Targeting Analysis of Lumenal Proteins of Chloroplast of Wheat using Proteomic Techniques

  • Plastid proteomics are essential organelles present in virtually all cells in plants and green algae. Plastids are responsible for the synthesis and storage of key molecules required for the basic architecture and functions of plant cells. The proteome of plastid, and in particular of chloroplast, have received significant amounts of attention in recent years. Various fractionation and mass spectrometry (MS) techniques have been applied to catalogue the chloroplast proteome and its sub-organelles compartments. To better understanding the function of the lumenal sub-organelles within the thylakoid network, we have carried out a systematical analysis and identification of the lumenal proteins in the thylakoid of wheat by using Tricine-SDS-PAGE, and LTQ-ESI-FTICR mass spectrometry followed by SWISS-PROT database searching. We isolation and fractionation these membrane from fully developed wheat leaves using a combination of differential and gradient centrifugation couple to high speed ultra-centrifuge. After collecting all proteins to eliminate possible same proteins, we estimated that there are 407 different proteins including chloroplast, chloroplast stroma, lumenal, and thylakoid membrane proteins excluding 20 proteins, which were identified in nucleus, cytoplasm and mitochondria. A combination of these three programs (PSORT, TargetP, TMHMM, and TOPPRED) was found to provide a useful tool for evaluating chloroplast localization, transit peptide, transmembranes, and also could reveal possible alternative processing sites and dual targeting. Finally, we report also sub-cellular location specific protein interaction network using Cytoscape software, which provides further insight into the biochemical pathways of photosynthesis. The present work helps understanding photosynthesis process in wheat at the molecular level and provides a new overview of the biochemical machinery of the thylakoid in wheat.

Effect of Sub- and Super-critical Water Treatment on Physicochemical Properties of Porcine Skin

  • Super- and sub-critical water treatments have been of interest as novel methods for protein hydrolysis. In the present study, we studied the effect of sub-critical water (Sub-$H_2O$, $300^{\circ}C$, 80 bar) treatment as well as super-critical water (Super-$H_2O$, $400^{\circ}C$, 280 bar) treatment on the physicochemical properties of porcine skin (PS), which has abundant collagen. Porcine skin was subjected to pre-thermal treatment by immersion in water at $70^{\circ}C$, and then treated with sub- or super-critical water. Physicochemical properties of the hydrolysates, such as molecular weight distribution, free amino acid content, amino acid profile, pH, color, and water content were determined. For the molecular weight distribution analysis, 1 kDa hydrolyzed porcine skin (H-PS) was produced by Super-$H_2O$ or Sub-$H_2O$ treatment. The free amino acid content was 57.18 mM and 30.13 mM after Sub-$H_2O$ and Super-$H_2O$ treatment, respectively. Determination of amino acid profile revealed that the content of Glu (22.5%) and Pro (30%) was higher after Super-$H_2O$ treatment than after Sub-$H_2O$ treatment, whereas the content of Gly (28%) and Ala (13.1%) was higher after Sub-$H_2O$ treatment. Super-$H_2O$ or Sub-$H_2O$ treatment affected the pH of PS, which changed from 7.29 (Raw) to 9.22 (after Sub-$H_2O$ treatment) and 9.49 (after Super-$H_2O$ treatment). Taken together, these results showed that Sub-$H_2O$ treatment was slightly more effective for hydrolysis than Super-$H_2O$ was. However, both Sub-$H_2O$ and Super-$H_2O$ treatments were effective processing methods for hydrolysis of PS collagen in a short time and can be regarded as a green chemistry technology.

Occurence of Zn-Pb Deposits in Danjang-Myeon, Milyang Area (밀양 단장면 일대에 발달하는 아연-연 광화대의 산출특성)

  • New occurrences of large-scaled Zn-Pb deposits are recently found in the Danjang-myeon, Milyang. They are skarn-type deposits which replaced the intercalated limestone beds in the Jeonggaksan Formation. This study aims at characterizing occurrences, mineralogy, and chemistry of Zn-Pb ores and skarn minerals. Skarn orebodies are mainly found in 3 areas, named Gukjeon-ri, Gorye-ri, and Gucheon-ri orebodies, where sphalerite found as main ore mineral in 200-300 m in height and amount of galena increases as altitude does. Ores are dark grey to dark green in color and closely related with clinopyroxene zone. They occur with hedenbergite, grossular, actinolite, epidote, and small amounts of axinite, calcite, and quartz. Main ore mineral is sphalerite which includes tiny spotted grains of galena and chalcopyrite and becomes rich in grade in association with clinopyroxene and epidote. FeS contents in sphalerite show relatively wide range between 1.53 and 23.07 mole%, whose contents intend to increase towards biotite granite known as ore-related igneous rocks. CdS contents are in the range of 0.22-0.93 mole%, showing decrease tendency from southwest (Gukjeon-ri) to northeast (Gucheon-ri). Zn-Pb deposits developed in Danjang-myeon reveal decrease in temperature with increase of altitude, leading to gradual changes in compositions of ore and skarn minerals.

