Changes in the concentration of total protein, albumin, ${\alpha}_1-,{\alpha}_2-,{\beta}-,$ and ${\gamma}-globulin$ in Serum from 138 healthy, normal pregnant woman were studied by the method of cellulose-acetate electrophoresis. 1. The concentration of total serum protein decreased gradually during the first 7 month, and showed a tendency to increase thereafter. 2. The concentration of serum albumin showed a steady continuous fall untill term. 3. During pregnancy,${\alpha}_1$ and ${\beta}-globulin$ value rose, ${\gamma}-globulin$ value fell and ${\alpha}_2-globulin$ value showed no significant change.

Background: The demand of recycling renewable agricultural by-products is increasing. Radiation breeding is a method used to improve saccharification efficiency. Thus, we investigated the effect of gamma ray irradiation on the pretreatment and enzymatic hydrolysis of the stalks of Senna tora, an important medicinal plants. Methods and Results: S. tora seeds were irradiated with gamma ray at doses of 100, 200, 300, and 400 Gy. In the pretreated biomass, glucan and lignin content were higher in the M1 ($1^{st}$ generations of irradiation) S. tora stalks than in the M2 ($2^{nd}$ generations of irradiation) stalks, this can be explained by the higher degradation rate in M1. After oxalic acid pretreatment, the concentration of total phenolic compounds (TPCs) in the hydrolysate increased in the gamma ray treated seeds. The highest relative increase rate in crystallinity in the pretreated biomass was observed in M1-400 Gy and M2-100 Gy. The cellulose conversion rate was higher in M1 than in M2, except for 200 Gy. Conclusions: Gamma ray irradiation at an appropriate dose can be used to improve the efficiency of pretreatment and enzymatic hydrolysis, thereby increasing biomass availability.
Fibrinolytic enzyme (FE-2) was purified from the fruiting bodies of Tricholoma saponaceum using DEAE-Cellulose chromatography and Mono-S column chromatography, The enzyme has a molecular weight of 18.23 kDa and include Zn$^{2+}$ ion as found by ICP/MS. The N-terminal amino acid sequence of the enzyme was A-L-Y-V-G-X-S-P-X-Q-Q-S-L-L-V It has a pH optimum at pH 7.5, suggested that FE-2 was a neutral pretense. The activity of FE-2 was highly inhibited by EDTA and 1,10-phenanthroline, indicating that the enzyme is a metalloprotease. The activity of FE-2 was increased by $Mg^{2+}$, Zn$^{2+}$, Fe$^{2+}$, and Co$^{2+}$, but the enzyme activity was totally inhibited by Hg$^{2+}$. No inhibition was found with PMSF, E-64, pepstatin and 2-mercaptoethanol. The enzyme hydrolyzed both $A\alpha$ and B$\beta$ chains of human fibrinogen. The $\gamma$ chain was resistant to hydrolysis by FE-2.
Typical radiation cross-linked hydrogels has the characteristic that high water content, but low emission efficiency of active ingredients. Therefore, the hydrogel was prepared by the addition to collagen, which is closely related to the formation of skin wrinkles in biocompatibility and highly water-soluble carboxymethyl cellulose sodium salt (CMC) in order to preparation of hydrogels has excellent emission efficiency of active ingredients. Hydrogels were prepared by dissolving CMC and collagen each of 0.5%, 10% concentration in deionized water. Then, prepared hydrogels are performed by gamma-radiation at 1, 3, 5 kGy irradiation dose. The results showed that the gel fraction of after irradiated 3 kGy hydrogel was higher than before irradiated gelation as long as the 55.3%. The swelling rate of irradiated 3 kGy hydrogel was lower than the non-irradiated sample. The compressive strength of 3 kGy irradiated hydrogel was the highest. The visco-elastic did not show any significant differences, even after irradiation. The CMC hydrogel in this study suggested a potential use as a material for the mask pack for improved emission efficiency of the active ingredient and anti-wrinkles.

