The genetic variabilities of second chromosomes of Drosophila melanogaster concealed in On-yang natural Population have been analyzed by the Cy/Pm method and an allelism test during two years(1993-1994). The mean frequencies of deleterious(lethal and semilethal) genes in On-yang natural population were estimated to be 23.97% in 1993 and 27.15% in 1994, respectively. The allelism rates between lethal genes in the population were 0.654%(1993) and 1.429%(1994). The mean values of elimination by frequencies of deleterious genes and allelism rates were 0.0004(1993) and 0.0010(1994), respectively. The frequencies of phenotypic sterility of males in 1994 were estimated to be 1.95%, and thoses of genotypic sterility of females and males were estimated to be 1.54% and 2.31%, respectively.

Bud sport (bud mutation) is a valuable source for existing new genotypes in mulberry (Morus spp.) as well as critical materials for studying the molecular mechanisms underlying essential traits. Thus, identification, collection, characterization, and conservation of such natural variants are prerequisites for enhancing the mulberry genetic resource in the germplasm. In this context, we identified and characterized three bud sports (VBS-1, VBS-2, and VBS-3) of a popular mulberry cultivar, Victory-1 (V-1). These bud sports are morphologically, anatomically, and genetically more distinct from their mother plant, Victory-1. Moreover, these bud sports display lower growth and yield potential. Furthermore, these showed remarkably lower 2C DNA contents of 0.74 pg (VBS-1), 0.78 pg (VBS-2), and 0.76 pg (VBS-3), when compared to their mother plant V-1 (2C = 0.81 pg). On the other hand, molecular characterization between the bud sports and their mother plant revealed the existence of genetic variation due to the natural bud mutation that occurred in the mulberry cultivar Victory-1.

Black sea bream, Acanthopagrus schlegelii, is a commercially important fish in Korea. As a preliminary investigation into the effect of hatchery rearing for stock enhancement, we examined genetic diversity between wild and farmed black sea bream populations from Jeju using six microsatellite markers. High levels of polymorphism were observed between the two populations. A total of 87 different alleles were found at the loci, with some alleles being unique. Allelic variability ranged from 8 to 22 in the wild population and from 7 to 17 in the farmed one. Average observed and expected heterozygosities were estimated at 0.87 and 0.88 in the wild sample. The corresponding estimates were 0.83 and 0.86 in the farmed sample. Although a considerable loss of rare alleles was observed in the farmed sample, no statistically significant reductions were found in heterozygosity or allelic diversity in the farmed sample, compared with the wild one. Significant genetic heterogeneity was found between the wild and farmed populations. These results suggest that more intensive breeding practices for stock enhancement may have resulted in a further decrease of genetic diversity. Thus, it is necessary to monitor genetic variation in bloodstock, progeny, and target populations and control inbreeding in a commercial breeding program for conservation. This information may be useful for fisheries management and the aquaculture industry.

The production of spring wheat, the major crop in Mongolia, is accounting for 98% of the cultivated area. Collection, conservation and utilization of wheat germplasm resources play an important role in wheat breeding and production in Mongolia. Understanding genetic variability in the existing genebank accessions is important for collection and conservation of wheat germplasms. To determine the genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with 3 to 5 alleles per locus and a mean genetic diversity value of 5.66. The average genetic diversity index was 0.68, with values ranging from 0.37 to 0.80. The 200 Mongolian wheat accessions were divided into two subgroups based on STRUCTURE, un-rooted NJ cluster and principal coordinate analyses. The results from this study will provide important information for future wheat germplasm conservation and improvement programs with Mongolian genebank.

