곰팡이 센서(Fungal detector)를 이용한 알러지 환자 가정의 실내 환경 평가 - 사례연구 (Assessment of the Environmental Conditions in Patient's Houses with Allergy by Use of a Fungal Index - A Case Study)

  • The indoor environmental condition was assessed in houses with allergy (asthma and atopy) patients by use of a fungal detector. The fungal index was calculated from the growth rate of the sensor fungi in a fungal detector encapsulating the spores, Alternaria alternata S-78, Eurotium herbariorum J-183 and Aspergillus penicillioides K-712. Fungal indices were higher in asthma patient's houses than in control houses and Eurotium herbariorum showed the highest growth response among the sensor fungi. Dust mites allergen, Der f1, was also significantly high in allergy patient's houses where fungal indices above 10 were detected. A correlation was observed between the fungal indices and dust mite allergen proliferations in examined houses. Therefore, the fungal index can be a useful tool as an indirect indication for detecting chronic dampness that brings both contaminations by fungi and dust mite.

무제치늪에 자생하는 식물의 뿌리에서 분리한 내생진균의 군집분석 및 다양성 분석 (Community Analysis of Endophytic Fungal strains Isolated from the Roots of Plants Inhabiting Mujechi-neup)

  • 이 논문은 무제치늪에서 채집한 기장대풀(Isachne globosa Kuntze), 솔방울고랭이(Scirpus karuisawensis Makino), 이삭귀개(Utricularia racemosa Wall.), 좀개수염(Eriocaulon decemflorum Maxim.)등 4종의 식물 뿌리에서 서식하는 내생곰팡이의 분포 및 다양성을 확인하기 위해 조사하였다. 총 226 균주가 분리되었고, 3문(Phyla), 7강(Class), 10목(Order), 22과(Family), 31(genera)속으로 분리되었다. 내생진균에서 가장 많이 분리된 속은 Acephala (19.9%), Tolypocladium (16.3%), Neopestalotiopsis (11.5%), Perenniporia (7.1%) 순으로 확인하였다. 다양성지수 분석에 있어서, 이삭귀개(Ur)가 다른 식물 종에 비해 종 풍부도 (Menhinick's index = 2.37), (Margalef's index = 4.46)와, 종 균등도 (Simpson's index diversity = 0.91), 그리고 종 다양성(Shannon's indx = 2.57)로 높게 나왔다. 이러한 분석 결과를 봤을 때, 이삭귀개(Ur)에서 내생진균이 가장 다양하게 서식할 수 있는 것으로 보여진다.

Effects of Fungicides and Bioagents on Seed Mycoflora, Growth and Yield of Watermelon

  • Fungicides and antagonists were tested for their efficacy in the management of fungal pathogens of watermelon. The fungal species in different genera were isolated from the seeds of watermelon and their vulnerability was assessed against an array of chemicals and bioagents. Among the fungal pathogens, Fusarium species were effectively controlled by Bavistin. Topsin also showed the promising effects against all the fungal pathogens, and Dithane M-45 effectively controlled Didymella bryoniae. Seed treatment with antagonists like Trichoderma harzianum and T. viride improved the seed germination, seedling vigour and reduced the incidence of seed-borne fungal pathogens. Bavistin and Topsin among chemicals increased significantly the seed germination and vigour index. Trichoderma harzianum showed its efficacy against all Fusarium species and even stood effective than Captan and Blitox. However, Pseudomonas fIuorescens also showed promising effect against Didymella bryoniae over fungicides. Under field condition, Topsin and Dithane M-45 showed better yield than Bioagents.

