Analysis of Genetic Variation in Botrytis cinerea Isolates Using Random Amplified Polymorphic DNA Markers

  • Random amplified polymorphic DNA (RAPD) markers were used to survey genetic variability among 34 Botrytis cinerea isolates from nine different host plants in Korea. For RAPD analysis, 115 arbitrary decamer primers were initially screened for polymorphic major DNA bands with 11 representative B. cinerea isolates. Eleven primers that initially detected polymorphisms were tested a second time with additional 23 isolates of B. cinerea as well as one isolate of Botrytis squamosa as an outgroup. The RAPD analyses revealed that all isolates except one showed different molecular phenotypes. Dendrograms obtained from dissimilarity matrices using the unweighted paired group method of arithmetic means (UPGMA) showed the 36.4% to 90.0% similarity among all B. cinerea isolates. The B. squamosa isolate showed the least similarity to all B. cinerea isolates. The cluster analyses indicated no correlation among all the characteristics examined including molecular phenotypes, host and geographic origins, year of isolation, or pathogenicity. The RAPD data suggest that a high level of genetic variation exists among Korean populations of B. cinerea and it seems to be caused by heterokaryosis among preexisting molecular phenotypes.

Adaptive User Profile for Information Retrieval from the Web

  • This paper proposes the information retrieval improvement for the Web using the structure and hyperlinks of HTML documents along with user profile. The method bases on the rationale that terms appearing in different structure of documents may have different significance in identifying the documents. The method partitions the occurrence of terms in a document collection into six classes according to the tags in which particular terms occurred (such as Title, H1-H6 and Anchor). We use genetic algorithm to determine class importance values and expand user query. We also use this value in similarity computation and update user profile. Then a genetic algorithm is used again to select some terms from user profile to expand the original query. Lastly, the search engine uses the expanded query for searching and the results of the search engine are scored by similarity values between each result and the user profile. Vector space model is used and the weighting schemes of traditional information retrieval were extended to include class importance values. The tested results show that precision is up to 81.5%.

Radiation Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus

  • To assess the effects of gamma-ray (Co-60) on radiation sensitivity and genetic similarity of the basidiospore and mycelium in oyster mushroom, Pleurotus ostreatus, the D$_{10}$ values and RAPD patterns were analysed. Three strains were isolated from basidiospores (PO-Bl, -B2, and - B3 from 2 kGy irradiation group) and five strains from mycelia (PO-Ml, -M2 from 1 key, PO-M3 from 2 kGy, and PO-M4 and -M5 from 2+1 key irradiation group). The D$_{10}$ values of basidiospore and mycelium of P. Preurotus ostreatus were 1,250 Gy and 500 Gy, respectively. The growth rates of the eight strains on the five media were various and the activities of extracellular chitinases of them were generally higher than those of the control. By the gamma-ray radiation, 22-25% of genetic similarities were changed in the basidiospore strains and 23-36% of them in the mycelium strains. From these results, it seems that the basidiospore could be more radio-resistant than the mycelium of P. ostreatus and that the genetic similarity of the mycelium of P ostrentus could be changed easier than that of the basidiospore by the gamma-ray radiation.ion.

Potential Risk of Genetically Modified Plants in Korean Ecosystem: a Proposal for Unintended Effects on Korean Wild Species

  • Introgression from genetically modified plants (GMPs) may be dependent on the genetic similarity to wild relative plants. In Korea, many wild plant species are botanically related to the cultivated plants that have a potential to be genetically transformed. The controversy for hazards of GMPs is continuing because the studies on gene flow or introgression are little. Based on the systematic criteria, we have surveyed Korean wild plant species that showed the similarity to cultivating crops. The consideration for feasibility of genetic pollution (introgression of transgene) is necessary for the successful accomplishment in the practical use of GMPs. Although the detrimental effects of GMPs on wild relatives have not been clearly verified, Korean wild plant species related to crop plant (potential GMP) have to be investigated with respect to the introgression. Korean flora consists of ca. 5,500 species. Among them, 1,448 species are classified as weed species (966 native, 325 naturalized, and 167 escaped ones), which is vulnerable to GMPs in term of introgression. We suggested the principal Korean wild plants related to major crops that might be affected by GMPs via introgression. The investigated species herein are selected based on the morphological and phenological relationship. It is necessary to verify the genetic relationship between cultivated plants and wild relatives sing more precise molecular techniques, which provide the information of likelihood for the introgression of transgene.

