High Glucose Inhibitory Effects on Bone Formation in UMR106 Cells

  • Recently, diabetes has been found to be associated with osteoporosis. Specially in IDDM. In both type I and type II diabetes, glucose levels are elevated. Thus, a linkage between high glucose and osteoporosis can not be ruled out. In this study, an attempt has been made to observe the effect of high glucose on bone formation; osteoblast like UMR 106 cells were treated with high glucose (22 mM, 33 mM) for 1, 3 or 7 days. The high concentration of glucose inhibited markers. of bone formation activity such as alkaline phosphatase and collagen synthesis. In addition, reduction in the level of total cellular protein in response to high glucose was also observed. This study showed high glucose concentration could alter the bone metabolism leading to a defective bone formation and thus paving the linkage of such situation to diabetic complications.

  • 적색육어류의 히스타만생성에 미치는 첨가물의 영향을 검토하기 위하여 고등어를 시료로 하여 이에 glucose, sucrose, glycine 및 sorbic acid 등을 첨가하여 저장하여 두고 저장기간에 따른 히스타민함량 및. 히스티딘탈탄산효소활성의 변화를 측정하였으며, 아울러 시료육의 성상이 히스타민생성에 미치는 영향을 조사하기 위하여 시료육의 역쇄정도를 달리 하였을 때의 히스타민함량의 변화를 비교, 검토하였다. 1. 균질화한 시료육에 glucose, sucrose 및 glycine을 각각 $5\%$$10\%$ 첨가하여 $25^{\circ}C$에 저장하였을 때 glucose 및 sucrose를 첨가한 것은 히스타민 생성을 다소 촉진하는 경향을 나타낸 반면, glycine을 첨가한 것은 히스타민생성 및 히스티딘탈탄산효소활성을 크게 억제하는 효과를 나타내어, $10\%$ 첨가한 것은 저장 5일 후에도 히스타민중독한계농도에 달하지 않았다. 2. 마쇄정도가 다른 시료육에 glucose 및 sucrose를 첨가하여 히스타민 생성에 미치는 영향을 조사한 결과 마쇄정도가 큰 것이 히스타민 생성을 촉진하는 경향을 나타내었다. 3. 시료육에 sorbic acid를 $0.1\%$$0.2\%$ 첨가하였을 때 히스타민 생성 및 히스티딘탈탄산효소활성은 크게 억제되었으며, $0.2\%$ 첨가한 것은 저장 6일째에 히스타민 함량이 최고치(275mg/100g)를 나타내었다.

방울토마토(Lycopersicon escylentnm mill ssp.)를 첨가(添加)한 Yogurt의 제조(製造)에 관한 연구 (A Study on the Preparation of Yogurt Added with Tomato Juice)

  • 본 연구는 토마토의 영양적인 효과와 유산균 배양시의 발효촉진 효과를 기대한 새로운 요구르트 개발의 일환으로, 환원(還元) 탈지유에 방울토마토(Lycopersicon escylentnm mill ssp.) juice를 1, 3, 및 5% 첨가하여 요구르트를 제조하였을 때와 방울토마토 juice에 glucose를 대치하여 요구르트를 제조하였을 때의 실험결과를 요약하면 다음과 같다. 1. 요구르트 발효에 있어 방울토마토 juice 첨가구(添加區)(pH 4.3, 적정산도 0.93)가 대조구(對照區)(pH 4.6, 적정산도 0.85)에 비하여 pH 저하 및 산생성의 증가가 빨랐으며, 방울토마토 juice 첨가 농도가 높을수록 pH저하 및 산 생성의 증가가 빨랐다. 2. 요구르트 발효에 있어 방울토마토 juice 첨가구(添加區)($4.57{\times}10^{12}cfu/ml$)가 대조구(對照區)($8.96{\times}10^{11}cfu/ml$)에 비하여 유산균수가 크게 증가 하였다. 3. 요구르트 발효에 있어 방울토마토 juice 첨가구(添加區)(15.7)가 대조구(對照區) (16.5)에 비하여 방울토마토 juice 첨가 농도가 높을수록 당도의 저하를 촉진시켰다. 4. 방울토마토 juice 시험구는 $5^{\circ}C$에서 14일간 저장시에 pH 4.05, 적정산도 1.092 유산균수가 $2.43{\times}10^{12}$로서 제품의 우수성을 확인할 수 있었다. 5. 요구르트 발효에 있어 방울토마토 juice 5% 첨가구(添加區)(pH 4.05, 적정산도 1.032)가 glucose만을 5% 첨가(添加)한 대조구(對照區)(pH 4.2, 적정산도 0.939)에 비하여 pH 저하 및 산생성의 증가가 빨랐다. 6. 요구르트 발효에 있어 방울토마토 juice 5% 첨가구(添加區)($3.16{\times}10^{11}cfu/ml$)가 glucose만을 5% 첨가(添加)한 대조구(對照區)($9.8{\times}10^{10}cfu/ml$)보다 유산균의 증식이 빨랐으며, 방울토마토 juice의 첨가농도가 높을수록 촉진되었다. 7. 요구르트 발효에 있어 방울토마토 juice 5% 첨가구(添加區)(15.0)가 glucose만을 5% 첨가(添加)한 대조구(對照區)(16.2)보다 빠른 당도의 저하로 발효를 촉진함을 알 수 있었다. 이상의 제 결과를 종합할 때 방울토마토의 첨가는 요구르트 발효에 있어 pH, 적정산도, 당도 및 유산균수 증가에 촉진효과를 나타냄을 알 수 있었다.