Species Diversity of Endophytic Fungi Isolated from Taxus cuspidata Inhabiting Mt. Hallasan, Korea

  • Several endophytic fungal strains were isolated from Taxus cuspidata and identified by molecular analysis of the internal transcribed spacer and RNA polymerase II second largest subunit. This study aimed to determine the relative abundance and compare the species diversity of endophytic fungal communities within needle leaves and twigs. We identified a total of 49 endophytic fungal species. Notably, two species, Trichoderma dingleyae and Xylaria cubensis, were discovered to be previously unrecorded in Korea. The fungal communities in both plant tissues demonstrated distinct species composition. Differences were observed in the relative abundance and species diversity index between needle leaves and twigs. Our findings suggest that the host plant tissues influence the species diversity of endophytic fungal communities.

  • The anti-fungal activity of plant extractives was investigated and the lacquer extractives were selected. The lacquer extractives were coated on anti-fungal paper and confirmed its effect. Water repellency, tensile index and brightness of anti-fungal paper treated with natural extractives were similar to commercial pesticide-treated paper. The incidence of Fusicaladium leuieri MAGNUS of commercial pesticide-treated paper and anti-fungal paper treated with lacquer were similar but, the incidence of Botryosphaeria dothidea (Moug.) Ces & De Not and Gloeodes pomigena of those were increased 10-60 % than commercial pesticide-treated paper.

Diversity of Endophytic Fungi Associated with the Roots of Four Aquatic Plants Inhabiting Two Wetlands in Korea

  • A total of 4 aquatic plants, Eleocharis kuroguwai Ohwi, Hydrocharis dubia Backer, Salvinia natans All., and Zizania latifolia Turcz., were sampled from representative two wetlands of South Korea. A total of 38 endophytic fungal strains were isolated from aquatic plants native to the Daepyeong wetland, and 27 strains were isolated from the Jilnal wetland. The internal transcribed spacer regions of fungal isolates were sequenced and a phylogenetic analysis was performed. In addition, endophytic fungal diversity from each wetland and host plant species was deduced. A total of 25 fungal genera were purely isolated, and 16 fungal genera were isolated from each of the two wetlands. Commonly isolated genera from both wetlands were Aspergillus, Cladosporium, Clonostachys, Fusarium, Leptosphaeria, Penicillium, and Talaromyces. This study revealed that fungal diversity varied with environmental conditions and by host plant in representative two wetlands.

  • The production of maltose utilizing swollen extrusion starch seems to have many technical advantages, such as, high reaction rate and high yield, production of high purity concentrated maltose, and low energy consumption, over the conventional method utilizing liquefied starch. The characteristics of maltose formation in heterogeneous enzyme reaction system comtaining swollen extrusion starch was investigated using fungal $\alpha $-amylase. The influence of extrusion conditions on structure of extruded starch, such as, degree of gelatinization, water absorption index, and water solubility index was analyzed. The relationship between the structural features and maltose forming reaction was investigated, and the result was analyzed in terms of surface reaction of insoluble extruded swollen starch. The characteristics of maltose formation from swollen sxtrusion starch was compared using endo-type fungal $\alpha $-amylase and exo-type $\beta $anylase, and the structural trasformation of extruded starch was also observed to clarify the reaction mechanism.

Diversity and Plant Growth-Promoting Effects of Fungal Endophytes Isolated from Salt-Tolerant Plants

  • Fungal endophytes are symbiotic microorganisms that are often found in asymptomatic plants. This study describes the genetic diversity of the fungal endophytes isolated from the roots of plants sampled from the west coast of Korea. Five halophytic plant species, Limonium tetragonum, Suaeda australis, Suaeda maritima, Suaeda glauca Bunge, and Phragmites australis, were collected from a salt marsh in Gochang and used to isolate and identify culturable, root-associated endophytic fungi. The fungal internal transcribed spacer (ITS) region ITS1-5.8S-ITS2 was used as the DNA barcode for the classification of these specimens. In total, 156 isolates of the fungal strains were identified and categorized into 23 genera and two phyla (Ascomycota and Basidiomycota), with Dothideomycetes and Sordariomycetes as the predominant classes. The genus Alternaria accounted for the largest number of strains, followed by Cladosporium and Fusarium. The highest diversity index was obtained from the endophytic fungal group associated with the plant P. australis. Waito-C rice seedlings were treated with the fungal culture filtrates to analyze their plant growth-promoting capacity. A bioassay of the Sm-3-7-5 fungal strain isolated from S. maritima confirmed that it had the highest plant growth-promoting capacity. Molecular identification of the Sm-3-7-5 strain revealed that it belongs to Alternaria alternata and is a producer of gibberellins. These findings provided a fundamental basis for understanding the symbiotic interactions between plants and fungi.

