Changes in the Myocardial Antioxidant Enzyme System by Post-Ischemic Reperfusion During Corontory Artery Bypass Operations (관상동맥우회술시 심근허혈후 재관류에 의한 활성산소 방어효소계의 변화)

  • Oxygen free radicals and their metabolites have been implicated as possible causes of reperrusion injury In animal models. Their role in the clinical setting is still controversial. The aim of this study was to evaluate the degree of tissue damage, oxidative stress. and changes in the antioxidant enzyme system in patients undergoing cor nary artery bypass graft operations(CABG) with myocardial protection by cold blood cardioplegia. In patients undergoing CABG(n:10). the levels of lactate dehydrogenate(LDH), creatine phosphokinase MB fraction(CK-MB), and malondialdehyde(M DA) were measured In the coronary sinus effluent before aortic cross clamping and 20 minutes after reperfusion. At the same time, the myocardial tissue activities of superoxide dismutase(SOD). catalase(CAT), glutathione peroxiddse(GSHPX), glutathione reductase (GSSGRd), and glucose 6-phosphate dehydrogenate(GfPDH ) were determined in the right atrial auricle excised before aortic cross clamping and in the left atrial auricle excised 20 minutes after reperfuslon. The levels of increased significantly after reperrusion(p< U.05). There were no significant changes in CAT and CfPDH levels. Western blot analysis was performed to study the induction of antioxidant enzyme and demonstrated increased amount of Cu,Zn-SOD.

Octyl Gallate Inhibits ATP-induced Intracellular Calcium Increase in PC12 Cells by Inhibiting Multiple Pathways

  • Phenolic compounds affect intracellular free $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) signaling. The study examined whether the simple phenolic compound octyl gallate affects ATP-induced $Ca^{2+}$ signaling in PC12 cells using fura-2-based digital $Ca^{2+}$ imaging and whole-cell patch clamping. Treatment with ATP ($100\;{\mu}M$) for 90 s induced increases in $[Ca^{2+}]_i$ in PC12 cells. Pretreatment with octyl gallate (100 nM to $20\;{\mu}M$) for 10 min inhibited the ATP-induced $[Ca^{2+}]_i$ response in a concentration-dependent manner ($IC_{50}=2.84\;{\mu}M$). Treatment with octyl gallate ($3\;{\mu}M$) for 10 min significantly inhibited the ATP-induced response following the removal of extracellular $Ca^{2+}$ with nominally $Ca^{2+}$-free HEPES HBSS or depletion of intracellular $Ca^{2+}$ stores with thapsigargin ($1\;{\mu}M$). Treatment for 10 min with the L-type $Ca^{2+}$ channel antagonist nimodipine ($1\;{\mu}M$) significantly inhibited the ATP-induced $[Ca^{2+}]_i$ increase, and treatment with octyl gallate further inhibited the ATP-induced response. Treatment with octyl gallate significantly inhibited the $[Ca^{2+}]_i$ increase induced by 50 mM KCI. Pretreatment with protein kinase C inhibitors staurosporin (100 nM) and GF109203X (300 nM), or the tyrosine kinase inhibitor genistein ($50\;{\mu}M$) did not significantly affect the inhibitory effects of octyl gallate on the ATP-induced response. Treatment with octyl gallate markedly inhibited the ATP-induced currents. Therefore, we conclude that octyl gallate inhibits ATP-induced $[Ca^{2+}]_i$ increase in PC12 cells by inhibiting both non-selective P2X receptor-mediated influx of $Ca^{2+}$ from extracellular space and P2Y receptor-induced release of $Ca^{2+}$ from intracellular stores in protein kinase-independent manner. In addition, octyl gallate inhibits the ATP-induced $Ca^{2+}$ responses by inhibiting the secondary activation of voltage-gated $Ca^{2+}$ channels.

A Study on the Crease Recovery Behavior of Core-spun Yarn Woven Fabrics (코어방적사직물의 구김회복거동에 관한 연구)

