Effect of Night Interruption with Mist and Shade Cooling Systems on Subsequent Growth and Flowering of Cymbidium 'Red Fire' and 'Yokihi'

  • Growth and flowering of Cymbidium 'Red Fire' and 'Yokihi' plants were examined in a greenhouse with cooling systems in summer, and with night interruption (NI) lighting in winter as a forcing culture system. The greenhouse was divided into two sections with separate cooling controls during the summer season. One section was cooled by a mist system (mist), while the other section was cooled by a shade screen (shade). During the winter, the greenhouse was redivided into three sections within each cooling system. Plants were grown with NI either at a low light intensity of $3-7{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(LNI) or a high l ight intensity of $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(HNI) u sing h igh-pressure sodium l amps during the 22:00-02:00 HR. The control plants were grown under 9 h short-day condition. NI for 16 weeks and cooling for 9 weeks were employed twice during the 2 years of the experimental period. The air temperature was approximately $2^{\circ}C$ lower in the mist than in the shade and the relative humidity was 80 ${\pm}5%$ in the mist compared to $55{\pm}5%$ in the shade. The daily light integral in the mist section was 48% higher than in the shade section. The time from initial planting to flowering pseudobulb emergence decreased with both LNI and HNI for both cultivars, regardless of the cooling treatments. Under NI conditions, however, between 60% and 1 00% of plants of both cultivars flowered in the mist, whereas no or 20% of 'Red Fire' or 'Yokihi' plants, respectively, flowered in the shade treatment over 2 years. Plants grown under the mist had bigger pseudobulbs than those grown in the shade under both NI treatments. These results show that commercial use of NI in winter and a mist cooling system in summer would decrease crop production time to 2 years and increase profits in Cymbidium forcing culture.

대장균 배양에서의 유기산 생성과 투석배양에 관한 연구 (A Study of Organic Acid Formation and Dialysis Culture in E. coli Fermentation)

  • 초산이온이 대장균 성장을 저해하는 현상을 규명 하기 위해서 M9 배지, LB 배지를 사용해서 플라스 크, 발효조 배양을 수행하였다. 대장균의 초산 생성 은 높은 포도당 초기 농도, LB 배지와 같은 복합 배 지에서 활발하였고, 플라스크 배양과 같은 pH 조절 이 되지 않는 배양방법에셔는 초산의 생성에 따른 pH 감소에 따라 세포성장이 많이 저해되었다. 초산 을 외부에서 첨가하여 세포성장에 마치는 영향을 조 사하여 보았는데 LB 배지를 사용한 플라스크 배양 에서는 첨가된 초산의 농도가 2g/l 이상, M9 배지 를 사용한 플라스크 배양에서는 초산의 농도가 4g/l 이상, 그리고 M9 배지를 사용한 발효조 배양에 셔는 8g/l 이상에서 초산의 성장 저해효과가 나타 다. LB 한천 배지를 볕에 깐 LB 액체 배지의 플라스 크 배양을 수행하였다. LB 한천의 양이 증가함에 따라서 한천에서 포도당의 방출이 일어나 최종 대장 균 세포 O.D.가 증가하였다.

Induction of Mac-2BP by nerve growth factor is regulated by the PI3K/Akt/NF-κB-dependent pathway in the HEK293 cell line

  • Mac-2BP is a ligand of the galectin family that has been suggested to affect tumor proliferation and metastasis formation. We assessed Mac-2BP expression at the transcriptional and translational levels to evaluate nerve growth factor (NGF)-induced Mac-2BP expression. A time kinetic analysis using reverse transcription-polymerase chain reaction showed that NGF-induced Mac-2BP transcript levels were 4-5 times higher than in controls. Mac-2BP enzyme-linked immunosorbent assay and immuno-fluorescence staining showed a 2-3-fold increase in intracellular and secreted Mac-2BP as a result of NGF stimulation. This increase was regulated by Akt activation and NF-${\kappa}B$ binding. p65 and p50-NF-${\kappa}B$ are major transcriptional factors in the Mac-2BP promoter region, and were shown to be regulated in accordance with the Akt activation states. Collectively, these results suggest that NGF induces Mac-2BP expression via the PI3K/Akt/NF-${\kappa}B$ pathway.

