Thermomechanical and surface chemical properties of composite films of poly(D, L-lactic-co-glycolic acid) (PLGA) were significantly improved by the addition of graphene oxide (GO) nanosheets as nanoscale fillers to the PLGA polymer matrix. Enhanced thermomechanical properties of the PLGA/GO (2 wt.%) composite film, including an increase in the crystallization temperature and reduction in the weight loss, were observed. The tensile modulus of a composite film with increased GO fraction was presumably enhanced due to strong chemical bonding between the GO nanosheets and PLGA matrix. Enhanced hydrophilicity of the composite film due to embedded GO nanosheets also improved the biocompatibility of the composite film. Improved thermomechanical properties and biocompatibility of the PLGA composite films embedded with GO nanosheets may be applicable to biomedical applications such as scaffolds.
The thermomechanical and surface chemical properties of nanocomposite of poly( D, L-actic-co-glycolic acid) (PLGA) were improved significant due to concentration of graphene oxide (GO) nanosheets as nanoscale fillers to PLGA film. Thermomechanical properties of the PLGA/GO (2wt.-%.) nanocomposite were decreased crystallization and melting temperature, weight loss. The storage and loss moduli of the nanocomposite were enhanced by chemical bonding between the oxygenated functional groups of the GO nanosheets and the polymer chains in the PLGA matrix. Enhanced hydrophilicity of nanocomposite caused by embedded GO nanosheets also improved for good biocompatibility. Our findings indicate that thermomechanical properties and biocompatibility of nanocomposite embedded with GO nanosheets are attractive candidates for use in biomedical applications such as scaffolds.
In order to clarify the pathogenesis of perirenal edema, pigs and rabbits were administered with oxalate and nitrate, with oxalate nitrate and glycolic acid, with oxalate, nitrate and ascorbic acid and with oxalate, nitrate and calcium, respectively. The results obtained are summarized as follows; The pigs and rabbits administered with oxalate and nitrate with oxalate, nitrate and glycolic acid and with oxalate, nitrate and ascorbic acid, respectively, were not showed perirenal edema despite of observing the abundant oxalate crystals in the proximal convoluted tubles. But pigs and rabbits administered with oxalate, nitrate and calcium were histopathologically showed perirenal edema similar to those of pigs fed Amaranthus retroflexus. Therefore, author considered that oxalate, nitrate and calcium are main factors to cause perirenal edema. It was regarded that perirenal edema in pigs was caused by the reciprocal reactions of those materials including oxalate, nitrate and calcium which may produce vascular damage, decreased osmotic pressure by hypoproteinemia and increased vascular permeability in kidney, rather than the mechanical obstructions by the oxalate crystals in the proximal convoluted tubule.
하이드록시 산(hydroxy acid, HA)은 피부 각질층에 대한 박리효과로 노화방지와 피부 보습을 높이는데 효과를 보여, 피부 외용제와 화장품으로서 많은 활용을 하고 있다. 그 중에서 가장 효과적으로 빠른 시간 내에 나타나는 각질 박리는 화장품 제형 pH에 의해 효과를 보이는 것으로 많은 보고가 있다. 그러나 pH에 의한 자극, 부작용에 의한 염려로 인해 사용에 많은 어려움이 있다. 본 연구의 목적은 피부 각질층에 (1) 하이드록시 산의 농도와 (2) 종류, (3) pH를 변화시킨 화장품을 인체 피부에 도포하여 각질 박리 효과에 대한 영향을 측정하는 데에 있다. 건강한 성인 22명을 대상으로 하박 내측에 DHA (dihydroxyacetone), DC (dansyl chloride)로 피부 표면 각질을 염색하여, 시험제품을 도포한 뒤 각질박리 효과를 측정하였다. (1) GA (glycolic acid)의 농도에 따라 각질 박리 효과는 농도 의존적으로 증가하는 것으로 나타났다. (2 )하이드록시 산의 종류에 따라 pH를 산성과 중성으로 제조한 제품을 대상으로 각질 박리 효과를 측정한 결과, 중성 pH의 GA는 각질 박리 효과가 나타나지 않았다. 이에 반해 SA (salicylic acid)는 산성 pH와 중성 pH에서 모두 통계적으로 유의한 각질 박리 효과가 나타났다. (3) 중성 pH의 SA는 DHA와 DC로 염색한 피부 표면에서 모두 우수한 각질 박리 효과를 나타내었다. 이러한 결과는 pH에 민감한 사람들에게 각질 박리 효과를 기대하는 화장품을 사용할 수 있는 기회를 제공하며, 피부 장벽의 손상 없이 안전한 화장품을 제조할 수 있을 것이라 사료된다.