  • In this paper, the fabric specimen undergoes repeated laundering under given condition. After this cyclic laundering was applied, the crease recoveries of the specimen were measured using shirley crease revovery tester in order to evaluate the effect of factors at given condition during crease deformation. 5 samples of grey plain cloth were desized, alkali-scoured, bleached, whased with water, and air-dried. All tests were made on samples preconditioned to $65\%\;RH\;and\;20^{\circ}C$. The experimental results were analysed statistically to relate crease recoveries and the properties of smaples, recovery periods (time) of crease. Furthermore, the crease recoveries of core-spun yarn woven fabrics were discussed in comparison with those values for $100\%$ combed cotton yarn woven fabric and $65\%$ polyester $35\%$ carded cotton blended yarn woven fabric. The results obtained are as follows; 1. Regardless of materials, remarkable decrease are observed in crease recoveries about 1-5 cycles of the repeated laundering, but slack decrease are observed in crease recoveries after 5 cycle of the re-peated laundering. 2. Crease recoveries ($\alpha$) of core-spun yarn woven fabrics are relate to recovery periods (t) of crease as follows; log$\alpha$=0.01415 log t+2.1168 ($r^2=0.94$) 3. Core-spun yarn woven fabrics were superior to $100\%$ combed cotton yarn woven fabrics and $65\%$ polyester $35\%$ carded cotton blended yarn woven fabric in crease recoveries. 4. Crease recoveries ($\alpha$) of core-spun yarn woven fabrics are relate to cover factor (CF), thickness (T) at pressure 0.5 $gf/cm^2$, weight (W) as follows; log$\alpha$=-0.3482 log CF-0.4924 log T-0.4727 W+2.4243 ($r^2=0.88$) 5. Crease recoveries ($\alpha$) of core-spun yarn woven fabrics are relate to 2HB/B, 2HB/W, $\sqrt[3]{B/W}$, WC/T which are concerning to formation of weared clothes and bending Iran formation behavior as follows: log $\alpha$=0.0091 2HB/B+0.4667 2HB/W+0.0185 $\sqrt[3]{B/W}$+0.0114 WC/T+1.8433 ($r^2=0.86$)

  • Microwave digestion and solid-state extraction were studied for determination of trace aluminum{Al(III)} in human urine samples. A mixed acid of nitric acid and hydrogen peroxide was added to urine samples, organic materials were destructed in a home microwave oven and dried in a drying oven. The dried residues were dissolved in a sulfuric acid solution. The solution was eluted through a XAD-4 resin column adsorbed with 8-hydroxyquinoline(Oxine, HQ). Al(III)-8-hydroxyquinolinate complex was formed in the column and eluted with 0.5 M nitric acid solution. The Al(III) eluted was determined by graphite atomic absorption spectrophotometry. Various experimental conditions of followings were investigated for the optimization : the type of acid to dissolve the residues, the amount of HQ adsorbed on the resin, the pH of sample solutions, the type and concentration of acid to elute the complex from column and so on. The contents of Al(III) in real samples were determinated by a calibration curve method. The recovery in standard spiked samples was 94~101% and the detection limit of this procedure was 0.05 ng/mL.

Relationship between Heavy Metal Concentrations in the Soil with the Blood and Urine of Residents around Abandoned Metal Mines (폐금속 광산지역 토양 중 중금속 농도와 주민의 혈액 및 요중 중금속 농도와의 관련성)

  • Objectives: This study was conducted in order to examine the relationship between heavy metal concentrations in the soil and the level of heavy metals in the blood or urine of 216 local residents living near abandoned metal mines. Methods: Residents around abandoned metal mines were interviewed about their dietary habits, including seafood consumption, medical history, cigarette smoking, and drug history. Metal concentrations in the soil were determined by atomic absorption spectrophotometer (AA-7000, Shimadzu, Japan). Lead (Pb) and cadmium (Cd) contents in the blood or urine were analyzed by GF-AAS (AA-6800, Shimadzu). Mercury (Hg) contents in the blood were determined by means of a mercury analyzer (SP-3DS, NIC). Arsenic (As) content in the soil and urine were measured by a HG-AAS (hydride vapor generation-atomic absorption spectrophotometer). Results: The heavy metal concentrations in the soil showed a log normal distribution and the geometric means of the four villages were 8.61 mg/kg for Pb, 0.19 mg/kg for Cd, 1.81 mg/kg for As and 0.035 mg/kg for Hg. The heavy metal levels of the 216 local residents showed a regular distribution for Pb, Cd, Hg in the blood and As in the urine. The arithmetic means were 3.37 ${\mu}g$/dl for Pb, 3.07 ${\mu}g$/l for Cd and 2.32 ${\mu}g$/l for Hg, 10.41 ${\mu}g$/l for As, respectively. Conclusions: As a result of multi-variate analysis for the affecting factors on the bodily heavy metal concentrations, gender and concentration in the soil (each, p<0.01) for blood lead levels; gender and smoking status (each, p<0.01) for blood cadmium levels; gender (p<0.01) for urine arsenic levels; gender, age and concentration in the soil (p<0.01) for blood mercury levels were shown to be the affecting factors.