당과 산류가 담배 판상엽의 품질에 미치는 영향 (Effects of added sugars and organic acid on the quality of reconstituted tobacco leaves)

  • (A series of cigarettes made from) RTL containing different leveles of added sugars (2.0. 5.0 and 8.0%) and acids(0.4, 0.8 and 1.2%) have been examined. Compared to the control cigarette there was reduced in the delivery of tar and nicotine in smoke on 5.0% addition of glucose there was a signification reduced the delivery of nicotine the addition of glucose or fructose decreased the delivery of nicotine tar and co. However, tar in smoke was an increased in the delivery of tar and nicotine was observed, when sucrose and lactose were the added sugars. The smoking taste by the addition of glucose and fructose on R.T.L. was found to be milder than by the others sugars. Tar and nicotine deliveries showed a decreasing trend with the addition of organic acid and tartaric acid among the organic acid described above had greater effect than the others acids 1.2% addition of tartaric acid gave some 11-31% reduction in tar and nicotine delivery and a fall of smoke pH from 4.49 was found. Smoking taste by the addition of lactic acid and tartaric acid had milder than the others and heat treatment for 10 minutes at 13$0^{\circ}C$ gave also better on smoking: taste. The addition of mixture of sugars and organic acid (glucose 5% + lactic acid 0.6% + tartaric acid 0.6%) and then the successive heat treatment for 10 minutes at 13$0^{\circ}C$ improved greatly the Quality of R.T.L. and nicotine were reduced to 11-28% and pH was changed from 5.40 to 3.89.

Effect of Glucose and Formic Acid on the Quality of Napiergrass Silage After Treatment With Urea

  • Urea as a silage additive increases crude protein but reduces fermentation quality of silage by increasing pH and enhancing clostridial bacteria growth, especially in low sugar forages. Glucose and formic acid might be expected to compensate these defects caused by urea addition to grass silage. Thus, in this experiment urea formic acid or urea with glucose was applied to improve N content and the quality of napiergrass (Pennisetum purpureum Schumach.) silage. The first growth of napiergrass was harvested at 85 days of age and about 700 g of the grass was ensiled in laboratory silos (1.0 liter polyethylene containers) for 2, 7, 14, and 30 days at room temperature ($28^{\circ}C$). The treatments were no additives (control), urea, urea+glucose or urea+formic acid. Urea was added before ensiling at 0.5% of fresh weight of napiergrass and glucose and formic acid were added at 1% of fresh weight, respectively. After opening the silo, pH, dry matter content (DM), contents on DM basis of total N (TN), volatile basic nitrogen (VBN), lactic acid (LA), acetic acid (AA) and butyric acid (BA) were determined. The control at 30 days of fermentation showed 5.89 for pH with 13.8% for VBN/TN and 1.51% for AA. The addition of urea increased TN by about 1.5% units but decreased the fermentation quality by increasing pH from 5.89 to 6.86, increasing VBN/TN from 13.8% to 24.63%, increasing BA from 0.02% to 0.56%, and decreasing LA from 1.03% to 0.02%. Glucose addition with urea significantly decreased VBN/TN from 13.8% to 4.44% by reducing pH from 6.86 to 4.83 because of higher production of LA (2.62%). Adding urea and formic acid resulted in a more pronounced depression of VBN/TN and fermentation than the addition of urea and glucose. This study suggested that the combination of 1% glucose or 1% formic acid with 0.5% urea will improve nutritive value and fermentation quality of napiergrass silage.

Streptomyces의 Aerial Mycelium 형성에 대한 Glucose 억제 기작에 관한 연구 (The Glucose Repression of Aerial Mycelium Formation in Streptomyces)

  • We have demonstrated that both L-histidine as an amino acid factor and dextrin as a carbon source were required for the glucose repression. 1% glucose was sufficient to the glucose repression of aerial mycelium formation in Streptomyces lavendulae and Streptomyces aureofacience. the synthesized medium, KK, which is lack of all orgnic nutrients except dextrin was able to induce glucose repression, but the addition of 0.003% or more L-histidiner recovers the capacity of glucose repression. 0.02% or more histidine was reuqired for glucose repression of aerial mycelium formation in the absence of dextrin. Treatments of $5{\mu}M$ ormore ethidium bromide (EtBr0 gave rise to bald mutants at high frequency in Streptomyces aureofaciens, and it is probable that the gene(s) for the function of aerial mycelium formation is linked to plasmed DNA in this species.

