Optimization of Maillard Reaction between Glucosamine and Other Precursors by Measuring Browning with a Spectrophotometer

  • The individual Maillard reactions of glucose, glucosamine, cyclohexylamine, and benzylamine were studied at a fixed temperature of $120^{\circ}C$ under different durations by monitoring the absorbance of the final products at 425 nm. Glucosamine was the most individually reactive compound, whereas the reactions of glucose, cyclohexylamine, and benzylamine were not significantly different from each other. Maillard reactions of reaction mixtures consisting of glucosaminecyclohexylamine, glucosamine-benzylamine, glucose-cyclohexylamine, and glucose-benzylamine were also studied using different concentration ratios under different durations at a fixed temperature of $120^{\circ}C$ and pH 9. Maillard reactions in the pairs involving glucosamine were observed to be more intense than those of the pairs involving glucose. Finally, with respect to the concentration ratios, it was observed that in most instances, optimal activity was realized, when the reaction mixtures were in the ratio of 1:1.

  • We have investigated the effect of glucosamine on the retinal cells after continuous infusion into cerebroventricle by using osmotic minipump to avoid peripheral effect. Continuous intracerebroventricular (i.c.v) infusion of glucosamine with the rate of 0.1 ${\mu}mol$/10 ${\mu}l$/hr for 7 days resulted in morphological changes of the optic nerve in electron microscopic level as well as morphological changes of the retina in light microscopic level. Retinal sections were immunostained for the detection of morphological changes of astrocytes. GFAP immunoreactivity appeared not only in the Muller cells but also many of the radial processes of Muller cells. The optic nerve showed deformed axon and slight lamellar separation of myelin sheath after continuous infusion of glucosamine in observing with electron microscope. Interestingly, vacuoles were observed in deformed axons and retinal layers were folded and detached. These results suggested that glucosamine plays a role in induction of morphological dysfunction in retina and optic nerves.

Anti-arthritic Effect of Glucosamine and Oriental Herbal Composition

  • This study was conducted to investigate the effect of an anti-inflammatory and analgesic action of the glucosamine HCl (Glucosamine) and SH-1 (Glucosamine + Oriental herbal composition combined group). Male sprague-Dawley rats $(200{\sim}250g)$ and ICR mice $(20{\sim}30g)$ were randomized and these experimental groups were divided into 4 groups. Two control group were given as negative control (saline) and positive control (Ibuprofen, 100 mg/kg) and two groups given as oral administration of Glucosamine (320 m/kg) and SH-1. Carrageenan induced paw edema test, hot plate method, croton oil induced granuloma, capillary permeability test and acetic acid writhing syndrome were also shown to be comparable in the SH-1 group to anti-inflammatory drug group such as positive control group (Ibuprofen). Although further studies should be performed to confirm the effects of SH-1, present results suggest that the combined administration of SH-1 have potential action in anti-inflammatory and analgesic action. It could be applicable for the improvement of arthritic symptoms as a new diet-supplement.

Optimization of Maillard Reaction in Model System of Glucosamine and Cysteine Using Response Surface Methodology

  • Sulfur-containing amino acids play important roles in good flavor generation in Maillard reaction of non-enzymatic browning, so aqueous model systems of glucosamine and cysteine were studied to investigate the effects of reaction temperature, initial pH, reaction time, and concentration ratio of glucosamine and cysteine. Response surface methodology was applied to optimize the independent reaction parameters of cysteine and glucosamine in Maillard reaction. Box-Behnken factorial design was used with 30 runs of 16 factorial levels, 8 axial levels and 6 central levels. The degree of Maillard reaction was determined by reading absorption at 425 nm in a spectrophotometer and Hunter's L, a, and b values. ${\Delta}E$ was consequently set as the fifth response factor. In the statistical analyses, determination coefficients ($R^2$) for their absorbance, Hunter's L, a, b values, and ${\Delta}E$ were 0.94, 0.79, 0.73, 0.96, and 0.79, respectively, showing that the absorbance and Hunter's b value were good dependent variables for this model system. The optimum processing parameters were determined to yield glucosamine-cysteine Maillard reaction product with higher absorbance and higher colour change. The optimum estimated absorbance was achieved at the condition of initial pH 8.0, $111^{\circ}C$ reaction temperature, 2.47 h reaction time, and 1.30 concentration ratio. The optimum condition for colour change measured by Hunter's b value was 2.41 h reaction time, $114^{\circ}C$ reaction temperature, initial pH 8.3, and 1.26 concentration ratio. These results can provide the basic information for Maillard reaction of aqueous model system between glucosamine and cysteine.

