Synthesis of Galactooligosaccharides in the Cheese Whey-based Medium by a Lactase from Lactobacillus paracasei YSM0308

  • An enzyme ${\beta}$-galactosidase or ${\beta}$-galactohydrolase [EC3.2.1.23], commonly called lactase, mediates galacto-oligosaccharide (GOS) synthesis under conditions of high substrate concentrations. Also, lactase hydrolyzes ${\beta}$($1{\rightarrow}4$) lactose into glucose and galactose, the latter is successively transferred to free lactose to make various oligosaccharides via transgalactosylation. GOS is non-digestible to human digestive enzymes and has been used as a functional prebiotics. Among the 24 lactic acid bacteria (LAB) strains used, Lactobacillus paracasei YSM0308 was selected based on its exhibition of the highest ${\beta}$-galactoside hydrolysis activity, and the crude lactase was prepared for examination of reaction conditions to affect the GOS synthesis. Lactase activity was measured with a spectrophotometer using ONPG (o-nitropheyl ${\beta}$-D-galactopyranoside) method. Lactase activity was not detected in the culture supernatant and was mostly present in the cell pellet after centrifugation. Activity of the crude lactase preparation ranges from102 to 1,053 units/mL, with the highest activity determined for L. paracasei YSM0308. Optimal conditions for GOS synthesis are as follows: concentration of whey powder, pH, temperature, and time were 30%, pH 6.5-7.0, $30^{\circ}C$, and 4 h, respectively. The final GOS concentration was 19.41% (w/v) by the crude YSM0308 lactase, which was obtained from strain YSM0308 grown in the 10% (w/v) reconstituted whey-based medium.

Influence of β 1-4 Galacto-oligosaccharides Supplementation on Nitrogen Utilization, Rumen Fermentation, and Microbial Nitrogen Supply in Dairy Cows Fed Silage

  • In a balanced incomplete block design, two dry Holstein cows were used to investigate the effect of $\beta$ 1-4 galactooligosaccharides (GOS) supplementation on nitrogen (N) utilization, rumen fermentation and microbial N supply in the rumen. During the experiment, cows were fed four diets: orchardgrass (Dactylis glomerata L.) silage (OS), OS with GOS supplementation (OSG), OS mixed with alfalfa (Medicago sativa L.) silage (MS) and MS with GOS supplementation (MSG). GOS was supplemented at 2% of dry matter intake. Diets were fed at maintenance level of protein and energy. Results showed that N digestion was affected by silage and interaction of silage and GOS supplementation. Cows fed OSG had the highest N digested (p<0.05) followed by MS, OS and MSG. Supplementation of GOS to OS or MS diets tended to improve N utilization through reducing the N losses on dairy cows. There was no effect of GOS supplementation on rumen fermentation parameters (i.e. pH, $NH_3$-N and total VFA) at 1 h and 6 h after feeding. Compared to cows fed MS, cows fed OS silage had higher (p<0.05) allantoin excretion (80.8 vs. 67.1 mmol/d) and higher (p<0.05) total purine derivatives excretion (92.9 vs. 78.5 mmol/d). The microbial N supply in cows fed OSG was higher (p<0.05) than those fed OS, MS and MSG.

Paenibacillus woosongensis의 만난분해 효소활성 (Mannanolytic Enzyme Activity of Paenibacillus woosongensis)

  • 만난의 분해활성을 갖는 Paenibacillus woosongensis의 배양 상등액으로부터 mannanase, ${\beta}$-mannosidase와 ${\alpha}$-galactosidase 활성이 관찰되었다. 배양상등액 내의 locust bean gum를 분해하는 mannanase는 pH 5.5와 $60^{\circ}C$, para-nitrophenyl-${\beta}$-D-mannopyranoside를 분해하는 ${\beta}$-mannosidase는 pH 6.5, $50^{\circ}C$, para-nitrophenyl-${\alpha}$-D-galactopyranoside를 분해하는 ${\alpha}$-galactosidase는 pH 6.0-6.5와 $50^{\circ}C$에서 각각 최대활성을 보였다. 배양상등액의 만난 분해효소는 mannotriose, mannotetraose, mannopentaose, mannohexaose와 같은 만노올리고당을 분해할 뿐 아니라 mannobiose도 분해하였다. 또한 melibiose, raffinose, stachyose 등의 ${\alpha}$-1,6 결합형 galacto-oligosaccharides를 분해하여 galactose를 생성하였다. 이러한 결과로 보아 P.woosongensis는 galactomannan을 완전히 분해하는데 필요한 3종류 효소를 모두 생산하는 것으로 판단된다.