Regeneration and Acclimatization of Plants Derived from Anther Cultures in Carrot (Daucus carota L.) (당근 약배양에 의한 식물체 재분화 및 순화)

  • Anthers from several lines of carrot (Daucus carota L.) were plated on the semi-solid B$_{5}$, basal medium supplemented with 2,4-D and NAA at two concentrations, 1.0 and 2.0 mg/L plus 0.2 mg/L BAP (benzylaminop-urine). Anthers of the most lines on the B$_{5}$ basal medium with 2,4-D showed higher percentages of callus formation than those with NAA. Particularly, in line 45477, highest percentages of callus formation (50%) were observed on B$_{5}$ medium with 1.0 mg/L 2,4-D plus 0.2 mg/L BAP. With 1.0 mg/L 2,4-D, two months was sufficient for initiation of callus development. Calli were regenerated into plantlets through embryogenesis onto regeneration medium without any growth regulators. When callus showing yellowish and soft structure was cultured, it yielded green plants at high regeneration rates, The response of anthers in callus induction and plant regeneration was different among lines investigated. Optimal callus induction and plant regeneration could be obtained through manipulating the concentration of growth regulators. Plantlets after transfer to perlite were grown successfully in greenhouse conditions. Anther culture of carrot will be used as a useful breeding tool in future.

New Alternate Host of Rice stripe virus - 'Deulmuksae' (벼 줄무의잎마름병의 새로운 중간기주 '들묵새')

  • Rice stripe virus (RSV) is one of Tenuivirus Group, which is carried by small brown planthopper. There is an outbreak of RSV in South Korea at 20Ot, and 2007. The infection caused by RSV had been investigated on weeds around the rice cultivated areas 13 region and 26 site including Jeonbuk Buan and Chungnam Seocheon. There have a doubt as to alternate host of RSV is total 15 Family and 50 Species including Gramineae 24 species of Duksaepul (Alopecurus aequalis), H. sativum var. vulgare etc.. There is identified the infected RSV in Festuca myuros, Alopecurus aequalis, Hordeum sativum var. vulgare, Trisetum bifidum, Echinochloa crus-galli var. crus-galli, Digitaria ciliaris among this species. Deulmuksae is the overwintering exotic weed which sprout in Autumn and wither in Spring and commonly growed as green manure crop or cover crop. In order to identify the infection rate furthermore, 111 samples which were collected at Buan Gyehwa-myeon region, and 50 samples from Seocheon Maseo-myeon in June, 2008, were ELISA tested. The results are 32 positives from Buan, 28.8% infection rate, 8 positives from Seocheon 16.0% infection rate. RSV infection of Deulmuksae is not reported currently, and follow report first describes the Deulmuksae as an alternate host of RSV.

Evaluation of Biomass and Nitrogen Nutrition of Tobacco under Sand Culture by Reflectance Indices of Ground-based Remote Sensors (지상원격측정 센서의 반사율 지표를 활용한 사경재배 연초의 생체량 및 질소영양 평가)

  • Remote sensing technique in agriculture can be used to identify chlorophyll content, biomass, and yield caused from N stress level. This study was conducted to evaluate biomass, N stress levels, and yield of tobacco (Nicotiana tabacum L.) under sand culture in a plastic film house using ground-based remote sensors. Nitrogen rates applied were 40, 60, 80, 100, 120, and 140 percent of N concentration in the Hoagland's nutrient solution. Sensor readings for reflectance indices were taken at 30, 35, 40, 45, 50 and 60 days after transplanting(DAT). Reflectance indices measured at 40th DAT were highly correlated with dry weight(DW) of tobacco leaves and N uptake by leaves. Especially, green normalized difference vegetation index(gNDVI) from spectroradiometer and aNDVI from Crop Circle passive sensor were able to explain 85% and 84% of DW variability and 85% and 92% of N uptake variability, respectively. All the reflectance indices measured at each sampling date during the growing season were significantly correlated with tobacco yield. Especially the gNDVI derived from spectroradiometer readings at the 40th DAT explained 72% of yield variability. N rates of tobacco were distinguished by sufficiency index calculated using the ratio of reflectance indices of stress to optimum plot of N treatment. Consequently results indicate that the reflectance indices by ground-based remote sensor can be used to predict tobacco yield and recommend the optimum application rate of N fertilizer for top dressing of tobacco.

  • The dispersion stability of nano $Y_2O_3-CeO_2$ system was investigated using colloid surface chemistry. Green body of $Y_2O_3$ doped $CeO_2$ was prepared by slip casting in and aqueous system. The dispersion stability of suspension between powders and organic additive was accomplished through electrokinetic behavior of suspension, which was done by ESA apparatus. The dynamic mobility of particles was enhanced when the anionic dispersant of the amount of 1wt% was added. The dissolution of $Y^{3+}$ ion in suspension occurred in the acidic region so that pH value in slurries did not move to below 7.0. In the $CeO_2-Y_2O_3$ system, optimal preparation of suspension was made after adding the anionic dispersant as the amount of 1wt% and pH value of 11.0, and then slip-cast and sintered at 1400$^{\circ}$C, 2 hrs. It appeared relative density of >98% and homogeneous distribution of Y element in depth direction as well as in the microstructure of surface.

