A Contrastive Study on the Change of Rural Houses of the Han Chinese and Korean-Chinese in Yanbian Area of China (중국연변지역 조선족과 한족 농촌주택의 변화에 관한 비교연구)

  • This study is aimed to suggesting a hypothesis about the process of improvement of houses of the Korean-Chinese, comparing how Han Chinese and Korean-Chinese houses changed in Yanbian Area of China. This draws the conclusion by analyzing the field survey about residence of the Han Chinese and the Korean-Chinese in Yanbian Area of China. First of all, the characteristic of residence style change and the plan of rural houses of the Han Chinese are in the following. The plan of houses has been changed from allocating rooms from side to side with the kitchen as the center to linking each functional spaces through the aisle. And the main bedroom of houses becomes the central space for family members instead of Kang (partial Ondol) by expanding the floor heating system under the whole floor in houses. Next, the feature of residence style changes of the Han Chinese and the Korean-Chinese is as follows. In 1970s barns disappeared from houses of the Korean Chinese, but houses of the Han Chinese were not much changed. From 1980s to 1990s, spaces linking each rooms with aisles had appeared for comfortableness of residence and respecting each individual's privacy. On the other hand, The houses of the Korean-Chinese make a whole big space by getting rid of walls between the rooms. And after 2 thousand years later, Han Chinese houses have a bedroom with floor heating system. Both houses of the Korean-Chinese and the Han Chinese are changed to have a residence style which taking sedentary style and standing style.

Composition of Isoflavone, Phytic Acid, and Saponins in Hypocotyls and Cotyledons of Six Traditional Korean Soybeans

  • The objective of present study was to examine the composition of functional components (isoflavones, phytic acid, and saponin) in hypocotyls and cotyledons of six traditional Korean soybeans, namely Cheongtae, Seoritae, Jinjoori, Subaktae, Yutae, and Huktae. Three 'family' of isoflavones, as four chemical structures were present in hypocotyls and cotyledons of six soybean cultivars: the aglycones genistein, daidzein, and glycitein; the glycosides genistin, daidzin, and glycitin; the acetylglycosides 6"-O-acetylgenistin, 6"-O-acetyldaidzin, and 6"-O-acetylglycitin; and the malonylglycosides 6"-O-malonylgenistin, 6"-O-malonyldaidzin, and 6"-O-malonylglycitin. Isoflavone contents of hypocotyls and cotyledons differed among the cultivars, and glucosides and malonylglucosides accounted for more than 90% of the total phytoestrogens, with the remaining $1\sim7%$ taken up by aglycones. Concentrations of isoflavones in cotyledons were approximately about $10\sim20%$ of respective hypocotyls. Contents of phytic acids in hypocotyls and cotyledons of the selected soybean cultivars were $1.21\sim1.70%$ and $2.59\sim3.01%$, respectively. Hypocotyls of Seoritae showed the lowest content of phytic acid with 1.21%, while cotyledons of Cheongtae showed the highest content with 3.01%. The sapogenol concentrations ranged from $13.58mg/100g\sim20.82mg/100g$ for hypocotyls and $0.95mg/100g\sim2.55mg/100g$ for cotyledons showing that concentrations of saponin are $7\sim10$ times higher in hypocotyls than in cotyledons of respective soybeans. For both hypocotyls and cotyledons the sapogenol A were present in higher concentrations than soyasapogenol B.

A Study on Serious Game Production Based on Jewish Play (유대인의 놀이를 주제로 한 기능성 게임 제작 방안에 관한 연구)

  • The Jews has been cited as the most influential nations in the world of politics, economy, and culture in many areas. For these reasons, the study of Jewish economic life such as child-rearing, family life, and religious life is getting expanded to various areas. In this paper, we deal with the Jews's play for the various areas. Play is the best way to see the consciousness of a certain age on learning the structure and lifestyle. Therefore, the research is described to produce serious games based on by study of Jewish play using information in the internet, researched paper. Then, the investigated play's role of technology for production of functional game plans are discussed. As a result, the Jews play: 1) the history, traditions of wisdom, tools, 2) a medium for delivering educational messages, 3) the formation of community tools emotions 4) promise to play a role as a tool for learning. The development and Release of serious games based on Jews play can lead to a positive social learning effect.

