In this paper it was attempted to observe the effect of defatted panax ginseng supplement of the growth rate, feed and protein efficiency ratios, and the contents of cholesterol, total lipid and protein in the serum, liver and aorta in Sprague-Dowley Albino male rat (weighing 83 ${\pm}$ 4 g). Seven kinds of experimental diets were prepared as follows : Stock (control) diet, ginseng control diets supplemented with 0.5, 1.0 and 3.0% of ginseng powder to the stock diet, and defatted ginseng Powder diets supplemented with 0.5, 1.0 and 3.0% of defatted ginseng powder to the stock diet. .All diets contained same level of lipid and protein, respectively. The results obtained are as follows : 1. The growth rate in the feeding group of 0.5% defatted ginseng powder diet for 16week were higher than other diet groups. 2. Feed and Protein efficiency in 0.5% defatted ginseng group showed similar tendency to that in body growth rate. 3. The total cholesterol contents in the serum of 0.5% defatted ginseng Powder diet$.$ group showed the tendency to decrease gradually for 4 to 12 weeks, maintaining higher level than other groups. The free cholesterol contents in the serum of defatted ginseng powder diet group in 8 the and 16th weeks were higher than all ginseng control diet group. The total and free cholesterol contents in the liver of all defatted ginseng diet groups in 16th week were higher than hose of all ginseng control groups. The total cholesterol content at 12h week and the free cholesterol content at 16th week i n the aorta of all defatted ginseng diet groups were lower than those of ginseng control groups, respectively. 4. The total lipid contents in the serum of 1.0 and 3.0% defatted ginseng diet groups at 2nd to I2th weeks were lower than other groups, and those in the liver and aorta of all defatted ginseng diet groups at 12th weeks were lower than those of ginseng control diet groups. 5. The protein contents of the serum and aorta were continuously increased throughout whole experimental period in all experimental groups. The protein content of the liver of all groups were decreased at 2nd week and after then no change was observed.
This study was divised to observe the effects of ginseng on the body components of Sprague-Dowley Albino male rats by alternating the diet. Just weaned rats (130 heads, weighing(83 ${\pm}$ 4g) were fed with each stock diets supplemented with 0.5, 1.0 or 3.0% ginseng powder for 4 weeks or 8 weeks, and after that fed with stock diet only for 8 or 4 weeks. Compared with the each corresponding group which was fed with ginseng control diet, the protein, total lipid, total cholesterol and free cholesterol contains in serum, liver and aorta of them were determined. The results obtained was summarized as follows ; Protein and total lipid contents of the rat were decreased, caused by the alternating diets, but total cholesterol and free cholesterol contents were increased in the serum, protein and free cholesterol contents increased, conversely total lipid contents decreased in the liver, and in aorta total lipid and total cholesterol contents decreased, In the view of the above results, it can be seen that the alternating diet (stock diet after feeding with ginseng diet) has an influence on the body components of rat.
Radioprotective ginseng protein fraction was isolated from Korean white ginseng and its cytotoxic effect on CHO-K1 cells was studied by the method of measuring the relative cell survival and total cellular protein content (FRAME method). When ginseng protein at the dose of 300, 600, 900, $1200{\mu}g/ml$ was treated to cells for 24 hrs, the relative survival was significantly decreased at the concentration of above $600{\mu}g/ml$, indicating the presence of cytotoxic effect of the protein at certain concentration. When cellular protein content was measured after ginseng protein at the dose of 300, 600, 900, $1200\;{\mu}g/ml$ was treated, the amount of cellular protein was significantly reduced at the concentration above $600{\mu}g/ml$ in the case of 24 hr treatment and at all concentrations including $300{\mu}g/ml$ in the case of 72 hr treatment. The data suggest that the protein may inhibit cell growth, resulting in the reduction of live cells in culture. $ID_{50}$ value which is the concentration of ginseng protein that reduces the total cellular protein content to 50% of the control was calculated as 2276.86 and $1323.32\;{\mu}g/ml$ in groups treated for 24 and 72 hr, respectively. Since $ID_{50}$ value of above $1000{\mu}g/ml$ indicates very weak cytotoxicity, the ginseng protein seems to exert very weak cytotoxic effect on CHO-K1 cells.
