Anti-stress effects of Gastrodia elata on catecholamine pathway in rat

  • Enzymes involved in catecholamine synthesis are present in the highest concentration in the adrenal medulla, however they were found also in other, mainly nervous tissues. Increased transcription of genes for catecholamine biosynthetic enzymes is an important mechanism to increase the capacity for epineprine/norepinephrine biosynthesis with stress. Gastrodia elata(Chinese name: Tienma), are very important Chinese herbal medicines used for the medical treatment of headaches, migraine, dizziness, epilepsy, rheumatism, neuralgia, paralysis and other neuralgic and nervous disorders. Immobilize stressed rat markedly increased tyrosine hydroxylase (TH) mRNA and dopamine-${\beta}$-hydroxylase (DBH) mRNA transcriptior level more than control group. But treated Gastrodia elata extracts in immobilized stressed rat slightly increased TH mRNA and DBH mRNA transcription level more than normal group. In addition, we are obtained identical results in PC12 cell line. Decrease of transcription level of TH mRNA and DBH mRNA is indicating that Gastrodia elata have a anti-stress effects which decrease the transcription level of TH and DBH mRNA on catecholamine biosynthesis pathway.

Effect of Dioscorea batatas and Gastrodia rhizoma on Fatty Acid Compositions of Serum, Liver and Brain in Rats (마와 천마 분말이 흰쥐의 혈청, 간장 및 뇌 조직의 지방산 조성에 미치는 영향)

  • This study evaluated the effects of Dioscorea batatas and Gastrodia rhizoma on fatty acid composition of serum, liver and brain in rats which were fed control diet adding 5%, 10%, 15% Dioscorea batatas powder and 5%, 10%, 15% Gastrodia rhizoma powder for 3 weeks, respectively. In the other total fatty acid composition of serum lipids, the percentage of SFA was higher in the control group than in the other groups. The percentages 18:2 and 20:4 in serum lipids were significantly higher in the group 5, 6 and 7 than control group. In phospholipid fatty acid composition of serum lipids, the percentage of 18:2 was significantly higher in the group 5, 6 and 7 than in the other groups. 16:0, 18:0, 18:1, 18:2 and 20:4 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of liver lipids. 16:0, 18:0, 18:1, 18:2, 20:4 and 22:6 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of brain lipids.

Volatile Flavor Constituents in the Rhizoma of Gastrodia elata (천마의 휘발성 향기성분)

  • Crude oils in the rhizoma of fresh and dried Gastrodia elata were obtained by a simultaneous steam distillation and extraction(SDE) method using n-pentane/diethyl ether (1 : 1) as solvent, and their volatile constituents were analyzed by gas chromatography-mass spectrometry(GC/MS) A total of 39 volatile flavor constituents (11 acids, 13 alcohols,6 hydrocarbons,7 carbonyls,2 esters) and 25 constituents (6 alcohols, 13 acids, 4 hydrocarbons, 1 carbonyl, ester) were identified in the fresh and dried Gastrodia elate respectively. The major volatile components of the fresh and dried sample were hexadecanoic acid(66.78%, 50.72%), 9-hexadecenoic acid(8.07%, 9.58%), heptadecanoic acid(2.01%, 0.13%), pentadecanoic acid(6.41%, 4.94%), p-cresol(1.43%,0.52%) and cyclododecene(1.83%, 6.00%).

  • Armillaria isolates (KNU-A110, KNU-A234, KNU-A1022 and KNU-A1030) were excellent isolates for producing Gastrodia tuber in farm cultivation. Depth of soil between $10\;cm{\sim}19\;cm$ was favorable for producing Gastrodia tuber and rhizomorph at cultivated area. Eighty nine isolates were collected from 5 countries; 16 from Japan, 22 from USA, 26 from France, 4 from Africa and 21 from Korea. Mycelial fan of most isolates were better formed on basal medium with lemon extract than without lemon.A. mellea (KNU-A997) were strongly pathogenic to Gastrodia but A. gallica (KNU-A110) were excellent symbiotic to Gastrodia tuber. Mycelial growth were good on basal medium containing $0.25%{\sim}0.5%$ ethanol and sawdust spawn added wheat bran and corncob. Liquid culture inoculation were not only fast growth of mycelium but also reduction of contamination.

  • This study aimed to evaluate the quality characteristics of Gastrodia elata Blume fermented by lactic acid bacteria after saccharifying by 3 methods including enzyme, malt, and rice-nuruk. The lactic acid bacteria (LAB), Pediococcus inopinatus BK-3, isolated from kimchi could reduce the unpleasant taste and odor of Gastrodia elata Blume. The total acidity value of Gastrodia elata fermented by LAB on the malt and rice-nuruk extract solution for 3 days was 2.23% and 2.33%, respectively. After saccharification by malt and rice-nuruk extract solution for 3 days, the viable cell number of fermented Gastrodia elata was 9.14 log cfu/mL and 9.27 log cfu/mL, respectively. The total acidity values were increased above 3.35% by malt and rice-nuruk extract solution for 8 days. Thus, the viable cell number was the highest by malt and rice-nuruk extract solution fermentation for 3 days. The amino acid content of Gastrodia elata fermented by LAB after saccharification by malt extract solution was higher than that of other saccharifying methods. The free sugar content and p-hydroxybenzyl derivatives induced by the enzyme method were higher than those of other saccharifying methods. The overall acceptability was the highest at 4.2 point in Gastrodia elata fermented by malt extract solution.