Protective Effect of Padina arborescens Extract against High Glucose-induced Oxidative Damage in Human Umbilical Vein Endothelial Cells

  • Dysfunction of endothelial cells is considered a major cause of vascular complications in diabetes. In the present study, we investigated the protective effect of Padina arborescens extract against high glucose-induced oxidative damage in human umbilical vein endothelial cells (HUVECs). High-concentration of glucose (30 mM) treatment induced cytotoxicity whereas Padina arborescens extract protected the cells from high glucose-induced damage and significantly restored cell viability. In addition, lipid peroxidation, intracellular reactive oxygen species (ROS), and nitric oxide (NO) levels induced by high glucose treatment were effectively inhibited by treatment of Padina arborescens extract in a dose-dependent manner. High glucose treatment also induced the overexpressions of inducible nitric oxide synthase (iNOS), cyclooxygenase- 2 (COX-2) and NF-${\kappa}B$ proteins in HUVECs, but Padina arborescens extract treatment reduced the over-expressions of these proteins. These findings indicate the potential benefits of Padina arborescens extract as a valuable source in reducing the oxidative damage induced by high glucose.

The Modulatory Role of Spinally Located Histamine Receptors in the Regulation of the Blood Glucose Level in D-Glucose-Fed Mice

  • The possible roles of spinal histamine receptors in the regulation of the blood glucose level were studied in ICR mice. Mice were intrathecally (i.t.) treated with histamine 1 (H1) receptor agonist (2-pyridylethylamine) or antagonist (cetirizine), histamine 2 (H2) receptor agonist (dimaprit) or antagonist (ranitidine), histamine 3 (H3) receptor agonist (${\alpha}$-methylhistamine) or antagonist (carcinine) and histamine 4 (H4) receptor agonist (VUF 8430) or antagonist (JNJ 7777120), and the blood glucose level was measured at 30, 60 and 120 min after i.t. administration. The i.t. injection with ${\alpha}$-methylhistamine, but not carcinine slightly caused an elevation of the blood glucose level. In addition, histamine H1, H2, and H4 receptor agonists and antagonists did not affect the blood glucose level. In D-glucose-fed model, i.t. pretreatment with cetirizine enhanced the blood glucose level, whereas 2-pyridylethylamine did not affect. The i.t. pretreatment with dimaprit, but not ranitidine, enhanced the blood glucose level in D-glucose-fed model. In addition, ${\alpha}$-methylhistamine, but not carcinine, slightly but significantly enhanced the blood glucose level D-glucose-fed model. Finally, i.t. pretreatment with JNJ 7777120, but not VUF 8430, slightly but significantly increased the blood glucose level. Although histamine receptors themselves located at the spinal cord do not exert any effect on the regulation of the blood glucose level, our results suggest that the activation of spinal histamine H2 receptors and the blockade of spinal histamine H1 or H3 receptors may play modulatory roles for up-regulation and down-regulation, respectively, of the blood glucose level in D-glucose fed model.

  • It was possible to produce W/O/W one step multiple emulsion on the system which satisfied following conditions. 1. 1-5% of hydrophilic liquid surfactant over HLB20 and lipophilic liquid surfactant which has HLB 3∼5 2. Non wax copolymers as oil thickener 3. More than 0.5% of carbomer as aqueous thickener 4. The manufacturing process which neutralize the dispersed carbomer (2.0% in water), after emulsifying. For the selective addition into inner and outer aqueous phase, we melted the glucose in water before emulsifying. Using an Anthrone analysis method, we analyzed the encapsulation yield of glucose in inner water phase. It was possible to raise the water encapsulation yield of the multiple emulsion through the following conditions. 1. Using of anionic hydrophilic surfactant(HLB 40) and lipophilic surfactant (HLB 3∼5) 2. Controlling the ratio of hydrophilic surfactant and lipophilic surfactant 3. Strengthening interface with increase of non wax oil thickener. When the separated adding process of glucose was adopted, approximately 85% of glucose was added selectively within inner aqueous phase.

  • Rhodopseudomonas K-7은 혐기 광조건과 호기암조건에서 glucose, xylose로 이용하면서 성 장할 수 있었는데, 종배양시 glucose를 첨가하는 것이 본 배양에서 glucose자화를 촉진시켰고, glucose 60mM 배지에서 배양한 것을 종균으로 했을 때, glucose이 용량이 가장 많았다. Glucose 60mM배지에서 배양한 것을 종균으로 했을 때, 배지의 glucose 농도가 높더라도 이용할 수 있는 glucose양은 제한되어 있었다. 혐기 광조건에서 보다는 호기 암조건에서 성장속도도 빨랐고, glucose, xylose자화량도 많았다.