This study was carried out to obtain the basic information for non-saponin contents that can be used to index fresh ginseng roots (Panax ginseng C. A. Meyer) cultivated in the Republic of Korea and China. Non-saponin components in fresh gingeng roots which were cultivated in various areas and ages in Korea were determined. Acidic polysaccharide, total polysaccharide, crude polyacetylene were quantitatively analyzed by using the method of spectrophotometric determination, while the total protein was analyzed by using Lowry method. The results show that there were no statistically significant differences for the average contents of four non-saponins among 4-years-old, 5-years-old, and 6-years-old fresh ginseng roots. Additionally, this study assessed the average contents of non-saponin components in 4-years-old fresh ginseng roots (Panax ginseng C. A. Meyer) which were cultivated in Korea and China. The result showed that the average contents of crude polyacetylene and acidic polysaccharide were statistically significant. Four-years-old fresh ginseng roots cultivated in Korea had the higher average contents of crude polyacetylene and acidic polysaccharide than those cultivated in China. However the average contents of total polysaccharide and total protein had no statistically significant difference.
In this study, the effects of red ginseng components [ginsenosides (total saponins and $Rg_1$) on the function of ryanodine receptor (RyR) -$Ca^{2+}$ release channel complex protein (named as RyR or $Ca^{2+}$ channel), a membrane protein in sarcoplasmic reticulum (SR) of rabbit skeletal muscle were examined at the SR vesicle's level and the molecular levels with Chaps-solubilized and purified $Ca^{2+}$ channel protein and with reconstituted proteoliposomes by dialysis. The results were as follows. 1. The binding of ryanodine known as inhibitor of muscle contraction to the RyR was decreased at the whole range of concentration ($10^2$~$10^7$%) by these two ginseng components. In heavy SR vesicles, Chaps-solubilized and purified $Ca^{2+}$ channel protein, and reconstituted vesicles, its maximal inhibition by total saponins was shown at the concentration of $10^3$, $10^3$%, and $10^5$% respectively, and by gin- senoside $Rg_1}$) each was $10^3$%, $10^3$%, and $10^4$%. 2. The release of $Ca^{2+}$ ion through $Ca^{2+}$ channel in heavy SR vesicles and reconstituted proteoliposomes was increased as a whole by these two ginseng components, and particularly maximal release by both of them was shown at the range of $10^4$~$10^6$%. These results were seemed to be caused by conformational change of $Ca^{2+}$ release channel protein (RyR) by red ginseng components [ginsenosides (total saponins and $Rg_1}$).
This study was investigated to evaluate the feasibility of protein concentrate for human food from ginseng leaf. The protein concentrate was prepared from ginseng green leaf by treating with cold acetone , followed by protein extraction with 0.2% NaOH containing 0.5% 2-mercaptoethanol and 0.5% sodium dodecyl sulfate. Proximate composition of the ginseng leaf protein concentrate (LPC) showed that fat and ash was less than 1%, protein was about 75%, total sugar and total saponin was 5% and 1.2%, respectively. As compared to the provisional amino acid pattern reported by FAO/WHO, ginseng LPC was found to be poor in S-containing amino acids, which were the first limiting amino acid. The amino acid score and E/T ratio of ginseng LPC were 43.1 and 3.02, respectively. Digestibility of ginseng LPC by pepsin and trypsin was lower than that of milk casein.
This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from the mixture of leaf and stem of ginseng radix(Panax ginseng CA. Meyer). This study measured the intake levels and efficiency ratio, growth rate, absorption ratio of carbohydrate, lipid and protein of rat by feeding with LSE during 6 weeks. We analyse the hematological and serum metabolic variables, serum lipid concentrations. Total diet and protein intake levels were low, but efficiency ratios were significantly high in LSE administered groups than the control group. Weight gain, liver and kidney weight of LSE groups were significantly higher than the control group. Blood RBC, Hct, Hb, total protein and albumin concentrations were reasonable levels in LSE administered groups compared to the basal diet group. Also serum total cholesterol, LDL-cholesterol, triglyceride contents of LSE groups were low, but HDL-cholesterol level was higher than the basal diet group. These results imply that leaf and stem of ginseng radix could be used as possible food resources, functional food material and feed stuff.
