Effect of Nutritional Requirements and Feeding Regimes at First Feeding on the Survival of the Larval Olive Flounder Paralichthys olivaceus

  • Despite the relatively high production of fingerlings of the olive flounder Paralichthys olivaceus, its larval rearing in terms of first feeding has not been fully analyzed. We evaluated the variations of amino acids and fatty acids of starved larvae over 96 hr after hatching. We also investigated depletion of the yolk and oil globule of starved larvae and those fed the rotifer Brachionus plicatilis. In addition, the optimum size of the rotifers according to the mouth size of the larvae, and the point of no return with delay of the first feeding, were also examined. The amino acids in the egg decreased abruptly during embryo development. At 48 to 72 hr after hatching, the amino acids of starved larvae decreased by $30-40\%$ from the level in newly hatched larvae. The concentrations of fatty acids in newly hatched larvae were lower than those of floating eggs and dropped sharply at 48 hr after hatching, when the yolk disappeared. The starved larvae depleted their yolksacs and oil globules earlier than the fed larvae did. At 84 hr after hatching, rotifers were detected for the first time in the guts of the larvae, which were about 3 mm in total length. The point of no return appeared to be close to the fourth day from the first feeding. For a high survival rate of P. olivaceus larvae, the first feeding should occur before the third day after hatching.

산양유의 산업적 이용 가치에 대한 연구 고찰 (The Commercial Value of Goat Milk in Food Industry)

  • In many countries, goat milk is an excellent nutrient source and is less allergenic for children and the elderly. The casein composition of goat milk consists largely of ${\beta}$-casein and lower amounts of ${\alpha}_{s1}$-casein, which may interfere with digestion by forming solid curds in the human stomach. Goat milk contains small fat globules and large amounts of medium chain fatty acids for, better digestibility, as well as abundant minerals and vitamins with high absorption rates. Recently, the medical benefits of goat milk in different human disorders have been recognized, leading to an increased interest in developing functional foods with goat milk, particularly for individuals with malabsorption syndrome. However, the physiological and biochemical properties of goat milk are largely unknown. We review the importance of goat milk as a potential functional food by providing scientific evidence confirming its health benefits.

Effects of Using Soybean Protein Emulsion as a Meat Substitute for Chicken Breast on Physicochemical Properties of Vienna Sausage

  • The aim of this study is to determine the effects of using emulsion manufactured with soybeans (ES) to substitute chicken breast in Vienna sausages. Four types of Vienna sausages (S1: 10% ES and 50% chicken, S2: 20% ES and 40% chicken, S3: 30% ES and 30% chicken, and S4: 40% ES and 20% chicken) for this study were made. The pH, color, proximate composition, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), microphotographs, cooking yields, and texture profile analysis of sausages were examined. The pH value of uncooked and cooked sausages increased significantly with increasing ES content (p<0.05). The crude protein contents of S2, S3, and S4 were significantly higher than that of the control (p<0.05). Furthermore, the SDS-PAGE results showed that α-conglycinin, β-conglycinin, and the acidic subunit of glycinin all increased with increasing ES content. Microphotographs revealed that increasing the ES content decreased the size of fat globules. The cooking yields of samples increased significantly with increasing ES content (p<0.05). The hardness values of ES treated samples were significantly lower than that of the control (p<0.05). Therefore, 30% substitute of chicken breast with ES can improve the quality and structure of Vienna sausage, without inducing critical defects.

우유 균질 조건 예측을 위한 반응표면방법론의 활용 (Applying Response Surface Methodology to Predict the Homogenization Efficiency of Milk)

  • Response surface methodology (RSM) is a statistical approach widely used in food processing to optimize the formulation, processing conditions, and quality of food products. The homogenization process is achieved by subjecting milk to high pressure, which breaks down fat globules and disperses fat more evenly throughout milk. This study focuses on an application of RSM including the logit transformation to predict the efficiency of milk homogenization, which can be maximized by minimizing the relative difference in fat percentage between the top part and the remainder of milk. To avoid a negative predicted value of the minimum of this proportion, the logit transformation is used to turn the proportion into the logit, whose possible values are real numbers. Then, the logit values are modeled and optimized. Subsequently, the logistic transformation is used to turn the predicted logit into the predicted proportion. From our model, the optimum condition for the maximized efficiency of milk homogenization was predicted as the combination of a homogenizer pressure of 30 MPa, a storage temperature of 10℃, and a storage period of 10 days. Additionally, with a combination of a homogenizer pressure of 30 MPa, a storage temperature of 10℃, and a storage period of 50 days, the level of milk homogenization was predicted to be acceptable, even with the problem of extrapolation taken into account.

