Assessment of the Environmental Conditions in Patient's Houses with Allergy by Use of a Fungal Index - A Case Study (곰팡이 센서(Fungal detector)를 이용한 알러지 환자 가정의 실내 환경 평가 - 사례연구)

  • The indoor environmental condition was assessed in houses with allergy (asthma and atopy) patients by use of a fungal detector. The fungal index was calculated from the growth rate of the sensor fungi in a fungal detector encapsulating the spores, Alternaria alternata S-78, Eurotium herbariorum J-183 and Aspergillus penicillioides K-712. Fungal indices were higher in asthma patient's houses than in control houses and Eurotium herbariorum showed the highest growth response among the sensor fungi. Dust mites allergen, Der f1, was also significantly high in allergy patient's houses where fungal indices above 10 were detected. A correlation was observed between the fungal indices and dust mite allergen proliferations in examined houses. Therefore, the fungal index can be a useful tool as an indirect indication for detecting chronic dampness that brings both contaminations by fungi and dust mite.

Influence of Moisture on Mold Growth in Building Materials (건축자재 내의 수분이 곰팡이 성장에 미치는 영향)

  • Recently, the indoor air pollution by microbes such as fungi and mites have become a concern as important research topic on indoor air quality. Fungal growth is significantly affected by humidity. In this study, we examined the influence of relative humidity on the surface of building materials and the water content of building materials on the fungal growth rate by measuring the mycelium length of fungi in the fungal detector placed on the surface of building materials. As a result, even if the relative humidity on the surface of building materials is identical, the more water content of building materials is, the more fungi grow faster. It was suggested that fungal growth rate depends on not only the relative humidity on the surface of building materials but also the water content of building materials.

  • Aflatoxin $B_1$, $B_2$, $G_1$, and $G_2$were quantitatively detected by the high pressure liquid chromatography on a Micropak-CN column, with Hexane-THF-IPA-water, using a Lichrosorbpacked flowceil in the fluorometric detector. Under those conditions, the minimum detectable amount of aflatoxin $B_1$ was 0.2 ng. HPLC was used in determining amount of aflatoxins in the commercially manufactured soybean food and home-made Meju. Aflatoxin producing abilities of strains used in the industrially fermented soybean food were also studied with the HPLC technique. Although aflatoxin-like substances were detected in a few samples on TLC, they were not identified with the HPLC retention times of standard aflatoxins. The commercial fungal strains used in Korea had no aflatoxin producing abilities.

Bioequivalence of Terbina Tablet to Lamisil Tablet (Terbinafine 125 mg) (라미실 정(테르비나핀 125 mg)에 대한 터비나 정의 생물학적 동등성)

  • Terbinafine is an orally active antifungal agent as it inhibits the fungal enzyme squalene epoxidase, which is important in the early biosynthetic pathway of ergosterol. This leads to abnormal development of the fungal cell membrane. Bioequivalence of two terbinafine tablets, $Lamisil^{TM}$ (Novartis Korea Ltd.) and $Terbina^{TM}$ (Korean Drug Co., Ltd.), was evaluated according to the guidelines of Korea Food and Drug Administration (KFDA). Sixteen normal male volunteers, $23.56{\pm}1.75$ years old and $65.60{\pm}8.54\;kg$ of body weight, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one tablet containing 125 mg of terbinafine was orally administered, blood was taken at predetermined time intervals and the serum concentrations of terbinafine were determined using an HPLC method with UV detector. The pharmacokinetic parameters $(AUC_t,\;C_{max}\;and\;T_{max})$ were calculated and ANOVA test was utilized for the statistical analysis of parameters. The results showed that the differences in $AUC_t,\;C_{max}\;and\;T_{max}$ between two tablets based on $Lamisil^{TM}$, tablet were -2.53%, -2.98% and 8.13%, respectively. The powers $(1-{\beta})$ for $AUC_t,\;C_{max}\;and\;T_{max}$ were 85.21%, 98.21% and 93.11%, respectively. Minimum detectable differences $({\Delta})$ at ${\alpha}=0.1\;and\;1-{\beta}=0.8$ were all less than 20%. The 90% confidence intervals were all within ${\pm}20%$. All the parameters above met the criteria of KFDA for bioequivalence, indicating that $Terbina^{TM}$ tablet is bioequivalent to $Lamisil^{TM}$ tablet.

