The Effects of Sasammaickmoondong-tang against Duodenal Mucosal Lesions (Mouse의 십이지장 점막 염증에 대한 사삼맥문동탕의 면역반응 연구)

  • Objectives : This study was carried out to investigate the effects of Sasammaickmoondong-tang(SME) on duodenal mucosal lesions induced by indomethacin in mouse. Methods : The normal group is that no inflammation elicitated mouse. Control group is that gastro-inflammation elicitated mouse. Sample group is that SME administered mouseice after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, control group was observed various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell-by hemorrhagic erosion, while sample group was as same as normal group. In the immunohistochemical change, the distributions of COX-1, Bcl-2, and BrdU treated with SME noticeably increased than control group(p<0.05). The distributions of TUNEL, $NF-{\kappa}B$, COX-2, $IL-2R-{\alpha}$, NK-1.1, ICAM-1, and CD11b/18 treated with SME noticeably decreased than control group(p<0.05). And the distribution of SBA was as same as normal group. Conclusion : According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to duodenal mucosal lesions.

Effect of mushroom (Schizophyllum spp.) derived β-glucan on low-fiber diet induced gut dysbiosis

  • Dietary pattern has paramount importance in shaping the gut microbiota and its associated host health. Herein this study, long term (12 weeks) impact of mushroom derived dietary fiber, ${\beta}-glucan$, is investigated for its effect on low fiber diet consumption. Inclusion of dietary fiber into the low fiber diet (LFD) increased the abundance of genera Lactobacillus and Anaerostipes, the microbes responsible for butyrate (major 'fuel source' of colonocytes) production. Mice fed LFD with ${\beta}-glucan$ showed significant increase in the length of small intestine compared to that of the LFD group without ${\beta}-glucan$. Further, dietary fiber consumption enhanced goblet cell density along with mucosal layer thickness. These results indicate promising effects of ${\beta}-glucan$ towards maintenance of healthy gut and gut microbiota.

Latilactobacillus curvatus BYB3 Isolated from Kimchi Alleviates Dextran Sulfate Sodium (DSS)-Induced Colitis in Mice by Inhibiting IL-6 and TNF-R1 Production

  • Recent studies have shown that probiotics have health-promoting effects, particularly intestinal immune modulation. In this study, we focused on the immunomodulatory properties of Latilactobacillus curvatus BYB3, formerly called Lactobacillus curvatus, isolated from kimchi. In a mouse model of 14-day dextran sulfate sodium (DSS)-induced colitis, treatment with L. curvatus BYB3 significantly decreased the disease activity index, colon length, and weight loss. Moreover, histological analyses showed that L. curvatus BYB3 protected the structural integrity of the intestinal epithelial layer and mucin-secreting goblet cells from DSS-induced damage, with only slight infiltration by immune cells. To evaluate the molecular mechanisms underlying L. curvatus BYB3-driven inhibition of interleukin 6 production, possible in vivo anti-inflammatory effects of L. curvatus BYB3 were examined in the same mouse model. In addition, significantly lower levels of IL-6 and tumor necrosis factor receptor 1 upregulation were seen in the DSS+BYB3 group (compared to that in the DSS group). These results indicate that L. curvatus BYB3 exhibits health-promoting effects via immune modulation; and therefore, it can be used to treat various inflammatory diseases.

Histochemistry of the Developing Chick Respiratory Tract (鷄胎呼吸氣道의 組織化學的 硏究)

  • A histochemical study of the respiratory tract in developing chick was done to demonstrate PAS-postivie materials, ribonucleic acid, phospholipid, and alkaline phosphatase activity. Following results are obtained: 1. The alkaline phosphatase activity was found to be high before the appearance of cartilage in mesenchyme surrounding the tracheal epithelium. The enzyme activity declined after the cartilage formation, followed by the restricted activity in epithelium in the postembryonic stage. 2. A moderate positive reaction of ribonucleic acid was found in the cytoplasm of the epithelium and undifferentiated mesenchyme. As the cartilage grew differentiated the reaction of ribonucleic acid was found to disappear in the mesenchyme surrounding the epithelim, but the cytoplasm of the glands showed a moderate positive reaction. 3. Goblet cells of the mucosa and glandular cells showed highly positive reaction, but the basement membrane exhibited slightly positive PAS-reaction. 4. Epithelial cells of the mucosa showed a weak to moderate reaction. However, the epithelia of bronchiol and alveoli in the differentiating period and glandular cells showed a strong positive reaction in Baker's hematein test.

