Purpose: Cytolethal distending toxin (CDT) is a family of heat-labile cytotoxins produced by several gram-negative mucosa-associated pathogens, including Aggregatibacter actinomycetemcomitans. CDT is well known to be capable of inducing growth arrest, morphological alterations, and eventually death in various cells. CDT belongs to a tripartite $AB_2$ toxin (CdtB: the enzymatic A subunit; CdtA and CdtC: the heterodimeric B subunit). Previous studies proposed that CdtA and CdtC together bind to a cell surface receptor and glycolipids act as a receptor for A. actinomycetemcomitans CDT (AaCDT). In this study, recombinant CdtA and CdtC proteins of AaCDT were co-expressed in a bacterial expression system and tested for their affinity for $GM_1$ ganglioside. Methods: The genes for CdtA and CdtC from A. actinomycetemcomitans Y4 were utilized to construct the expression vectors, pRSET-cdtA and pET28a-cdtC. Both CdtA and CdtC proteins were expressed in Escherichia coli BL21(DE3) and then purified using hexahistidine (His6) tag. The identity of purified protein was confirmed by anti-His6 antibody and monoclonal anti-CdtA antibody. Furthermore, the affinity of recombinant protein to $GM_1$ ganglioside was checked through ELISA. Results: Recombinant CdtA and CdtC proteins were expressed as soluble proteins and reacted to anti-His6 and monoclonal anti-CdtA antibodies. ELISA revealed that purified soluble CdtA-CdtC protein bound to $GM_1$ ganglioside, while CdtA alone did not. Conclusions: Co-expression of CdtA and CdtC proteins enhanced the solubility of the proteins in E. coli, leading to convenient preparation of active CdtA-CdtC, a critical material for the study of AaCDT pathogenesis.
비방사구와 방사구 달걀 난황을 가열 압착하여 얻은 난황유를 정제한 후 화학적 성질과 지질조성에 관하여 실험한 결과는 다음과 같다. 난황의 일반성분은 비방사구, 방사구가 각각 수분이 49.50%, 47.06%,조단백질은 16.53%, 16.98%,조 지방은 31.05%, 33.34%로 주성분을 이루었고, 조지방, 조단백의 함량은 방사구가 비방사구 보다 더 높았다 난황유의 화학적 성질은 비방사구, 방사구가 각각 산가 8.95, 9.85, 요오드가 57.64, 58.15, 비누가 240.14, 223.92로. 나타났다. 난황유 총지질의 조성은 비방사구, 방사구 각각 중성지질 76.60%, 71.23%,당지질 3.95%, 5.03%, 인지질 19.45%, 23.74%였다. 중성지질 중에는 비방사구, 방사구 각각 triglycride가 59.3%, 56.3%로 주성분을 이루었고 그 외에 monoglyceride와 diglyceridr의 함량이 높았다. 당지질은 비방사구, 방사구 각각 digalactosyl diglyceride가 98.3%, 97.8%로 대부분을 차지 하였다. 인지질은 방사구, 비방사구 각각 phosphatidiyl choline + phosphatidyl serine이 58.6%, 59.8%로 주성분을 이루었고, 그 외에 lecithin + sphingomyelin과 미확인 성분이 존재하였다.
Serratia marcescens를 $30^{\circ}C$에서 진탕배양했더니, 적색색소인 prodigiosin이 초로기(senescent phase of growth)에서 생성되었다. 그리고 이조건에서 배양한 세포를 polystyrene dish를 사용하여 세포의 hydrophobicity를 측정한 결과 상당한 소수성 성질이 발현되어 대부분의 세포가 비극성 성질의 polystyrene dish에 흡착되었다. 그러나 이 박테리아를 $37^{\circ}C$에서 배양했더니, 적색 색소인 prodigiosin도 생성되지 않았을 뿐 아니라 소수성 성질도 발현되지 않음으로서 세포가 polysyrene dish에 흡착되지 않고 pre-washing 단계에서 모두 씻겨져 났다. 또한 $30^{\circ}C$와 $37^{\circ}C$에서 배양한 serratia marecescens의 지질성분을 분석한 결과, $30^{\circ}C$에서 배양한 세포의 지질은 phospholipid, glycolipid 및 확인되지 않은 지질 등이 생성되었으나 $37^{\circ}C$에서 배양한 세포의 경우는 주로 양쪽성 성질의 aminolipid인 serratamolide가 생성되어, 배양한 온도조건에 따라 뚜렷한 차이를 보였다.