Effects of Glutamine, Glucosamine and Glutathione on the In Vitro Maturation of Porcine Oocytes

  • This study was carried out to investigate the effects of the supplementation of glutamine, glucosamine and glutathione on the porcine oocytes on IVM rates. Cocs were incubated in NCSU-23 supplemented with at $2.0{\sim}10.0\;mM$ glucosamine, $0.5{\sim}4.0\;mM$ glutamine and $0.1{\sim}1.0\;mM$ glutathione for 48 hrs. Oocytes were transferred to 50 ul drops of maturation medium covered with mineral oil and cultured in a $CO_2$ incubator ($38^{\circ}C$, 5% $CO_2$, 95% air). The IVM rates of oocytes cultured in NCSU-23 supplemented with 0.5, 1.0, 2.0 and 4.0 mM glutamine for 48 hrs were $46.0{\pm}4.5%$, $52.0{\pm}4.8%$, $50.0{\pm}4.2%$ and $44.0{\pm}4.5%$, respectively. The IVM rates of oocytes cultured in NCSU-23 supplement with 2.0, 5.0, 7.0, 10.0 mM glucosamine for 48 hrs were $44.0{\pm}4.5%$, $42.0{\pm}4.5%$, $38.0{\pm}4.6%$ and $24.0{\pm}4.8%$, respectively. The IVM rates of oocytes cultured in NCSU-23 supplemented with glucosamine were no significantly increased compare to the control ($42.5{\pm}4.0%$). The IVM rate of oocytes cultured in NCSU-23 supplemented with 3.0, 5.0, 7.0, 10.0 mM glutathione for 48 hrs were $40.0{\pm}3.2%$, $54.0{\pm}4.2%$, $48.0{\pm}4.5%$, $44.0{\pm}4.8%$, respectively. The IVM rate of oocytes cultured in NCSU-23 supplemented with glutamine and glutathione were significantly increased co~pared to those control ($42.5{\pm}4.0%$). Glucosamine did not affect the IVM rates of oocytes. IVM rates of oocytes cultured in NCSU-23 medium for 48 hrs were significantly increased compared to the cultured for 40 hrs.

  • 23종(種)의 대표적(代表約)인 세균(細菌)에 대(對)하여 glucosamine 함량(含量)을 측정(測定)하여 당지질(糖脂質)의 분포(分布)를 조사(調査)하고 당지질(糖脂質)의 함량(含量)과 Gram염색(染色)과의 상관관계(相關關係)를 규명(究明)한바 다음과 같은 결과(結果)를 얻었다. 1. Gram음성균(陰性菌) 및 varieable균(菌)은 그 균체(菌體) 당지질중(糖脂質中) glucosamine함량(含量)이 최소(最少) Proteus vulgaris의 $0.04{\mu}g$에서 부터 최고(最高) Aerobacter aerogenes 의 $2.48{\mu}g$의 범위(範圍)로 분포(分布)되여 있었다. Gram positive균(菌) 및 Bacillus속중(屬中)의 Gram양성균(陽性菌)은 전부(全部) $0.02{\mu}g$ 이하(以下)의 함량(含量)을 갖고 있었다. Gram양성(湯性)인 Corynebacterium sepedonicum 및 Staphylococcus aureaus는 특(特)히 glucosamine함량(含量)이 최소(最少)였다. 이들보다 Gram음성균(陰性菌)은 양성균(湯性菌)과 명확(明確)히 구별(區別)할 수 있을만큼 glucosamine함량(含量)이 많았다. 2. Gram염색(染色)은 당지질(糖脂質) 추출전(抽出前)보다 추출후(抽出後)가 염색(染色)이 더 잘되는 경향(傾向)이였다. 3. Aerobacter aerogenes, Bacillus circulans, Pseudomonas fluorescens 및 Salmonella typhirurium의 4균주(菌株)에서 얻은 당지질(糖脂質)의 적외선(赤外線) spectra는 대체(大體)로 같았으며 다 OH-group C-O, C-O-C, $CH_2+CH_3$, amide 결합(結合), 지방산(脂肪酸) ester 결합(結合), ester carbonyl 결합(結合)의 존재(存在)를 확인(確認)하였다.

  • 본 연구는 해양 갑각류의 껍질의 주요 구성성분인 키틴/키토산의 단량체인 glucosamine을 고체 산 촉매를 이용하여 화학중간체인 levulinic acid와 5-hydroxymethyl furfural (5-HMF)을 생산하기 위한 전환 반응을 수행하였다. 반응 결과, glucosamine 50 g/L, 반응온도 $180^{\circ}C$, 촉매량 5%, 그리고 반응시간 60분의 조건에서 약 36.86%의 levulinic acid를 얻을 수 있었다. 반면에 5-HMF는 약 0.91%의 낮은 수율로 전환되었다.

  • 골관절염은 관절연골의 퇴행성 변화로 연골기질의 분해로 인하여 연골 강도와 cushion으로서의 능력이 감소되는 질환이다. 골관절염의 대부분 약물치료는 통증과 염증을 감소시키는 목적으로 사용되며, 근본적인 치료효과를 주는 약물은 현재까지 개발되어 있지 않다. 영양약학 제품에 대한 질병의 예방적, 치료 보조적 차원에서 인체에 대한 기초적인 생리활성의 중요성을 인식하게 됨으로써 그 역할이 중요하게 인식되고 있다. 특히, 골관절염 치료에 쓰이는 glucosamine은 proteoglycan (PG)와 glucosaminoglycans(GAGs)의 합성의 전구물질로서 연골세포 생성을 자극하며, 통증, 염증의 경감 및 진행과정을 억제시키고, 관절기능 회복을 촉진하는 것으로 알려져 있다. 그러나 glucosamine이 어떤 기전에 의해 관절연골세포에 직접적인 영향을 주는지 밝혀져 있지 않다. 본 연구에서는 glucosamine sulfate가 연골세포에 미치는 영향을 규명하고자 하였다.

Glucosamine Hydrochloride and N-Acetylglucosamine Influence the Response of Bovine Chondrocytes to TGF-β3 and IGF in Monolayer and Three-Dimensional Tissue Culture

  • BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.
