Cloning and Phylogenetic Characterization of Coat Protein Genes of Two Isolates of Apple mosaic virus from ¡?Fuji¡? Apple

  • Apple mosaic virus (ApMV), a member of the genus Ilarvirus, was detected and isolated from diseased 'Fuji' apple (Malus domestica) in Korea. The coat protein (CP) genes of two ApMV strains, denoted as ApMV-Kl and ApMV-K2, were amplified by using the reverse transcription and polymerase chain reaction (RT-PCR) and were analyzed thereafter. The objectives were to define the molecular variability of genomic information of ApMV found in Korea and to develop virus-derived resistant gene source for making virus-resistant trans-genic apple. RT-PCR amplicons for the APMVS were cloned and their nucleotide sequences were determined. The CPs of ApMV-Kl and ApMV-K2 consisted of 222 and 232 amino acid residues, respectively. The identities of the CPs of the two Korean APMVS were 93.1% and 85.6% at the nucleotide and amino acid sequences, respectively. The CP of ApMV-Kl showed 46.1-100% and 43.2-100% identities to eight different ApMV strains at the nucleotide and amino acid levels, respectively. When ApMV-PV32 strain was not included in the analysis, ApMV strains shared over 83.0% and 78.6% homologies at the nucleotide and amino acid levels, respectively. ApMV strains showed heterogeneity in CP size and sequence variability. Most of the amino acid residue differences were located at the N-termini of the strains of ApMV, whereas, the middle regions and C-termini were remarkably conserved. The APMVS were 17.(1-54.5% identical with three other species of the genus Ilarviyus. ApMV strains can be classified into three subgroups (subgroups I, II, and III) based on the phylogenetic analysis of CP gene in both nucleotide and amino acid levels. Interestingly, all the strains of subgroup I were isolated from apple plants, while the strains of subgroups II and III were originated from peach, hop, or pear, The results suggest that ApMV strains co-evolved with their host plants, which may have resulted in the CP heterogeneity.

주간 절단시기 및 생장조절제를 이용한 '후지'/M9 사과나무 수관 상단부 생장조절 (Growth Control of Upper Part in 'Fuji'/M.9 Apple Tree Canopy by Cutting Time of Trunk and Plant Growth Regulators)

  • BACKGROUND: The vigorous shoot growth in upper part of apple tree canopy leads to poor fruit quality and flower bud formation in lower part of canopy. So, this study was conducted to develop the proper control method about the shoot growth in upper part of apple tree canopy. METHODS AND RESULTS: Trunks of 'Fuji'/M9 apple trees were cut (back pruned) to 2.5 m in tree height on 11 February (dormant) or 12 April (full bloom). Naphthalene acetic acid (NAA) was applied at 2.0% to cut surface when trunk was pruned. Prohexadione-calcium (Pro-Ca) was sprayed at 250 mg/L above 2.0 m in tree height at 23 April (petal fall). The NAA or Pro-Ca application after trunk was pruned at dormant (TR-2 and TR-3) significantly reduced shoot growth in upper part of canopy compared with the control (tree was only pruned at dormant, TR-1), but the percent of shoots showing the secondary growth of TR-3 was higher over 2 times than that of TR-2. The reduction of shoot growth in upper part of canopy by TR-2 and TR-3 increased the fruit red color from the lower part in the treating year and blooming of the lower part in the following year. CONCLUSION: Applying 2.0% NAA to cut surface of pruned apple trunk at dormant was the most effective way for stabilization of the tree vigor in upper part of the canopy in a high density apple orchard.

영양진단을 위한 신속한 엽분석 방법으로서 근적외분광분석기의 이용 (Application of Near Infrared Reflectance Spectroscopy as a Rapid Leaf Analysis Method to Evaluate Nutritional Diagnosis in Apple (Malus Domestica Borkh, Fuji) and grape(Vitis Labrusca, Campbell Early))

  • 근적외 분광분석기를 이용하여 과수의 영양진단을 위한 신속한 엽분석 수단으로 활용하고자 본 시험을 수행하였다. 사과 '후지'품종 잎 177점, 포도 '캠벨얼리' 품종 잎130점의 시료를 이용하여 1,100~2.500nm까지 근적외영역의 흡수스펙트럼을 측정하였다. 총질소 분석은 살리실산-황산으로 분해후 켈달 증류하여 측정하였다. 근적외 스펙트럼과 총 질소함량과의 상관분석을 이용, 검량식을 개발하였고 화학분석한 결과에 대한 예측식을 구하였다. 사과는 중회귀계수 0.965, 검량식의 표준오차 0.086, 포도는 중회귀계수 0.926, 검량식의 표준오차 0.152이었다. 개발된 검량식을 이용하여 예측식을 작성한 결과는 사과의 경우 0.360, 포도 0.210의 결과를 얻었다. 지역간, 년차간 발생되는 변이가 보정된다면 신속한 엽분석 수단이 될 것으로 판단된다.

