섬유질계 동시당화발효를 위한 내열성 융합 효모, Kluyveromyces marxianus CHY1612의 개발 (Development of Thermostable Fusant, CHY1612 for Lignocellulosic Simultaneous Saccharification and Fermentation)

  • To develop thermostable ethanol fermentative yeast strain for lignocellulosic simultaneous saccharification and fermentation, high ethanol producing yeast, Saccharomyces cerevisiae CHY1012 and thermostable yeast, Kluyveromyces marxianus CHY1703 were fused by protoplast fusion. The thermostable fusant, CHY1612 was identified as a Kluyveromyces marxianus by phenotypic and physiological characteristics, as well as molecular analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1 + 2 regions. For lignocellulosic ethanol production, AFEX pretreated barley straw at $150^{\circ}C$ for 90 min was used in a simultaneous saccharification and fermentation (SSF) process using thermotolerant CHY1612. The SSF from 16% pretreated barley straw at $43^{\circ}C$ gave a saccharification ratio of 90.5%, a final ethanol concentration of 38.5 g/L, and a theoretical yield of 91.2%. These results show that K. marxianus CHY1612 has potential for lignocellulosic ethanol production through simultaneous saccharification and fermentation with further development of process.

THP-1 세포에서 융합 발효 독활의 항염증 효과 (Anti-Inflammatory Effects of Fusion-Fermented Aralia continentalis Radix (fACR) on THP-1 cells)

  • 본 연구는 독활의 기능을 부가시키고자 발효과정을 통해 융합-발효 추물물을 얻어 항염증효과를 확인 하고자 한다. 독활을 이용하는데 한계가 있어 유산균주들을 활용한 발효과정을 통해 이용 가능한 균주를 선별하고, 독활의 열수 추출물과 비교하여 총 폴리페놀, 아미노산 총량, 주요 생리활성 미네랄 성분 분석 결과 발효과정을 통해 증가함을 확인하였다. 독활의 발효추출물의 항염증 효과를 확인하고자 관절염 염증반응과 관련된 사이토카인들을 측정하였고, 관절염시 주로 동반되는 연골조직 파괴를 억제 효능을 평가하기 위해 MMP-9 활성도를 확인하였다. 이러한 결과들은 유산균을 이용한 발효과정을 통해 독활의 주요 생리활성이 증가하고 염증반응 억제 효과도 증가하는 것으로 보여진다. 이 연구는 독활의 관절염 예방적 효과를 연구하는데 있어 기초 연구가 될 것으로 기대된다.

Development of Monacolin K-Enriched Ganghwayakssuk (Artemisia princeps Pamp.) by Fermentation with Monascus pilosus

  • Monacolin K-enriched ganghwayakssuk (Artemisia princeps Pamp.) was developed by fermentation with Monascus sp. Among the 15 Monascus spp. isolated previously from Monascus fermentation products, Monascus pilosus KMU108 produced 2,219 mg/kg of monacolin K during ganghwayakssuk fermentation with no detectable citrinin. The optimum concentrations of ganghwayakssuk and glucose determined from the response surface methodology (RSM) design were 2.2% and 3.8%, respectively. By applying these conditions, the monacolin K productivity was increased to 3,007 mg/kg after 15 days of fermentation. On the other hand, other characteristics such as the total content of flavonoids and phenolic compounds, and the antioxidant activity were relatively unchanged. Therefore, Monascus-fermented ganghwayakssuk is an excellent biomaterial for the development of functional foods because of its high level of monacolin K, known to lower cholesterol levels.

효모의 에탄올 생산능 및 세포 생존능의 증진을 위한 Rare-mating과 원형질체 융합 (Rare-Mating and Protoplast Fusion for the Improvement of Ethanol Producibility and Cell-Viability of Yeast)

  • 에탄올 발효능이 우수한 Saccharomyces에 속하는 4 균주를 가지고 여러 조합의 mating-pair 또는 fusion-pair를 만들고 이들 pair들로부터 만들어진 hybrid주들의 에탄올 생성능과 생존능을 통계적으로 분석한 결과, 에탄올 생성능에서는 차이가 없었으나, 생존능의 경우는 [S. kluveri $khl{\times}S.$ cerevisiae cp3]의 균주조합이 가장 우수한 hybrid를 낼 수 있는 것으로 나타났다. 실제로 에탄올 생성능과 잔당, 효율, 생존능에서 두루 우수한 균주는 [S. kluveri $khl{\times}S$ cerevisiae cp3] 조합에서 얻어진 융합주 clone No. 3가 에탄올 생성능 10.11%(w/v) 또는 12.81%(v/v), 잔당 3.53%(w/v), 생존능 62.65%, 발효 효율 92.2%로서 가장 발효능과 생존능이 우수한 균주로 선정되었다.

