The metabolic disturbance and nephrotoxicity induced by sodium dichromate (20 mg/kg, SC) have been diminished by the administration of Korean red ginseng extract (100 mg/kg, PO). Red ginseng has a powerful potency on the blood urea nitrogen (BUN) increment shown in the early 2h after dichromate intoxication. It normalized the dichromate induced hepatic glycogenolysis. The effect of red ginseng on dichromate induced nephrotoxicity was investigated by hematological analysis, and urinalysis. Ginseng treatment significantly reduced the increases in the urinary excretion of protein and glucose. These effects were dose dependent. Ginseng protected the accumulation of BUN and cretonne in the blood, caused by dichromate intoxication. Unlike CaEDTA, ginseng did not change the urinary excretion chromium. And it could not convert htxavalent chromium to trivalent chromium. These results suggest that ginseng treatment is effective in decreasing the metabolic disturbance, one of the earliest signs of dichromate toxicity, resulting in the protective effect of dichromate induced renal damage.

Background: Helicobacter pylori increases reactive oxygen species (ROS) and induces oxidative DNA damage and apoptosis in gastric epithelial cells. DNA damage activates DNA damage response (DDR) which includes ataxia-telangiectasia-mutated (ATM) activation. ATM increases alternative reading frame (ARF) but decreases mouse double minute 2 (Mdm2). Because p53 interacts with Mdm2, H. pylori-induced loss of Mdm2 stabilizes p53 and induces apoptosis. Previous study showed that Korean Red Ginseng extract (KRG) reduces ROS and prevents cell death in H. pylori-infected gastric epithelial cells. Methods: We determined whether KRG inhibits apoptosis by suppressing DDRs and apoptotic indices in H. pylori-infected gastric epithelial AGS cells. The infected cells were treated with or without KRG or an ATM kinase inhibitor KU-55933. ROS levels, apoptotic indices (cell death, DNA fragmentation, Bax/Bcl-2 ratio, caspase-3 activity) and DDRs (activation and levels of ATM, checkpoint kinase 2, Mdm2, ARF, and p53) were determined. Results: H. pylori induced apoptosis by increasing apoptotic indices and ROS levels. H. pylori activated DDRs (increased p-ATM, p-checkpoint kinase 2, ARF, p-p53, and p53, but decreased Mdm2) in gastric epithelial cells. KRG reduced ROS and inhibited increase in apoptotic indices and DDRs in H. pylori-infected gastric epithelial cells. KU-55933 suppressed DDRs and apoptosis in H. pylori-infected gastric epithelial cells, similar to KRG. Conclusion: KRG suppressed ATM-mediated DDRs and apoptosis by reducing ROS in H. pylori-infected gastric epithelial cells. Supplementation with KRG may prevent the oxidative stress-mediated gastric impairment associated with H. pylori infection.
It has been well known that extended exposure to reactive oxygens causes severe damage to susceptible biomolecules. In this study, the effects of flavonoids including trifolin and kaempferol from Ginseng leaves on singlet oxygen induced photohemolysis of erythrocytes and free radical scavenging activities were investigated. Each flavonoid aglycone (5-50$\mu$M) such as kaempferol, quercetin or baicalein exhibited a high protective effect against the photohemolysis. They protected the cells by scavenging $^1O_2$ and free radicals Although the free radical scavenging activities of the flavonoid glycosides were not much lower than those of their corresponding aglycones, their insolubility into lipid bilayers of membrane made them less effective in preventing the photohemolysis induced by $^1O_2$. The $^1O_2$ and free radical scavenging activities of flavonoids were estimated by the decomposition of the flavonoid by $^1O_2$ and the bleaching of free radicals by the flavonoid, respectively. The solubilization of the flavonoid into micells or erythrocytes was deduced from spectrophotometric and microscopic observations. The cooperation of L-ascorbic acid and a flavonoid, and a possible involvement of lipoxygenase or cyclooxygenase in the photohemolysis mechanism were discussed.
Objectives : This study was carried out to examine protective effect of wild ginseng extract on HepG2 human hepatoma cell line against tert-Butyl hydroperoxide (t-BHP)-induced oxidative damage. Methods : To evaluate protective effect of wild ginseng extract against t-BHP induced cytotoxicity, LDH level and activity of glutathione peroxidase and reductase were measured. Gene expression was also measured using DNA microarray. Results : Wild ginseng extract showed a significant protective effect against t-BHP-induced cytotoxicity in HepG2 cell line. It is not, however, related with the activities of glutathione peroxidase and glutathione reductase. Analysis of gene expression using DNA chip, demonstrated that 28 genes were up-regulated in t-BHP only group. Five genes - selenoprotein P, glutathione peroxidase 3, sirtuin 2, peroxiredoxin 2, serfiredoxin 1 homolog - may be related with the protective effect of wild ginseng extract. Conclusions : Based on the results, a protective effect of wild ginseng extract against t-BHP-induced oxidative damage in HepG2 cell line is not associated with the activities of glutathione peroxidase and glutathione reductase, but with the expression of selenoprotein P, glutathione peroxidase 3, sirtuin 2, peroxiredoxin 2, and serfiredoxin 1 homolog.