Enhanced Production of Galactooligosaccharides Enriched Skim Milk and Applied to Potentially Synbiotic Fermented Milk with Lactobacillus rhamnosus 4B15

  • In the current study, we first investigated a method for directly transforming lactose into galacto-oligosaccharides (GOS) for manufacturing low-lactose and GOS-enriched skim milk (GSM) and then evaluated its prebiotic potential by inoculating five strains of Bifidobacterium spp. In addition, fermented GSM (FGSM) was prepared using a potentially probiotic Lactobacillus strain and its fermentation characteristics and antioxidant capacities were determined. We found that GOS in GSM were metabolized by all five Bifidobacterium strains after incubation and promoted their growth. The levels of antioxidant activities including radical scavenging activities and 3-hydroxy-3-methylglutaryl-CoA reductase inhibition rate in GSM were significantly increased by fermentation with the probiotic Lactobacillus strain. Moreover, thirty-nine featured peptides in FGSM was detected. In particular, six peptides derived from ${\beta}$-casein, two peptides originated from ${\alpha}s_1$-casein and ${\kappa}$-casein were newly identified, respectively. Our findings indicate that GSM can potentially be used as a prebiotic substrate and FGSM can potentially prevent oxidative stress during the production of synbiotic fermented milk in the food industry.

Effects of Combination of Nitrate with ${\beta}$1-4 Galacto-oligosaccharides and Yeast (Candida kefyr) on Methane Emission from Sheep

  • The objective of the present study was to determine whether ${\beta}$1-4 galacto-oligosaccharides (GOS) and Candida kefyr combined with nitrate as manipulators could suppress rumen methanogenesis without nitrate poisoning in sheep. Four rumen fistulated wethers were allocated to a $4{\times}4$ Latin square design. Nitrate (1.3 g $NaNO_3$ $Kg^{-0.75}$body weight) with and without GOS and Candida kefyr were administered into the rumen through fistula as a single dose 30 min after the morning meal. GOS and Candida kefyr were supplemented by sprinkling onto the feed and through rumen fistula, respectively. The four treatments consisted of saline, nitrate, nitrate plus GOS and nitrate plus GOS plus Candida kefyr. Physiological saline was used as the control treatment. Compared to saline treatment, the administration of nitrate alone resulted in a very marked decrease in rumen methanogenesis and an increase in rumen and plasma nitrite production and blood methaemoglobin formation consequently causing a decline in oxygen consumption, carbon dioxide production and metabolic rate. When compared to nitrate alone, the simultaneous administration of nitrate with GOS decreased nitrite accumulation in rumen and plasma and nitrate-induced methaemoglobin, while retaining low methane production. However, GOS could not fully restore metabolic parameters reduced by nitrate. When compared to the simultaneous administration of nitrate with GOS, the simultaneous administration of nitrate with GOS plus Candida kefyr lowered rumen methanogenesis to a negligible level, but did not decrease rumen and plasma nitrite accumulation as well as blood methaemoglobin formation. Thus, these results suggest that combination of nitrate with GOS may be a potent manipulator to suppress rumen methanogenesis with abating the hazards of nitratenitrite toxicity in ruminants.

