Diffusion study for chloride ions and water molecules in C-S-H gel in nano-scale using molecular dynamics: Case study of tobermorite

  • Porous materials such as concrete could be subjected to aggressive ions transport. Durability of cement paste is extremely depended on water and ions penetration into its interior sections. These ions transport could lead different damages depending on reactivity of ions, their concentrations and diffusion coefficients. In this paper, chloride diffusion process in cement hydrates is simulated at atomistic scale using molecular dynamics. Most important phase of cement hydrates is calcium silicate hydrate (C-S-H). Tobermorite, one of the most famous crystal analogues of C-S-H, is used as substrate in the simulation model. To conduct simulation, a nanopore is considered in the middle of simulation cell to place water molecules and aggressive ions. Different chloride salts are considered in models to find out which one is better for calculation of the transport properties. Diffusion coefficients of water molecules and chloride ions are calculated and validated with existing analytical and experimental works. There are relatively good agreements among simulation outputs and experimental results.

  • The effective diffusion coefficients of glucose, sucrose and fructo-oligosaccharides in Ca-alginate gel beads at high concentration of sucrose solutions were investigated at $50^{\circ}C$. A mathematical model for the kinetics of fructo-oligosaccharide production using immobilized cells was proposed and compared with experimental results varying the bead size, the substrate concentration and the bead ratio. Very low values of diffusion coefficients ranging $1.2-7.6\times10^{-7}\textrm{cm}^2$/sec were obtained, and the predicted results were in good agreement with experimental ones in all cases tested.

  • In this study, mesoporous titania/alumina membranes were prepared by sol-gel method. Pore structure and phase composition of titania/alumina membranes could be changed by calcination temperature. The addition of alumina into titania membranes retarded anatase-to-rutile phase transformation, resulting in stabilization of pore structures. The 5 time dip-coated membrane calcined at $450^{\circ}C$ is about $10.3{\mu}m$ in thickness with an average pore size of 5 nm. Hydrogen and nitrogen permeances through the membrane were $17.1{\times}10^{-7}mol/m^2{\cdot}s{\cdot}Pa$ and $4.7{\times}10^{-7}mol/m^2{\cdot}s{\cdot}Pa$, respectively. These data were explained by the Knudsen diffusion mechanism.

Simultaneous Purification of Enterotoxin A and C by Fast Protein Liquid Chromatography (FPLC에 의한 Staphylococcal Enterotoxin A와 C의 동시분리)

  • A new method developed for simultaneous purification of enterotoxin A and C from Staphylococcus aureus strain L 350/1 consisted of chromatography on carboxymethyl (CM)-cellulose using a buffer of variable pH, gel filtration on Ultro gel, and fast protein liquid chromatography(FPLC) using a buffer of variable pH. The enterotoxin A and C were purified by three steps: batchwise adsorption from culture supernatant on Amberlite CG-50; chromatography on CM-cellulose using a buffer of constant pH and molarity; and gel filtration on Sephadex G-75. The purified enterotoxin appeared homogeneous by gel diffusion and polyacrylamide gel electrophoresis. Upon treatment with CM-cellulose using a elution of variable pH, enterotoxin A and C were so close that they were not separated completely. After elution from gels, the enterotoxins appeared as a single peak at the same position. Gel filtration gave a reaction of complete identity to enterotoxin A and C in Ouchterlony immunodiffusion. In FPLC using a CM-cellulose, enterotoxin A and C were simultaneously separated at pH 8.6 and 6.8. When each fraction was performed to gel immunodiffusion, at peak of enterotoxin A and C were not detected each other. In a method of elution by pH-gradient was to be more efficient as a simultaneous separation method in terms of speed, yields and simplicity. The purified toxin A and C were identical to type A and C reference enterotoxin on both disc electrophoresis and Ouchterlony gel diffusion.

Serovars of Xanthomonas campetris pv. oryzae Collected from Korea and Serological Diagnosis of Bacterial leaf Blight (우리나라 벼 흰빛잎마름병균 (Xanthomonas campestris pv. oryzae)의 혈청학적 분류 및 진단)

  • Seventy-one strains collected from Korea were classified into three serovars (designated A, B-I and B-II) by using agar gel diffusion test with the antisera produced against Xanthomonas campestris pv. oryzae isolates Q7472 and Q7502. Of 71 isolates tested, 65 isolates belonged to serovar A, 5 isolates were serovar B-I, and one isolate was serovar B-II. The isolates of serovar B-I and B-II could be distinguished clearly from those of serovar A showing marked autoagglutination. Xanthomonas campestris pv. oryzae was serologically diagnosed in rice leaves by agar gel diffusion tests, possibly being distinguished from Xanthomonas campestris pv. olyzicola and E. herbicola. The pathogen could be also serologically detected from the extracts of diseased leaves, squeezed immediately, heated at $100^{\circ}C$ or incubated in PSA. Serological detection of Xanthomonas campestris pv. oryzae is a more reliable and less time-comsuming method.

