MhMTS and MhMTH are trehalose ($\alpha$-D-glucopyranosyl-[1,1]-$\alpha$-D-glucopyranose) biosynthesis genes of the thermophilic microorganism Metallosphaera hakonensis, and encode a maltooligosyltrehalose synthase (MhMTS) and a maltooligosyltrehalose trehalohydrolase (MhMTH), respectively. In this study, the two genes were fused in-frame in a recombinant DNA, and expressed in Escherichia coli to produce a bifunctional fusion enzyme, MhMTSH. Similar to the two-step reactions with MhMTS and MhMTH, the fusion enzyme catalyzed the sequential reactions on maltopentaose, maltotriosyltrehalose formation, and following hydrolysis, producing trehalose and maltotriose. Optimum conditions for the fusion enzyme-catalyzed trehalose synthesis were around $70^{\circ}C$ and pH 5.0-6.0. The MhMTSH fusion enzyme exhibited a high degree of thermostability, retaining 80% of the activity when pre-incubated at $70^{\circ}C$ for 48 h. The stability was gradually abolished by incubating the fusion enzyme at above $80^{\circ}C$. The MhMTSH fusion enzyme was active on various sizes of maltooligosaccharides, extending its substrate specificity to soluble starch, the most abundant natural source of trehalose production.
Demineralized boneparticle (DBP) has been widely used as and a powerful promoter of new bone growth. In this study, DBP sponges were chemically crosslinked and characterized for the potential application of tissue engineered scaffolds. The DBP sponges prepared by crosslinking with EDC. 0.1, 0.2 or 0.3% pepsin was applied to DBP dissolved in 3% (v/v) acetic acid aqueous solution for 48 hrs. The prepared sponges were crosslinked by 1, 5, 10, 50 or 100 mM of EDC solution concentration and then were lyophilized. The DBP sponges were characterized by SEM, FT-IR and DSC and analyzed in terms of their porosity and water absorption ability. The cellular viability and proliferation were assayed by MTT assay. Our investigation revealed that 0.2$\sim$0.3% of pepsin and 50$\sim$100 mM of EDC produced DBP sponges with good physical characteristics. In conclusion, DBP sponge prepared under these conditions is potentially useful for the applications of tissue construction.
Enzymatic glucosylation with glycosyltransferases can be used to regulate the water solubility of aglycone. The drawback of this process is the demand of UDP-glucose as a sugar donor. We made an in-frame fusion of the flavonoid O-glucosyltransferase (OsUGT-3) and sucrose synthase (AtSUS) genes. The resulting fusion protein, OsUGT3-AtSUS, was expressed in E. coli and purified. When sucrose and UDP were supplied, the fusion protein was able to convert quercetin into quercetin O-glucoside without the addition of UDP-glucose. In addition, UDP-glucose was recycled when sucrose was added to the reaction mixture. This fusion protein is useful for the enzymatic production of flavonoid O-glucosides.

The Center for Technology Fusion in Construction was established on Sep. 30, 2005, with the support of Korea Ministry of Construction and Transportation and Korea Institute of Construction and Transportation Technology Evaluation and Planning. It aims to develop the next generation of economic growth engine through the fusion of traditional construction technology and cutting-edge emerging technologies. To achieve this vision, the center tries to establish a system for the systematic construction research based on the fusion approach. The scope of the center focuses on improving the performance of construction project, including planning, design, construction, and maintenance. Along with the newly developed Korea Construction Technology Road Map, the center is expected to significantly contribute to the development of innovative construction technologies for the world-class Korean society.

It has been widely accepted that costal cartilage cells (CCs) have more excellent initial proliferation capacity than articular cartilage cells as well as the easiness for isolation and collection. This study demonstrated that CCs might be one of the substitutes for articular cartilage cells by tissue engineered cartilage. Poly(lactic-co-glycolic acid) (PLGA) has been extensively tested and used as scaffold material but it was limited by the low attachment of cells and the induction of inflammatory cells. Base on previous our studies, we confirmed demineralized bone particle (DBP) had the power of the reduction of inflammatory reaction and the stimulation proliferation of cells. We fabricated PLGA scaffold loaded with 10, 20, 40 and 80 wt% DBP and then tested the possibility of the regeneration of cartilage using CCs. Assays of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and scanning electron microscope (SEM) carried out to evaluate the attachment and proliferation of CCs in DBP/PLGA scaffolds. Glycosaminoglycan (sGAG) and collagen contents assay were conducted to confirm the effects of DBP on formation of extracellular matrix. This study demonstrated that DBP/PLGA scaffolds showed significant positive effects on cell growth and proliferation due to the vitality of DBP as well as the possibility of the application of CCs for tissue engineered cartilage.

