Genetic and Phenetic Differentiation among Three Forms of Korean Salamander Hynobius Ieechii

  • Genetic and morphological variations of Korean Hynobius leechii were surveyed. Starch gel electrophoresis was used to study genetic variation at 23 loci. H. leechii is genetically divided into three groups, each of which is named as Form-A, Form-B, and Form-C. These forms are clearly separated with 4-5 fixed or nearly fixed allelic differences. At a sympatry near Jangseong, Jollanam-do, a significant deviation from random mating between Form-A and Form-B was observed. Therefore, they are considered as two distinct taxi of specific rank. Form-A is widely distributed in the inland of Korea, whereas Form-B is distributed in the islands or near the coastal regions of Western and Southern Korea. Form-B had, in addition, a significant morphological difference from Form-A. On the other hand, Form-C was restricted to an area near Kori atomic power plant, Kyongsangnam-do, and the genetic similarities between this form and Form-A and Form-B were 0.742 and 0.736, respectively. Moreover, Form-C had a unique color pattern, short body and small coccyx. Thus, it seems reasonable to assign this form as another new species.

지능 알고리즘 기반의 이족 보행로봇의 보행 구현 (A Gait Implementation of a Biped Robot Based on Intelligent Algorithm)

  • This paper deals with a human-like gait generation of a biped robot with a balancing weight of an inverted pendulum type by using genetic algorithm. The ZMP (Zero Moment Point) is the most important index in a biped robot's dynamic walking stability. To perform a stable walking of a biped robot, a balancing motion is required according to legs' trajectories and a desired ZMP trajectory. A dynamic equation of the balancing motion is nonlinear due to an inverted pendulum type's balancing weight. To solve the nonlinear equation by the FDM (Finite Difference Method), a linearized model of equation is proposed. And GA (Genetic Algorithm) is applied to optimize a human-like balancing motion of a biped robot. By genetic algorithm, the index of the balancing motion is efficiently optimized, and a dynamic walking stability is verified by the ZMP verification equation. These balancing motion are simulated and experimented with a real biped robot IWR-IV. This human-like gait generation will be applied to a humanoid robot, at future work.

Analysis genetic diversity of Plasmodiophora brassicae using RFLP and RAPD(oral)

  • Genetic diversity of Plasmodiophora brassicae from major chinese cabbage cultivating areas in Korea was analyzed by using PCR-RFLP and RAPD. Single spores of P brassicae isolated from galls of club root made induce lesion on chinese cabbage successfully. The PCR-RFLP and RAPD by primers PbITS, URP 3, 6 and OPA 7 revealed that single spore isolates showed various DNA polymorphisms among them unrelated geographic origins. These results indicate that P. brassicae population in Korea showed genetic difference among them. This study could be facilitate to identify genetic characteristics ofP. brassicae based on DNA polymorphisms between single spore isolates and to get basic information which can be used to advanced resistance breeding against club root of chinese cabbage.

Genetic Discrimination of Catharanthus roseus Cultivars by Multivariate Analysis of Fourier Transform Infrared Spectroscopy Data

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts of higher plants is applied to discriminate plants genetically, leaf samples of eight cultivars of Catharanthus roseus were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR fingerprint region data were analyzed by principal component analysis (PCA). Major peaks as biomarkers were identified as the most significant contributors to distinguish samples by using genetic programming. A hierarchical dendrogram based on the results from PCA separated the eight cultivars into two major groups in the same manner as the dendrograms based on genetic fingerprinting methods such as RAPD and AFLP. A slight difference between the dendrograms was found only in branching pattern within each subgroup. Therefore, we conclude that the hierarchical dendrogram based on PCA of the FT-IR data represents the most probable chemotaxonomical relationship between cultivars, which is in general agreement with the genetic relationship determined by conventional DNA fingerprinting methods.

Replicated Association Study between Tuberculosis and CLCN6, DOK7, HLA-DRA in Korean

  • Tuberculosis is a global public health problem and manifests itself as a difference in the genetic susceptibility of the host, along with the properties of Mycobacterium tuberculosis (MTB). The single nucleotide polymorphisms (SNPs) and candidate genes proposed in the Genome-wide association study (GWAS) on tuberculosis in a recently published Chinese population were reported. In this study, we investigated whether the genetic polymorphism of candidate genes related to tuberculosis is reproduced when targeting Koreans. The CLCN6 (rs12404124, rs198391, rs535107), DOK7 (rs1203104, rs1203103) and HLA-DRA (rs1051336) gene polymorphisms showed statistically significant results. In addition, it was also found whether it acts as an expression quantitative trait loci (eQTL) that can influence gene expression. This study confirmed that the genetic polymorphism of the three genes (CLCN6, DOK7, HLA-DRA) affects the development of tuberculosis and will help to understand the genetic specificity of tuberculosis and the interaction between pathogens and hosts.