Effect of Reaction Conditions for n-Butane Dehydrogenation over Pt-Sn/θ-Al2O3 Catalyst (Pt-Sn/θ-Al2O3 촉매상에서 반응조건에 따른 n-부탄의 탈수소화 반응)

  • Pt-Sn/${\theta}-Al_2O_3$ catalyst for n-butane dehydrogenation reaction was prepared by incipient wetness method. To confirm the physicochemical properties of Pt-Sn/${\theta}-Al_2O_3$ catalyst, the characterization was performed using X-ray diffraction (XRD), $N_2$ sorption analysis, temperature programmed desorption of $NH_3$ ($NH_3$-TPD), temperature programmed reduction of $H_2$ ($H_2$-TPR) techniques. Also, the catalytic activities of Pt-Sn/${\theta}-Al_2O_3$ for n-butane dehydrogenation was tested as a function of pretreatment temperature, pretreatment time, reaction temperature, and the partial pressure of n-butane and hydrogen. The sum of selectivities to n-butenes consisting of 1-butene, cis-2-butene, and trans-2-butene was almost constant 95% in the range of conversion of n-butane 5-55%. The activation energy calculated from Arrhenius equation was $82.4kJ\;mol^{-1}$ and the reaction orders of n-butane and hydrogen from Power's law were 0.70 and -0.20, respectively.

Hepatotoxicity in treatment of canine dermatophytosis with ketoconazole (피부사상균 감염개에서 Ketoconazole 경구투여시의 간독성에 관한 연구)

  • The purpose of this study is to compare hepatotoxicity of each treatment for dermatophytosis; one is the administration of the ketoconazole only and the other, ketoconazole with diphenyl-dimeththyl-dicarboxylate. Have chosen the range of 14-24 months of healthy dogs divided by two groups (group 1 and group 2) for the experiment of which test proved positive in dermatophytosis diagnosis and showed normal reaction in terms of physical examination, blood chemistry and especially of liver function. Group 1 was administrated ketoconazole orally at 10 mg/kg/day and of same dose of ketoconazole with diphenyl-dimethyl-dicarboxylate for group 2. After administering, we have tested two groups by blood collecting every one week in order to check the differences of hepatotoxicity state through AST, ALT and r-GTP, the barometers of liver function which lasted for 12 weeks. Moreover, tested Indocyanine Green (ICG), known as susceptible gauge of function of excretion before starting the experiment and tested ICG as well after 12 weeks. The experiment of result the value of group 1 in AST, ALT and r-GTP has been highly rised after administering ketoconazole for 10 weeks meanwhile, of group 2 has shown a steady state troughout the whole experiment. For ICG test, we injected 0.5 mg/kg of ICG into a vein for both groups and tested the retention rate at regular interval of 15, 30, 45 minutes. The results of retention rate in two groups were similar to before the drug administration. However, after 12 weeks the retention rate of group 1 has been delayed, on the other hand, retention rate of group 2 were a steady state. In conclusion, the administration of ketoconazole only for a long period of time induced hepatotoxicity where as, the administration of ketoconazole with diphenyl-dimethyl-dicarboxylate didn't induce hepatotoxicity. Therefore, when doctors prescribes for a dog with dermatophytosis should not administrate ketoconnazole itself but with diphenyl-dimethyl-dicarboxylate and one who has abnormal condition of liver function should not be prescribed ketoconazole treatment. If there is a case needed to prescribe ketoconazole treatment, the regular monitoring should be accompanied by at the same time.

The Effect of Microalgal Growth on Nutrient Sources Using Microalgal Small Scale Raceway Pond (SSRP) for Biodiesel Production (바이오디젤 생산을 위한 미세조류 옥외배양 시스템의 영양원에 따른 미세조류 성장 특성 비교)

  • The world is in need of sustainable and eco-friendly energy sources such as microalgal biodiesel due to global warming and fossil fuel shortages. In this study, we compared the effectiveness of liquid fertilizer produced from swine manure and agriculture grade solid fertilizers as nutrient sources for microalgal biomass production. Mixed culture (Chlorella spp., Scenedesmus spp., Stigeoclonium spp.; CSS) was cultivated for 28 days in Small Scale Raceway Pond (SSRP) using various nutrient sources (swine manure liquid fertilizer, agricultural solid fertilizer, and mixture of these two fertilizers). Biomass and lipid productivity of fertilizer mixture were the highest at 0.8 g/L and 5.8 mg/L/day, respectively. These results indicate that the fertilizer mixture can provide microalgae necessary nutrient sources for stable biodiesel production and biomass growth. In addition, overall cost of microalgal cultivation and subsequently biodiesel production would be significantly reduced.