분자생물학적 방법을 이용한 Moniliella 속과 Trichosporonoides 속의 분류학적 고찰 (Are the genus Moniliella and Trichosporonoides closely related in molecular taxonomic relationship\ulcorner)

  • The molecular taxonomic relationship of nine species in the genus Moniliella Stolk & Dakin and Trichosporonoides Haskins & Spencer and six species of other yea나-like fungi was examined by sequencing analysis of large subunit rDNA D1/D2 variable domain. The fifteeen species fell into two major groups corresponding with their genetic relationships. The nine species of the genus Moniliella and Trichosporonoides were placed at the same cluster. similarity values based on the D1/D2 domain sequences were 45.4-100% among species of genus Moniliella, 45.2-84.4% among genus Trichosporonoides species, and 45.6-90.1% among species of genus Moniliella and Trichosporonoides. Identical sequence similarity was observed between M. suaveolens var. nigra and M. suaveolens. A colse relationship of M. mellis. and M. acetoabutens is observed. The result of this study provided and insight into the genetic origins of genus Moniliella and Trichosporonoides species as well as their genetic relationships. Genus Moniliella and Trichosporonoides are closely related to each other based on sequence analysis of the large subunit rDNA D1/D2 region and we suggest combination of the genus Moniliella and Trichosporonoides to single genus.

  • Group technology(GT) is a manufacturing philosophy which identifies and exploits the similarity of parts and processes in design and manufacturing. A specific application of GT is cellular manufacturing. the first step in the preliminary stage of cellular manufacturing system design is cell formation, generally known as a machine-part cell formation(MPCF). This paper presents and tests a grouping gentic algorithm(GGA) for solving the MPCF problem and uses the measurements of e(ficacy. GGA's replacement heuristic used similarity coefficients is presented.

RAPD와 URP를 이용한 심비디움 유전자원 유연관계 분석 (Analysis of Genetic Relationship among Cymbidium germplasms Using RAPD and URP)

  • 심비디움속 유전자원 48품종에 대하여 RAPD와 URP를 이용하여 유전적 유연관계를 분석하였다. RAPD분석에는 10mer에 해당하는 random primer (Operon사) 80개를, URP는 20 mer에 해당하는 12종의 상용 primer를 이용하였다. 48 품종의 심비디움에는 34종의 동양 심비디움, 7종의 동서양란 교잡종, 7종의 서양 심비디움이 포함되어 있다. 선별된 41개의 random primer와 6개의 URP primer로부터 각각 407, 56개의 다형성 밴드를 획득하여 총 463개의 마커를 이용하였다. 이들 마커의 크기 범위는 0.4 kb 에서 1.5 kb 에 해당하였다. 유전적 유사도를 바탕으로 UPGMA clustering 프로그램을 이용하여 dendrogram을 작성하였는데 유전자원 48품종은 유사도 0.638 수준에서 총 4그룹으로 구분되었다.

  • Shortnecked clam is a commercially important mollusks species, which is distributed all over the Yellow Sea. Consequent of the rapid increase in seed production, there is a need to understand the genetic composition of wild shortnecked clam populations in order to evaluate exactly the latent genetic effects induced by seed production operations. Many genetic and molecular researches were made because RAPD-PCR is a simple and rapid method for determining genetic diversity and similarity in various organisms with the advantage that no prior knowledge of the genome under research is needed (Fischer et at., 2000). 북극권 Svalbard 지역과 한국에 분포하는 Saxifraga 속 식물의 유전적 다형성 비교 (Comparison of Genetic Diversity of Saxifraga Species Distributed in the Arctic Svalbard and Korea)

  • 북극지역에 분포하는 Saxifraga 속에 식물종들은 분류가 까다롭고 연구보고가 매우 적다. 북극권인 노르웨이령 Svalbard와 한국에서 수집된 16종의 Saxifraga 속 식물의 RAPD를 통하여 유전적 다형성을 비교하였다. 실험에 활용한 12 종류의 URP primer 중 4종의 primer를 이용하여 총 79개(44.8%)의 다형성을 가진 DNA 밴드를 선발하여 UPGMA 분석으로 유전적 다형성을 비교하였다. 그 결과 북극 Svalbard에서 수집된 9종의 Saxifraga 속 식물들이 한국에서 수집된 종들에 비해 유전적 다형성이 큰 것으로 확인되었다. 유사도 매트릭스를 활용한 분석 결과는 두 지역에 수집된 종들간에 비교적 뚜렷한 구룹을 형성하였으며, 한국 분포종의 RAPD는 북극 Svalbard 지역에 분포하는 종들에 비해 유사도가 높은 것으로 분석되었다. 본 연구를 통하여 우리는 두 지역에 분포하는 종들 간의 형태적 유사성이 유전적 유사성과 일치하고 있음을 확인하였다. 또한 분석 결과를 통하여 두 지역간의 지리적 격리, 집단내 영양 생식, 그리고 두 지역간의 환경조건의 차이가 높은 수준의 유전적 다형성을 형성하는 요인이 될 것으로 추론할 수 있었다.