To study e(feces of Korean Binseng (Parfax ginseff C. A. hfeyer) total saponin fraction on spermidine and spermine metabolism in rat reproductive systems, we administrated the saponin fractation to rats for 2 years. Then, we determined the activities of S-adenosylmethionine decarboxylase (SAMDC), the quantitation of the enzyme protein and the amounts of spermidine and spermine contents In prostate and testis. In young sexually immature stage, administration of Korean ginseng saponin fraction showed no effect on SAMDC activities. The stimulatory effect on the activities of SAMDC gradually increased and reached maximal activities in test groups of prostate and testis at sexually mature stage. The amounts of SAMDC protein in test groups were paralleled by the changes of SAMDC activities in test groups, indicating that all of the increased activity occurring in administration of ginseng saponin fraction was not due to the activation of SAMDC activity but to the Increase in enzyme protein. However, the spermidine and spermine contents of test groups showed small increase in compared to that of control groups. From these results, we suggest that administration of ginseng saponin fraction alter the spermidine and spermine metabolism in sexually mature and aged reproductive systems in rats.
This study was carried out to Investigate the effects of Panax ginseng, Ganoderma lucidum extract and crude polysaccharide of G. lucidum on the growth of lactic acid bacteria. p. ginseng extract contained 60.7% carbohydrate and 27.5% protein, whereas G. lucidum contained 35.9% carbohydrate and 46.3% protein. The total sugar and protein content of crude polysaccharide of G. lucidum were 47.2% and 15.2%, respectively. Two amino acids(hg, Trp) were detected in p. ginseng extract and 11 amino acids (hg, Trp, Ua, Lys, Ser, etc.) in C. lucidum extract. By the addition of p. ginseng, 5. lucidum extract and crude polysaccharide, the cia. p. ginseng was more effective on the growth of 1. casei an: G. lucidum was more effective on that of S. thermophilus. The effect of free amino acids on the growth of tactic acid bacteria was also examined. Arginine and lysine stimulated the growth of L. casei, whereas Lysine, serine, arginine, and glutamic acid stimulated the growth of 5. thermophilus.
Nitrogen compounds of Panax ginseng and their biological activities in plant and animal were reviewed. Major nitrogen compounds found in P. ginseng are free amino acids. Water solilble proteins, indouble proteins and peptides. Minor nitrogen compounds are dencichine. Glycolyroteins, amines, alkaloides, methoxy or alkyl pyrazine derivatives, free nucleosides and nucleic acid bases. 4-methyl-i-thiazoltethanol and pyroglutamic acid the contents of total nitrogen and protein in root Increased until 13 years old which was the highest age tinder investigation. Soluble protein content increased with the root weight and was higher in xylem pith than cortex-epidermis indicating the close relation with root growth. Arginine, which covered 58% of total free amino acids, may serve as storage nitrogen. Arginine seems to be changed into proline in rhizome. threonine in stem and again threonine and arginine in leaf. The greater the root weight the higher the polyamine stimulated Polyamine stimlllated the growth of root callus. Physiological roles of other minor nitrogen compounds are unknown although content is relatively high ((1.if) 6.w). Biochemical and pharmacological activities of some nitrogen compounds for animal were more investigated than physiological role there plant itself. Radiation and U.V protective function (heat stable protein). insulin-like activity in lipogenesis and livolysis (adenosine and pyroglutamic acid), depression of blood sugar content (glycopevtide). htmostatic and nellrotoxic activity (dencichine) and, sedative and hypnotic activity (4-methyl-i-thiazoleethanol) are reported. Heat stable protein increased with root age. The traditional quality criteria appear to be well in accordance with biological activities of nitrogen compounds. Chemical studies of nitrogen compounds seem relatively rare, probably due to difficulty of isolation, subsequently the investigations of biological activities are little.
