Efficacy of Soil Amendment with Medicinal Plant Materials for the Control of Root-knot Nematode (Meloidogyne incognita) in Tomato

  • Soil amendments with oriental herbal medicines such as fruit of Anethum graveolens, flower buds of Syzygium aromaticum, rhizome of Cnidium officinale, rhizome of Coptis chinensis, root bark of Paeonia suffructicosa, stem bark of phellodendron amurense, and stem bark of Cinnamomum cassia at the rate of 0.2% (weight by volume of soil) significantly reduced Meloidogyne incognita infection (root gall formation) of tomato seedlings compared with the control. The most effective treatments were root bark of p. suffructicosa and stem bark of C. cassia as they gave minimum numbers of galls on tomato roots (4.7% and 8.9%, respectively, relative to control) as compared to other treatments. Another study with root bark of p. suffructicosa and C. cassia at different application doses also showed consistent results in reducing gall number. The control efficacy decreased as the application doses were lower-ed, indicating their dose-dependent control activities. These treatments significantly enhanced aboveground plant growths (total masses).

  • These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

Effective Combination of Agrobacterium tumefaciens Strains and Ti Plasmids for the Construction of Plant Vector System

  • The purpose of this study is to obtain the most efficient combination of Agrobacterium tumefaciens strains and Ti plasmids for the construction of dicotyledonous plant vector system. Ti plasmid-curing A. tumefaciens A136 and KU12C3 were transformed with four kinds of Ti plasmids, pTiBo542, pTiA6, pTiKU12 and pTiAch5, respectively. The stems of 28 species of dicotyledonous plants were then inoculated with these transformants and examined for crown gall formation. The different combination of A. tumefaciens strains and Ti plasmids showed quite a difference in terms of the crown gall formation. Agrobacterium strins A136 and KU12C3 have a same plant host range in case that both strains harour the same kind of Ti plasmid, pTiBo542 or pTiAch5. However, the above-mentioned both strains have quite different host range in the event of containing the same Ti plasmid, pTiKU12 or pTiA6. In case that KU12C3 contains pTiA6 or pTiKU12, this strain has a wider plant host range than A136. The plant host range of pTiBo542 is the widest, followed by pTiA6, pTiKU12 and pTiAch5. Twelve plants among 28 tested plants are not transformed by any virulent Agrobacterium strains used in this study. In conclusion, A. tumefaciens KU12C3 and A136 harboring pTiBo542 showed the widest host range for transforming dicotyledonous plants. Also, it was acertained that the host range of Ti plasmids is affected by chromosomal level.

공단지역 주변 소나무림의 솔잎혹파리 형태적 특성 (Morphological Characteristics of Thecodiplosis japonensis (Diptera: Cecidomyiidae) Larvae in Pine Forests Around Onsan Industrial Complex in Ulsan, Korea)

  • 환경오염원으로부터 지역별 솔잎혹파리 피해도의 차이를 조사하기 위하여 실시한 결과, 솔잎혹파리 충영형성율은 석유화학공단에 의한 환경오염이 심각한 상황에 있는 공단지역은 충영형성률 47.94%이었고, 대조지역은 9.94%였다. 솔잎혹파리 피해엽은 공단지역 피해엽의 길이는 평균 4.5 cm 였으며, 대조지역은 평균 4.9 cm 이었다. 솔잎혹파리 피해엽의 충영내 유충의 수는 공단지역은 평균 3.4개체이었고, 대조지역은 2.4개체 이었다. 성숙유충의 크기는 공단지역이 유충체장은 2.40 mm, 체폭은 0.7 mm 이었으며, 대조지역의 유충체장은 2.45 mm, 체폭은 0.71 mm 이었다.

