Halenia coreana is an endangered, endemic species that is distributed in only a few locations in Korea, such as Mts. Hwaaksan and Daeamsan. It has been recently segregated from H. corniculata, broadly distributed in cold temperate regions that include northern Japan, the Russian Far East, northeastern China, Mongolia, and eastern Europe, where population sizes are usually large. To examine the genetic diversity of H. coreana and evaluate the level of genetic differentiation of the species compared with that of H. corniculata, we surveyed 183 candidate simple sequence repeats (SSR) motif markers for H. coreana and H. corniculata from sequence data of amplified fragments of a specific length in the genome. A total of 17 genomic-SSR markers were selected to examine the levels of genetic diversity and differentiation using 17 samples of H. coreana and 60 samples of three populations of H. corniculata. The results here suggest that the genetic diversity of H. coreana is very low with a high frequency of inbreeding within its population. We found that H. coreana is genetically differentiated from H. corniculata, supporting the recognition of the geographically isolated H. coreana as a distinct species.
The effects of a polyamine, spermine, on the differentiation of Naegleria gruberi amebas into flagellates were tested. Addition of spermine at early stages of differentiation (until 40 min after the initiation of differentiation) completely inhibited the differentiation. To understand the inhibition mechanism, we examined the effect of spermine treatment on the transcription and translation of differentiation-specific genes during differentiation. Addition of spermine at early stages did not inhibit the accumulation of two differentiation-specific mRNAs, ${\alpha}$-tubulin and Class I mRNA, significantly, but rather prevented the rapid degradation of the mRNAs in later overall protein synthesis partially and gradually. However, translation of the ${\alpha}$-tubulin mRNA was completely inhibited. These data suggest that the inhibition of differentiation of N. gruberi by spermine treatment did not result from the inhibition of transcription of differentiation-specific genes but from the specific inhibition of translation of the mRNAs during the differentiation.
Tumor suppressor genes have received much attention for their roles in the development of human malignancies. Gelsolin has been found to be down-regulated in several types of human cancers, including leukemias. It is, however, expressed in macrophages, which are the final differentiation derivatives for the monocytic myeloid lineage, implicating this protein in the differentiation process of such cells. In order to investigate the role of gelsolin in leukaemic cell differentiation, stable clones over-expressing ectopic gelsolin, and a control clone were established from U937 leukaemia cells. Unlike the control cells, both gelsolin-overexpressing clones displayed retarded growth, improved monocytic morphology, increased NADPH and NSE activities, and enhanced surface expression of the ${\beta}$-integrin receptor, CD11b, when compared with the parental U937 cells. Interestingly, RT-PCR and western blot analysis also revealed that gelsolin enhanced p21CIP1 mRNA and protein expression in the overexpressing clones. Moreover, transient transfection with siRNA silencing P21CIP1, but not the control siRNA, resulted in a reduction in monocytic differentiation, accompanied by an increase in proliferation. In conclusion, our work demonstrates that gelsolin, by itself, is capable of inducing monocytic differentiation in U937 leukaemia cells, most probably through p21CIP1 activation.
국내 소나무 19개 집단을 대상으로 cpSSR 표지자 분석을 통해서 관찰된 28개 변이체로부터 총 167개의 독특한 haplotype이 확인되었고, 동일한 haplotype을 보인 13개체가 10집단에 고르게 분포하였으며, 각 집단에서 관찰된 유효 haplotype의 수는 평균 13.37개로 나타났다. cpSSR haplotype의 집단내 다양도(He)는 0.987로 계산되어 기존의 임목을 대상으로 한 연구에서 보고된 수치와 유사하거나 약간 높은 수치를 보였다. 각 haplotype을 구성하고 있는 cpSSR 변이체를 대상으로 각 집단에서의 다양성(S.I.)을 계산한 결과 강원도 영월집단이 1.109로 계산되어 가장 높은 수치를 보였으며, 경북 문경집단이 0.411로 가장 낮은 수치를 보였다(평균 0.887), 관찰된 cpSSR 변이체들의 대부분이 19개 집단에 공통적으로 존재하는 것으로 나타났으며(97.62%), 집단간에 cpSSR 변이체 분화는 미약한 것으로 나타났는데(${\Phi}_{ST}=0.024$) cpSSR 표지자의 높은 돌연변이 발생빈도가 주요 원인인 것으로 추정된다. 반면에 비교 가능한 173개 집단 쌍 간에 동일한 haplotype이 전혀 존재하지 않는 집단 쌍이 39쌍으로 나타나 집단간의 유전적 유연관계에 대한 직접적인 비교가 불가능했으며, 따라서 분석된 19개 집단간에 유전적 교류가 자유롭게 일어나지 않는 것으로 나타났다. cpSSR 표지자 변이체의 분포양상과 기존의 I-SSR 표지자 변이체의 분포양상을 비교 고찰해 볼 때 국내 소나무 유전자원의 효율적인 관리를 위해서는 분석된 소나무 집단의 현재 위치 정보와 유전정보가 함께 고려되어야할 것으로 생각된다.