Bacillus sp.유래 ${\beta}-mannanase$에 의한 $Gal^3Man_4(6^3-mono-{\alpha}-D-galacto-pyranosyl-{\beta}-mannotetraose)$ 조제 및 장내세균에 대한 생육활성 (Preparation of $Gal^3Man_4(6^3-mono-{\alpha}-D-galacto-pyranosyl-{\beta}-mannotetraose)$ by Bacillus sp. ${\beta}-mannanase$ and Growth Activity to Intestinal Bacteria)

  • Bacillus sp. 유래 ${\beta}-mannanase$의 brown copra meal에 대한 기질 특이성을 규명하기 위하여 정제효소 150 ml를 0.5% 기질에 $50^{\circ}C$, 24 hrs 가수분해후 TLC, FACE로 비교 검토한 결과 2종류의 galactosyl mannooligosaccharide로 구성되어 1차 activated carbon column chromatography을 이용해 250 ml/hr 유속으로 tube당 50ml씩 ethanol $0{\sim}30%$ linear gradient로 당을 분리하였다. Activated carbon column chromatography에 의한 당용액 0.2 ml와 5% phenol 0.2 ml를 가하여 혼합 후 $Conc.H_2SO_4$ 1 ml를 가하여 혼합한 후 20분간 방치하여 490 nm로 흡광도를 측정하여 TLC로 pattern을 검토한 후 $Gal^3Man_4$ 및 DP 7의 galactosyl manooligosaccharide의 fraction을 회수하여 2차 activated carbon column chromatography를 이용해 1차와 동일한 조전에서 분리 회수하여 중합도 5는 $Gal^3Man_4(63-mono-{\alpha}-D-galactopyranosyl-{\beta}-mannotetraose)$로 동정되었고, 중합도 7은 현재 동정중에 있다. Bifidobacterium속 균주(B. longum, B. bifidum)에 대한 생육활성을 비교 검토하기 위하여 MRS media에서 탄소원을 dextrose대신에 조제된 $Gal^3Man_4$를 첨가후 측정한 결과 $Gal^3Man_4$이 첨가되지 않은 MRS broth에 비해 생육촉진 활성을 보였다. 특히 B. longum에 대해서는 $Gal^3Man_4$를 dextrose대체 탄소원으로 처리시 10배의 생육활성이 증가하였다.

Food contributing to fermentable oligosaccharide, disaccharide, monosaccharide, and polyols intake in Korean adults

  • BACKGROUND/OBJECTIVES: The dietary intake of foods with fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAP) is known to adversely affect patients with irritable bowel syndrome (IBS). However, the effects of FODMAP have been studied predominantly among Western populations. This study aimed to identify foods high in FODMAP content which form a part of the Korean adult diet and obtain basic data for the preparation of IBS guidelines. SUBJECTS/METHODS: An online survey of 1,000 adults from the general population in the age group of 20 to 40 years was performed. Data from 787 participants (men, 386; women, 401) were analyzed. The general characteristics of the participants, health status, IBS diagnosis using the Rome III diagnostic criteria, semi-quantitative food frequency questionnaire findings, and food items causing symptoms were analyzed. RESULTS: Overall, 169 participants (21.5%) had IBS. The contribution of the FODMAP nutrients in both IBS and healthy groups was as follows: fructan > lactose > excess fructose > sorbitol > mannitol > galacto-oligosaccharides (GOS). The fructan intake was 4.6 ± 2.2 g/day and 4.3 ± 2.5 g/day in the IBS and healthy groups (P = 0.014), respectively. In the IBS group, the ratio of the intake of fructan to the total FODMAP intake was 39.5%, 29.8%, and 5.8% through onions, garlic, and bananas, respectively. CONCLUSIONS: Fructan was the FODMAP nutrient most consumed by Korean adults. Therefore, given the difference in the dietary habits of each country's population, the dietary guidelines for IBS should be country specific.