황토의 인흘착 성능평가

  • 우리나라의 대부분 하천과 인공댐의 경우 조류발생에 기여하는 영양염은 질소, 인 그리고 규소 등인데 특히 인이 생산제한 인자로 작용하고 있다. 따라서 인을 적절히 제거할 경우에는 조류 발생에 의한 수질의 악화와 수이용의 저해요인을 배제할 수 있어 이에 대한 연구가 많이 진행되어 왔다. 지금까지와 연구는 인의 화학적 응집 침전법, 생물학적 처리법에 대하여 수행되어 왔으나 설비자금, 운영비, 운전기술, 슬러지 생성 그리고 제거효율 등에서 만족스런 결과가 도출되지 못하여 현장 적용을 하지 못하고 있는 실정이다. 이러한 현실을 감안하여 우리의 자연 환경에서 쉽게 자할 수 있는 황토를 모재로하여 Al3-과 Fe3-, Ca2-을 적절히 배합하여, 주로 인을 선택적으로 제거하 고자 개발된 황토의 흡착능력 그리고 흡착 메카니즘에 대한 기본적인 연구를 하고자 (1) 등온흡착실험을 통하여 흡착용량을 평가하고 (2) 흡착제거속도를 평 가하였고 (3)파과시간 및 흡착특성을 파악하기 위해 column 흡착실험을 하였다 또한 (4) 황토내의 Al3-, Fe3-과 Ca2- 등이 인의 화학적 흡착에 기여하는 정도를 파악하여 흡착메카니즘을 규명하고자 하였다. 먼저 흡착용량실험을 위하여 PO3-4-P 농도 3ppm의 용액 200mf에 황토 0.2g, 0.5g, 1.0g, 2.0g을 각각 투여한 후 충분한 흡착평형이 일어나게 24시간 동 안 130rpm으로 $25^{\circ}C$ 등온반웅조에서 저어주어 흡착평형에 도달하면 상등액을 GF/C Filter로 여과한 후, 여액에 대해 PO3-4-P의 농도를 Ascrobic he건법으로 측정한 결파, Freundlich 등온흡착식에 의하면 K값은 17.34와 16.28이었으며 1/n 값은 1.32와 1.42로 인흡착 성능이 뛰어난 것으로 평가되었다. 둘째, 흡착속도 실험은 PO3-4-P 농도 1.5ppm의 용액 2f에 259의 황토를 투여하고 충분한 혼합이 일어날 수 있도록 170rpm으로 교반하면서 시간별 용액 의 농도 변화를 측정한 결과, 0.45mg/g/m교의 속도로 15분만에 94.3%의 인 제거 효율을 보였다. 셋째, 직경 12mm의 glass column에 황토를 209 채우고 1.5ppm의 PO34P 용액을 2.Sne11in의 유량으로 통수 시킨 후, 시간에 따른 농도 변화를 측정한 결과, 원수 농도의 50%에 해당하는 파과점까지 약 70시간 만에 도달하였다. 넷째, Al3-, Fe3-과 Ca2- 등이 화학적 흡착에 기여하는 정도를 파악하기 위 하여 황토 2g에 대하여 Hieltijes and Lijklema 방법에 의해 Adsorbed-p, Nonapatite inorganic-P(NAI-P), Apatite-p, Organic-P로 구분하여 분석하고, 총인(Total Phosphorus)을 Standard Methods에 따라 Persulfate digestion후 0.45 m membrane 여지 여과하여 여액에 대해 PO3-4-P의 농도를 Ascorbic Acid 법으로 측정한 결과, NAI-P가 가장 큰 비율을 차지하였고, 부원료로 첨가된 금속 양이온 중 Fe3-이온이 흡착에 기여하는 정도가 가장 큰 것으로 평가되었다.

Design of a Small-Area Finite-Field Multiplier with only Latches (래치구조의 저면적 유한체 승산기 설계)

  • An optimized finite-field multiplier is proposed for encryption and error correction devices. It is based on a modified Linear Feedback Shift Register (LFSR) which has lower power consumption and smaller area than prior LFSR-based finite-field multipliers. The proposed finite field multiplier for GF(2n) multiplies two n-bit polynomials using polynomial basis to produce $z(x)=a(x)^*b(x)$ mod p(x), where p(x) is a irreducible polynomial for the Galois Field. The LFSR based on a serial multiplication structure has less complex circuits than array structures and hybrid structures. It is efficient to use the LFSR structure for systems with limited area and power consumption. The prior finite-field multipliers need 3${\cdot}$m flip-flops for multiplication of m-bit polynomials. Consequently, they need 6${\cdot}$m latches because one flip-flop consists of two latches. The proposed finite-field multiplier requires only 4${\cdot}$m latches for m-bit multiplication, which results in 1/3 smaller area than the prior finite-field multipliers. As a result, it can be used effectively in encryption and error correction devices with low-power consumption and small area.