  • 본 연구는 Brevibacterium flavum ATCC 14067 및 Corynebacterium glutamicum ATCC 13032간의 protoplast fusion을 행하여 L-methionine의 생산성을 검토하고 발효조건을 개선하기 위하여 연속배양을 행하였다. N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) $500{\mu}g/ml$로 처리하였으며 B. flavum K-104($Thr\;Met\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}$)와 C. glutamicum B-70($Thr\;Hos\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}$)의 변이주를 분리하였다. 이들 변이주에 $500{\mu}g/ml$의 lysozyme을 처리하였을 때 원형질체 형성을 및 재생율은 각각 99% 및 $64{\sim}66%$를 나타내었으며 융합 빈도는 3% PVP를 함유한 35% PEG 용액에서 $3.5{\times}10^{5}$을 나타내었다. Sodium alginate로 고정화시킨 융합주 BFCG 37은 72시간 회분배양에서 0.89g/l의 methionine을 생산하였고 연속배양에서는 $18.75mg/^{1}hr\;^{1}$의 L-methionine를 안정적으로 생산할 수 있었다.

Ethanol Fermentation in Lactose Medium Using a Fusant Strain of Saccharomyces cerevisiae and Kluyveromyces fragilis

  • The fermentative characteristics in ethanol production from lactose, with increased ethanol tolerance, of a fusant yeast strain constructed by protoplast fusion of Saccharomyces cerevisiae and Kluyveromyces fragilis were studied. The ethanol tolerance of this strain was increased to 8.0%, compared with the parent K. fragilis. During batch ethanol fermentation the optimal cultivation conditions for this fusant yeast were an initial pH of 4.5, a culture temperature $30^\circ{C}$. stirring at 100 rpm without aeration in 10% lactose medium (supplied with 1.0% yeast extract). Using this fusant strain in whey fermentation to ethanol, maximum ethanol production reached 3.41% (w/v) (theoretical yield; 66.7%) after a 48 hour cultivation period.

  • An interspecific fusant strain, Streptomyces MS1 was obtained by protoplast fusion between S. peucetius subsp. caesius and S. platensis. We studied on the fermentation characteristics of the fusant strain. The fermentation products of the fusant MS1 was identical with S. peucetius, but its production of anthracycline was more stable than S. peucetius under various fermentation conditions in regard to acidogenesis of fermentation broth. The optimal medium composition for anthracycline production by fusant MS1 as follows: sucrose 2.0%, glucose 1.0%, soytone 0.7%, $CaCO_3$ 0.2%, $KH_2PO_4$ 0.013%, casamino acids 0.01%, $K_2SO_4$ 0.025%, $MaCl_2\;6H_2O$ 1.024%, 5M $CaCl_2\;5H_2O$ 0.4%, 1N NaOH 0.7%, 20% L-proline 1.5%. In this condition, the productivity of anthracycline was $80{\sim}100\;{\mu}g/ml$.

Studies on Improved Amylases Developed by Protoplast Fusion of Aspergillus species

  • Improved amylases were developed from protoplast fusants of two amylase-producing Aspergillus species. Twenty regenerated fusants were screened for amylase production using Remazol Brilliant Blue agar. Crude enzyme extracts produced by solid state fermentation of rice bran were assayed for activity. Three variable factors (temperature, pH and enzyme type) were optimized to increase the amylase activity of the parents and selected fusants using rice bran medium and solid state fermentation. Analysis of this optimization was completed using the Central Composite Design (CCD) of the Response Surface Methodology (RSM). Amylase activity assays conducted at room temperature and 80℃ demonstrated that Aspergillus designates, T5 (920.21 U/ml, 966.67 U/ml), T13 (430 U/ml, 1011.11 U/ml) and T14 (500.63 U/ml, 1012.00 U/ml) all exhibited improved function making them the preferred fusants. Amylases produced from these fusants were observed to be active over the entire pH range evaluated in this study. Fusants T5 and T14 demonstrated optimal activity under acidic and alkaline conditions, respectively. Fusants T13 and T14 produced the most amylase at 72 h while parents TA, TC and fusant T5 produced the most amylase after 96 h of incubation. Response surface methodology examinations revealed that the enzyme from fusant T5 was the optimal enzyme demonstrating the highest activity (1055.17 U/ml) at pH 4 and a temperature of 40℃. This enzyme lost activity with further increases in temperature. Starch hydrolysis using fusant T5 gave the highest yield of glucose (1.6158 g/100 ml). The significant activities of the selected fusants at 28 ± 2℃ and 80℃ and the higher sugar yields from cassava starch hydrolysis over their parental strains indicate that it is possible to improve amylase activity using the protoplast fusion technique.

Demethylteracycline 발효조건에 관한 연구 (A Study on the Conditions of Demethyltetracycline Fermentation)

  • Demeclocycline 생산균주를 세포 융합하여 얻은 변이주를 이용하여 demethyltetracycline을 생산하기 위한 발효조건에 대해 연구한 결과, 발효 최적온도는 $25^{\circ}C$였고, 초기pH는 6.7였으며 발효기간 중 pH를 일정하게 조절하면 생산성이 저하되었다. 또한 식균량이 많을 수록 초기 당 소모와 점도가 높았는데, 이는 식균량이 증가함에 따라 발효가 급격히 진행됨을 나타냈다. 식균량은 5(v/v)일 때 생산성이 가장 높았으며 전배양 시간과 전배양 pH가 중요한 요인임을 밝혔다.
