis and M intracellulare (serotype 8) for 2 to 8 weeks were incubated with homologous and heterologous PPD antigens, mean values of LAI test were $67.9{\pm}2.9$ and $66.9{\pm}5.0%$ in homologous PPD antigens, while $27.4{\pm}7.4$ and $24.4{\pm}7.1%$ in heterologous PPD antigens, showing the prominently high value of LAI in the homologous system (p<0.01). 4. Comparing with the specificity of LAI and skin tests on the basis of the value obtained from the homologous system, deviation of reaction was revealed to be 49.5 to 100.2 in LAI test, and -15.9 to 52.0 in skin test.

Antigenicity Tests of BR92021, a Vi polysaccharide Typhoid Vaccine, in Guinea Pigs (기니픽을 이용한 BR92021(정제 브이아이 장티푸스 백신)의 항원성 평가)

  • To study the antigenicity of BR92021(Vi polysaccharide typhoid vaccine), active systemic ana-phylaxis and passive cutaneous anaphylaxis were tested in guinea pigs. The groups were as follows: group I(low dose, 30 $\mu\textrm{l}$/kg), group II(high dose, 300 $\mu\textrm{l}$/kg), group III(300 $\mu\textrm{l}$/kg plus complete Freund's adjuvant), group IV(positive control, ovalbumin plus complete Freund's adjuvant) and group V(saline-treated control). Male Hartley guinea pigs at 7 weeks of age were sensitized subcutaneously with the test article or saline three times per week for three weeks(j.e., total 9 times). For groups III and IV, animals were sensitized subcutaneously with either the test article or ovalbumin plus complete Freund's adjuvant once per three week for 6 weeks(i.e., total 3 times). Twelve days after the last sensitization, the blood was collected from the sensitized animals for the passive cutaneous anaphylaxis test. In addition, the sensitized animals were subjected to the active systemic anaphylaxis test on fourteen days after the last sensitization by an intravenous challenge with either the test article or ovalbumin. In group I, mild(1/5) or moderate(4/5) symptoms of anaphylactic shock were observed. In group II, no sign(1/5), moderate(3/5) and severe(1/5) symptoms were observed. In group III, four animals of revealed moderate signs and one of 5 showed no signs of anaphylactic shock. In group IV, all 5 animals showed severe signs of shock. In group V, one of 5 revealed moderate and four of 5 showed no signs. The necropsy findings related to the active systemic anaphylaxis were observed in most animals of groups I to V In the passive cutaneous anaphylaxis test, the antiserum was diluted 10- to 5120- fold and was injected intradermally on the clipped back of recipient animals, followed by an intravenous challenge with either the test article or ovalbumin. No animals in groups I, II, III and V showed the positive reaction, whereas all animals in group IV, the positive control, showed the positive reaction at the dilution range of x1280 to x5120. Our results indicate that the test article, BR92021, may have weak antigenic potential in male guinea pigs.

  • The pathogenicity of Br abortus RB51 strains, producted by commercial vaccine companies in Republic of Korea and USA, were evaluated in mouse and guinea pig. BALB/c and ICR mice were intraperitoneally inoculated with RB51 vaccines or virulent field strain and the existence of RB51 including its ratio of spleen to weights and persistence in spleens were examined. Groups of guinea pigs on day 55-58 of received subcutaneously with various RB51 vaccines, RB51 field isolates (Daehungjin) or virulent field isolates(Sangju) to compare the histopathogenicity in uterus. All the mice received RB51 vaccines or RB51 field isolates survived for 10 days, but the groups of mice received virulent field isolates died 5 from 11 (45.5%) in case of BALB/c mice and 12 from 12 (100%) in ICR mice, respectively. The number of RB51 in the groups of mice given with vaccine strains and RB51 field isolates were declined rapidly were in spleens between 12 and 20 days after inoculation. In contrast the mice given with the virulent field isolates rose in number of bacteria up to 20 days after inoculation. In the groups of mice infected with virulent field isolates, the ratio of spleen weights to body weight were significantly higher than those in control or in the groups inoculated with RB51 strain, including RB51 field isolates, at 12 and 20 days after inoculation. At ten days after inoculation, placentas of both the pregnant and non-pregnant guinea pigs were conducted for histopathological examination. Although any abnormal lesions were not observed in non-pregnant guinea pigs, all the strains caused the inflammation of the placenta, implying pathogenecity of RB51 in pregnant guinea pigs.
