Post-conditioning Periods and Seed Orientation Affects the Vigor of Cucurbit Seeds with Dry-heat Treatment (건열처리에 따른 박과채소종자의 활력 회복)

  • We investigate the effect of post-conditioning periods and seed orientation on the vigor of cucurbit seeds with dry heat treatment (DHT). All the dry-heat treated seeds exhibited varying degree of seed vigor decreases. In general, pumpkin seeds showed less vigor decreases than the bottle gourd seeds. When the dry heat treated seeds were germinated after post-conditioning for 0, 30, and 120 days, the percentage of germination was enhanced by increasing the period of post-conditioning and the efficiency of post-conditioning differed by crop and cultivar. In both bottle gourd and pumpkin, the vigor of seeds placed in vertically upward and horizontal orientations was higher than that of the seeds placed in the vertically downward orientation. The results suggested that the vigor of dry-heat treated seeds could be improved by applying the proper post-conditioning and seed orientation.

Effect of Seed Coat Softening, Washing and Drying on Seed Germination of Gourd (종피의 연화처리, 세척 및 건조가 박 종자의 발아에 미치는 영향)

  • The present experiment was carried out to determine the effect of seed-coat softening by various chemicals, washing, chilling, and light treatment alone and in combination during desiccation of seeds on germination of gourd (Lagenaria siceraria Standl.) seeds. Potassium hydroxide, sodium hydroxide, acetone, acetonitrile, and acetamide were used as chemicals for the softening. Washing, prechilling, drying and light treatment of the softened seeds were done for a hour with tap water, for one or three weeks at 3$^{\circ}C$, 12 hr at 35$^{\circ}C$, and 14 hr a day at 3$0^{\circ}C$ with red light illumination or darkness, respectively. FR-yongjadaemok and FR-kunghap were used as test cultivars. Seed-coat softening with 10% acetone and acetonitrile for 1 hour enhanced and accelerated seed germination compared to the other chemicals and water imbibition. KOH treated for the softening was necessary to increase the germination rate of seeds for following chilling, but acetone did not influence their germinability regardless of chilling. Washing the softened seeds in tap water increased the germination rate in comparison with no-washing. During desiccation of the seeds sequentially treated with the softening, washing and chilling, red light treatment showed higher germination rate than dark one, suggesting that red light treatment are necessary during desiccation.

Sing1e or Combined Treatment Effect of Priming, Gibberellin and Prechilling on Germination of Gourd Seeds

  • The present experiment was done to determine the effect of priming, $GA_3$ and prechilling treatment alone and in their combination on germination of gourd (Lagenaria siceraria Standl.) seeds. Priming using $KNO_3$ and $GA_3$ were treated with different concentrations and periods whereas prechilling was done at only different periods. FR-yongjadaemok and FR-kunghap were used as test cultivars and their daily germination rates were measured at treatment levels. The best germination rate of each treatment was shown in a week prechilling at $3^{\circ}C$, a day priming with 100 mM $KNO_3$ and a day treatment with 0.01 mM $GA_3$ although the prechilling had the highest rate among them. The rate of priming treated after the prechilling was continuously decreased with its longer treatment periods and was less than no-priming. In addition, the rate of $GA_3$ treatment after the prechilling was not affected by its concentrations and treatment periods. Priming and $GA_3$ treatments before the prechilling, however, showed similar germiabilities between their treatment levels including no-treatment. The results mean that sequential order of available presowing treatments is important to enhancing the rate.

Vinegar Produced from Chrysanthemum zawadskii and Pearl Shell (구절초진주식초)

  • With the addition of ethanol to wax gourd extract and by acetic fermentation, 5.0% acidity vinegar was produced. After putting 10% extract(10% concentration) of Chrysanthemum zawadskii in this, and by dissolving shell, Chrysanthemum zawadskii-pearl vinegar was produced. When a 1% of ark shell, oyster shell, or ear shell was added to wax gourd vinegar, 95.6~98.4% of the shell dissolved, and when a 2% content of shell was added, 97.2~98.4% was dissolved. The acidity of vinegar which dissolved 1% shell was pH 3.0~3.17, and the acidity of vinegar which dissolved 2% shell was pH 1.11~ 1.20. The pH values of vinegar which dissolved 1%, and 2% shell contents were 4.54~4.55, and 4.86~4.95, respectively. When 1% shell was dissolved, the acidity was higher than that of commercial vinegar, with a high pH value and low level of free acid. This shows that when Chrysanthemum zawadskii 1% is added during acetic acid fermentation, the inhibition was 44.4%, and 22.2% respectively. In this regard, Chrysanthemum zawadskii should be added after the fermentation of acetic acid. The calcium content of 1% shell vinegar is 0.4%, and that of 2% vinegar is 0.78%. Non-heated native wax gourd shows an angiotensin converting enzyme inhibition rate of 21.7%, an antioxidant activity of 5.23%, and a tyrosinase inhibition rate of 5.5%. In the case of heated-extracted wax gourd, the angiotensin converting enzyme inhibition rate was 16.1%, superoxide dismutase activity was 20.5%, antioxidant activity was 23.2%, and the tyrosinase inhibition rate was 7.1%. Also, in the case of Chrysanthemum zawadskii, the angiotensin converting enzyme inhibition rate was 28.8%, the xanthine oxidase inhibition rate was 28.2%, the superoxide dismutase activity was 14.5%, the antioxidant activity was 3.2%, and the tyrosinase inhibition rate was 9.2% Data also revealed that when a 10% sample of the heated-wax gourd extract was added to A549 human lung cancer epithelial cells of, the number of cancer cells declined by 80% in 72 hours, When a 10% native extract was added, the number of cells declined by, 74% in 48 hours, and when a heated-extract of Chrysanthemum zawadskii was added, 100% of the cells died after 72 hours.

