  • Treatment of mice with 0.04% anthranilic acid (AnA) as drinking water resulted in an apparent stimulation of pup's growth and food intake of mothers in their first lactations associated with an increased rearing rate and no alteration in plasma prolactin level. AnA showed no significant effects on the day of vaginal opening, the pattern of estrous cycles, plasma growth hormone level and endocrine organ weights of the female and male offspring. The growth of male offspring was significantly retarded by AnA, however, their reproductivity was quite normal. The results indicate the AnA can simulate lactation of mice through little modulation of endocrine systems.

  • We have synthesized acetylsalicylic acid derivative of eugenol. And also, physico-chemical properties and analysis on this compound were examined. The correlation coefficient of the calibration curve in methanol solution was 0.9998. The hair growth stimulation of eugenol derivative on the hair of black mouse (C57BL/6), was also carried out using Implant method. When its ethanol solution was administered to the black mouse by route of skin, eugenol derivative promoted the growth of hair.

  • Ceramide. a product of sphingomyelin hydrolysis, has been proposed as a lipid second messenger mediating antiproliferative activation. In this study, we examined the role of the cell cycle-related proteins in the ceramide-mediated growth suppression. Treatment of U-937 cells with C$_2$-ceramide(N-acetylsphingosine) resulted in growth suppression in a time- and concentration dependent manner. Ceramide induced concentration dependent dephosphorylation of retinoblastoma gene product (Rb). Rb remains hypophosphorylated in synchronized cells even after serum stimulation in the presence of ceramide. Ceramide decreased the expression of cyclin D$_1$ and cyclin E levels. These results suggest that antiproliferative effect of ceramide is associated with hypophosphorylation of Rb and decreased expression of cyclin D1 and cyclin E.

Udenafil Induces the Hair Growth Effect of Adipose-Derived Stem Cells

  • Udenafil, which is a $PDE_5$ inhibitor, is used to treat erectile dysfunction. However, it is unclear whether udenafil induces hair growth via the stimulation of adipose-derived stem cells (ASCs). In this study, we investigated whether udenafil stimulates ASCs and whether increased growth factor secretion from ASCs to facilitate hair growth. We found that subcutaneous injection of udenafiltreated ASCs accelerated telogen-to-anagen transition in vivo. We also observed that udenafil induced proliferation, migration and tube formation of ASCs. It also increased the secretion of growth factors from ASCs, such as interleukin-4 (IL-4) and IL12B, and the phosphorylation of ERK1/2 and $NF{\kappa}B$. Furthermore, concomitant upregulation of IL-4 and IL12B mRNA levels was attenuated by ERK inhibitor or $NF{\kappa}B$ knockdown. Application of IL-4 or IL12B enhanced anagen induction in mice and increased hair follicle length in organ culture. The results indicated that udenafil stimulates ASC motility and increases paracrine growth factor, including cytokine signaling. Udenafil-stimulated secretion of cytokine from ASCs may promote hair growth via the ERK and $NF{\kappa}B$ pathways. Therefore, udenafil can be used as an ASC-preconditioning agent for hair growth.

Effect of KCl and NaCl on Uptake of Proline in Staphylococcus aureus

  • Staphylococcus aureus, the most salt-tolerant food-borne pathogen, produces enterotoxins which may cause symptoms such as vomiting, diarrhea, nausea, and cramps. Since this bacterium has been able to grow at extremely high osmolarity its identity in foods with low water activity values such as salted or dried foods is of great concern. In this study, the growth of S. aureus at high osmolarity has been studied and the doubling time of S. aureus grown at TSB medium containing 15% NaCl has been found to be increased to 4∼5 hours. The stimulation of proline uptake after exposure of cells to high concentration of both extracellular KCl and sucrose was not increased. Stimulation of proline uptake at these environment only occured when 25mM NaCl was present I transport buffer. In additional experiments, the time required to reach mid-logarithmic phase in defined medium of high osmolarity found to be reduce by the presence of glycine betaine, proline, and choline.

  • A protein-polysaccharide fraction of a Korean wild mushroom Psathyrella velutina, PVP, was prepared and its antitumor immunomodulatory activity was investigated. When PVP was administered once daily for seven days from day 1 to day 7 into male ICR mice implanted with 1 $\times$ 10$^{5}$ cells of sarcom 180 tumor cells into the peritoneum on day 4, it inhibited the growth of sarcoma 180 cells by 92.8%. In XTT assay, PVP also exhorted in vitro anti-proliferation activity on U-937, a human monoblastoid cell line, as well as sarcoma 180 cells. PVP showed marked stimulatory activity on the immune system in that it induced the accumulation of PEC (the stimulation index, Sl=4.90 at 100 mg/kg), stimulated the BALB/c mouse splenic lymphocytes to form lymphoblasts (Sl=5.75 at 100$\mu\textrm{g}$/ml), and upergulated the expression of CD25 molecules. All these results strongly support that PVP exhorts its antitumor activity through stimulation of the immune system as well as anti-proliferative activity on the tumor cells.