Anger Expression Style by the Enneagram Personality Types of College Students (남녀대학생의 에니어그램 성격유형에 따른 분노표현 방식)

  • The purpose of this study is to investigate the relationship between the Enneagram personality types and the anger expression styles, and to see how the relationship depends on the gender. The subjects, selected by the convenience sampling method, are 315 college students. The instrument is the Korean Enneagram Personality Types(Yoon 1999), which categorizes the personal types into the gut-center, the heart-center and the head-center. On the other hand, the anger expression styles, which is the anger-control as a functional role, and the anger-out and the anger-in as a dysfunctional role, is measured by the Korean version of STAXI(Chon et al 1998). The major findings of the study are as follows. First, comparing the personality types of students, the ratio of the students of the gut-center, that of the head-center and that of the heart-center types are in descending order. Second, It is found that students rely on the anger-out more than the other two expression styles. There also exist the gender differences in terms of the level of the anger expression: female students tend to express the anger-out and the anger-control significantly more than male students. Third, the relationship between the Enneagram personality types and the anger expression styles of students are statistically significant such that students of the gut-center style express the anger-out more severely than the head-center and the heart-center. Therefore, the findings from the study may become the basis on which the education program is designed for the sake of the psychological adjustment of college students, especially taking into account the gender differences.

Sturcture of the Rice Glutelin Multigene Family and Its Expression (쌀 Glutelin 유전자군의 구조 및 발현조절)

  • Plants store a significant amount of their nitrogen, sulfur and carbon reserves as storage proteins in seed tissues. The major proteins present in rice seeds are the glutelins. Glutelins are initially synthesized at 4-6 days postanthesis and deposited into protein bodies via Golgi apparatus. Based on nucleic acid sequences and Southern blot analysis, the three isolated glutelin genomic clones were representative members of three gene subfamilies each containing 5 to 8 copies. A comparison of DNA sequences displayed by relevant regions of these genomic clones showed that two subfamilies, represented by clones, Gt1 and Gt2, were closely, related and probably evolved by more recent gene duplication events. The 5' flanking and coding sequences of Gt1 and Gt2 displayed at least 87% homolgy. In contrast, Gt3 showed little or no homolgy in the 5' flanking sequences upstream of the putative CAAT boxes and exhibited significant divergence in all other portions of the gene. Conserved sequences in the 5' flanking regions of these genes were identified and discussed in light of their potential regulatory role. The derived primary sequences of all three glutelin genomic clones showed significant homology to the legume 11S storage proteins indicating a common gene origin. A comparison of the derived glutelin primary sequences showed that mutations were clustered in three peptide regions. One peptide region corresponded to the highly rautable hypervariable region of legume peptide region of legume 11S storage proteins, a potential target area for protein modification. Expression studies indicated that glutelin mRNA transcripts are differentially accumulated during endosperm development. Promoterss of Gt2 and Gt3 were functional as they direct transient expression of chloramphenicol acetyltransferase in cultured plant cell.

Effect of Sambucus sieboldiana Extract on the Cell Growth and Extracellular Matrix Formation in Osteoblast Cells

  • Sambucus sieboldiana (SS) is a member of the family Caprifoliaceae and has been recommended as a functional material because of its several bioactivities. Although numerous literatures are available on the pharmacological and biological activities, the biological activity of SS in bone regeneration process has not yet been well-defined. Therefore, in this study, the effect of SS was investigated in the proliferation and differentiation of MC3T3-E1 osteoblastic cell line. The treatment of SS did not significantly affect the cell proliferation in MC3T3-E1 cells. SS significantly accelerated the mineralization and significantly increased the expression of alkaline phosphatase (ALP) and osteocalcin (OC) mRNAs, compared to the control, in the differentiation of MC3T3-E1 cells. SS significantly accelerated the decrease of osteonectin (ON) mRNA expression as compared with the control in a time-dependent manner in the differentiation of MC3T3-E1 cells. These results suggest that the SS facilitate the osteoblast differentiation and mineralization in MC3T3-E1 osteoblastic cells. Therefore, there may be potential properties for development and clinical application of bone regeneration materials.

cis-Prenyltransferase interacts with a Nogo-B receptor homolog for dolichol biosynthesis in Panax ginseng Meyer

  • Background: Prenyltransferases catalyze the sequential addition of isopentenyl diphosphate units to allylic prenyl diphosphate acceptors and are classified as either trans-prenyltransferases (TPTs) or cis-prenyltransferases (CPTs). The functions of CPTs have been well characterized in bacteria, yeast, and mammals compared to plants. The characterization of CPTs also has been less studied than TPTs. In the present study, molecular cloning and functional characterization of a CPT from a medicinal plant, Panax ginseng Mayer were addressed. Methods: Gene expression patterns of PgCPT1 were analyzed by quantitative reverse transcription polymerase chain reaction. In planta transformation was generated by floral dipping using Agrobacterium tumefaciens. Yeast transformation was performed by lithium acetate and heat-shock for $rer2{\Delta}$ complementation and yeast-two-hybrid assay. Results: The ginseng genome contains at least one family of three putative CPT genes. PgCPT1 is expressed in all organs, but more predominantly in the leaves. Overexpression of PgCPT1 did not show any plant growth defect, and its protein can complement yeast mutant $rer2{\Delta}$ via possible protein-protein interaction with PgCPTL2. Conclusion: Partial complementation of the yeast dolichol biosynthesis mutant $rer2{\Delta}$ suggested that PgCPT1 is involved in dolichol biosynthesis. Direct protein interaction between PgCPT1 and a human Nogo-B receptor homolog suggests that PgCPT1 requires an accessory component for proper function.