미생물의 크롬내성과 크롬환원의 상호 비교 (A Comparison between Bacterial Cr(VI) Resistance and Cr(VI) Reduction among Environmental Isolates)

  • 미생물에 의한 6가이온 크롬의 3가이온으로의 환원은 세균에게 크롬내성을 제공할 수 있는 것으로 알려져 있다. 본 연구에서는 이러한 가설을 확인하기 위하여 크롬오염 토양과 오염되지 않은 토양으로부터 형태적으로 다른 20종의 세균을 순수분리하고 각각의 균주에서 크롬내성과 크롬환원능을 상호 비교하였다. 전자의 공여체로 glucose를 사용했을 때 세포현탁액에 의한 크롬의 환원은 시간당 0.014-0.305mM Cr(VI)의 범위로 관찰되었으며, 액체배지에서 2mM의 크롬에 대한 내성은 크롬을 첨가하지 않은 배지에서 배양한 경우와 혼탁도를 비교한 결과 전혀 저해를 받지 않는 경우부터 93.4%의 성장저해를 받는 경우까지 다양하게 관찰되었다. 이러한 결과로부터 상관관계를 분석한 결과 크롬내성과 크롬환원은 전혀 상관이 없는 것으로 계산되었다. 한편 군집수준에서의 크롬내성비교는 크롬오염 토양과 오염되지 않은 토양에서 각각 19.1%와 0.4%로 나타나 크롬의 존재는 내성균주의 선택적 성장을 유도하는 것으로 밝혀졌다. 그러나 이때 획득된 크롬에 대한 내성은 크롬의 환원에 의해서 제공되는 것이 아님을 상관관계 분석으로부터 알 수 있었다.

사과 겹무늬썩음병균(Botryosphaeria dothidea)의 Benomyl에 대한 저항성 (Resistance of Botryosphaeria dothidea to Benomyl)

  • 1980년경부터 Botryosphaeria dothidea에 의한 사과겹무늬썩음병이 전국적으로 만연하였는데 그 동안 화학적 방제를 하였으나 병해는 감소되지 않고 현재도 그 피해손실이 극심하다. 본 병의 주요 방제제인 benomyl에 대한 본 병원균의 저항성을 조사한 결과는 다음과 같다. 1. 감수성균은 $150\;{\mu}g/ml$에서 균사생장을 할 수 없었으나, 저항성균은 $300\;{\mu}g/ml$에서 7-l3mm, $2,400\;{\mu}g/ml$에서도 균총직경 6-8mm의 균사생장을 보였다. 후자의 고농도에서 저항성균은 5-9%의 포자발아를 보였으나 감수성균은 포자발아가 없었다. 2. $20-2,400\;{\mu}g/ml$의 모든 공시농도에서 감수성균은 병자각을 형성할 수 없었는데 반하여 저항성균은 저농도에서는 많은 병자각을 형성하였으나 약제농도가 높아질수록 적게 형성하였다.

Genistein Suppression of Matrix Metalloproteinase 2 (MMP-2) and Vascular Endothelial Growth Factor (VEGF) Expression in Mesenchymal Stem Cell Like Cells Isolated from High and Low Grade Gliomas

  • Objective: Brain tumors cause great mortality and morbidity worldwide, and success rates with surgical treatment remain very low. Several recent studies have focused on introduction of novel effective medical therapeutic approaches. Genistein is a member of the isoflavonoid family which has proved to exert anticancer effects. Here we assessed the effects of genistein on the expression of MMP-2 and VEGF in low and high grade gliomas in vitro. Materials and Methods: High and low grade glioma tumor tissue samples were obtained from a total of 16 patients, washed with PBS, cut into small pieces, digested with collagenase type I and cultured in DMEM containing 10% FBS. When cells reached passage 3, they were exposed to genistein and MMP-2 and VEGF gene transcripts were determined by quantitative real time PCR (qRT-PCR). Results: Expression of MMP-2 demonstrated 580-fold reduction in expression in low grade glioma cells post treatment with genistein compared to untreated cells (P value= 0.05). In cells derived from high grade lesions, expression of MMP-2 was 2-fold lower than in controls (P value> 0.05). Genistein caused a 4.7-fold reduction in VEGF transcript in high grade glioma cells (P value> 0.05) but no effects were evident in low grade glioma cells. Conclusion. Based on the data of the present study, low grade glioma cells appear much more sensitive to genistein and this isoflavone might offer an appropriate therapeutic intervention in these patients. Further investigation of this possibility is clearly warranted.