Synbiotic of Pediococcus acidilactici and Inulin Ameliorates Dextran Sulfate Sodium-Induced Acute Ulcerative Colitis in Mice

  • Colitis is a major gastrointestinal disease that threatens human health. In this study, a synbiotic composed of inulin and Pediococcus acidilactici (P. acidilactici) was investigated for its ability to alleviate dextran sulfate sodium (DSS)-induced colitis. The results revealed that the synbiotic, composed of inulin and P. acidilactici, attenuated the body weight loss and disease activity index (DAI) score in mice with DSS-mediated colitis. Determination of biochemical indicators found that the synbiotic increased anti-oxidation and alleviated inflammation in mice. Additionally, histopathological examination revealed that colonic goblet cell loss and severe mucosal damage in the model group were significantly reversed by the combination of inulin and P. acidilactici. Moreover, synbiotic treatment significantly reduced the levels of IL-1β, TNF-α, and IL-6 in the serum of mice. Thus, a synbiotic composed of inulin and P. acidilactici has preventive and therapeutic effects on DSS-induced colitis in mice.

Histochemical study on the tracheal development in fetuses and neonates of Korean native goats (한국재래산양의 태아 및 신생아 기관의 조직발달에 관한 조직화학적 연구)

  • The present study was performed to investigate the patterns of appearance of lectin in trachea of fetuses of 60, 90 and 120 days old and neonates of Korean native goat. Carbohydrate markers were used in histochemistry for the determination of the lectin by staining of avidin-biotin-peroxidase complex(ABC), and the markers consisted of biotin-labeled concanavalin A(Con A), dolichos biflorus agglutinin(DBA), rincinus communis agglutinin(RCA-I), ulexeuropalus communis agglutinin(UEA) and wheat germ agglutinin(WGA). 1. The Con A-binding reactions appeared moderately on the apical surface of the tracheal epithelia in 60 days old fetuses, and the reactions were similar on the tracheal epithelia and glands in 90 and 120 days old fetuses and neonates. 2. Reaction of the DBA appeared as the strongest meanwhile the DBA-binding reactions were determined strongly on the apical surface of the tracheal epithelia in the 60 days old fetuses. Reaction for the DBA on the tracheal epithelia and glands of 90 and 120 days old fetuses and neonates were in same manner. 3. The RCA-I-binding reactions appeared very strongly on the apical surface of the tracheal epithelia in 60 and 90 days old fetuses. Reaction to the RCA-1 appeared moderately on certain apical surface of tracheal epithelia and glands in 120 days old fetuses and neonates. 4. No reactions provoked for the UEA in trachea of 60 days old fetuses and neonates, but the UEA-binding reactions appeared moderately in the tracheal epithelia of 90 days old fetuses and weakly in 120 days old fetuses. 5. The WGA-binding reactions appeared very strongly on the apical surface of the tracheal epithelia in 60 and 90 days old fetuses, and moreover, the reactions were determined on the luminal surface of the tracheal gland in 90 days old fetuses. On the other hand, goblet cells of the tracheal epithelia and glands in neonates reacted moderately to the WGA.

Enhancement of Allergen-induced Airway Inflammation by NOX2 Deficiency

  • Background: NADPH oxidase (NOX) modulates cell proliferation, differentiation and immune response through generation of reactive oxygen species. Particularly, NOX2 is recently reported to be important for regulating Treg cell differentiation of CD4+ T cells. Methods: We employed ovalbumin-induced airway inflammation in wild-type and NOX2-deficient mice and analyzed tissue histopathology and cytokine profiles. Results: We investigated whether NOX2-deficiency affects T cell-mediated airway inflammation. Ovalbumin injection which activates T cell-mediated allergic response increased airway inflammation in wild-type mice, as evidenced by increased immune cell infiltration, allergic cytokine expression, and goblet cell hyperplasia in the lung. Interestingly, NOX2 knockout (KO) mice were more susceptible to allergen-induced lung inflammation compared to wild-type mice. Immune cells including neutrophils, lymphocytes, macrophages, and eosinophils were drastically infiltrated into the lung of NOX2 KO mice and mucus secretion was substantially increased in deficiency of NOX2. Furthermore, inflammatory allergic cytokines and eotaxin were significantly elevated in NOX2 KO mice, in accordance with enhanced generation of inflammatory cytokines interleukin-17 and interferon-${\gamma}$ by CD4+ T cells. Conclusion: These results indicate that NOX2 deficiency favorably produces inflammatory cytokines by T cells and thus increases the susceptibility to severe airway inflammation.