꽃게(Portunus trituberculatus)는 세계적으로 넓게 분포하는 갑각류로 모래나 돌멩이가 있는 해저에 서식한다. 본 연구는 서해의 4개 지점(영광, 태안, 소래, 연평도)에서 채집된 P. trituberculatus 281 개체에 대해 10 종류 microsatellite 좌위의 유전적 다형성을 조사하였다. 좌위당 대립유전자 수는 50-129개로 평균 69.5개였으며, 관측 및 예상 이형접합도는 각각 0.111-1.000 및 0.609-0.979 범위에 있었다. 좌위별 근친계수((Fis)는 -0.0207에서 0.8175 범위였다. 유전적 분화도(Fst)는 0.05보다 낮게 나타났는데, 이것은 4 꽃게 간의 유전적 분화(genetic differentiation)가 매우 낮은 이루어진 것으로 추정하게 한다. UPGMA을 이용한 계통도 작성에서도 4 그룹 간의 유전적 거리는 매우 가깝다는 결과를 얻을 수 있었다. 매우 높은 다형성과 집단간의 낮은 유전적 분화는 서해안의 꽃게 집단은 활발한 유전적 흐름(gene flow)이 일어나며, 그룹간 지리적 경계가 없음을 제시한다
파밤나방(Spodoptera exigua(Hubner))의 유전변이가 다형 동위효소를 이용하여 분석되었다. 집단들은 다른 기주, 지리적 위치, 그리고 시기에 따라 세분화되었다. 평균 이형접합자빈도($0.443\pm$0.013)는 파밤나방 전체 집단의 높은 유전변이를 나타냈다. 각 소집단들은 모두 Hardy-Weomberg 균형에서 벗어난 뚜렷한 동계교배 양식을 보였다. 이러한 높은 동계교배 효과는 지역고립에 의한 집단간 분화보다는 대부분 소집단 내 표본 추출 효과에 기인된 비임의교배에 의해 야기되었다. Wright($F_{ST}$ ) 와 Nei의 유전거리(D)는 소집단들간 유전적 분화가 낮음을 나타내지만, 일부 남쪽지역의 소집단들(해남과 사천)은 상대적으로 북쪽의 소집단들(안동과 군위)과 차이를 보였다. 유전적 거리를 기초하여 추정된 세대당 이주 개체수는 서로 다른 기주 집단간 5.9마리, 지역 집단간 10.6마리, 시기 집단간 31.8마리였다. 이러한 유전분석 결과들을 보아 국내 파밤나방은 이들의 탁월한 집단간 이주 능력 때문에 집단간 유전분화가 적은 것으로 나타냈다.

A human gene T-complex protein 10 like (TCP10L) was cloned in our lab. A previous study showed that it expressed specifically in the liver and testis. A transcription experiment revealed that TCP10L was a transcription factor with transcription inhibition activity. In this study, the human MAD4 was identified to interact with TCP10L by a yeast two-hybrid screen. This finding was confirmed by immunoprecipitation and subcellular localization experiments. As MAD4 is a member of the MAD family, which antagonizes the functions of MYC and promotes cell differentiation, the biological function of the interaction between TCP10L and MAD4 may be to maintain the differentiation state in liver cells. Also, we propose that the up-regulation of Myc is caused by the down-regulation of TCP10L in human hepatocarcinomas.

Agaricus bisporus is a popular edible mushroom that is cultivated worldwide. Due to its secondary homothallic nature, cultivated A. bisporus strains have low genetic diversity, and breeding novel strains is challenging. The aim of this study was to investigate the genetic diversity and population structure of globally collected A. bisporus strains using simple sequence repeat (SSR) markers. Agaricus bisporus strains were divided based on genetic distance-based groups and model-based subpopulations. The major allele frequency (MAF), number of genotypes (NG), number of alleles (NA), observed heterozygosity (HO), expected heterozygosity (HE), and polymorphic information content (PIC) were calculated, and genetic distance, population structure, genetic differentiation, and Hardy-Weinberg equilibrium (HWE) were assessed. Strains were divided into two groups by distance-based analysis and into three subpopulations by model-based analysis. Strains in subpopulations POP A and POP B were included in Group I, and strains in subpopulation POP C were included in Group II. Genetic differentiation between strains was 99%. Marker AB-gSSR-1057 in Group II and subpopulation POP C was confirmed to be in HWE. These results will enhance A. bisporus breeding programs and support the protection of genetic resources.
북방종개(Iksookimia pacifica)의 유전적 다양성과 구조적 특징을 밝히기 위해 동해 독립하천들에 서식하는 10개의 집단들을 대상으로 핵유전자와 미토콘드리아 유전자에 기반한 집단유전학적 분석을 실시하였다. 일부 예외적인 경우를 제외하고, 미토콘드리아와 핵유전자 모두 통계적으로 유의미한 집단 간 유전적 분화가 관찰되었다. 핵유전자들의 DNA 서열자료에서 추출한 유전자형 자료를 Bayesian 방법으로 분석한 결과 북방종개는 천진천과 양양남대천을 기준으로 북쪽과 남쪽의 두 개의 그룹으로 나뉘는 구조를 보였다. 현재 동해 하천들이 지리적으로 단절되어 온 독립 수계라는 것을 감안했을 때, 남북으로 구별되는 집단유전적 구조는 북방종개가 한반도에 정착했던 초기 조상 집단이 남북으로 갈라지는 지리적인 분리 사건과 관련되었을 것으로 해석되며, 이러한 초기 조상집단의 지리적 분리 이후, 두 조상그룹들은 남북의 지리적인 범위 내에서 하천 별 고립에 따른 추가적인 분화 과정을 거쳤을 것으로 추정된다. 주목할 점으론, 자산천 집단의 많은 개체들이 지리적 거리가 먼 양양남대천 및 강릉남대천 집단과 하나의 유전적 cluster를 형성하고 있는 것이다. 이와 함께 미토콘드리아 유전자의 경우 몇몇 이웃하는 집단들 사이에 현저히 낮은 유전적 분화도 그리고 일부 집단들에서 매우 낮은 유전적 다양성이 관찰되었다. 본 집단유전학적 결과는 향후 북방종개의 보존 및 관리를 위한 기초자료로 제시될 것이다.