복령과 황태껍질 발효물 혼합물의 고지방식이 유도 비만 동물모델에서의 항비만효과 (Anti-Obesity Effect of Fermented Pollack Skin and Wolfiporia extensa Ginns in High Fat-induced Obesity Animal Model)

  • This study aimed to determine the anti-obesity effect of adding Wolfiporia extensa Ginns (W) to fermented pollack skin products in an obesity-induced animal model. The experimental groups were the normal diet group (C), high-fat diet group (HF), dried pollack skin (H1), fermented pollack skin (H2), and W of 0.1 (F2-WL), 0.3 (F2-WM), and 0.5 (F2), respectively. It was confirmed that adding W to fermented pollack skin reduced blood triglycerides, total cholesterol, and LDL levels, while increasing HDL levels. Wolfiporia extensa Ginns was effective in controlling weight and improving blood lipids in a dose-dependent manner. In histological analysis, findings of fatty liver induced by a high-fat diet were improved by the addition of H2 and W. Size and density of fat globules in the epididymis were decreased. In addition, the concentration of TNF-α was increased in the high-fat diet group, but decreased by the addition of fermented pollack skin and W. In conclusion, adding fermented dried pollack skin and Wolfiporia extensa Ginns was effective for weight control and blood lipid improvement. Thus, the use of by-products in functional foods is expected to have a high value in the future.

양식산 벤자리(Parapristipoma trilineatum) (농어목, 하스돔과) 자치어의 형태 및 골격발달 (Skeletal and Morphological Development of Larvae and Juveniles in Cultured Threeline Grunt Parapristipoma trilineatum (Perciformes: Haemulidae) from Korea)

  • Morphological and skeletal development in the cultured threeline grunt Parapristipoma trilineatum was studied from 1.96 to 61.39 mm notochord length (NL) or standard length (SL) specimens (n=360). After two days post-hatching (dph, 2.13 mm NL), the mouth opened, and melanophores formed in the eyes and egg yolk were absorbed. Oli globules were absorbed at 3 dph (2.71 mm NL). The total number of fin rays reached that of adults at 28 dph (10.58 mm SL). During skeletal development, the premaxillaray, dentary, opercle, preopercle, and cleithrum first began to ossify at 6 dph (3.51 mm NL). Ossification of all skeletal elements was completed at 28 dph (14.57 mm SL). The vertebrae began to ossify at 17 dph (5.63 mm SL) and were completed at 28 dph (8.48 mm SL), progressing from cranium to caudal bone. The centrum ossified from the dorsal to ventral direction. Vertebral fusion and separation were found to be common skeletal abnormalities.

멸종위기에 처한 얼룩새코미꾸리 Koreocobitis naktongensis (Cobitidae)의 난 발생 및 자치어 형태 발달 (Morphological Development of Egg and Larvae of Koreocobitis naktongensis (Cobitidae))

  • 멸종위기 담수어류인 얼룩새코미꾸리 Koreocobitis naktongensis의 난 발생 과정 및 자치어 형태 발달을 조사하였다. 인간 융모성 성선자극호르몬(HCG)을 10 IU/g의 농도로 암컷의 복강에 주사하여 성숙란을 얻었으며, 건식법으로 정자와 수정시켰다. 수정란의 난경은 1.0~1.2 mm로 유구는 존재하지 않았다. 배체는 수온 $20{\pm}1^{\circ}C$에서 수정 38시간 후부터 부화하기 시작하였다. 부화 직후 자어의 전장은 2.5~2.8 mm로 입과 항문은 아직 열리지 않았다. 부화 후 7일째 자어는 전장 4.5~5.2 mm로 난황은 완전히 흡수되었다. 부화 후 50일째 모든 지느러미가 완성되는 치어기로 이행하였고, 이때의 전장은 21~27 mm였다.