Monitoring of Aflatoxins in Herb Medicines (한약재 중 아플라톡신 Monitoring)

  • Our paper shows the results of 302 samples of herb medicines about fungal contamination at Yakyeang markets in Seoul. The sample medicines were treated VICAM pretreatment and analysed by post column derivatisation procedure(PHRED-HPLC) with a fluorescence detector. Aflatoxin B1 was founded from 50.3% of samples, aflatoxin B2 was 39.7%, aflatoxin G1 was 21.2% and aflatoxin G2 was 23.5%. The detected ranges of aflatoxin B1, B2, G1 and G2 were from 0.1 to $57.2\;{\mu}g/kg$, 0.1 to $42.6\;{\mu}g/kg$, 0.1 to $23.5\;{\mu}g/kg$ and 0.1 to $9.5\;{\mu}g/kg$ respectively. Among total samples, 26 samples contained aflatoxin B1 violated the regulation (less than 10 ug/kg) for aflatoxin B1 of KFDA. From the result, we could presumed that more than a half of samples were contaminated by aflatoxins. Therefore, it seems to be necessary that the new safety giudeline will be established aflatoxin B2, G1 and G2 from herb medicines as aflatoxin B1.

Bioequivalence of Mycosil Tablet to Lamisil Tablet (Terbinafine 125 mg) (라미실 정(테르비나핀 125 mg)에 대한 미코실 정의 생물학적 동등성)

  • Terbinafine has a primary fungicidal action mediated by squalene epoxidase inhibition. Treated fungi accumulate squalene while becoming deficient in ergosterol, an essential component of fungal cell membranes. Bioequivalence of two terbinafine tablets, $Lamisil^{TM}$ (Novartis Korea Ltd., Seoul, Korea) and $Mycosil^{TM}$ (Daewon Pharmaceutical Co., Ltd., Seoul, Korea), was evaluated according to the guidelines of Korea Food and Drug Administration (KFDA). Sixteen normal male volunteers ($20\sim29$ years old) were randomly divided into two groups and a randomized $2\times2$ cross-over study was employed. After oral administration of $Mycosil^{TM}\;or\;Lamisil^{TM}$ (125 mg terbinafine), blood samples were taken at predetermined time intervals and the serum terbinafine concentrations were determined using an HPLC method with UV/VIS detector. The pharmacokinetic parameters $(AUC_t,\;C_{max}\;and\;T_{max})$ were calculated and ANOVA was utilized for the statistical analysis. The results showed that the differences in $AUC_t,\;C_{max}\;and\;T_{max}$ between two tablets based on the $Lamisil^{TM}$ tablet were $-2.24\%,\;-7.68\%\;and\;2.92\%$, respectively. The powers %(1-\beta)\;for\;AVC_t,\;C_{max}\;and\;T_{max}\;were\;87.11\%,\;95.36\%\;and\;99.99\%$, respectively. Minimum detectable differences $(\Delta)\;and\;90\%$ confidence intervals were all less than $\pm20\%$. All these parameters met the criteria of KFDA for bioequivalence, indicating that $Mycosil^{TM}$ tablet is bioequivalent to $Lamisil^{TM}$ tablet.