  • The spot detection phase of the 2-DE image analysis program segments a gel image into spot regions by an image segmentation algorithm and fits the spot regions to a spot shape model and quantifies the spot informations for the next phases. Currently the watershed algorithm is generally used as the segmentation algorithm and there are the Gaussian model and the diffusion model for the shape model. The diffusion model is closer to real spot shapes than the Gaussian model however spots have very various shapes and especially an asymmetric formation in x-coordinate and y-coordinate. The reason for asymmetric formation of spots is known that a protein could not be diffused completely because the 2-DE could not be processed under the ideal environment usually. Accordingly we propose an asymmetric diffusion model in this paper. The asymmetric diffusion model assumes that a protein spot is diffused from a disc at initial time of diffusing process, but is diffused asymmetrically for x-axis and y-axis respectively as time goes on. In experiments we processed spot matching for 19 gel images by using three models respectively and evaluated averages of SNR for comparing three models. As averages of SNR we got 14.22dB for the Gaussian model, 20.72dB for the diffusion model and 22.85dB for the asymmetric diffusion model. By experimental results we could confirm the asymmetric diffusion model is more efficient and more adequate for spot matching than the Gaussian model and the diffusion model.

Skin Permeation of Indomethacin from Gels (겔제제로부터 인도메타신의 피부투과)

  • In order to reduce the systemic side effects and the gastrointestinal irritation of indomethacin following its oral administration, the drug was formulated as a transdermal gel using poloxamer 407. In vitro diffusion cells fitted with excised rat skins were used to evaluate the effects of formulation variables on skin permeation of indomethacin from poloxamer gels. The formulation variables were the concentrations of indomethacin, poloxamer 407 and ethanol, and the gel pH. The increase of the drug amount in the gel from 0.5% to 2.0% induced a direct but nonlinear increase in the skin permeation rate of indomethacin. The increase of poloxamer concentration from 17.5% to 25% in the gel resulted in a decrease of skin permeation rate of indomethacin, which was due to a reduction in the amount of free drug molecules available for permeation through skin by entrapping more drug molecules within the micelles formed by poloxamer. The increase of ethanol concentration from 10% to 20% in the gel resulted in a linear increase of permeation rate of indomethacin through skin, possibly due to the penetration enhancing effect of ethanol. The skin permeation of indomethacin was substantially influenced by the gel pH, exhibiting a maximum at pH 4.

Synthesis and Swelling Kinetics of a Cross-Linked pH-Sensitive Ternary Copolymer Gel System (pH-민감성 삼성분계 공중합체 젤의 합성 및 팽윤 속도론)

  • A pH sensitive ternary copolymer gel was synthesized in the presence of ethylene glycol dimethacrylate (EGDMA) as a crosslinking agent through radical polymerization of vinyl acetate (VA), acrylic acid (AA) and methyl acrylate(MA) with a weight ratio of 1 : 1.3 : 1. A number of experiments were carried out to determine the swelling behavior of the gel under a variety of pH conditions of the swelling medium. As the pH of the swelling medium was changed from 1.0 to 8.0 at $37^{\circ}C$, the gel showed a shift in the pH-dependent swelling behavior from Fickian (n=0.3447) to non-Fickian (n=0.9125). The resulting swelling parameters were analyzed using graphical and statistical methods. The results showed that the swelling of the gel was controlled by the pH of the medium, i.e. $n=n_o{\exp}(S_{C}pH)$, where n is the diffusion exponent, $n_o(=28.9645{\times}10^{-2})$ is the pre-exponential factor and $S_C$(=0.1417) is pH sensitivity coefficient. The swelling behavior of the gel was also examined in aliphatic alcohols. The results showed that the rate of swelling increased with increasing number of carbon atoms in the alcoholic molecular chain.

Nanoparticulate Co-Ferrite Thin Films on Glass Substrate Prepared by Sol-Gel Method (유리기판에 sol-gel법으로 제조된 나노입자 Co-ferrite 박막의 특성)

  • Cobalt ferrite thin films on Corming glass substrate were fabricated by a sol-gel method. Cobalt ferrite thin films with the grain size of 20-35 nm and thickness of 50nm were obtained. Rapid thermal annealing (RTA) and Annealing processes were adopted for comparison of characteristics of the films. Coercivity values were changed with thermal condition and magnetization values were increased as a function of soaking time. With prolonged soaking time, however, it was decreased because of the diffusion of cations from the glass substrate. The RTA process in preparation of cobalt ferrite thin film was the effective way to prevent and to form a single spinel phase in reduced soaking time. The film heated at 600$^{\circ}C$ for 30 minutes by RTA had coercivity of 2,600 Oe, saturation magnetization 460 emu/㎤, and Mr$.$$\delta$ of 1.43 memu/$\textrm{cm}^2$.

Skin Permeation of Flurbiprofen through Excised Rat Skin from Poloxamer 407 Gel

  • In order to reduce systemic side effects following oral administration, flurbiprofen was formulated as transdermal gels consisting of the drug, poloxamer 407 and ethanol in buffer solutions. The effect of formulation variables in the preparation of flurbiprofen gels on skin permeation of the drug was evaluated using Keshary-Chien diffusion cells fitted with excised rat skins. The permeation rate of flurbiprofen through rat skin was directly proportional to initial drug concentration (between 0.1% and 1.0%) in the gel while it was inversely proportional to poloxamer 407 concentration (between 17.5% and 25%). The skin permeation of flurbiprofen was substantially influenced by the gel pH between 3 and 7, exhibiting a maximum at pH 4. The concentration effect of ethanol on the permeation of the drug was negligible in the concentration range of $10{\sim}20%$.