Examination of the xanthosine response on gene expression of mammary epithelial cells using RNA-seq technology

  • Background: Xanthosine treatment has been previously reported to increase mammary stem cell population and milk production in cattle and goats. However, the underlying molecular mechanisms associated with the increase in stem cell population and milk production remain unclear. Methods: Primiparous Beetal goats were assigned to the study. Five days post-partum, one mammary gland of each goat was infused with xanthosine (TRT) twice daily ($2{\times}$) for 3 days consecutively, and the other gland served as a control (CON). Milk samples from the TRT and CON glands were collected on the 10th day after the last xanthosine infusion and the total RNA was isolated from milk fat globules (MEGs). Total RNA in MFGs was mainly derived from the milk epithelial cells (MECs) as evidenced by expression of milk synthesis genes. Significant differentially expressed genes (DEGs) were subjected to Gene Ontology (GO) terms using PANTHER and gene networks were generated using STRING db. Results: Preliminary analysis indicated that each individual goat responded to xanthosine treatment differently, with this trend being correlated with specific DEGs within the same animal's mammary gland. Several pathways are impacted by these DEGs, including cell communication, cell proliferation and anti-microbials. Conclusions: This study provides valuable insights into transcriptomic changes in milk producing epithelial cells in response to xanthosine treatment. Further characterization of DEGs identified in this study is likely to delineate the molecular mechanisms of increased milk production and stem or progenitor cell population by the xanthosine treatment.

Temperature-dependent Index of Mitotic Interval (${\tau}_0$) for Chromosome Manipulation in Korean Bullhead, Pseudobagrus fulvidraco

  • Korean bullhead (Pseudobagrus fulvidraco) was collected from the Kum River areas of Kangkyung-eup, Nonsan city, Chungcheongnam-do, Korea, from April to June, 2012 and was fertilized in order to observe egg development and temperature-related cleavage rates and mitotic intervals (${\tau}_0$). The fertilized eggs were separative, demersal and light yellowish with $1.5{\pm}0.06mm$ in diameter, and did not contain oil globules. The first cleavage stages were 90 min, 80 min, 60 min and 50 min at $21^{\circ}C$, $24^{\circ}C$, $27^{\circ}C$ and $30^{\circ}C$, respectively. At higher temperatures, eggs developed faster and underwent further identical development. For Korean bullhead, ${\tau}_0$ were $33.4{\pm}2.08$ min at $21^{\circ}C$, $31.5{\pm}3.06$ min at $24^{\circ}C$, $28.1{\pm}2.11$ min at $27^{\circ}C$ and $26.4{\pm}3.35$ min at $30^{\circ}C$. There were strong negative correlations between the $\tau_0$ and water temperatures at all points studied (Y=-1.13X+58.15, $R^2$=0.98, n=30, where Y is ${\tau}_0$ and X is temperature). The results obtained in this work will be helpful for chromosome manipulation by use of cleavage frequency data and ${\tau}_0$ data in Korean bullhead.

대두입자가 분산된 두유에서 기름입자의 입도분포 측정 (The Measurement of Oil Globule Size Distribution in the Soymilk Suspended with the Soybean Particle)

  • 두유에는 기름의 양이나 입도를 분석하는데 장해를 주는 대두단백질, 미세한 대두입자들이 공존하고 있어서 기존의 microscopic image analysis, coulter counter, 또는 Photo-analysis 등의 사용이 많은 문제점을 노출했다. 본 실험에서 Stokes 법칙을 응용하여, 중력장과 원심력장하에서 homogeneous suspension과 cumulative method를 이용하여 입도분포를 분석했다. 선정된 두유에서 기하학적 평균지름은 중력장하에서는 $0.33{\mu}m$ 표준편차 $1.73{\mu}m$이고, 원심력 장에서는 평균지름은 $0.31{\mu}m$, 표준편차는 $1.81{\mu}m$으로, 매우 유사한 결과를 얻었다. 균질압이 유화안정성에 크게 영향을 주는 인자이기 때문에 균질압을 변화했을 때, 균질압이 높을수록 평균입도는 작은 쪽으로 이동하였으며, 입자의 분산 형태는 평균지름을 중심으로 밀도 있게 분포하는 쪽으로 변화하여, 균질압이 기름 입도분포에 영향을 미쳐 유화 안정과 깊은 관계가 있는 것을 확인하였다.
