The Distribution and Position of Fatty Acids in Glycerides Hydrolyzed from Fish Oil by Lipase

  • In order to determine the position and the content of fatty acids sttached to glycerides and the migration degree of fatty acids in the migration reaction, fish oil was hydroyzed with lipolase-100T which was derived from Aspergillus oryzae. The content of fatty acids in the glyceride mixture was analyzed and compared with that of fish oil. The amounts of fatty acid in a 2-position and the migration degree of the fatty acid in 2,3-DG (diglyceride) and 2-MG (monolyceride) were carefully calculated. The results showed that approximately 95% (w/w) of DHA (docosahexanoic acid) and 65% of EPA(eicosapentaenoic acid) were attached to the 2-position of glycerides in fish oil. Approximately 87% (w/w) of DHA and 75% of EPA remained in 2,3-DG, and 88% of DHA and 65% of EPA in 2-MG were not involved in the migration reaction.

  • Bacillus polyfermenticus SCD which is commonly called as Bisroot (equation omitted) has been appropriately used for the treatment of long-term intestinal disorder's. This strain strongly inhibited against methicillin resistance Staphylococcus aureus (MRSA) and various pathogenic microorganisms. Effects of B. polyfermentius SCD administration on death rates and egg-laying rates in two groups of hens were investigated. This strain decreased the death rates of two groups by 16.26% and 11.72%, respectively. Also this strain increased the egg-laying rates of those 2.74% and 2.66%, respectively Clinical tests of B. polyfermenticus SCD administration to healthy adults showed not adverse effects but decreased glyceride concentration from 154.52 mg/dl to 135.41 mg/dl after two week administrations.

  • Mono alkyl glycerides have bean obtained in good yield by enzyme catalyst from soybean oil. The reaction was carried out in an oil rich microemulsion formula. Best results were obtained with sodium bis(2-ethyl hexyl) sulfo succinate(AOT), isooctane as hydrocarbon component and buffer of pH 7. The enzyme used was a 1,3-specific lipase which leaves the 2-position intact. However, the 2-monoglyceride formed slowly undergoes long chain acyl migration to 1-mono-glyceride. Optimal reaction time at $35^{\circ}C$ reaction temperature was found to be three hour.

  • Pasting characteristics and amylose-surfactant complex forming ability of sweet potato starch were investigated after defatting and the addition of surfactants, such as SSL (sodium steamyl-2-lactylate) Dimodan (mono/di glyceride) and SE (sucrose ester) with different concentrations. All starch granules were smooth and round, there were no damages to starch granules after defatting and surfactant addition. amylose content of surfactant added stach decreased and me order of decrease was SSL, SE and Dimodan. The cornplex forming ability of SE added starch increased according to increasing HLB value. As surfactants concentration increased, amylose complex formig ability increased. In case of gelatinization patterns by amylograph, the initial pasting temperature of surfactant added starches was higher than mat of untreated or defatted starches, but viscosity at each temperature were all decreased. Soluble carbohydrate and leached amylose of starches increased at increasing temperature, those of surfactant added starches decreased at each temperature in the order of SSL, SE and Dimodan.

Optimization of Pre-treatment of Tropical Crop Oil by Sulfuric Acid and Bio-diesel Production (황산을 이용한 열대작물 오일의 전처리 반응 최적화 및 바이오디젤 생산)

  • In this study, the feasibility of using vegetable oil extracted from tropical crop seed as a biodiesel feedstock was investigated by producing biodiesel and analysing the quality parameters as a transport fuel. In order to produce biodiesel efficiently, two step reaction process(pre-treatment and transesterificaion) was required because the tropical crop oil have a high content of free fatty acids. To determine the suitable acid catalyst for the pre-esterification, three kinds of acid catalysts were tested and sulfuric acid was identified as the best catalyst. After constructing the experimental matrix based on RSM and analysing the statistical data, the optimal pre-treatment conditions were determined to be 26.7% of methanol and 0.982% of sulfuric acid. Trans-esterification experiments of the pre-esterified oil based on RSM were carried out, then discovered 1.24% of KOH catalyst and 22.76% of methanol as the optimal trans-esterification conditions. However, the quantity of KOH was higher than the previously established KOH concentration of our team. So, we carried out supplemental experiment to determine the quantity of catalyst and methanol. As a result, the optimal transesterification conditions were determined to be 0.8% of KOH and 16.13% of methanol. After trans-esterification of tropical crop oil, the produced biodiesel could meet the major quality standard specifications; 100.8% of FAME, 0.45 mgKOH/g of acid value, 0.00% of water, 0.04% of total glycerol, $4.041mm^2/s$ of kinematic viscosity(at $40^{\circ}C$).

