13 weeks repeated oral dose toxicity studies with LMK02-Jangwonhwan in SD rats (LMK02의 Sprague-Dawley 랫드를 이용한 13 주간 반복 경구투여 독성시험)

  • Objectives : The oriental medicine Jangwonhwan, a boiled extract of 12 medicinal herbs/mushrooms, has been prescribed to patients with cognitive dysfunction, as originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan (LMK02-Jangwonhwan) consisting of seven medicinal plants/mushrooms, was shown to reduce the ${\beta}$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer's disease. The toxicity of LMK02-Jangwonhwan was investigated in SD rats, by a daily oral administration for 13 weeks and NOAEL(No observed adverse effect dose), a definite toxic dose and target organ, as well. Methods : Quality control of the tablet form of LMK02-Jangwonhwan was established by estimating the indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02-Jangwonhwan was investigated in 6 week old, specific pathogen free (SPF), Sprageu-Dawley rats by oral administration. Each test group consisted of 10 male and 10 female rats. The groups received doses of 500, 1,000 or 2,000 mg/kg/day of test substance for 13 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of the control group. Results : The 13-week repeated oral treatment doses didn't result in any specific symptoms or death. There were no significant changes in the rat's weight and food consumption. Further, ophthalmic examination, urinalysis, hematological, serum biochemistry test and organ weight revealed no significant differences. Conclusions : The no-observed-adverse-effect level(NOAEL) of LMK02 for male and female Sprague-Dawley rats was determined as 2,000mg/kg/day and the target organ wasn't confirmed. Because no significant adverse effects were observed, the target organ could not be determined.

Quality control of tablet form of LMK02 was established by estimating indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02 was investigated in 6 weeks old specific pathogen free (SPF) Sprageu-Dawley rats by oral administration. Each test group were consist of 5 male and 5 female and they received doses of 500, 1,000 and 2,000 mg/kg/day of test substance for 4 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of control group. Urinalysis : We observed increase of PRO(p<0.01), SG(p<0.01) in female rats of 1,000 mg/kg/day and 2,000 mg/kg/day(p<0.01). Also, we observed increase of pH and KET in female rats of 1,000 mg/kg/day(p<0.05) and of 2,000 mg/kg/day(p<0.01). WBC in female rats in 1,000 mg/kg/day and 2,000 mg/kg/day were on increase. Hematological test : We observed increase of MCV in male rats of 250 mg/kg/day. (p<0.05) Serum biochemistry test : We found increase of CHO in female rats of 2,000 mg/kg/day(p<0.05). During the experimental period, there were no animals dead or moribund. There were no treatment related changes of general symptom, food and water consumption, organ weight and autopsy According to the results of 4-week repeated dose range finding study, the highest dose was established as 1000 mg/kg for 13-week repeated dose toxicity study and we determined to put 2 more groups by common ratio two.

Changes in Rice Yield and Soil Properties under Continued Application of Chemical Fertilizer for 50 Years in Paddy Soil (화학비료 50년 연용에 따른 벼수량과 논토양 특성 변화)

  • This study was conducted to investigate the effect of long-term continuous application of fertilizers for rice cultivation. Changes of physical and chemical properties of paddy soil and the rice yield by continuous application of NPK fertilizers, ammonium sulphate, and urea over fifty years, from 1954 to 2003, were investigated. The rice yield index of each treatment were 100 of NPK plots, 84 of ammonium sulphate plots, 81 of urea plots, and 62 of no fertilizer plots. The variance of yield was large according to the quality of irrigation water. Nutrient uptakes by rice plants in ammonium sulphate and urea plots were significantly smaller than those in NPK plots; 86 and 75% in T-N, 79 and 82% in $P_2O_5$, 64 and 58% in $K_2O$, and 94 and 90% in $SiO_2$, respectively. Bulk density of soil in NPK plots significantly decreased compared to those in no fertilizer, ammonium sulphate, and urea plots, whereas CEC in NPK plots increased compared to other plots. Soil pHs of all plots were higher than that before experiment which was 5.2; 6.0 in no fertilizer, 5.9 in urea and NPK, and 5.4 in ammonium sulphate plots. The available phosphate in soil increased by $2.5mg\;kg^{-1}\;yr^{-1}$ when $70kg\;ha^{-1}$ of P fertilizer applied for rice cultivation, and decreased by $1.8mg\;kg^{-1}\;yr^{-1}$ when no P fertilizer applied.

Quality characteristics and physiological activities of mulberry (Morus alba) vinegar (오디 식초의 품질 특성 및 생리활성)

  • This study aimed to develop high value-added mulberry (Morus alba) vinegar by fermenting mulberry with yeast and acetic acid bacteria, for using it in various foods. To select the optimal strain for mulberry fermentation, different strains were tested and Saccharomyces cerevisiae SRCM101756 and Acetobacter pasteurianus SRCM102419, exhibiting excellent alcohol and acetic acid production ability during mulberry fermentation, were selected for fermentation. Mulberry vinegar was prepared using mulberry wine and the selected acetic acid bacteria, and the physicochemical properties and physiological effects were measured. The pH was 2.98 and total acidity was 4.70% by day 9 of fermentation, establishing the possibility of developing them into vinegars for industrial use. The angiotensin-glucosidase inhibition activity of mulberry vinegar increased from 13.22% to 19.19% in the 100-fold dilution, and from 42.35% to 46.11% in the 50-fold dilution, from before fermentation to after fermentation, respectively. The angiotensin-converting enzyme inhibition activity of mulberry vinegar was found to significantly increase from 44.82% before fermentation to 63.88% after fermentation in the 25-fold dilution. Moreover, a significant increase in pancreatic lipase inhibition activity after fermentation was observed. Thus, mulberry vinegar can be used as a functional material in vinegar and other foods.

