The rhizospheric zone abutting plant roots usually clutches a wealth of microbes. In the recent past, enormous genetic resources have been excavated with potential applications in host plant interaction and ancillary aspects. Two Pseudomonas strains were isolated and identified through 16S rRNA and rpoD sequence analyses as P. fluorescens QAU67 and P. putida QAU90. Initial biochemical characterization and their root-colonizing traits indicated their potential role in plant growth promotion. Such aerobic systems, involved in gluconic acid production and phosphate solubilization, essentially require the pyrroloquinoline quinine (PQQ)-dependent glucose dehydrogenase (GDH) in the genome. The PCR screening and amplification of GDH and PQQ and subsequent induction of mutagenesis characterized their possible role as antioxidants as well as in growth promotion, as probed in vitro in lettuce and in vivo in rice, bean, and tomato plants. The results showed significant differences (p ≤ 0.05) in parameters of plant height, fresh weight, and dry weight, etc., deciphering a clear and in fact complementary role of GDH and PQQ in plant growth promotion. Our study not only provides direct evidence of the in vivo role of GDH and PQQ in host plants but also reveals their functional inadequacy in the event of mutation at either of these loci.
The processing conditions for producing and edible alginate film from sea tangle were investigated by measuring water vapor permeability (WVP), tensile properties and colors of film. The alginates were extracted with $3\%$ sodium carbonate ($Na_2CO_3$) solution for 5 hrs, and the alginate films were prepared with extracted sodium alginates, D-gluconic acid lactone, $CaCO_3$ and various plasticizers, As the concentration of alginate in making the film was increased, the WVP and elongation of the film were decreased and the tensile strength was increased. The higher amount of plasticizer showed the higher WVP and elongation, and the order of various plasticizers in increasing the WVP and elongation of the film was glycerol, polyethylene glycol, and sorbitol. The addition of sorbitol in the film showed the highest value (89.57) in lightness of Hunter color system and the film added polyethylene glycol had the highest values (-1,12 and 11.4) in redness and yellowness.
Background: Panax notoginseng is a highly valued medicinal herb used widely in China and many Asian countries. Its root and rhizome have long been used for the treatment of cardiovascular and hematological diseases. Imaging the spatial distributions and dynamics of metabolites in heterogeneous plant tissues is significant for characterizing the metabolic networks of Panax notoginseng, and this will also provide a highly informative approach to understand the complex molecular changes in the processing of Panax notoginseng. Methods: Here, a high-sensitive MALDI-MS imaging method was developed and adopted to visualize the spatial distributions and spatiotemporal changes of metabolites in different botanical parts of Panax notoginseng. Results: A wide spectrum of metabolites including notoginsenosides, ginsenosides, amino acids, dencichine, gluconic acid, and low-molecular-weight organic acids were imaged in Panax notoginseng rhizome and root tissues for the first time. Moreover, the spatiotemporal alterations of metabolites during the steaming of Panax notoginseng root were also characterized in this study. And, a series of metabolites such as dencichine, arginine and glutamine that changed with the steaming of Panax notoginseng were successfully screened out and imaged. Conclusion: These spatially-resolved metabolite data not only enhance our understanding of the Panax notoginseng metabolic networks, but also provide direct evidence that a serious of metabolic alterations occurred during the steaming of Panax notoginseng.
Burkholderia contaminans PSB-A and Burkholderia ambifaria PSB-B were isolated from button mushroom bed to estimate their phosphate solubility. The phosphate-solubilizing abilities of these strains were assessed by measuring the phosphorus content in a single and co-inoculation medium for 7 days. The co-inoculation of these two strains released the highest content of soluble phosphorus (166.3 ㎍ mL-1) into the medium, followed by single inoculation of B. contaminans PSB-A (143.73 ㎍ mL-1) and B. ambifaria PSB-B (127.1 ㎍ mL-1). The highest pH reduction, organic acid production, and glucose consumption were also observed in the co-inoculation medium. According to the plant growth promotion bioassay, co-inoculation enhanced the growth of romaine lettuce much more than the single inoculation (20.4% for leaf widths and 16.6% for root lengths). Although no significant difference was noted between single and co-inoculation of bacterial strains in terms of phosphorous release and plant growth, co-inoculation of PSB may have a beneficial effect on crop growth due to a synergistic effect between the strains.
This study investigated the synergistic effect of single inoculation and co-inoculation of phosphate-solubilizing bacteria (PSB) Burkholderia metallica JH-7 and Burkholderia contaminans JH-15. Phosphate-solubilizing abilities of these strains were assessed by measuring phosphorus content in culture media that were singly inoculated or co-inoculated with these strains for 7 days. B. metallica JH-7 was found to release the highest content of soluble phosphorus ($140.80{\mu}g\;mL^{-1}$ ) into the medium, followed by single inoculation of B. contaminans JH-15 ($135.95{\mu}g\;mL^{-1}$ ) and co-inoculation of two strains ($134.84{\mu}g\;mL^{-1}$ ). The highest pH reduction, organic acid production, and glucose consumption were observed in the medium inoculated with B. metallica JH-7 alone compared with that in the medium co-inoculated with both the strains. Results of a plant growth promotion bioassay showed 17.4% and 7.48% higher leaf and root growth, respectively, in romaine lettuce inoculated with B. metallica JH-7 alone than in romaine lettuce inoculated with a control strain. However, no significant difference was observed between single inoculation and co-inoculation of these strains with respect to phosphorus release and plant growth. Although the results of the present study did not show the synergistic effect of phosphate solubilization by the PSB strains examined, these results indicate that treatment with PSB exerts a beneficial effect on crop growth.