Effects of Growth Factors and Gut Regulatory Peptides on Glucose Uptake in HC 11 Mouse Mammary Epithelial Cells

  • The large and rapid changes of glucose utilization in lactating mammary tissue in response to changes in nutritional state must be largely related by external signal of insulin. This also must be related with the quantity and composition of the diet in vivo. To characterize the mode of growth factors and gut regulatory peptides with insulin, in vitro experiment was conducted with HC11 cells. All the growth factor alone and the combinations of growth factors significantly (p<0.05) increased in glucose uptake. Insulin, EGF and IGF-1 exhibited a stimulation of glucose uptake for at least 24 h. Furthermore, the highest (p<0.05) synergistic effect was shown in EGF plus IGF-1 and the second synergistic effect in insulin plus EGF while no synergistic effect was found between insulin and IGF-1. However, the gut regulatory peptides neither potentiated nor inhibited the action of insulin on glucose uptake. Although growth factors did not modulates glucose uptake via increasing the rate of translation of the GLUT1 protein, RT-PCR analysis indicated that the growth factors significantly (p<0.05) increased the expression of GLUT1. The growth factors are therefore shown to be capable of modulating glucose uptake by transcription level with insulin in HC 11 cells.

Porcine growth hormone induces the nuclear localization of porcine growth hormone receptor in vivo

  • Objective: Recent studies have challenged the traditional paradigm that growth hormone receptor (GHR) displays physiological functions only in the cell membrane. It has been demonstrated that GHR localizes to the cell nucleus and still exhibits important physiological roles. The phenomenon of nuclear localization of growth hormone (GH)-induced GHR has previously been described in vitro. However, until recently, whether GH could induce nuclear localization of GHR in vivo was unclear. Methods: In the present study, we used pig as an animal model, and porcine growth hormone (pGH) or saline was injected into the inferior vena cava. We subsequently observed the localization of porcine growth hormone receptor (pGHR) using multiple techniques, including, immunoprecipitation and Western-blotting, indirect immunofluorescence assay and electronmicroscopy. Results: The results showed that pGH could induce nuclear localization of pGHR. Taken together, the results of the present study provided the first demonstration that pGHR was translocated to cell nuclei under pGH stimulation in vivo. Conclusion: Nuclear localization of pGHR induced by the in vivo pGH treatment suggests new functions and/or novel roles of nuclear pGHR, which deserve further study.

Growth Stimulation of Lactic Acid Bacteria by a Radish Component (무의 젖산균 증식촉진물질과 촉진작용)

  • Growth stimulatory material for lactic acid bacteria was extracted from radish and radish green juice and its growth stimulatory effect was tested. Dried methanol-precipitated growth stimulatory material was lightly grayish white powder, Its ash content is 44% and approximately 50% of the ash is sulfur. It has reddish brown color upon solubilization in water. The material had unchanged stimulatory effect when it was treated with proteinase or pectinase, or ashed. The growth stimulatory activity was dialyzable. The material was able to counteract the growth inhibitory effect of EDTA. When selected lactic acid bacteria were grown at $30^{\circ}C$ for 24 hours in peptone(0.5%)-yeast extract(0.5%)-glucose(2%) broth with and without 0.5% growth stimulatory material, the material stimulated the growth of Lactobacillus plantarum, L. fermentum, L. leichmanii, L. sake, L. brevis, L. acidophilus, L. casei, Pediococcus pentosaceus, Leuconostoc mesenteroides, Streptococcus faecalis, S. lactis, S. cremoris and S. thermophilus by 19, 1833, 133, 444, 840, 32, 14, 18, 6, 17, 4, 5 and 4 times, respectively.

A Study on Physicochemical Properties of Artificial Substrates and Changes of Plant Growth in Tropical Plant Resources Research Center of Korea National Arboretum (국립수목원 열대온실 내 인공배합토의 물리화학적 특성 및 식물 생육 변화에 관한 연구)

  • This study was carried out in the Tropical Plant Resources Research Center of Korea National Arboretum to determine the optimum edaphic environment for the growth of tropical and subtropical plants. The physicochemical properties of artificial substrates and the growth characteristics of tropical and subtropical plants were investigated. Subtropical plants exhibited a high growth rate when cultivated in a substrate of Dry Zone that had physical properties similar to those of arid native soil. Mediterranean plants showed a low growth rate when grown in a substrate of Subtropical Zone that required changes in acidity. The substrate of Tropical Zone had high organic matter and mineral contents and therefore had good physical properties:this substrate has a good environment for the stimulation of the growth of tropical plants. Our results indicate that the chemical properties such as pH and mineral contents of most artificial substrates need to be more urgently improved than their physical properties in order to ensure better growth of tropical and subtropical plants. Initial management strategies for the construction of new tropical greenhouses were formulated, and data from monitoring studies will be continuously gathered and incorporated in the manual to keep it updated.