Crosstalk of Zn in Combination with Other Fertilizers Underpins Interactive Effects and Induces Resistance in Tomato Plant against Early Blight Disease

  • The present study was undertaken to evaluate the integrated effect of zinc (Zn) with other nutrients in managing early blight (EB) disease in tomato. A pot experiment was carried out with basal application of the recommended level of macronutrients [nitrogen, phosphorus and potassium (NPK)] and micronutrients [magnesium (Mg) and boron (B)] in bilateral combination with Zn (2.5 and 5.0 mg/kg) in a completely randomized deigned in replicates. Results revealed that interactive effect of Zn with Mg or B was often futile and in some cases synergistic. Zn with NPK yield synergistic outcome, therefore EB disease was managed significantly (disease incidence: 25% and percent severity index: 13%), which resulted in an efficient signaling network that reciprocally controls nutrient acquisition and uses with improved growth and development in a tomato plant. Thus, crosstalk and convergence of mechanisms in metabolic pathways resulted in induction of resistance in tomato plant against a pathogen which significantly improved photosynthetic pigment, total phenolics, total protein content and defense-related enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL)]. The tremendous increase in total phenolics and PAL activity suggesting their additive effect on salicylic acid which may help the plant to systemically induce resistance against pathogen attack. It was concluded that interactive effect of Zn (5.0 mg/kg) with NPK significantly managed EB disease and showed positive effect on growth, physiological and biochemical attributes therefor use of Zn + NPK is simple and credible efforts to combat Alternaria stress in tomato plants.

  • We investigated the effects of a highly saline solution (HS) containing hypochlorous acid, calcium chloride ($CaCl_2$), and phosphoric acid ($H_3PO_4$) on cell death and growth rate of laver Porphyra yezoensis and green laver Ulva intestinalis. Cell death rates of laver treated with HS and HS plus hypochlorous acid (HS + HOCl) in the harvesting stage were less than 0.5%, and there were no significant differences between the HS and HS + HOCl treatments. However, cell death of green laver treated with HS + HOCl in the harvesting stage was greater than 81.2%. These results indicate that the addition of HOCl is highly effective to eradicate noxious green laver without causing damage to laver. The addition of HOCl and $H_3PO_4$ to HS did not increase the area or weight of laver blades. A combination treatment of $CaCl_2$ and HS, however, significantly increased the area and weight of laver lades compared to controls (P<0.05).

반자동화 평가워크플로우 관리 시스템 설계 및 구현 (Design and Implementation of the Semi-automated Evaluation Workflow Management System(Sa-EWMS))

  • 정보화 역기능을 해결하기 위한 정보보호의 중요성이 높아짐에 따라 정보보호제품 및 시스템의 평가인증 수요가 증가하고 있으며 비용-효과적인 평가관리가 필요하다. 따라서, 본 논문은 평가자가 정보보호시스템에 대한 평가를 수행할 때 일련의 절차에 따라 평가업무를 수행 및 관리할 수 있는 CC(Common Criteria :공통평가기준)기반의 반자동화 평가워크플로우 관리 시스템(Sa-EWMS : Semi-automated Evaluation Workflow Management System)을 제시하였다. 본 시스템은 종래 수동적인 평가업무로 인한 시간과 노력의 소비 문제점을 해결하고 효율적인 평가수행을 위한 것으로, 각 엔진별 업무에 대한 워크플로우 프로세스를 추적하고 수행을 조정하는 역할을 하며 평가수요 및 시장창출에 대응하는 민간평가기관에서 유용하게 이용할 수 있을 것이다.

주요 밭작물의 생육단계에 따른 관개 스케줄링의 효율성 평가 (Quality Evaluation of Irrigation Scheduling on Upland Crops by Crop Development Rates)

  • Irrigation scheduling is a water management strategy of applying the proper amount of water in a plant's root zone at the right time to maximize profit. We conducted an experimental evaluation of the response of soybean, sorghum, and sesame to an irrigation scheduling scheme. The soil water contents were adjusted in the root zone between 20% and 28% to reflect changes in crop water consumption. The other ones fixed at 25% during the whole growing season were compared to evaluate the effectiveness of irrigation scheduling. Surface drip irrigation (SDI) were employed as an irrigation method. For all three crops, the evapotranspiration (ET) was the greatest at flowering stage (6.93 mm), followed by vegetative growth stage (5.00 mm) and maturity stage (2.53 mm). The irrigation amount was significantly reduced by 21.8% (soybean), 22.2% (sorghum), and 16.1% (sesame), respectively, compared with the ones at constant soil water content treatment. Their water use efficiency (WUE) were also much higher than the controls: 2.65-fold increase at soybean, 1.82-fold increase at sorghum, and 1.47-fold increase at sesame. These results showed that an effective irrigation scheduling on upland crops (soybean, sorghum, sesame) could increase crop yield while minimizing water use.