A Scalable Hardware Implementation of Modular Inverse (모듈러 역원 연산의 확장 가능형 하드웨어 구현)

  • This paper describes a method for scalable hardware implementation of modular inversion. The proposed scalable architecture has a one-dimensional array of processing elements (PEs) that perform arithmetic operations in 32-bit word, and its performance and hardware size can be adjusted depending on the number of PEs used. The hardware operation of the scalable processor for modular inversion was verified by implementing it on Spartan-6 FPGA device. As a result of logic synthesis with a 180-nm CMOS standard cells, the operating frequency was estimated to be in the range of 167 to 131 MHz and the gate counts were in the range of 60,000 to 91,000 gate equivalents when the number of PEs was in the range of 1 to 10. When calculating 256-bit modular inverse, the average performance was 18.7 to 118.2 Mbps, depending on the number of PEs in the range of 1 to 10. Since our scalable architecture for computing modular inversion in GF(p) has the trade-off relationship between performance and hardware complexity depending on the number of PEs used, it can be used to efficiently implement modular inversion processor optimized for performance and hardware complexity required by applications.

Epidermal Growth Factor Induces Vasoconstriction Through the Phosphatidylinositol 3-Kinase-Mediated Mitogen-Activated Protein Kinase Pathway in Hypertensive Rats

  • We investigated whether increased contractile responsiveness to epidermal growth factor (EGF) is associated with altered activation of mitogen-activated protein kinase (MAPK) in the aortic smooth muscle of deoxycorticosterone acetate (DOCA)-salt hypertensive rats. EGF induced contraction and MAPK activity in aortic smooth muscle strips, which were significantly increased in tissues from the DOCA-salt hypertensive rats compared with those from sham-operated rats. AG1478, PD98059, and LY294002, inhibitors of EGF receptor (EGFR) tyrosine kinase, MAPK/extracellular signal-regulated kinase (ERK) kinase, and phosphatidylinositol 3-kinase (PI3K), respectively, inhibited the contraction and the activity of ERK1/2 that were elevated by EGF. Y27632 and GF109203X, inhibitors of Rho kinase and protein kinase C, respectively, attenuated EGF-induced contraction, with no diminution of ERK1/2 activity. Although EGF also elevated the activity of EGFR tyrosine kinase in both sham-operated and DOCA-salt hypertensive rats, the expression and the magnitude of activation did not differ between strips. These results strongly suggest that EGF induces contraction by the activation of ERK1/2, which is regulated by the PI3K pathway in the aortic smooth muscle of DOCA-salt hypertensive rats.

Research on the Effect of Gardeniae Fructus on Regulatory T Cell Stimulation (조절 T세포에 미치는 치자(梔子)의 효과)

  • Objectives : Regulatory T cells can reduce inflammation and allergic reactions through their inhibitory functions. Gardeniae Fructus(GF) is a Heat-clearing herb used in traditional Korean medicine, and a wide range of studies on its antiinflammatory effects are being carried out. The authors investigated the effect that Gardeniae Fructus has on regulatory T cells. Methods : The authors screened 14 herbs for their effects on regulatory T cells. 100mg of each herb were separately dissolved in 1ml of sterile saline and the supernatant was harvested after 10 minutes of centrifuge at 15,000 rpm. The supernatant was filtered through a 0.2 ${\mu}m$ syringe filter, and the resulting stock was refrigerated at $4^{\circ}C$. The stock was diluted before testing and used at a final concentration of $0.01{\mu}g/ml$. CD4+CD25+ T cells from healthy BALB/c spleens were used as natural regulatory T cells (nTreg), and CD4+CD25- T cells were used as reactive T cells. CD4+CD25+ and CD4+CD25- T cells were activated with anti-CD3e ($10{\mu}g/m{\ell}$)/anti-CD28 ($1{\mu}g/m{\ell}$) and cultured. IL-10 from supernatant of the culture medium was measured by IL-10 cytokine ELISA. The percentages, cell numbers, phenotype and function of CD4+CD25+ Treg cells were determined by flow cytometry. Results : Gardeniae Fructus was shown to be the most potent herb among the 14 herbs tested for suppressing CD4+CD25- reactive T cell proliferation by stimulating CD4+CD25+ natural regulatory T cells. Gardeniae Fructus induces IL-10 secretion increase by stimulating CD4+CD25+ natural regulatory T cells, and indirectly suppresses CD4+CD25- reactive T cell proliferation through increasing CD25 (IL-2 receptor $\alpha$) expression and thus promoting bonding with IL-2. Gardeniae Fructus did not directly affect CD4+CD25- reactive T cell proliferation. Conclusions : Gardeniae Fructus suppressed reactive T cell proliferation through inducing increases in IL-10 secretion and CD25 (IL-2 receptor $\alpha$) expression.