Residual characteristic of tebuconazole and fludioxonil in Allium victorialis (Allium ochotense Prokh.) (소면적 재배작물 산마늘(Allium ochotense Prokh.) 중 살균제 Tebuconazole 및 Fludioxonil의 잔류특성)

  • In recent years, Allium victorialis has been extensively used as a pharmacological agent for various diseases in the form of anti-arteriosclerotic, anti-diabetic and anti-cancer. Allium victorialis is severely affected by various fungal diseases since it naturally grow in the shady and humid environments in Korea. In this case, different types of fungicides are applied to control the fungal diseases in Allium victorialis. The present study was aimed to determine the residual characteristics of two fungicides namely tebuconazole and fludioxonil on Allium victorialis. For this study, the fungicides were drenched soil on Allium victorialis in the cultivation area Pyeongchang by the standard (two thousand fold) and double (thousand fold) dilutions. At the end of $15^{th}$, $30^{th}$ and $40^{th}$ days samples were collected for residue analysis. Residues of tebuconazole and fludioxonil were analyzed using GC/NPD (Gas Chromatography/Nitrogen Phosphorus Detector) and their recovery were found to be 108.8~119.5% and 91.3~104.8%, respectively. The method of limits of quantification for both fungicides was $0.01mg\;kg^{-1}$. Further, the results of this study shows that the residue levels of both fungicides on Allium victorialis were <$0.01{\sim}0.12mg\;kg^{-1}$ and $0.01{\sim}0.09mg\;kg^{-1}$ and their % ADI (% Acceptable Daily Intake) were 17.44% and 25.75%, respectively. Based on the results obtained in this study, we suggest that the residue levels of both of the fungicides on Allium victorialis are safe and these fungicides can also be used to control fungal diseases in Allium victorialis.

Bioequivalence of Terbinex Tablet to Lamisil Tablet (Terbinafine 125mg) (라미실 정(테르비나핀 125mg)에 대한 터비넥스 정의 생물학적동등성)

  • Terbinafine is a synthetic allylamine that is available in an oral formulation and is used at a dosage of 250mg/day. It is used as an active antifungal agent and inhibits the fungal enzyme squalene epoxidase, which leads to the accumulation of the sterol squalene, which is toxic to the organism. The purpose of the present study was to evaluate the bioequivalence of two terbinafine tablets, Lamisil (Novartis Korea Ltd.) and Terbinex (C-TRI Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). Eighteen normal male volunteers, 26.00$\pm$2.57 year in age and 70.51$\pm$9.36 kg in body weight, were divided into two groups and a randomized 2${\times}$2 cross-over study was employed. After one tablet containing 125 mg of terbinafine was orally administered, blood was taken at predetermined time intervals and the concentrations of terbinafine in plasma were determined using HPLC with UV detector. Pharmacokinetic parameters such as AUC, $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in AUC, $C_{max}$ and $T_{max}$ between two tablets were -4.191%, 5.223% and -25.720%, respectively when calculated against the Lamisil, tablet. The powers (1-$\beta$) for AUC, $C_{max}$ and $T_{max}$ were 81%, 87% and below 60%, respectively. Minimum detectable differences(.il) at alpha=O.1 and 1-/3=0.8 were less than 20% (e.g., 19.72% and 17.77% for AUC and $C_{max}$, respectively). But minimum detectable differences($\Delta$) at alpha=0.1 and 1-$\beta$=0.8 for $T_{max}$ were more than 20% (e.g., 26.25%). The 90% confidence intervals were within $\pm$20% (e.g., -17.440∼9.06 and -6.713∼17.160 for AUC and $C_{max}$ respectively). But 90% confidence intervals for $T_{max}$ were not within $\pm$20% (e.g., -43.346∼8.083). Another ANOVA test was conducted for logarithmically transformed AUC and $C_{max}$. These results showed that there are no significant differences in AUC and $C_{max}$ between the two formulations: The differences between the formulations in these log transformed parameters were all for less than 20% (e.g., -4.19% and 5.22% for AUC and $C_{max}$, respectively). The 90% confidence intervals for the log transformed data were not the acceptance range of log 0.8 to log 1.25 in AUC but the acceptance range of log 0.8 to log 1.25 in $C_{max}$ (e.g., log 1.13∼log 1.50 and log 0.94-log 1.22 for AUC and $C_{max}$ respectively). The major parameters, AUC and $C_{max}$ met the criteria of KFDA for bioequivalence although $T_{max}$ did not meet the criteria of KFDA (1998 year) for bioequivalence, indicating that Onfran tablet is bioequivalent to Zofran tablet. But in another ANOVA test AUC did not meet the criteria of KFDA (2002) for bioequivalence but $C_{max}$ met the criteria of KFDA (2002 year) for bioequivalence.or bioequivalence.equivalence.equivalence.equivalence.