Production Techniques for the Metal Artifacts Excavated in Hasong-ri, Yeongwol and Their Conservation Treatment (영월 하송리 출토 금속유물 보존처리 및 제작기법 연구)

  • This study describes the processes undertaken for the conservation treatment of metal artifacts excavated in Hasong-ri, Yeongwol, Gangwon-do and the results of the related surface composition analysis. X-ray fluorescence analysis (XRF) was applied to analyze the surface composition of a small gilt-bronze wind chime, two clappers, and a small gourd-shaped bronze bottle. The gourd-shaped bronze bottle was investigated using radiography to examine its internal structure. The wind chime and clappers were excavated from the same location. A gilt layer was identified on the wind chime, but surface corrosion made it difficult to identify any such layers on the clappers. The element analysis revealed that the wind chime was made of bronze in a Cu-Sn-Pb ternary system and was gilt-plated using mercury amalgam. The clappers were made from copper with a small amount of gold detected, but did not show any evidence of mercury. Since a thick corrosion layer was affixed to the surface of each clapper, it was impossible to identify the surface composition and determine if the clappers had been gilded. It is possible that the gold detected from the clappers was a foreign substance or had detached from the giltbronze wind chime buried alongside them. The small gourd-shaped bronze bottle was investigated through surface element analysis and radiography to verify if it was a type of silver bottle used as currency during the Goryeo dynasty. The radiography photography identified that a small hole in the middle of the base had been stopped up. The general surface analysis did not detect silver, which suggests that the bottle was made of bronze instead and was not one of the silver bottles used as a means of currency.

Studies on Pulping of Sponge Gourd Net Fiber - Analysis of Morphology and Characteristics of Pulps - (수세미외 섬유의 펄프화에 관한 연구-섬유의 구조와 펄프화별 특징 분석-)

  • Studies were carried out on the preparation of several kinds of pulps from Sponge gourd fiber by KP, ASP, SP PAP, AP and addition of AQ pulping process. These unbeaten and beaten pulping fibers were observed their characteristics and fiber structure by SEM, FQA, Image analyzer and Micro projector. The results were summarized as follows; 1) The cooking condition which is the possible defibrilation of Shives are KP base($160^{\circ}C$, 2hr.), ASP base($155^{\circ}C$, 4hr.), PAP base($160^{\circ}C$, 1hr.). From the results, the kappa no. had the range of 12, 25, 10 each other. 2) The pulp yields of sponge gourd fiber obtained the range of KP 50~55%, ASP&60~70% and PAP 45~50%. SP base have the highest and contnets of KP&PAP base are much the same as woods. 3) Increasing amount of NaOH on Pulping was accelerated the defibrilation of Shives and was changed a morphology of pulping fiber quality such as fiber length, curl and kink index. 4) Addition of AQ on pulping process of sponge gourd fiber had a affect to raise the rate of delignification while protecting cellullosic components against degradation, especially defibrilation was very excellent, beated pulp much more easily and increased the fibrilation. 5) ASP system have higher bulk density, fiber bonding and protecting cellullosic components against degradation than KP or PAP. 6) The color reactions of the "C" stain solution showed blue or blue-gray with clean and transparency thin cell wall.

Charantin Contents and Fruit Characteristics of Bitter Gourd (Momordica charantia L.) Accessions (여주의 유전자원별 과실특성과 Charantin 성분 함량)

  • Bitter gourd (Momordica charantia L.) has long been used for food and medicinal plant in Korea, China and Japan. This study aimed at evaluating productivity, and vitamin-C and charantin contents in bitter gourd (Momordica charantia L.) accessions. The contents of charantin of these two accessions were analyzed using HPLC with the UV-diode array detection. The highest fruit yield was observed in accessions, 'BG1' and 'BG7.' The vitamin-C contents of fruits in these two high-yield bitter gourd accessions, 'BG1' and 'BG7,' depended on days after fruit set and were highest in 24 days and 17 days after fruit set, respectively. The charantin contents of the two accessions were different according to the number of days after fruit set. The charantin content of 'BG1' was highest in fruits harvested at 24 days and followed by 15 days after fruit set. The charantin content of 'BG7' was highest in fruits harvested at 13 days and followed by 16 and 19 days after fruit set. The charantin contents of 13 M. charantia accessions with relatively high yield potential were analyzed and three accessions, 104615, K169995 and NS454, were selected based on their relatively high yield and charantin content. These accessions will be used for breeding program and processed foods.