본 연구는 dermal equivalent를 이용하여 AHA류(lactic acid, glycolic acid, citric acid 및 malic acid) 및 보습 원료(sodium hyaluronate, glycerine, natural carbohydrate complex)에 대한 wound healing 효과 및 그 기작을 알아보기 위한 것이다. AHA류 및 보습 원료의 독성은 dermal equivalent를 이용하여 m assay를 실시한 후 적정 농도를 petri dish위에 조성한 dermal equivalent에 처리하였다. 그리고 wound healing 효과를 알아보기 위하여 collagen 수축률을 측정하였다. 또한 collagen 합성을 촉진하는 것으로 잘알려진 vitamin C도 함께 처리하였다. AHA류의 경우 평균 pH 2-3으로 매우 낮기 때문에 4N NaOH를 이용하여 pH를 6-7사이로 조절한 sample를 함께 실험하였다. 보습 원료의 경우 전반적으로 10% 이상의 높은 NR$_{50}$보여 주었다. 특히 sodium hyaluronate(1% stock solution)의 경우에는 16% NR$_{50}$를 보여 주었다. pH를 6-7로 조절한 AHA류의 경우에는 전반적으로 0.6% 내외의 NR$_{50}$를 보이는 가운데 lactic acid는 상대적으로 높은2% NR$_{50}$를 보여 주었다. Collagen 수축률 측정 실험 결과에서는 2% sodium hyaluronate(1% stock solution)가 대조군에 비하여 처리 후 2일째 25%내외의 향상된 수축률을 보여 주었다. pH를6-7로 조절한 AHA류 중 0.1%의 malic acid의 경우에서는 대조군에 비하여 처리 후 1일째 및 2일째 각각 28% 및 35%의 수축률을 보여 주었으며 pH를 6-7로 조절한 0.1% vitamin C에서도 유사한 결과를 보여 주었다. 반면에 pH를 6-7로 조절한 0.1% citric acid의 경우에는 10-20%의 낮은 수축률을 보여 주었다. MTT assay를 이용한 UV 조사 후 pH를 6-7로 조절한 AHA류의 repairing UV damage 효과에 대한 실험에서 0.1% 및 0.01%의 malic acid와 0.01% citric acid은 irradiation control에 비하여 약 10% 이상 세포수를 증가시켰다. 그러나 예외적으로 citric acid의 경우 0.1% 농도에서 오히려 20%내외로 세포수가 감소되는 경향을 보여주었다. 그리고 lactic acid 및 glycolic acid는 두드러지는 효과를 나타내지 않았다. Collagen 합성을 측정 실험에서는 pH를 6-7로 조절한 AHA류에서는 대조군에 비하여 상대적으로 12-19% 더 합성을 촉진하였다. 반면에 pH가 2-3인 AHA류의 경우에는 대조 군과 유사하거나 조금 낮은 합성율을 보여 주었다.