Fungal Community Analyses of Endophytic Fungi from Two Oak Species, Quercus mongolica and Quercus serrata, in Korea

  • Fungal endophytes have been recorded in various plant species with a richness of diversity, and their presence plays an essential role in host plant protection against biotic and abiotic stresses. This study applied the Illumina MiSeq sequencing platform based on the amplification of fungal ribosomal ITS2 region to analyze fungal endophytic communities of two oak species (Quercus mongolica and Q. serrata) with different oak wilt disease susceptibilities in Korea. The results showed a total of 230,768 sequencing reads were obtained and clustered at a 97% similarity threshold into 709 operational taxonomic units (OTUs). The OTUs of Q. serrata were higher than that of Q. mongolica with the number of 617 OTUs and 512 OTUs, respectively. Shannon index also showed that Q. serrata had a significantly higher level of fungal diversity than Q. mongolica. Total of OTUs were assigned into 5 fungal phyla, 17 classes, 60 orders, 133 families, 195 genera, and 280 species. Ascomycota was the dominant phylum with 75.11% relative abundance, followed by Basidiomycota with 5.28%. Leptosillia, Aureobasidium and Acanthostigma were the most abundant genera detected in Q. serrata with the average relative abundance of 2.85, 2.76, and 2.19%, respectively. On the other hand, Peltaster, Cladosporium and Monochaetia were the most common genera detected in Q. mongolica with the average relative abundance of 4.83, 3.03, and 2.87%, respectively. Our results indicated that fungal endophytic communities were significantly different between two oak species and these differences could influence responses of host trees to oak wilt disease caused by Raffaelea quercus-mongolicae.

Cutaneous Microflora from Geographically Isolated Groups of Bradysia agrestis, an Insect Vector of Diverse Plant Pathogens

  • Larvae of Bradysia agrestis, an insect vector that transports plant pathogens, were sampled from geographically isolated regions in Korea to identify their cutaneous fungal and bacterial flora. Sampled areas were chosen within the distribution range of B. agrestis; each site was more than 91 km apart to ensure geographical segregation. We isolated 76 microbial (fungi and bacteria) strains (site 1, 29; site 2, 29; site 3, 18 strains) that were identified on the basis of morphological differences. Species identification was molecularly confirmed by determination of universal fungal internal transcribed spacer and bacterial 16S rRNA gene sequences in comparison to sequences in the EzTaxon database and the NCBI GenBank database, and their phylogenetic relationships were determined. The fungal isolates belonged to 2 phyla, 5 classes, and 7 genera; bacterial species belonged to 23 genera and 32 species. Microbial diversity differed significantly among the geographical groups with respect to Margalef's richness (3.9, 3.6, and 4.5), Menhinick's index (2.65, 2.46, and 3.30), Simpson's index (0.06, 0.12, and 0.01), and Shannon's index (2.50, 2.17, and 2.58). Although the microbial genera distribution or diversity values clearly varied among geographical groups, common genera were identified in all groups, including the fungal genus Cladosporium, and the bacterial genera Bacillus and Rhodococcus. According to classic principles of co-evolutionary relationship, these genera might have a closer association with their host insect vector B. agrestis than other genera identified. Some cutaneous bacterial genera (e.g., Pseudomonas) displaying weak interdependency with insect vectors may be hazardous to agricultural environments via mechanical transmission via B. agrestis. This study provides comprehensive information regarding the cutaneous microflora of B. agrestis, which can help in the control of such pests for crop management.