Experiments were carried out to investigate the selection of industrial medium and optimization of fermentation process for the production of erythritol by Candida magnoliae SR101. In the batch fermentation, light steep water(LSW) was the best nitrogen source for the industrial production of erythritol. For the optimization of culture condition, the batch culture was performed. When the concentration of LSW was 65 mL/L in the defined medium containing 250 g/L of glucose, 44% of erythritol yield with 110 g/L of erythritol concentration and 0.66 g/L-hr of productivity, respectively were obtained. Two-stage fed-batch culture was performed to improve the volumetric productivity of erythritol. High density cell culture in the growth stage was performed by batch type with 100 g/L glucose and 500 mL/L LSW concentration, respectively. The cell yield was 0.72 g-cell/g-glucose. Productivity of erythritol was increased and concentration of organic acids such as gluconic acid and acetic acid were decreased when initial pH of 6.5 controlled by 28% ammonia water For increasing yield of erythritol, glucose concentration in the production stage was tested. 37% of total erythritol yield with 186 g/L of erythritol concentration and 1.66 g/L-hr of erythritol productivity were obtained when 820 g of glucose powder was directly added for making up 450 g/L of glucose at production stage.
This study investigated the synergistic effect of single inoculation and co-inoculation of phosphate-solubilizing bacteria (PSB) Pantoea rodasil LH-1and Burkholderia stabilis HS-7. Phosphate-solubilizing abilities of these strains were assessed by measuring phosphorus content in culture media that were singly inoculated or co-inoculated with these strains for 7 days. The co-inoculation of P. rodasil LH-1and B. stabilis HS-7 was found to release the highest content of soluble phosphorus ($783.41{\mu}g\;mL^{-1}$) into the medium, followed by single inoculation of B. stabilis HS-7B ($743.90{\mu}g\;mL^{-1}$) and P. rodasil LH-1 ($736.59{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production, and glucose consumption were also observed in the medium inoculated with both the strains, compared with that in the medium inoculated with the strain alone. Results of a plant growth promotion bioassay showed 7.7% and 15.5% higher leaf and root growth, respectively, in romaine lettuce co-inoculated with P. rodasil LH-1and B. stabilis HS-7 than those inoculated with the strain alone. However, no significant difference was observed between single inoculation and co-inoculation of these strains with respect to phosphorus release and plant growth. Although the results of the present study did remarkdly not show the synergistic effect of phosphate solubilization by co-inoculation of the PSB strains examined, these results indicate that treatment with PSB exerts a beneficial effect on crop growth.
The present study was conducted to investigate the synergistic effects caused by single and co-inoculation of the phosphate solubilizing bacteria (PSB), Paraburkholderia phenazinium YH3 and Paraburkholderia metrosideri YH4. Phosphate solubilization was assessed by measuring the phosphorus contents for 7 days in a single and co-inoculation medium. Co-inoculation of the two strains was found to release the highest content of soluble phosphorus ($1,250{\mu}g\;mL^{-1}$) into the medium, followed by the single inoculation of P. metrosideri YH4 ($1196.59{\mu}g\;mL^{-1}$) and P. phenazinium YH3 ($994.34{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production and glucose consumption was also observed in the co-inoculation medium of the two strains. A plant growth promotion bioassay revealed that co-inoculation with the two strains enhanced the growth of romaine lettuce more than single inoculation with either of the two strains (28.5% for leaf and 16.6% for root). Although there was no significant difference between single and co-inoculation of bacterial strains in terms of phosphorous release and plant growth, the synergistic effects of co-inoculation with PSB could be beneficial for crop growth.
Glucose fed-batch culture was studied to improve cellulose productivity by Acetobacter xylinum BRC5. When initial glucose concentrations in batch cultures were less than 20 g/L, yield coefficients of cellulose (Yp/s) remained a constant value of 0.21 g cellulose/g glucose. But a low yield coefficient, Yp/s=0.13 was obtained from an initial glucose concentration of 40 g/L. Since initial high glucose concentrations in batch culture resulted in low yields of cellulose, constant fed-batch cultures were carried out. The optimal feed rate for fed-batch culture was 2.22 g glucose/L.h. In constant fed-batch culture without DO control, 10 g/L of cellulose was obtained from 40 g/L of glucose with this feed rate, which was approximately two fold higher than that of the batch culture with the same initial glucose concentration. In DO stat plus fed-batch culture, the highest cellulose productivity could be obtained when dissolved oxygen level was controlled at 10% of air saturation, and cellulose productivity increased about 1.5 times compared with that of the culture without DO control.
In order to produce high concentration of sodium gluconate, optimization of the fermentation conditions, such as glucose concentration, inoculum size, dissolved oxygen concentration and glucose feeding method, was examined. When the glucose concentration was maintained in the range of 30∼50 g/L during the batch fermentation, glucose conversion yield and productivity were 92.2% and 6.0 g/L/hr, respectively. In the case of the low concentration below 30 g/L, the yield decreased by about 25%. As the inoculum size increased above 20%(w/v), lag phase was shortened but the productivity decreased. The dissolved oxygen level of 60∼70% was shown to be the threshold point for 75% of increase in the productivity of sodium gluconate. Finally, optimal glucose feeding rate was determined using various feeding methods such as exponential feeding, feeding based on the average glucose consumption rate and was determined using various feeding methods such as exponential feeding, feeding based on the average glucose consumption rate and on the oxygen uptake rate and etc. Our result shows that glucose feeding, based on the oxygen uptake rate is a very simple, efficient and robust method, especially when oxygen is consumed as a substrate for the bioconversion. Using the above glucose feeding strategy under the optimized condition, 255 g/L of sodium gluconate concentration, 12 g/L/hr of productivity and 95% of glucose conversion yield were achieved with A. niger ACM53.