Purpose: An ideal bony construct can be divided into two broad categories: (1) the design and fabrication of biodegradable, biomimetic scaffolds that provide correct signals to induce osteogenesis: (2) the identification of an ideal source of osteoprogenitor cells to seed onto the scaffold. We selected poly-glycolic acid as a synthetic scaffold among various scaffolds because of these properties. Meanwhile, culture medium is supplemented with fetal bovine serum(FBS): such serum contains essential elements such as proteins, hormones, growth factors and trace minerals. The composition of FBS can be ideal for various cell growth in vitro. We supposed that we could enhance bone growth at a fractured site if FBS was mixed with synthetic scaffold-PGA. Methods: We cultured human osteoblasts in five different prepared culture dishes made with FBS and PGA mixture. The mixtures contained different ratio of FBS, that is, 0, 1.5, 3, 7, and 10%. We cultured human osteoblasts for seven days and examined the growth and attachment of the cells at the 1st, 3rd, 5th, 7th days, respectively. Results: In the mixture of 0% FBS and PGA, the growth of the cells lasted for one day. In 1.5 and 3% FBS and PGA, the growth of the cells was examined at the 3rd day, then minimally declined at the 5th and 7th days. In 7% FBS and PGA, the growth of the cells lasted for 5 days, then declined at the 7th day. In 10% FBS and PGA, the growth of the cells lasted for 5 days, then declined at the 7th day. Staining status of the osteoblasts with alkaline phosphatase showed pale pink color in 0% FBS and PGA groups, but bright pink color in 1.5, 3, 7, 10% FBS and PGA groups, especially in 3%, 7%. Conclusion: In consequence, the growth of human osteoblast was higher in the mixture of FBS and PGA groups than in pure PGA ones. It is assumed that the mixture of FBS and PGA affects the proliferation of human osteoblasts.
담홍색 열매체유를 포도주로 오인하고 음용하여 사망한 변사사건에서 열매체유 중 39%의 에틸렌글리콜을 확인하였다. 열매체유는 물을 열전달 매체로 사용하였을 경우 고온에서 발생되는 고압 수증기에 의한 문제를 피하기 위하여 사용되는 열전달 매체로 주로 광유계 또는 에틸렌글리콜을 사용하는 것으로 알려져 있다. 부검을 통한 혈액 및 기타 조직 등의 변사자 생체시료에서 중수소로 치환된 에틸렌글리콜을 내부표준물질로 하여 아세토니트릴로 추출, 정제 및 질소 기류하에서 농축한 다음 1% TMCS가 함유된 BSTFA로 유도체화하여 에틸렌글리콜과 그 대사체인 글리콜산을 GC/MS에 의하여 동시 분석하였다. 약 0.2 g의 생체시료에 대하여 0.5 M의 HCl 용액 $50\;{\mu}L$를 이용한 산성조건에서 추출하였으며 무수 황산나트륨을 단계적으로 처리하여 수분제거를 하였고 혈액에서 $2,755\;{\mu}g/mL$의 에틸렌글리콜과 $174\;{\mu}g/mL$의 글리콜산 및 기타 시료에서 $860\;{\mu}g/g\sim1,290\;{\mu}g/g$의 에틸렌글리콜과 $93\;{\mu}g/g\sim134\;{\mu}g/g$의 글리콜산을 검출하였다. 특히 신장 조직에서 이와 관련된 대사체인 옥살산염으로 추정되는 결정이 관찰되었다.
탈미네랄화된 골분(demineralized bone particle, DBP)은 연골 형성의 유도인자로 사용되기 때문에 조직공학에서 널리 사용되는 생체재료이다. 본 연구에서는 in vivo 환경에서 디스크 재생 효과를 연구하기 위해 DBP를 poly(lactic-co-glycolic acid) (PLGA)에 첨가하여 다공성 지지체를 제조하였다. 디스크의 섬유륜 조직을 반으로 절개한 후, 수핵 조직을 제거하여 디스크 결손을 유발시켰다. 빈 공간에 PLGA, DBP/PLGA 지지체를 이식하여 in vivo 환경에서 조직공학적 디스크 재생을 관찰하였다. 1, 2 및 3개월 후 디스크를 적출하여 글리코스아미노글라이칸(glycosaminoglycan, sGAG) 및 콜라겐 합성 정도를 측정하였으며 조직학적 평가로 H&E, Safranin-O, Alcian blue 염색과 면역조직학적 평가로 제 I형 콜라겐, 제 II형 콜라겐 염색을 수행하였다. 그 결과 DBP/PLGA 지지체에서 sGAG 및 콜라겐 함량이 높은 것을 확인하였으며 추간판 디스크 재생의 가능성을 확인하였다.