The Effects of Sunbanghwalmyung-eum Extract on Acute Sinusitis-induced Mouse (선방활명음(仙方活命飮)이 알레르기 비염을 동반한 급성 비부비동염 마우스에 미치는 영향)

  • Background and Objectives: Sinusitis, referring to an inflammatory disease that occurs on the mucus membrane inside the sinus, is one of the most common diseases in the otorhinolaryngology area. In oriental clinic, Sunbanghwalmyungeum (SHE) has been used as a primary prescription to treat inflammatory diseases and intumescence and to treat sinusitis. The aim of this study was to investigate the anti-inflammatory and anti-allergic effects of SHE on acute sinusitis induced mice. Materials and Methods: BALB/c mice were divided into three groups: the normal group, the group inoculated with S. pneumoniae which caused them allergic rhinitis (control group), and the group treated with the SHE extract after it was treated the same as the control group (sample group). We investigated the inhibition of Th 2 cell differentiation by SHE and the suppression of NF-${\kappa}B$ activation. Results: NF-${\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE in acute sinusitis. IL-4 and STAT 6 also appeared to be suppressed. The number of eosinophils in the sample group noticeably decreased when compared to the control group. In the general morphologic change, the increase of damaged respiratory ciliated epithelium & eosinophil's infiltration were decreased in the sample group. Goblet cells were maintained in the sample group. MIP-2 and HSP-70 decreased in the sample group. Apocrine secretion decreased in the sample group. Conclusion: The results suggest that SHE is significantly effective in the treatment of inflammation caused by acute sinusitis through the suppression of NF-${\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

  • Experimental infection and histopathological study using a deleterious aquatic fungus (Saprolegnia diclina type I) were carried out in the eels (Anguilla japonica) to know what pathological changes the fungus would produce in the affected eels. The eels in group A and B which were treated with fish net were not susceptible to experimental infection. In group C which the cuticle was scraped by artificial treatment, the aquatic fungi were invading all the eel bodies including the desquamated lesions. Histopathologically, the affected eels were markedly reduced in number of goblet cells and showed moderate damage of the clavate cells. Nodular formation involving ecchymotic hemorrhage was seen in some affected areas. The eels survived to 25 days after experimental infection.

Effects of Inhalable Microparticles of Seonpyejeongcheon-Tang in an Asthma Mouse Model - Effects of Microparticles of SJT -

  • Objectives: Allergic asthma generally presents with symptoms of wheezing, coughing, breathlessness, and airway inflammation. Seonpyejeongcheon-tang (SJT) consists of 12 herbs. It originated from Jeong-cheon-tang (JT), also known as Ding-chuan-tang, composed of 7 herbs, in She-sheng-zhong-miao-fang. This study aimed to evaluate the effects of local delivery of SJT via inhalable microparticles in an asthma mouse model. Methods: Microparticles containing SJT were produced by spray-drying with leucine as an excipient. SJT microparticles were evaluated with respect to their aerodynamic properties, in vitro cytotoxicity, in vivo toxicity, and therapeutic effects on ovalbumin (OVA)-induced asthma in comparison with orally-administered SJT. Results: SJT microparticles provided desirable aerodynamic properties (fine particle fraction of $48.9%{\pm}6.4%$ and mass median aerodynamic diameter of $3.7{\pm}0.3{\mu}m$). SJT microparticles did not show any cytotoxicity against RAW 264.7 macrophages at concentrations of 0.01 - 3 mg/mL. Inhaled SJT microparticles decreased the levels of IL-4, IL-5, IL-13, IL-17A, eotaxin and OVA-IgE in bronchoalveolar lavage fluid (BALF) in mice with OVA-induced asthma. These effects were verified by histological evaluation of the levels of infiltration of inflammatory cells and collagen, destructions of alveoli and bronchioles, and hyperplasia of goblet cells in lung tissues. The effects of SJT microparticles in the asthma model were equivalent to those of orally-administered SJT extract. Conclusion: This study suggests that SJT is a promising agent for inhalation therapy for patients with asthma.