  • 솔잎혹파리 내충성(耐蟲性) Pinus thunbergii의 선발목(選拔木) 7~15개체(個體)와 피해목(被害木) 8~15개체(個體)에 대(對)하여 침엽내(針葉內) monoterpenes 조성(組成)을 1월(月)과 6월(月)에 각각(各各) 조사(調査)하고, 그리고 솔잎혹파리의 산란(産卵) 선택성(選擇性)에 대(對)하여 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. 1. 1월중(月中)에 선발목(選拔木)이 증가(增加)하는 성분(成分)은 myrcene, limonene, ${\beta}$-phellandrene과 terpinolene이고, 감소(減少)하는 성분(成分)은 ${\alpha}$-pinene, camphene과 ${\beta}$-pinene이다. 그리고 6월(月)에 선발목(選拔木)이 증가(增加)하는 성분은 ${\alpha}$-pinene, camphene, limonene, ${\beta}$-phellandrene과 terpinolene이고, 감소(減少)하는 성분(成分)은 ${\beta}$-pinene과 myrcene이다. 2. 6월(月)에 선발목(選拔木)이 피해목(被害木)보다 limonene은 6.8%가 증가(增加)하고, ${\beta}$-pinene은 9.2%가 감소(減少)한다. 3. 선발목(選拔木)에도 피해목(被害木)에서와 같이 상당(相當)한 수(數)의 산란(産卵)을 하나 충영형성율(蟲癭形成率)은 대단(大端)히 저하(低下)한다. 이상(以上)을 요약(要約) 결론(結論)하면 본(本) 연구(硏究)로 Pinus thunbergii 침엽내(針葉內)의 monoterpene 중(中) limonene과 ${\beta}$-pinene의 양성분(兩成分)은 계절(季節)에 관계(關係)없이 내충성(耐蟲性) 검정(檢定)에 유효(有效)하다고 할 수 있다.

  • 솔잎혹파리 유충(幼蟲)은 대부분(大部分) 강우시(降雨時)에 낙하(落下)하는 생태적(生態的) 특성(特性)을 이용(利用), 추기(秋期)의 자연낙하기(自然落下期) 동안 피해임지내(被害林地內)의 수관상부(樹冠上部)에 분수장치(噴水裝置)를 설치(設置)하고 강수량별(降水量別), 강수시기별(降水時期別) 및 강수시각별(降水時刻別)로 인공강수(人工降水)를 실시(實施)하여 인공강수(人工降水)가 솔잎혹파리 유충낙하(幼蟲落下)에 미치는 영향(影響)을 분석검토(分析檢討), 인공강수(人工降水)에 의(依)한 새로운 천적사육용(天敵飼育用) 솔잎혹파리 유충채집법(幼蟲採集法)을 개발(開發)코자 하였으며 얻어진 결과(結果)는 다음과 같다. 1. 인공강수(人工降水)에 의(依)한 솔잎혹파리 천적사육용(天敵飼育用) 유충채집법(幼蟲採集法)은 매우 효과적(效果的)이었으며 유충채집(幼蟲採集)에 필요(必要)한 적정강수량(適正降水量)은 5.3-9.4mm, 이때에 소요(所要)된 살수량(撒水量) 및 살수시간(撒水時間)은 각각(各各) $8,000-16,000{\ell}$와 180-360분(分) 이었다. 2. 인공강수(人工降水)에 의(依)한 효과적(效果的)인 유충채집시기(幼蟲採集時期)는 중부지방(中部地方) 소나무림(林)의 경우 11월(月) 초순(初旬)부터 중순(中旬)까지 약(約) 20일간(日間)이며 이 기간(期間)에 낙하(落下)한 유충수(幼蟲數)는 전체(全體) 낙하수(落下數)의 93.4%였다. 3. 하루중에 있어서의 인공강수(人工降水)에 의(依)한 유충낙하(幼蟲落下)는 강수시각(降水時刻)에 영향(影響)을 받지 않았다. 4. 인공강수(人工降水)에 의(依)한 유충(幼蟲)의 채집시기별(採集時期別) 천적기생율(天敵寄生率)은 11월(月)의 것이 비교적(比較的) 높았으며 12월(月) 채집분(採集分)은 다소 떨어지는 경향(傾向)을 보였다. 5. 인공강수(人工降水)에 의(依) 유충채집적지(幼蟲採集跡地)의 1년후(年後)의 밀도변동(密度變動)은 채집전(採集前)과 비교(比較)하여 약(約) 34%의 밀도감소효과(密度減少效果)가 있었다. 6. 인공강수(人工降水)에 의(依)한 유충채집방법(幼蟲採集方法)은 현행방법(現行方法)인 충영채집법(蟲癭採集法)에 비(比)하여 약(約) 14-50%의 채집비용(採集費用)(경제적(經濟的)) 절감효과(節減效果)가 있었다.