유청의 갈락토올리고당을 이용한 Bifidobacteria 의 생육촉진 (Use of Galactooligosaccharides from Cheese Whey for Growth of Bifidobacteria)

  • Effect of galactooligosaccharides produced by the $\beta$-galactosidase from Aapwefillua niger CAD 1 on the growth of Bifidobacterium infantis KCTC 3127 Bifidobacterium longum KCTC 3128 and Bifidobacterium bif-idum ATCC 11863 were investigated. Bifidbacterium infantis Bifidobacterium longum and Bifidobacterium bif-idum were in the logarithmic growth phase after 6hr incubation at 37$^{\circ}C$. Bifidobacterium infantis was in the stationary phase after 24hr incubation at 37$^{\circ}C$. The growth rate of B. bifidum containing galactooligo-saccharides and raffinose in MRS broth increased up to 18%, 8% and 7% compared to glucose galac-tose and lactose during 48hr incubation. The growth rate of B. infantis and B. longum contatining galacto-oligosaccharides and raffinose in MRS broth increased up to both 6% and 8% and both 13% and 10% compared to glucose and galactose during 48hr incubation.

Innovative Dietary Intervention Answers to Baby Colic

  • Purpose: The purpose of this paper is to evaluate the efficacy of a lactose- reduced synbiotic partial whey hydrolysate in formula fed infants presenting with colic and the impact of this dietary intervention in mean crying time and quality of life. Methods: Forty infants with infantile colic were treated during one month with parental reassurance and the intervention formula (partial whey hydrolysate, reduced lactose, Bifidobacterium lactis BB12 and galacto-oligosaccharides) and were compared to a control group of 20 infants with infantile colic treated with parental reassurance and a standard infant formula. Parents completed a quality of life (QoL) questionnaire assessing the burden of infantile colic. Wilcoxon test, t-test and Mann-Whitney test were used to compare QoL scores before and after intervention as well as between the intervention and control group. Results: At inclusion, duration of crying did not differ between both groups. Crying duration decreased with 2.7 hours (from 3.2 to 0.5 hours) in the intervention group while duration of crying decreased only with 1.2 hours in the control group (p<0.001). Stool composition became looser in the intervention group, but defecation frequency did not change. The median scores of the QoL questionnaire improved significantly in the intervention group for all parameters. In the control group, parameters improved significantly also but not for the parent-child and social interaction. The score changes were significantly greater in the intervention than in the control group. Conclusion: The intervention formula (partial whey hydrolysate, synbiotic, reduced lactose) significantly reduced the duration of crying and improved QoL of the parents and infants.

Effect of Galacto-mannan-oligosaccharides or Chitosan Supplementation on Cytoimmunity and Humoral Immunity in Early-weaned Piglets

  • Immunomodulatory feed additives might offer alternatives to antimicrobial growth promoters in pig production. This experiment was designed to determine the effects of dietary galacto-mannan-oligosaccharide (GMOS) and chitosan oligosaccharide (COS) supplementation on the immune response in early-weaned piglets. Forty 15-day-old piglets (Duroc$\times$Landrace$\times$Yorkshire) with an average live body weight of $5.6{\pm}0.51kg$ were weaned and randomly assigned to 4 treatment groups that were fed maize-soybean meal diets containing either basal, 110 mg/kg of lincomycin, 250 mg/kg of COS or 0.2% GMOS, respectively, over a 2-week period. Another six piglets of the same age were sacrificed on the same day at the beginning of the study for sampling, in order to obtain baseline values. Interleukin (IL)-1${\beta}$gene expression in peripheral blood monocytes, jejunal mucosa and lymph nodes, as well as serum levels of IL-1${\beta}$ IL-2 and IL-6, IgA, IgG, and IgM, were evaluated for 5 pigs from each group at 15 and 28 days of age. The results indicate that weaning stress resulted in decreases in serum antibody and cytokine levels. Dietary supplementation with GMOS or COS enhanced (p<0.05) IL-1${\beta}$gene expression in jejunal mucosa and lymph nodes, as well as serum levels of IL-1${\beta}$ IL-2, IL-6, IgA, IgG and IgM compared to supplementation with lincomycin. These findings suggest that GMOS or COS may enhance the cell-mediated immune response in early-weaned piglets by modulating the production of cytokines and antibodies, which shows that GMOS or COS have different effects than the antibiotic on animal growth and health.