In this study, the effects of gamma-radiation treatment on cell wall composition and physiological characteristics of astringent persimmon fruit were investigated. The soluble tannin contents of gamma-radiated samples were reduced by the radiation treatment. The hardness of the radiated fruit was decreased when compared to non-radiated fruit. Alcohol-insoluble component of the cell wall in the radiated fruit was decreased from 39.3 mg/g to 37.2 mg/g. The water-soluble content of the radiated fruit was increased from 11.4 mg/g to 13.9 mg/g. The cell wall content of the non-radiated fruit was 26.6 mg/g whereas the cell wall content of radiated fruit was decreased to 23.1 mg/g. Due to the maturation of astringent persimmon fruit by gamma-radiation, water-soluble compounds were increased whereas decreasing in cell wall compounds. The contents of lignin, pectin, and cell wall were decreased from 0.82 mg/g and 3.56 mg/g to 0.77 mg/g and 3.14 mg/g, respectively. Acid-soluble hemicellulose content was decreased by gamma-radiation, while alkali-soluble hemicellulose and cellulose contents were increased. Activities of sotening enzyme as polygalacturonase, pectinesterase and $\beta$-galactosidase existed in persimmon fruit were increased by gamma-radiation. In the sensory evaluation, gamma-radition treated persimmon showed very low astringent taste when compared to the non-radiated fruit. In hardness test, the non-radiated persimmon maintained the hardness while gamma-treated persimmon showed softened outer layer due to the condensation of tannin during radiation treatment. Therefore, gamma-radiation treatment will be used for the removal of its astringency of persimmon fruit and for enhancement of its maturation.
For elucidating excessive growth of biofilm that subsequently leads to the clogging problem in a small town's sewer lines of Wisconsin, the FISH method was employed. At the beginning of the simulated experiments, ${\beta}$-subclass proteobacteria prevailed in runs fed with industrial wastewater, while ${\gamma}$-subclass proteobacteria dominated in runs with domestic wastewater. However, the bacterial community structure changed significantly over six weeks; Cytophaga-Flavobacterium (CF)group bacteria dominated in most runs fed with the small town's wastewater regardless of their source, while CF-group decreased strongly in run fed with domestic sewage from another city (Madison). It was also microscopically confirmed that most of those clogging materials was toilet tissue, which in turn may lead to vigorous growth of cellulose-degrading CF-group bacteria. This dominant presence of CF-group bacteria in the small town's sewer indicates that the main constituent of biofilm, toilet tissue (cellulose) in sewage, might have induced the unique pattern of their microbial community structure. Therefore, it suggests that molecular technique is useful for monitoring the clogging problems in sewer lines.
Electron spin resonance (ESR) spectroscopy of gamma-irradiated fresh broccoli and kimchi cabbage was conducted to identify their irradiation history. Different pretreatments, such as freeze-drying (FD), oven-drying (OD), alcoholic-drying (ALD), and water-washing and alcoholic-drying (WAD) were used to lower the moisture contents of the samples prior to ESR analysis. The non-irradiated samples exhibited a single central signal ($g_0$=2.0007) with clear effect of $Mn^{2+}$, especially in kimchi cabbage. Upon irradiation, there was an increase in the intensity of the central signal, and two side peaks, mutually spaced at 6 mT, were also observed. These side peaks with $g_1$ (left)=2.023 and $g_2$ (right)=1.985 were attributed to radiation-induced cellulose radicals. Leaf and stem in broccoli, and root and stem in kimchi cabbage provided good ESR signal responses upon irradiation. The signal noise was reduced in case of ALD and WAD pretreatments, particularly due to $Mn^{2+}$ signals. The ALD treatment was found most feasible to detect the improved ESR spectra in the irradiated samples.