Japanese quail is still used as a model for poultry research because of their usefulness as laying, meat, and laboratory animals. Microsatellite markers are the most widely used molecular markers, due to their relative ease of scoring and high levels of polymorphism. The objective of the research was to determine genetic diversity and population genetic structures of selected Japanese quail lines (high body weight 1 [HBW1], HBW2, low body weight [LBW], and layer [L]) throughout 15th generations and an unselected control (C). A total of 69 individuals from five quail lines were genotyped by fifteen microsatellite markers. When analyzed profiles of the markers the observed ($H_o$) and expected ($H_e$) heterozygosity ranged from 0.04 (GUJ0027) to 0.64 (GUJ0087) and 0.21 (GUJ0027) to 0.84 (GUJ0037), respectively. Also, $H_o$ and $H_e$ were separated from 0.30 (L and LBW) to 0.33 (C and HBW2) and from 0.52 (HBW2) to 0.58 (L and LBW), respectively. The mean polymorphic information content (PIC) ranged from 0.46 (HBW2) to 0.52 (L). Approximately half of the markers were informative ($PIC{\geq}0.50$). Genetic distances were calculated from 0.09 (HBW1 and HBW2) to 0.33 (C and L). Phylogenetic dendrogram showed that the quail lines were clearly defined by the microsatellite markers used here. Bayesian model-based clustering supported the results from the phylogenetic tree. These results reflect that the set of studied markers can be used effectively to capture the magnitude of genetic variability in selected Japanese quail lines. Also, to identify markers and alleles which are specific to the divergence lines, further generations of selection are required.
“The Regional complex long-term program of restoration (reintroduction) of Primoryes ginseng population up to 2005” elaborated by Primorye governor administration, Regional Committee of Natural Resources and Russian Academy of Sciences operates in Russian Primorye. The Institute of Biology and Soil Science (IBSS) provides the scientific implementation of the program including the genetic analysis of extant ginseng populations, plant reproduction and off-spring identification. According to our investigations, the genetic resource of P. ginseng in Primorye is represented by three populations of wild-growing ginseng and a few pritate plantations. The results obtained by RAPD allowed concluding that the resource is dispersed among the wild and cultivated ginseng sub-populations in such a way that each of sub-populations studied has to be represented as a stock material to maintain species genetic variability. The allozyme analyses also showed that the small sub-populations of wild ginseng are characterized by unique genetic diversity and, therefore, they all need to be represented in reintroduction centers. Additionally the allozyme analysis discovered that the Blue Mountain and Khasan populations possess the most genetic diversity. So, at least one more reproductive ginseng unit has to be created besides two already existing reintroduction centers representing the Sikhote-Alin and the Blue Mountain populations.
The oily bittering Acheilognathus koreensis is a freshwater species that is endemic to Korea and is experiencing severe declines in natural populations as a result of habitat fragmentation and water pollution. For the conservation and restoration of this species, it is necessary to assess its genetic diversity at the population level. We developed 13 polymorphic microsatellite loci that were used to analyze the genetic diversity of two populations collected from the Kum River and the Tamjin River in Korea. All loci exhibited Mendelian inheritance patterns when examined in controlled crosses. Both populations revealed high levels of variability, with the number of alleles ranging from 3 to 20 and observed and expected heterozygosities ranging from 0.500 to 0.969 and from 0.529 to 0.938, respectively. None of the loci showed significant deviation from Hardy-Weinberg equilibrium, and one pair of loci showed significant linkage disequilibrium after Bonferroni correction. Pairwise $F_{ST}$ and genetic distance estimation showed significant differences between two populations. These results suggest that the microsatellites developed herein can be used to study the genetic diversity, population structure and conservation measure of A. koreensis.

AFLP marker의 유용성 을 알아보고자 재배콩과 야생콩을 대상으로 유전적 다양성과 유전분리 현상을 분석하였다. 공시 재료들의 polymorphism은 재배 콩과 야생 콩에서 각각 평 균 2 9%와 12.2%의 polymorphism을 보였으며, 재배 콩과 야생 콩에서 공히 유전적 다양성을 보인 DNA단편은 11개 primer 평균 24개를 나타내었다. Primer 조합별로도 polymorphism에 다양한 차이가 있었는데 평균 22.9%로 13.0-38.5%의 변이를 나타내었다. 재배 콩 간의 교잡후대(화엄풋콩 ${\times}$ PI417479) F$_2$집단에서 AFLP marker의 유전분리 양상을 분석한 결과 3 : 1의 분리 비를 따르는 것으로 판단되었다.
Random amplified polymorphic DNA (RAPD) markers were used to survey genetic variability among 34 Botrytis cinerea isolates from nine different host plants in Korea. For RAPD analysis, 115 arbitrary decamer primers were initially screened for polymorphic major DNA bands with 11 representative B. cinerea isolates. Eleven primers that initially detected polymorphisms were tested a second time with additional 23 isolates of B. cinerea as well as one isolate of Botrytis squamosa as an outgroup. The RAPD analyses revealed that all isolates except one showed different molecular phenotypes. Dendrograms obtained from dissimilarity matrices using the unweighted paired group method of arithmetic means (UPGMA) showed the 36.4% to 90.0% similarity among all B. cinerea isolates. The B. squamosa isolate showed the least similarity to all B. cinerea isolates. The cluster analyses indicated no correlation among all the characteristics examined including molecular phenotypes, host and geographic origins, year of isolation, or pathogenicity. The RAPD data suggest that a high level of genetic variation exists among Korean populations of B. cinerea and it seems to be caused by heterokaryosis among preexisting molecular phenotypes.