Residual Characteristic of Chlorpyrifos in Squash and Estimation of Its Residues Before Harvest (애호박 중 Chlorpyrifos의 잔류특성 및 수확전 잔류량 예측)

  • This study was carried out to survey the residual characteristic of chlorpyrifos and estimate their residues in squash before harvest. The pesticide was sprayed onto the crop at the recommended and its double rates 10 days before the prearranged harvest and sampling was done at 0, 2, 3, 5, 6, 7 and 10 days after spraying. The amounts of the chlorpyrifos residue in the crop was analyzed by chromatographic method. Limit of detection (LOD) of chlorpyrifos was 0.005 mg/kg and its recovery ranged from 95.21 to 102.69%. The initial concentration of chlorpyrifos sprayed with recommended dose exceeded its MRL of 0.1 mg/kg but its concentration was less than its MRL 10 days after application. However its concentration in case of the double dose was over its MRL both immediately and 10 days after application. Biological half-lives of chlorpyrifos sprayed onto squash was 2.5 and 2.9 days at the recommended and double doses, respectively. Ten days later, the residual concentration of chlorpyrifos in squash was decreased substantially. The concentration of chlorpyrifos was estimated in squash at the given day using its regression equations. The estimated concentration of chlorpyrifos in case of application with recommended dose was below its MRL at 10 days after application but its concentration in case of application with double dose was over its MRL at 10 days of the prearranged harvest. The rate of the estimated daily intake (EDI) of chlorpyrifos to its acceptable daily intake (ADI) was 282% right after application but it decreased to less than 18% at 10 days of the prearranged harvest.

Changes of Chemical Components during Fermentation of Pear Wine (배술 발효 과정 중 화학 성분의 변화)

  • We used pears to manufacture wine, and analyzed changes in pH, acidity and ethanol and sugar content during fermentation. Pear wine with added ginger (to improve quality) did not differ from ginger-free wine in pH or acidity level. The ethanol content of pear wine was the highest (13.0%, v/v) inpear wine with 0.1% (w/v) added ginger compared to pear wine with no ginger, and sensory tests examining taste and color yielded the highest scores for pear wine with 0.2% (w/v) ginger. To assess storage stability, pear wine was treated for 30 minutes at $55^{\circ}C$, $60^{\circ}C$, $65^{\circ}C$, or $70^{\circ}C$. Unheated pear wine showed rapid changes in pH and acidity level after 30 days of storage, whereas pear wine treated for 30 minutes at $60^{\circ}C$ did not show such changes. Total organic acid levels in pear wine increased by 0.71% and 0.89% (v/v), respectively. The free sugar level in pear wine decreased from 12.05% to 3.13% (w/v). Turning to phenolic compounds, caffeic acid, catechin, and epicatechin contents in pears were 1.64, 1.40, and 0.23 mg/100mL, respectively, with diverse compositions. Caffeic acid levels in pear wine decreased sharply to 0.12 mg/100 mL upon fermentation, whereas free catechin inpear wine increased to 1.16 mg/100 mL compared with 0.28 mg/100 mL in pears. Free arbutin increased from 8.34 mg/100 mL in pears to 10.39 mg/100 mL in pear wine. The free amino acid content of pear wine was 118.5 g/100 mL, but the levels of serine, alanine, glutamic acid, and aspartic acid decreased sharply upon fermentation, with corresponding increases in tyrosine, GABA, lysine, and arginine.

Effects of the Soil Properties and N, P, K Application on the Contents of Inorganic Constituents in the Rhizoma and Leaf of Rehmannia glutinosa (토양이화학성(土壤理化學性) 및 삼요소(三要素) 시용량(施用量)이 지황(地黃) (Rehmannia glutinosa) 근경(根莖) 및 엽중(葉中)의 무기성분함량(無機成分含量)에 미치는 영향(影響))

  • The effects of soil properties and N, P and K application rates on the contents of inorganic constituents in the rhizomas of Ji-whang were investigated to know the condition for producing the better quality rhizoma of Ji-whang. Most of soil texture for Ji-whang cultivating belongs to sandy clay loam and sandy loam soils. The nitrogen contents in the rhizomas of Ji-whang was negatively correlated with the contents of organic matter and total nitrogen in surface soils, whereas positively correlated with the sand contents in subsoils. The calcium contents in the rhizomas of Ji-whang were negatively correlated with the sand contents, however, positively correlated with the contents of clay and organic matter in soils. The iron contents was positively correlated with the contents of clay, available phosphorus and exchangeable potassium in surface soils. Much more contents of the inorganic constituents in the leaf of Ji-whang was generally detected than that in the rhizoma of Ji-whang. The contents of iron and copper in the leaf was determined to be more than that in the rhizomas by ten times. In the results of field experiments, it is apparent that the nitrogen contents in the rhizoma and leaf increased with increasing N application rate more than 20kg/10a and 10kg/10a, respectively.