An improved detection of radiation-induced paramagnetic faults was developed to identify the irradiation status of basil and clove. The effectiveness of different sample pretreatments, including freeze-drying (FD), oven-drying (OD), alcoholic-extraction (AE), and water-washing and alcoholic-extraction (WAE), were examined. All non-irradiated samples showed a single central signal ($g_0$=2.006), whereas radicals representing two additional side peaks ($g_1$=2.023 and $g_2$=1.986) with a mutual distance of 6 mT were detected in the irradiated samples. AE and WAE produced the best results for irradiated clove in terms of intensities of radiation-specific ESR signals and their ratios to the central signal. However, FD provided the highest intensities of radiation-specific ESR signals for basil, whereas their ratios to the major signal were better in the cases of AE and WAE. Signal noise, particularly due to $Mn^{2+}$ signals, was observed, whereas it decreased in AE and WAE pretreatments. Based on our results, AE and WAE can improve the detection conditions for radiation-specific ESR signals in irradiated samples.
This study was designed to investigate the effects on anti-oxidative activities and increasing extraction yield of Aralia elata Cortex by gamma irradiation. Electron spin resonance (ESR) analysis as physical techniques for irradiation identification of Aralia elata showed that a pair of peak appeared on a space of 6.0 mT at the left and right of symmetric unspecific central ESR spectrums, confirming that the plant was gamma-irradiated. The optimum extracting conditions for preparing gamma irradiated samples from Aralia elata Cortex were to extract with 50% ethanol for 15 hrs after 10 kGy irradiation. DPPH scavenging activity and ABTS radical cation inhibitory activity of the water and 50% ethanol extracts from non irradiated and irradiated Aralia elata Cortex was very high as over 80% and 98%, respectively, at tested low concentration of $50{\mu}g/mL$. Antioxidant protection factor (PF) as anti-oxidation indicator of lipophilic compounds showed a very high level of activity as 2.18~2.78 PF. As for TBARs, water and ethanol extracts showed high level. Increase of TBARs inhibitory activity of water extracts was not shown by gamma-ray irradiation but ethanol extracts showed slight increasement of TBARs inhibitory activity with 10 kGy gamma-ray irradiation. These results shown confirmed increasement of extraction yield for phenolic compounds and anti-oxidative activity from Aralia elata. Thus, the treatment of gamma-irradiation can be used a way to amplify a solubility for biological active compounds and anti-oxidative activity in plants.
The intestinal microflora of humans is an extraordinarily complex mixture of microorganisms, the majority of which are anaerobic microorganisms. The distribution of amylolytic microorganisms in the human large intestinal tract was investigated in various individuals of differing ages using anaerobic culture techniques. A large percentage of the amylolytic microorganisms present belonged to the Genus Bifidobacteria. The number of Bifidobacteria increased significantly at two years of age. Adults and children above 2 years old carried about $0.8{\times}10^9-2.0{\times}10^{10}$ colony forming units (CFU/gram) of amylolytic Bifidobacteria. Among these amylolytic Bifidobacteria, Int-57 was chosen for further studies. Between 65% and 85% of the amylase produced was secreted and the remaining amylase was bound to the cell wall facing the outside. Amylase production could be induced by starch in a stable form. When cells were grown on maltose or glucose, amylase production was much lower than on starch and amylase activity disappeared after 24 hours growth on these media. Partially purified enzymes showed optimum activity at a temperature of $50^{\circ}C$ and at an optimum pH of 5.5, respectively. Heat treatment at $70^{\circ}C$ for 30 minutes almost completely inactivated amylase. The hydrolysis products of starch were mainly maltose and maltotriose. Soluble starch, amylose, amylopectin, and $\gamma$-cyclodextrin($\gamma$-CD) were easily hydrolyzed. The rate of hydrolysis of $\alpha$-CD and $\beta$-CD was slower than that of $\gamma$-CD. Carboxymethyl cellulose, $\beta$-1, 3-glucan and inulin were not hydrolyzed.