Suppression of Autophagy and Activation of Glycogen Synthase Kinase 3beta Facilitate the Aggregate Formation of Tau

  • Neurofibrillary tangle (NFT) is a characteristic hallmark of Alzheimer's disease. GSK3β has been reported to play a major role in the NFT formation of tau. Dysfunction of autophagy might facilitate the aggregate formation of tau. The present study examined the role of GSK3${\beta}$-mediated phosphorylation of tau species on their autophagic degradation. We transfected wild type tau (T4), caspase-3-cleaved tau at Asp421 (T4C3), or pseudophosphorylated tau at Ser396/Ser404 (T4-2EC) in the presence of active or enzyme-inactive GSK3${\beta}$. Trehalose and 3-methyladenine (3-MA) were used to enhance or inhibit autophagic activity, respectively. All tau species showed increased accumulation with 3-MA treatment whereas reduced with trehalose, indicating that tau undergoes autophagic degradation. However, T4C3 and T4-2EC showed abundant formation of oligomers than T4. Active GSK3${\beta}$ in the presence of 3-MA resulted in significantly increased formation of insoluble tau aggregates. These results indicate that GSK3${\beta}$-mediated phosphorylation and compromised autophagic activity significantly contribute to tau aggregation.

형질전환(形質轉換)에 의한 Ti Plasmid의 Bradyrhizobium japonicum에의 도입(導入) (Introduction of Ti Plasmid into Bradyrhizobium japonicum by Spheroplast Transformation)

  • Bradyrhizobium japonicum ROKS 26 균주를 glycine및 Iysozyme을 처리하여 spheroplast를 형성시킨 후 Agrobacterium tumefaciens에서 분리한 Ti plasmid를 분리하여 형질전환 시키고 octopine 영양 요구성에 의하여 transformants를 분리하였다. 그 결과 Ti plasmid가 도입된 transformants의 형질전환율은 $1{\times}10^{-7}$ 정도 되었다. 얻어진 transformant의 plasmid를 조사한 결과 도입된 Ti plasmid 존재가 확인되었으며, 2차원 전기영동법으로 균체 단백질을 분석한 결과 수용세포인 Bredyrhizobium japonicum ROKS 26과 transformant간의 단백질 구성 차이도 화인되었다. 또한 transformant의 근류 형성력을 조사한 결과 본래의 근류의근류형성력을 가지고 있었으나, crown gall형성에 있어서는 Agrobacterium tumefaciens 15955에 의한 gall보다는 크기에 있어 차이가 있었다.

Gene Expression and Secretion of the Anticoagulant Hirudin in Saccharomyces cerevisiae

  • Hirudin, a 65-amino acid protein isolated from the salivary gland of the bloodsucking leech, Hirudo medicinalis, is a potent thrombin-specific inhibitor and blocks the thrombin-mediated conversion of fibrinogen to fibrin in clot formation. We have studied the gene expression and secretion of hirudin in yeast. Saccharomyces cerevisiae. A gene coding for hirudin was synthesized based on the amino acid sequence and cloned into a yeast expression vector $YEG{\alpha}-1$ containing the ${\alpha}-mating$ factor pre-pro leader sequence and galactose-inducible promoter, GALl0. Recombinant S. cerevisiae was found to secrete biologically active hirudin into the extracellular medium. The secreted recombinant hirudin was recovered from the culture medium and purified with ultrafiltration and reverse phase high performance liquid chromatography. Approximately 1 mg of hirudin per liter was produced under suboptimal culture conditions and brought to about 90% purity in two steps of purification.

Ceratomyxa oplegnathus n. sp. from the gallbladder of cultured rock bream, Oplegnathus fasciatus

  • A new myxosporean species Ceratomyxa oplegnathus n. sp. was found from the gallbladder of cultured rock bream, Oplegnathus fasciatus. Mature spores were 44.14 ± 2.41 (39.04-48.41) ㎛ in length, 8.52 ± 0.36 (7.9-9.26) ㎛ in width in sutural view. Spores contained two spherical polar capsules measuring 3.04 ± 0.2 (2.71-3.37) ㎛ in diameter and a binucleated sporoplasm. Each polar capsule had a 5-6 coiled polar filament, and was situated near the sutural line. In ultrastructural observation, asynchronous divisions of generative cells without pansporoblast formation gave arise to 2 spores within a trophozoite. Sporoplasmic cell in the sporoblast was binucleated, and capsulogenic cells had large amounts of rough endoplasmic reticulum, external tubules, and capsular primordia.