Anti-platelet and Anti-thrombotic Effects of Gastrodia elata (천마(Gastrodia elata Blume)의 항혈소판, 항혈전활성)

  • MeOH extract of Gastrodia elata was fractionated to five solvent fractions, hexane fr (fr I), 90% MeOH fr (fr II), EtOAc fr (fr III), BuOH fr (fr IV) and $H_2O$ fr (fr V). Among the five fractions, fr II, III and IV showed platelet anti-aggregating effects against ADP or collagen induced rat platelet aggregation in vitro. Fr II , III and IV were also tested in vivo, in the mouse and rat models of thrombosis. Oral administration of fr II, III or IV enhanced the recovery from the thrombotic shock in the mouse model of thrombosis to 32-40% from 17% of recovery with the control group of mice. Treatment with fr II, III or IV also attenuated the sudden reduction in the blood platelet count following intravenous collagen injection to rat. The above results were indicative of the presence of anti-platelet and anti-thrombotic components in this plant.

Combination with Korean Red Ginseng and Gastrodia rhizoma Enhances Vascular Protective Effects in Hyperlipidemic Rats (고지혈증 동물모델에서 홍삼과 천마 혼합투여에 의한 혈관 염증 개선연구)

  • Objectives : This experimental study was designed to investigate the inhibitory effects of combination with Korean red ginseng and Gastrodia rhizoma on vascular dysfunction in high-fat/cholesterol diet-induced hyperlipidemia. Methods : Sprague-Dawley rats were fed with 7.5% cocoa butter and 1.25% cholesterol for 10 weeks, with Panax ginseng (PG), and mixtures of Panax ginseng and Gastrodia rhizoma (PGM), respectively. Results : Chronic treatment with PG and PGM significantly decreased body weight. The aortic expression of cell adhesion molecules such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin were markedly increased in hyperlipidemia rats. Interestingly, PGM significantly decreased cell adhesion molecules expression. However, there was no significant decrease in PG group. In addition, PG and PGM group inhibited high-fat/cholesterol diet-induced cytokine such as monocyte chemoattractant protein (MCP-1) mRNA expression. Furthermore, PG and PGM group significantly decreased c-reactive protein protein (CRP) level. Especially, PGM significantly accentuated the decrease of MCP-1 mRNA expression and CRP level. Conclusions : the present study provides an evidence that combination with Panax ginseng and Gastrodia rhizoma enhances anti-vascular protective effects through suppression of vascular inflammation in hyperlipidemic rats.

Effects of Elevated CO2 Concentration and Increased Temperature on the Growth of Gastrodia elata Blume, Parasitic Medicinal Plant (CO2 농도와 온도 변화가 기생 약초인 천마의 생육반응에 미치는 영향)

  • Gastrodia elata (Orchidaceae) is a perennial parasitic plant that has a unique lifestyle of being in a symbiotic interaction with fungi. The underground root of Gastrodia is used for medicinal treatment to cure high blood pressure, stroke, leukemia, headaches, and especially neurasthenia. This study is intended to investigate the effect of the global warming on the ecological responses of Gastrodia. We cultivated the small tuber of Gastrodia with the oak wood lot and mulberry fungus under control (ambient $CO_2$ concentration + ambient temperature, ACAT), temperature treatment (ambient $CO_2$ concentration+elevated temperature, ACET), and $CO_2$+temperature treatment (elevated $CO_2$ concentration+elevated temperature, ECET). The elevated $CO_2$ concentration was about twice in the ambient air while the elevated temperature was about $2^{\circ}C$ higher than the control group. And then we observed the growth and production of reproductive organs and the underground root. The observation showed that the number of flower stalk was highest at ACET and lowest at ECET. The flower stalk was longest at ACET and shortest at ECET. The inflorescence was longest at ACAT and shortest at ECET. The seed capsule was heaviest at ACET and lightest at ECET. The aboveground biomass was highest at ACET and lowest at ECET. The number of rhizomes was highest at ACET and lowest at ECET. The total rhizome biomass was highest at ACET and lowest at ECET. The average rhizome biomass was highest at ACET and lowest at ECET. The results showed that the growth of Gastrodia increased because of more active growth of Gastrodia elata when only the temperature increased and decreased when both $CO_2$ concentration and temperature increased, indicating the poor growth of Gastrodia elata under the global warming condition. Therefore, the Gastrodia elata plantation should be maintained at the temperature of $20-25^{\circ}C$ and not be exposed to a high $CO_2$ concentration.