Evaluation of Genetic Relationship and Fingerprinting of Rice Varieties using Microsatellite and RAPD Markers

  • Genetic diversity of 31 rice varieties including 25 japonica and 6 indica varieties was evaluated using a combination of 19 microsatellite or simple sequence repeats (SSRs) and 28 random decamer oligonucle-otide primers. All 19 microsatellite primer sets representing 19 loci in the rice genome showed polymorphisms among the 31 varieties and revealed 91 alleles with an average of 4.80 bands per primer. Also all 28 random decamer primers used were informative and generated 114 non-redundant bands with a mean of 4.07 bands. Microsatellite markers detected higher number of alleles than random primers .although the mean difference was not statistically significant. A cluster analysis based on Nei's genetic distances calculated from the 205 bands resolved the 31 varieties into two major groups that correspond to indica and japonica subspecies, which is consistent with the genealogical information. As few as six random decamer primers or a combination of one microsatellite and four random decamer primers were sufficient to uniquely differentiate all 31 varieties. These combinations would be potentially useful in rice variety protection and identification considering that 25 out of 31 varieties used in this study are japonica rices with high grain quality and have close make up.

Genetic Distances between Two Echiuran Populations Discriminated by PCR

  • Genomic DNA extracted from representatives of two populations, Gunsan and Chinese, of Urechis spp. was amplified using PCR with several primers. The band-sharing (BS) value between individuals no. 05 from the Gunsan population and no. 22 from the Chinese population was 0.206, which was the lowest recognized value. Oligonucleotides primer OPC-04 revealed 44 unique loci, which distinguished the Chinese population. Primer OPB-17 allowed the discovery of 22 loci shared by the two populations, which were present in all samples. Based on the average BS results, individuals from the Gunsan population demonstrated lower BS values (0.661±0.012) than did those from the Chinese population (0.788±0.014; p<0.05). The shortest genetic distance (GD) displaying a noteworthy molecular difference was between individuals CHINESE no. 12 and no. 13 (GD=0.027). Individual no. 06 from the Gunsan population was most distantly related to CHINESE no. 22 (GD=0.703). A group tree of the two populations was constructed by UPGMA Euclidean GD analysis based on a total of 543 fragments generated using six primers. The explicit markers recognized in this study will be used for genetic analysis, as well as to evaluate the species security and proliferation of echiuran individuals in intertidal regions of the Korean Peninsula.

단감나무로부터 분리한 탄저병 병원균 Colletotrichum spp.의 RAPD와 PCR-RFLP를 이용한 유연관계 분석 (Analyses of Genetic Relationships of Collectorichum spp. Isolated from Sweet Persimon with RAPD and PCR-RFLP.)

  • Colletotrichum spp.는 광범위한 기주범위를 갖는 다범성균으로 각종작물에 피해를주는 중요한식물병원진균이다. 최근 국내에서 널리 재배되고 있는 단감, 사과, 복숭아, 포도 등에 탄저병 이 발생하여 많은 경제적 손실을 초래하고 있다. 탄저병원균의 경우 기존에는 주로 형태적 특징이나 배지 상에서의 특성, 기주에 대한 병원성의 차이에 의존하여 분류를 해 왔다. 그러나 최근에는 병원균의 분류에 있어 문제점을 해결하기 위하여 분자생물학적 방법을 이용하고 있다. 이에 본 실험에서는 Random Amplified Polymorphism DNAs (RAPD)와 Ploymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) 기법을 이용하여 단감나무에 탄저병을 일으키는 균들 간에 유연관계를 밝혔다. 유연관계 분석결과 크게는2개의 그룹으로 나뉘었고 작게는5개의 그룹으로 나뉘는 것을 알 수 있었다.

Gene Expression Analysis of Pregnant Specific Stage in the Miniature Pig Ovary

  • The miniature pig is considered to be a better organ donor breed for xenotransplantation than other pig breeds because the size of the organs of the miniature pig is similar to that of humans. In this study, we aimed at identifying differentially expressed genes in the miniature pig ovary during pregnancy. For this, we used the miniature pig ovary model, annealing control primer-based reverse transcription polymerase chain reaction (PCR), quantitative real-time PCR (qRT-PCR), and northern blotting analysis. We identified 13 genes showing differential expression on the based of pregnancy status and validated 8 genes using qRT-PCR. We also sequenced the full-length cDNA of ephrin receptor A4 (EphA4), which had a significant difference in expression level, and validated it by northern blotting. These genes may provide a better understanding of the cellular and molecular mechanisms during pregnancy in miniature pig ovary.

Association of the Genetic Polymorphisms for CD247 Gene and Tuberculosis Case

  • Tuberculosis is airborne disease caused by Mycobacterium tuberculosis (MTB). Host genetic factors of these tuberculosis play an important role in determining individual difference in susceptibility or resistance to infectious diseases including tuberculosis. CD247 is named CD3zeta chain or CD3ζ. CD247 gene is a protein-coding gene involved in phagocytosis and signal transduction of the T cell receptor (TCR). Also, downregulation of the CD3ζ chain has been associated to chronic inflammation. The aim of this study was to research association of the genetic polymorphisms for CD247 gene and tuberculosis. We analyzed association of CD247 and Mycobacterium tuberculosis using 149 imputed single nucleotide polymorphisms (SNPs) with Korean population. And the results of this study show that seven SNPs of CD247 were identified to associate with tuberculosis. The most significant SNP was rs858545 (OR=1.22, CI: 1.05~1.42, P=0.009481). This study suggests that polymorphisms of CD247 may affect the T cell receptor signaling pathway, which may associate the infection of tuberculosis.