가열처리 및 건조방법이 새우의 지방질 함량과 중성지방질에 미치는 영향을 밝히고자 하였다. 생새우의 총지방질 함량은 건물기준으로 6.0%였으며, 가열 및 건조방법에 따라 총지방질의 함량은 유의적인 변화가 없었다. 생새우의 중성, 당 및 인지방질의 함량비는 각각 36.8:21.5:41.7% 이었고 동결건조한 것은 열풍건조한 것보다 중성시방질의 함량이 유의적으로 많은 반면에 당 및 인지방질의 함량은 적었다. 중성지방질의 주요성불은 triglyceride, free sterol, free fatty acid 및 esterified sterol이었으며, 동결건조한 것은 열풍건조한 것보다 triglyceride 함량은 유의적으로 많았고 free fatty acid 함량은 적었는데, 이는 열풍건조중 triglyceride가 분해된 것으로 생각되었다. 생새우의 총지방질의 구성 주요지방산은 palmitoleic acid, eicosapentaenoic acid 및 docosahexaenoic acid이었고, 중성지방질의 구성 주요 지방산은 palmitic acid, palmitoleic acid, oleic acid, eicosapentaenoic acid 및 docasahexaenoic acid이었다.
시료로부터 chloroform-methanol(2:1/v:v)혼합용매로 추출하여 얻은 조, 기장, 수수의 총지질 함량은 각각 3.9%, 2.7% 및 2.3%였다. 추출한 총지질을 silicic acid column을 이용하여 중성지질, 당지질, 인지질로 분획하고 그 함량을 구한 결과 총지질에서 중성지질, 당지질, 인지질의 함량은 조에서 72.4%, 18.8% 및 8.8%였고, 기장은 69.2%, 21.1% 및 9.6%였으며, 수수는 77.4%, 16.1% 및 6.6%였다. 모든 시료에서 중성지질이 가장 많은 양을 함유하고 있었으며, 각 지방질의 지방산을 GC로 분석한 결과, 모든 시료에서 가장 많은 지방산은 linoleic acid 였고, oleic acid와 palmitic acid가 그 다음으로 많은 양을 나타내었다. 조의 인지질에서 behenic acid가 12.77%로 상당한 양이 검출된 것과 중성지질에서 극히 소량이지만 myristic acid가 검출된 것이 특징적이다. Linolenic acid는 모든 시료에서 검출되지 않았다.
우리나라에서 재배한 4가지 품종의 보리를 디 에틸에테르로 유리지질을, 그 찌꺼기로부터 클로로포름-메탄올-물의 혼합용매로 결합지질을 추출한 각 지방질의 조성을 분석하여 다음과 같은 결과를 얻었다. 1. 시료 보리중 유리 및 결합지질의 평균함량은 각각 1.57% 및 0.48%였다. 2. 유리 및 결합지질중의 중성지질은 81.1% 및 16.4%, 당지질은 11.4% 및 6.8%, 인지질은 6.7% 및 75.5%였다. 3. 유리 및 결합지질중의 중성지질중 TG가 각각 92.3% 및 68.5%로 주성분이었고, 그 외에 FFA, MG, DG, FS 및 SE가 부성분으로 함유되어 있었으며 이들 함량은 유리 및 결합지질간에 다소 차이가 있었다. 4. 총 유리 및 총 결합지질의 주요지방산은 리놀레산, 올레산, 팔미트산, 리놀렌산이었다. 또 중성지질, 당지질 및 인지질획분의 주요 지방산 함량은 유리 및 결합지질간에 차이가 있었다.