Retinal pigment epithelium (RPE) plays an important role in maintaining the visual function and the degeneration of the RPE causes several retinal degeneration disease. In order to fabricate the suitable carrier for RPE transplantation, the hybrid poly(lactide-co-glycolide) (PLGA) film with hesperidin was prepared. Hesperidin has an anti-inflammatory and antioxidant characteristics. ARPE-19 was seeded on hesperidin/PLGA film and then, cell proliferation was determined by the MTT assay, and cell adhesion and cell morphology were confirmed by SEM. Also, RT-PCR was performed to confirm the expression of the specific genes, and AEC immunohistochemical staining was performed to determine the expression of RPE65. As a result, we confirmed that attachment, proliferation and phenotype maintenance of RPE cells were more excellent on hesperidin/PLGA film than PLGA film, thereby we were able to confirm the potential applications of hesperidin/PLGA film as tissue engineering carrier for regeneration of retina.
Because of insufficiency of energy resources and pollution of environment, it is necessary to develop alternative energy sources. Nuclear fission energy is used widely for source of electric Power but being restricted due to radioactivity problem. Nuclear fission is highlighted as the new generation of nuclear energy and researched worldwide because of low risk of radiation effect. The representatives of fusion research is China's EAST, KSTAR of Korea and ITER of world. Korea Superconducting Tokamak Advanced Research(KSTAR) project is on progress for the completion in August, 2007. In this study, the research of utility system for KSTAR be carried out. The utility system of KSTAR is consist of water cooling & heating system, $N_2$ gas system, DI water system, service water system and instrument air & auto control system. The progress of KSTAR utility system is under commissioning state after construction completion. The optimal operation scenario will be verified during commissioning and adopted to the KSTAR operation.

Objective : To identify and investigate differences in spinal fusion between the normal and osteopenic spine in a rat model. Methods : Female Sprague Dawley rats underwent either an ovariectomy (OVX) or sham operation and were randomized into two groups: non-OVX group and OVX group. Eight weeks after OVX, unilateral lumbar spinal fusion was performed using autologous iliac bone. Bone density (BD) was measured 2 days and 8 weeks after fusion surgery. Microcomputed tomography was used to evaluate the process of bone fusion every two weeks for 8 weeks after fusion surgery. The fusion rate, fusion process, and bone volume parameters of fusion bed were compared between the two groups. Results : BD was significantly higher in the non-OVX group than in the OVX group 2 days and 8 weeks after fusion surgery. The fusion rate in the non-OVX group was higher than that in the OVX group 8 weeks after surgery (p=0.044). The bony connection of bone fragments with transverse processes and bone formation between transverse processes in non-OVX group were significantly superior to those of OVX group from 6 weeks after fusion surgery. The compactness and bone maturation of fusion bed in non-OVX were prominent compared with the non-OVX group. Conclusion : The fusion rate in OVX group was inferior to non-OVX group at late stage after fusion surgery. Bone maturation of fusion bed in the OVX group was inferior compared with the non-OVX group. Fusion enhancement strategies at early stage may be needed to patients with osteoporosis who need spine fusion surgery.

Objective : Although iliac crest autograft is the gold standard for lumbar fusion, the morbidity of donor site leads us to find an alternatives to replace autologous bone graft. Ceramic-based synthetic bone grafts such as hydroxyapatite (HA) and b-tricalcium phosphate (b-TCP) provide scaffolds similar to those of autologous bone, are plentiful and inexpensive, and are not associated with donor morbidity. The present report describes the use of Polybone$^{(R)}$ (Kyungwon Medical, Korea), a beta-tricalcium phosphate, for lumbar posterolateral fusion and assesses clinical and radiological efficacy as a graft material. Methods : This study retrospectively analyzed data from 32 patients (11 men, 21 women) who underwent posterolateral fusion (PLF) using PolyBone$^{(R)}$ from January to August, 2008. Back and leg pain were assessed using a Numeric Rating Scale (NRS), and clinical outcome was assessed using the Oswestry Disability Index (ODI). Serial radiological X-ray follow up were done at 1, 3, 6 12 month. A computed tomography (CT) scan was done in 12 month. Radiological fusion was assessed using simple anterior-posterior (AP) X-rays and computed tomography (CT). The changes of radiodensity of fusion mass showed on the X-ray image were analyzed into 4 stages to assess PLF status. Results : The mean NRS scores for leg pain and back pain decreased over 12 months postoperatively, from 8.0 to 1.0 and from 6.7 to 1.7, respectively. The mean ODI score also decreased from 60.5 to 17.7. X-rays and CT showed that 25 cases had stage IV fusion bridges at 12 months postoperatively (83.3% success). The radiodensity of fusion mass on X-ray AP image significantly changed at 1 and 6 months. Conclusion: The present results indicate that the use of a mixture of local autologous bone and PolyBone$^{(R)}$ results in fusion rates comparable to those using autologous bone and has the advantage of reduced morbidity. In addition, the graft radiodensity ratio significantly changed at postoperative 1 and 6 months, possibly reflecting the inflammatory response and stabilization.