Analysis of trans-Resveratrol Contents of Grape and Grape Products Consumed in Korea (포도와 포도 가공품에 함유되어 있는 trans-resveratrol의 함량 분석)

  • Resveratrol is natually occurring phytoalexin compounds produced by grape berries, peanuts, and their products in response to stress such as fungal infection, heavy metal ions or UV irradiation. The objective of this study was to develop a reliable high-performance liquid chromatographic method for the quantitative determination of trans-resveratrol in grape and its products. The trans-resveratrol was separated isocratically on Nucleosil 100-5 C18 column, using a mobile phase containing acetonitrile : water (40 : 60, v/v), detected by UV detector at 306 nm and the flow rate was 0.3 mL/min. Under this analytical condition, the recoveries of trans-resveratrol in grape, wine, and grape juice were 92.35, 104.72, and 91.08, respectively. Limit of detection in grape, wine, and grape juice were 14.5 ng/g, 3.62 ng/mL, and 4.02 ng/mL. Also, limit of quantitation in grape, wine, and grape juice were 14.8 ng/g, 3.69 ng/mL, and 4.10 ng/mL. Assay values of 32 grape varieties, 9 wines, and 9 grape juices were ranged from trace amount to $207.1\;{\mu}g/100\;g$, from 5.4 to $275.7\;{\mu}g/L$, and from 63.3 to $751.6\;{\mu}g/L$, respectively.

Varietal Analysis and Quantification of Resveratrol in Mulberry Fruits (뽕나무 계통별 오디의 레스베라트롤 함량 분석)

  • Resveratrol is naturally occurring phytoalexin compounds produced by grape berries, peanuts, and their products in response to stress such as fungal infection, heavy metal ions or UV irradiation. The objective of this study was to develope a reliable high performance liquid chromatographic (HPLC) method for the quantitative determination of trans-resveratrol in mulberry fruit. Samples were extracted in 80% MeOH and filtered with $0.45{\mu}m$ syringe filter. The transresveratrol was separated Waters $C_{18}$ column, using a mobile phase containing 0.025% trifluroacetic acid in 5% acetonitril and 0.035% trifluroacetic acid in 50% acetonitril, detected by photodiode array detector (PDA) at 254 nm and the flow rate was 1ml/min. Under this analytical condition, the mean content of mulberry fruits (fifty varieties) was $777.3{\pm}585.9ppm$. Among the tested samples, 'Mansaengbaekpinosang (II)' was the highest level in 3450.6 ppm. However four accessions including 'Gukbu', 'Sabangso (I)', 'Simseol' and yield mulberry fruit were not able to detected. Eight suitable varieties selected for the production of fruit were 'Jeolgokchosaeng (Chungbuk)' 777.8 ppm, 'Dangsang 7' 771.1 ppm, 'Jangsosang' 133.9 ppm, 'Susungppong' 31.1 ppm, 'Suwonnosang' 639.7 ppm, 'Palcheongsipyung' 1475.9 ppm, 'Kangsun' 864.0 ppm, and 'Jukcheonchosaeng' 1458.5 ppm. 'Daesungppong' which was the first authorized variety for the production of mulberry fruit was 1236.7 ppm. In conclusion, these results suggest that mulberry including fruit and leaf may a good new resource for resveratrol production.
