Protein Engineering of an Artificial Intersubunit Disulfide Bond Linkage in Human Dihydrolipoamide Dehydrogenase

  • Dihydrolipoamide dehydrogenase (E3) belongs to the protein family of pyridine nucleotide-disulfide oxidoreductases, including glutathione reductase (GR). The two subunits of human GR are covalently linked by an intersubunit disulfide bond between the pair of the Cys-90 residues. The corresponding residue (Ser-79) in human E3 was substituted to Cys using site-directed mutagenesis. The mutant was expressed in Escherichia coli and highly purified using an affinity column. About 40% of the mutants formed a spontaneous intersubunit disulfide bond linkage. This result implies that Ser-79 and possibly surrounding residues constitute one of the several intersubunit contact regions in human E3. It provides another good piece of evidence for the predicted high degree of the structural homology between human E3 and GR. Spectroscopic studies indicate conformational changes in the mutant.

  • A new stereoscopic PTV was developed using two CCD cameras, stereoscopic photogrammetry based on a 3D-PTV principle. The arrangement of the two cameras was based on angular position. The calibration of cameras and the pair-matching of the three-dimensional velocity vectors were based on Genetic Algorithm based 3D-PTV technique. The constructed Stereoscopic W technique was tested on the standard images of the impinged jet proposed by VSJ. The results on the turbulent properties of the jet obtained by the constructed system showed a good agreement with the original LES data.

  • The embedded atom method based on density functional theory was developed as a new means for calculating ground state properties of realistic metal system by Murray S. Daw, Stephen M. Foiles and Michael I. Baskes. In the paper, we had corrected constitutive formulae and parameters on the nickel for the purpose of doing Embedded Atom Method analysis. And then we have computed the properties of the nickel on the fundamental scale of the atomic structure. In result, simulated ground state properties, such as the lattice constant, elastics constants and sublimation energy, show good agreement with Daw's simulation data and with experimental data.

  • A fuzzy control scheme has been proposed for a supercritical extraction process which has attracted much attention recently as a new separation technology. Based on the manual operation experience, three control pairs between manipulated and output variables are selected first and then seven membership functions are defined for control error and time rate of the error, respectively for each control pair, resulting in forty nine Fuzzy control rules. In addition to these, the membership functions are defined in two steps (coarse and fine) to enhance control performance. Fuzzy inference is performed using MAX-MTN composition rule and defuzzified control output is calculated based on center of gravity method. The prosed Fuzzy control scheme has been assessed through numerical simulation. As a result, the proposed scheme shows good control performance comparable with that by INA(inverse nyquist array) which usually requires complicated design procedure.

  • Curretly, the development of MEMS(Micro Electronic Mechanical System) technology awakes many research's interest for the aerodynamics. This work presents the development of a compact synthetic jet actuator for flow separation control at the flat plate. The formation and evolution of fluidic actuators based on synthetic jet technology are investigated using Reynolds-Averaged Navier-Stokes equations. Also, 2-Dimensional, unsteady, incompressible Navier-Stokes equation solver with single partitioning method for Multi-Block grid to analyze and a modeled boundary condition in developed fo. the synthetic jet actuator. Both laminar and turbulent jets are investigated. Results show very good agreement with experimental measurements. A jet flow develops, even though no net mass flow is introduced. Pair of counter-rotating vortices are observed near the jet exit as are observed in the experiments.

  • Surface roughness is one of the most important parameters to estimate quality of products. As this reason. so many studies were carried out through various attempts that were contact or non-contact using computer vision. Even though these efforts, there were few good results in this research. However, texture analysis is making a important role to solve these problems in various fields including universe, aviatiion, living thing and fibers. In this study, texture parameter was obtained by means of position operator according to variation of angle direction and distance. As a result, it was found that surface texture was more effected by direction then distance

  • To be reversible as a data hiding method, the original content and hidden message should be completely recovered. One important objective of this approach is to achieve high embedding capacity and low distortion. Using predicted errors is very effective for increasing the embedding capacity. Sorting the predicted errors has a good influence on decreasing distortion. In this paper, we present an improved reversible data hiding scheme using upgraded histogram shifting based on sorting the predicted errors. This new scheme is characterized by the algorithm which is able to find the optimum threshold values and manage the location map effectively. Experimental results compared with other methods are presented to demonstrate the superiority of the proposed method.

  • Emotion-Specific Physiology may still remain to bean elusive entity even to many of the proponents and seekers, but an ever-growing body of experimental evidence sheds much brighter prospects for the future researches in that direction. Once such Emotion-Physiology pairs are identified, there exist a high hope that some Sense-Friendly Features that are causally related, or highly correlated, to each pair may be identifiable in the nature or man-made objects. On the premise that certain emotions, if and when engendered by a consumer good, may be conducive to an urge “to own or to identify oneself with the product”, presented here is a model of Sensory Engineering that is oriented objectively towards identifying the Emotion-Specific Physiology in order to have the Sense-Friendly Features reproduced in product designs. Relevant and complementary concepts and some suggested procedures in implementing the proposed model are offered.

A Revocable Fingerprint Template for Security and Privacy Preserving

  • With the wide deployment of biometric authentication systems, several issues pertaining security and privacy of the biometric template have gained great attention from the research community. To resolve these issues, a number of biometric template protection methods have been proposed. However, the design of a template protection method to satisfy four criteria, namely diversity, revocability and non-invertibility is still a challenging task, especially performance degradation when template protection method is employed. In this paper, we propose a novel method to generate a revocable minutiae-based fingerprint template. The proposed method consists of feature extraction from fingerprint minutiae pairs, quantization, histogram binning, binarization and eventually binary bit-string generation. The contributions of our method are two fold: alignment-free and good performance. Various experiments on FVC2004 DB1 demonstrated the effectiveness of the proposed methods.

Performance Improvement of Sound Direction of Arrival Estimation by Applying Threshold to CPSP (CPSP 문턱값 설정을 통한 음원도달 방향 추정 성능 개선)

  • To estimate sound direction of arrival with a pair of microphones, a method based on Time Difference of Arrival (TDOA) estimation using the Cross Power Spectrum Phase (CPSP) function is largely used due to its simplicity and good performance. In this paper, we investigate CPSP maximum values for various SNRs and adverse environments, and propose a novel method to improve the estimation performance of sound direction of arrival. The proposed method applies a threshold to the CPSP values and increases the reliability of the estimated sound direction. Through computer simulation for various SNRs, we validate the effectiveness of the proposed method. When the threshold was set to 0.1, more than 90% of success rate of sound direction of arrival estimation has been achieved for directions of $10^{\circ}$, $40^{\circ}$, $70^{\circ}$ from the source location even with reverberation times of 0.1s.