Overexpression of ginseng patatin-related phospholipase pPLAIIIβ alters the polarity of cell growth and decreases lignin content in Arabidopsis

  • Background: The patatin-related phospholipase AIII family (pPLAIIIs) genes alter cell elongation and cell wall composition in Arabidopsis and rice plant, suggesting diverse commercial purposes of the economically important medicinal ginseng plant. Herein, we show the functional characterization of a ginseng pPLAIII gene for the first time and discuss its potential applications. Methods: pPLAIIIs were identified from ginseng expressed sequence tag clones and further confirmed by search against ginseng database and polymerase chain reaction. A clone showing the highest homology with pPLAIIIβ was shown to be overexpressed in Arabidopsis using Agrobacterium. Quantitative polymerase chain reaction was performed to analyze ginseng pPLAIIIβ expression. Phenotypes were observed using a low-vacuum scanning electron microscope. Lignin was stained using phloroglucinol and quantified using acetyl bromide. Results: The PgpPLAIIIβ transcripts were observed in all organs of 2-year-old ginseng. Overexpression of ginseng pPLAIIIβ (PgpPLAIIIβ-OE) in Arabidopsis resulted in small and stunted plants. It shortened the trichomes and decreased trichome number, indicating defects in cell polarity. Furthermore, OE lines exhibited enlarged seeds with less number per silique. The YUCCA9 gene was downregulated in the OE lines, which is reported to be associated with lignification. Accordingly, lignin was stained less in the OE lines, and the expression of two transcription factors related to lignin biosynthesis was also decreased significantly. Conclusion: Overexpression of pPLAIIIβ retarded cell elongation in all the tested organs except seeds, which were longer and thicker than those of the controls. Shorter root length is related to auxinresponsive genes, and its stunted phenotype showed decreased lignin content.

  • Raf protein kinases and protein kinase C belong to serine/threonine-specific proteins in the cytoplasin, and are similar to each other in functional structure and the aspect of the distribution of celI. The distribution of raf protein kinase in the cerebrum of Geoclemys reevesfi as studied by using the antibodies against a-raf and c-raf protein kinase which induce the expression of raf fainily oncogenes. In general, raf protein kinases were distributed in such restricted regions as the general pallium, hippocampal formation, pdmordiuin hippocampi,nucleus of lateral olfactory tract, basal amygdaloid nucleus, and bed of stria terminalis. Immunological labeling of c-raf protein kinase was more widespread than that of a-raf. However, the intensity of the labeling of c-raf was lower than that of a-raf. The spherical cells of basal amygdaloid nucleus is a ring-like form, because only the cytoplasm was imunolabeled. Especially, c-raf protein kinase occurred in the cells which contained protein kinase C abundandy such as pyramidal cells and Purkinje cells. This suggests that a- and e-raf protein kinases may synegistically induce carclnoma with myc gene which is activated by protein kinase C.

Crystal Structure of Hypothetical Fructose-Specific EIIB from Escherichia coli

  • We have solved the crystal structure of a predicted fructose-specific enzyme $IIB^{fruc}$ from Escherichia coli ($EcEIIB^{fruc}$) involved in the phosphoenolpyruvate-carbohydrate phosphotransferase system transferring carbohydrates across the cytoplasmic membrane. $EcEIIB^{fruc}$ belongs to a sequence family with more than 5,000 sequence homologues with 25-99% amino-acid sequence identity. It reveals a conventional Rossmann-like ${\alpha}-{\beta}-{\alpha}$ sandwich fold with a unique ${\beta}$-sheet topology. Its C-terminus is longer than its closest relatives and forms an additional ${\beta}$-strand whereas the shorter C-terminus is random coil in the relatives. Interestingly, its core structure is similar to that of enzyme $IIB^{cellobiose}$ from E. coli ($EcIIB^{cel}$) transferring a phosphate moiety. In the active site of the closest $EcEIIB^{fruc}$ homologues, a unique motif CXXGXAHT comprising a P-loop like architecture including a histidine residue is found. The conserved cysteine on this loop may be deprotonated to act as a nucleophile similar to that of $EcIIB^{cel}$. The conserved histidine residue is presumed to bind the negatively charged phosphate. Therefore, we propose that the catalytic mechanism of $EcEIIB^{fruc}$ is similar to that of $EcIIB^{cel}$ transferring phosphoryl moiety to a specific carbohydrate.