Background: Niemann-Pick disease type C (NPC) is caused by the mutation of NPC genes, which leads to the abnormal accumulation of unesterified cholesterol and glycolipids in lysosomes. This autosomal recessive disease is characterized by liver dysfunction, hepatosplenomegaly, and progressive neurodegeneration. Recently, the application of induced neural stem cells (iNSCs), converted from fibroblasts using specific transcription factors, to repair degenerated lesions has been considered a novel therapy. Objectives: The therapeutic effects on NPC by human iNSCs generated by our research group have not yet been studied in vivo; in this study, we investigate those effects. Methods: We used an NPC mouse model to efficiently evaluate the therapeutic effect of iNSCs, because neurodegeneration progress is rapid in NPC. In addition, application of human iNSCs from NPC patient-derived fibroblasts in an NPC model in vivo can give insight into the clinical usefulness of iNSC treatment. The iNSCs, generated from NPC patientderived fibroblasts using the SOX2 and HMGA2 reprogramming factors, were transplanted by intracerebral injection into NPC mice. Results: Transplantation of iNSCs showed positive results in survival and body weight change in vivo. Additionally, iNSC-treated mice showed improved learning and memory in behavior test results. Furthermore, through magnetic resonance imaging and histopathological assessments, we observed delayed neurodegeneration in NPC mouse brains. Conclusions: iNSCs converted from patient-derived fibroblasts can become another choice of treatment for neurodegenerative diseases such as NPC.
Mannosylerythritol lipids (MELs) are glycolipids and have several pharmacological efficacies. MELs also show skin-moisturizing efficacy through a yet-unknown underlying mechanism. Aquaporin-3 (AQP3) is a membrane protein that contributes to the water homeostasis of the epidermis, and decreased AQP3 expression following ultraviolet (UV)-irradiation of the skin is associated with reduced skin moisture. No previous study has examined whether the skin-moisturizing effect of MELs might act through the modulation of AQP3 expression. Here, we report for the first time that MELs ameliorate the UVA-induced downregulation of AQP3 in cultured human epidermal keratinocytes (HaCaT keratinocytes). Our results revealed that UVA irradiation decreases AQP3 expression at the protein and messenger RNA (mRNA) levels, but that MEL treatment significantly ameliorated these effects. Our mitogen-activated protein kinase inhibitor analysis revealed that phosphorylation of c-Jun N-terminal kinase (JNK), but not extracellular signal-regulated kinase or p38, mediates UVA-induced AQP3 downregulation, and that MEL treatment significantly suppressed the UVA-induced phosphorylation of JNK. To explore a possible mechanism, we tested whether MELs could regulate the expression of peroxidase proliferator-activated receptor gamma ($PPAR-{\gamma}$), which acts as a potent transcription factor for AQP3 expression. Interestingly, UVA irradiation significantly inhibited the mRNA expression of $PPAR-{\gamma}$ in HaCaT keratinocytes, whereas a JNK inhibitor and MELs significantly rescued this effect. Taken together, these findings suggest that MELs ameliorate UVA-induced AQP3 downregulation in HaCaT keratinocytes by suppressing JNK activation to block the decrease of $PPAR-{\gamma}$. Collectively, our findings suggest that MELs can be used as a potential ingredient that modulates AQP3 expression to improve skin moisturization following UVA irradiation-induced damage.
Mycobacterial cell walls comprise thick and diverse lipids and glycolipids that act as a permeability barrier to antibiotics or other chemical agents. The use of OH radicals from a non-thermal plasma jet (NTPJ) for the inactivation of mycobacteria in aqueous solution was adopted as a novel approach. Addition of water vapor in a nitrogen plasma jet generated OH radicals, which converted to hydrogen peroxide ($H_2O_2$) that inactivated non-pathogenic Mycobacterium smegmatis and pathogenic Mycobacterium tuberculosis H37Rv. A stable plasma plume was obtained from a nitrogen plasma jet with 1.91 W of power, killing Escherichia coli and mycobacteria effectively, whereas addition of catalase decreased the effects of the former. Mycobacteria were more resistant than E. coli to NTPJ treatment. Plasma treatment enhanced intracellular ROS production and upregulation of genes related to ROS stress responses (thiolrelated oxidoreductases, such as SseA and DoxX, and ferric uptake regulator furA). Morphological changes of M. smegmatis and M. tuberculosis H37Rv were observed after 5 min treatment with $N_2+H_2O$ plasma, but not of pre-incubated sample with catalase. This finding indicates that the bactericidal efficacy of NTPJ is related to the toxicity of OH and $H_2O_2$ radicals in cells. Therefore, our study suggests that NTPJ treatment may effectively control pulmonary infections caused by M. tuberculosis and nontuberculous mycobacteria (NTM) such as M. avium or M. abscessus in water.