The Effect of Acyl Chain Structure on the Hydrolysis of Fatty Acids from Fish Oil by Lipase-OF 360,000 (아실체인 구조가 효소 Lipase-OF 360,000에 의한 지방산의 가수분해 특성에 미치는 영향)

  • The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.

  • Seven terpenoids, trans-phytol (1), ${\alpha}-spinasterol$ (2),${\beta}-sitosterol$ (3), oleanolic acid (4), traxasterol (5), ${\alpha}-spinasterol\;3-O-{\beta}-D-glucopyranoside$ (8), ${\beta}-spinasterol\;3-O-{\beta}-D-glucopyranoside$ (9), and three glycerides, 3-O-(9Z, 12Z, 15Z-octadecatrienoyl) glycerol (6), 3-O-(9Z, 12Z-octadecadienoyl) glycerol (7), 1, 2-O-(9Z, 12Z, 15Z-dioctadecatrienoyl)-3-O-${\beta}$- D-galactopyranosyl glycerol (10) were isolated from n-hexane fraction of the aerial parts of Saussurea nutans which was used as Korean traditional medicine to σeat rheumatic arthritis and dysmenorrhea. Their structures were established by chemical and spectroscopic methods.

  • One of the popular formulations is microemulsion in cosmetics. A kind of microemulsions is liquid crystal emulsion. Liquid crystal emulsion is used by wrapping functional ingredients. As the meaning of the name, Liquid crystals (LCs) are a state of matter that has properties between those of a conventional liquid and those of a solid crystal. There are various types of Liquid crystals (LCs) consisting of micelles or two layer structures of surfactants. Recently, microemulsion has been studied to improve its stability in thermodynamics of colloid science field. In this study, we prepared the microemulsion with PEG-8 Capryliccapric Glycerides and Polyglyceryl-3 Diisostearate. We studied the microemulsion with different oil types and different and cosurfactant content ratio. Also, the aim of this study is to develop a facial oil or a cleansing oil containing liquid crystal emulsion with functional ingredients.

Glyceryl Esterification of Fibroin Peptide by Papain (파파인을 이용한 피브로인 펩타이드의 글리세릴 에스터화 반응)

  • Papain hydrolysate of fibroin was found to be mainly composed of several even-numbered peptides that can be produced at a large scale and can be used as a precursor for biological fine-chemicals such as peptide detergents. Thus, the hydrolysate was further modified to synthesize a peptide mixture of glyceryl esters using the identical enzyme for the production of such chemicals. Formation of glyceryl ester of each peptide was confirmed by identifying peaks of the nominal mass shift of +74 Da in mass spectrometry. Analysis of the mass spectra indicated that glyceryl esters of di- and tetra-peptides were the major constituents of the mixture and that alanylglycine was most preferentially esterified. It also suggests that papain prefers dipeptide to tetrapeptide and alanine to serine or tyrosine at $P_2$ position as substrate for glyceryl esterification. The glyceryl esters were recovered using ion exchange resin and the yield of glyceryl esterification recorded was 17.8% by weight.

Comparison of LDL-Cholesterol direct measurement with the estimate using various formula

  • Low-Density Lipoprotein cholesterol (LDLC) is the most important marker for the treatment of hyperlipidemia in NCEP-ATP III(National Cholesterol Education Program-Adult Treatment Panel III) guideline. Therefore, LDL cholesterol is pathologically meaningful, accurate measurement should be a top priority. Currently, LDLC is directly measured in most cases, but, the estimate is still used in mass health examination or screening test. This study is about the comparison of LDL-Cholesterol direct measurement with the estimate using various formula (Friedewald: [LDL-F=TC-HDL-TG/5], Nakajima: [LDL-N=TC-HDL-TG/4], Hattori: [LDL-H =0.94TC-0.94HDL-0.19TG], Puavilai: [LDL-P=TC-HDL-TG/6], Carvalho: [LDL-C=3(TC-HDL)/4]) for calculating more accurate value. We analyzed total cholesterol (TC), try-glyceride (TG), high-density lipoprotein cholesterol (HDLC), and LDLC levels of 210 subjects between June and November in 2011. Until now, the Friedewald formula is the most commonly used estimate for the LDLC. When Friedewald formula was applied, the correlation coefficient (r) was 0.940, showing high correlation. But, the result of the direct method was significantly different, compared with those of the Friedewald formula in triglyceride levels ${\geq}400mg/dL$(p<0.05). There was the highest correlation when we used LDL-P formula(r=0.947) in triglyceride levels <400 mg/dl. Also there was the lowest mean difference regardless of triglyceride level. Therefore, the study showed that TG/6 is more precise means of calculation than TG/5. On the other hand, the calculation of LDL-Cholesterol was underestimated, compared with direct measurement. It is necessary to have more data and modified Friedewald formula should be used for the accurate calculation.