Lately, the Korean ginseng was confronted with the sale crisis in world markets from the various circumstance. One of important difficulties is overspreading of Korean ginseng imitative in foreign markets. In Korean markets also, chinese ginseng was imported illegally and manufactured as Korean ginseng, which was resulted in the damage of Korean ginseng brand. The most essential problem is misunderstanding of Korean ginseng's information in the foreigner. In this paper, we developed a genuine guarantee system to identify the true Korean ginseng through internet and geographic cording system of Korean ginseng products. This system can be applied to the enforcement of Korean ginseng sale to international markets.
Objectives: The study investigated the efficacy of oral Korean red-ginseng (RG) on sCRP response mediated by eccentric contraction-induced muscle damage and discussed its mechanism. Methods: Nineteen healthy young subjects (aged 24.4$\pm$2.07 yr) volunteered for this double blind test. They were classified into either the RG group (N=10) or placebo (P) group (N=9) during the 10-d experimental protocol. Blood samples were collected on T1 (baseline), T1 (1h), T2 (1d), T3 (2d) and T4 (3d) after eccentric contraction-induced injury on the seventh day. Statistical analyses were conducted using nonparametric methods (p<0.05). Results: The RG group increased sCRP more than P group and didn't increase soreness peak at T2, not significant. There were no correlations between soreness intensity and sCRP. Oral RG had little efficacy for reducing sCRP at the muscle damage-mediated acute phase; rather, it increased because of its proinflammatory cytokine production. Conclusions: Oral RG could stimulate proinflammatory cytokine production, and occasionally bal-pyo-beob could be helpful for the efficient recovery of muscle injury.
This study investigated the antioxidant effects of ginseng coffee in L6 muscle cells. Ginseng coffee was prepared by coating and digesting coffee beans with ginseng concentrate. The ginseng coffee water extract potently protected against hydrogen peroxide-induced L6 cell death and adenosine triphosphate reduction in a dose-dependent manner; in fact, these cytoprotective effects were significantly greater than those of normal coffee. However, ginseng coffee did not exhibit significant radical scavenging or catalase-like activity. These results suggest that ginseng coffee might act as a cytoprotective agent in muscles, but that the protective effects are not due to a direct radical-reduction property but rather to another intracellular signaling factor.

Background: High temperature damage in ginseng is influenced by shading materials related not only to temperature, but also to light intensity and light quality. To address, this green-colored khaki shading sheet is widely used. As they are recently, developed, there is limited research information about their attributes and use. Methods and Results: The four-layered shading net (FLSN), blue-colored shading sheet (BCSS), aluminum-coated shading board (ACSB), and green-colored shading sheet (GCSS) were installed in the wooden A type of sun-block facilities. Two layered black, shading net was additionally used to cover the facilities since the beginning of June. The average temperature at the facility where different shades were tested was in the order of BCSS ($28.9^{\circ}C$) > FLSN ($27.7^{\circ}C$) > GCSS ($27.6^{\circ}C$) > ACSB ($27.1^{\circ}C$). However, high temperature injury rates were in order: FLSN > ACSB > GCSS > BCSS. Root weight vaired and was in the order: ACSB > GCSS > BCSS > FLSN. Conclusions: High temperature damage is possible not only because of temperature increase, but also due to various environmental light factors. Ginseng high temperature injury was minimal when BCSS or GCSS were used by difference of light quality. Although the root weight was higher in ACSB, it could be vulnerable to high temperature damage. Therefore, we propose using GCSS for ginseng shading.
The monoxenic culture of the fungivorous nematode, Aphelenchus avenae, was applied for the control of soil-borne ginseng pathogens such as Fusarium solani and Rhizoctonia solani. Fungivorous nematode populations were measured in a field to examine relationships between the nematode populations and suppression of ginseng root diseases. Inoculation of A. avenae (5000 nematodes per petri-dish) reduced the colonization of the Fusarium mycelium on root discs of ginseng and carrot by 80.0% and 60.5%, respectively. A. avenae also significantly reduced the occurrence of damping-off of ginseng by R. solani pathogenic to ginseng, and no plant damage by the nematode was noted. In a 3-year-old ginseng field infested with Cylindrocarpon destructans, plant missing caused by root rot positively correlated to the density of potato rot nematode, Ditylenchus destructor, but it was reduced with the population of A. avenae, suggesting that A. avenae might inhibit the occurrence of ginseng root rot.