CA 저장을 위한 사과의 최적 수확시기 결정 (Determination on the Optimal Harvest Date of Apples for CA Storage)

  • This study was carried out for the establishment of optic harvest date and storage condition of 'Fuji' apple for CA storage. Apples were picked at 10 day intervals from 17 Sept to 27 Oct.. Apples were analyzed for respiration rata, weight flesh firmness, titratable acidity, soluble solid, total sugar, reducing sugar, surface color and thiault value. In 1990, the optimal harvest season could be judged between 17 Oct. and 27 Oct. from the results of respiration rate rise and change of quality properties. Apples harvested on 27 Oct. wert stored at 2$^{\circ}C$ and 90~95% RH in 1%O2 + 3%CO2, 3%O2 + 3%CO2 and air for 9 months. After 9 months of storage, apples kept in CA storages had beater quality than apples kept in refrigeration storage. Especially, apples kept in 1%O2, + 3%CO2 were fumer and more acid than those kept in 3%O2 + 3%CO2 At the sensory evaluations, apples stored in CA were rated higher quality than apples stared refrigeration.

First Report of Two Colletotrichum Species Associated with Bitter Rot on Apple Fruit in Korea - C. fructicola and C. siamense

  • Bitter rot caused by the fungal genus Colletotrichum is a well-known, common disease of apple and causes significant yield loss. In 2013, six fungal strains were isolated from Fuji apple fruits exhibiting symptoms of bitter rot from Andong, Korea. These strains were identified as Colletotrichum fructicola and C. siamense based on morphological characteristics and multilocus sequence analysis of the internal transcribed spacer rDNA, actin, calmodulin, chitin synthase, and glyceraldehyde-3-phosphate dehydrogenase Pathogenicity tests confirmed the involvement of C. fructicola and C. siamense in the development of disease symptoms on apple fruits. This is the first report of C. fructicola and C. siamense causing bitter rot on apple fruit in Korea.

First Report of Fruit Rot Caused by Fusarium decemcellulare in Apples in Korea

  • In 2014, abnormal brown spots were observed on Hongro apples in fields in Gyeongsangbuk-do Province and during low-temperature storage. The spots were round, blight brown, and different from the symptoms of previously reported apple diseases. A fungal pathogen was isolated and cultured on potato dextrose agar, and it was morphologically similar to Fusarium decemcellulare. A pathogenicity test showed the same brown spots on both wounded and unwounded Hongro and Fuji apple cultivars. RPB1 and RPB2 sequences of F. decemcellulare KNU-GC01 matched with those of F. decemcellulare NRRL 13412 (98.3% and 97.6% similarities, respectively); both strains clustered together in the phylogenetic tree, indicating their close relationship at the species level. Therefore, F. decemcellulare is a newly reported pathogen that causes brown spots on apples in Korea.

Unreported Post-harvest Disease of Apples Caused by Plenodomus collinsoniae in Korea

  • This study was conducted to isolate and identify the fungal pathogen caused unreported post-harvest disease on apples (cv. Fuji) fruit in Korea. The disease symptoms on apples appeared as irregular, light to dark brown, slightly sunken spots. The three fungal strains were isolated from infected tissues of apple fruits and their cultural and morphological characteristics were completely consistent with those of Plenodomus collinsoniae. The phylogenetic analysis using the internal transcribed spacer (ITS) regions, beta-tubulin (TUB), and the second largest subunit of RNA polymerase II (RPB2) sequences revealed the closest relationship of the isolates with Plenodomus collinsoniae at the species level. The pathogenicity test showed the same dark brown spots on Fuji apple cultivar. Therefore, P. collinsoniae is a newly reported fungal agent causing post-harvest disease on apples in Korea.

굴 껍데기에서 개미산으로 추출한 칼슘화합물과 활성제의 수관살포가 사과 '후지' 과실의 칼슘농도, 과피 형태 및 품질에 미치는 영향 (Effects of Tree-spray of Calcium Formate Compound Extracted from Oyster Shell and Active Agent on the Calcium Concentration, Fruit Skin Shape, and Quality of 'Fuji' Apple Fruit)

  • 본 연구는 굴 껍데기로 제조한 개미산 칼슘화합물에 몇 가지 활성제를 첨가한 용액 수관살포가 사과 '후지' 과실의 칼슘농도, 과점의 발육 및 품질에 미치는 영향을 구명코자 실시하였다. 굴 껍데기로부터 추출한 칼슘 화합물(Os-CaF, $52.4mg{\cdot}kg^{-1}$)을 수관살포하였을 때 조직으로의 칼슘전이 효과는 무처리에 비하여 잎, 과피 및 과육에서 현저한 증가를 보였다. 그리고 Os-CaF에 활성제를 첨가하여 살포하였을 때 '후지' 과실의 칼슘함량은 증가되었다. 특히, 잎에서는 ascorbic acid, polyvinyl alcohol을, 과피에서는 Ag-colloidal, polyvinyl alcohol을, 과육에서는 ascorbic acid, Ag-colloidal, polyvinyl alcohol을 첨가한 칼슘화합물이 칼슘 흡수량을 현저히 증가시켰다. 칼슘화합물의 농도 및 활성제 종류에 따른 과실품질(과중, 경도, 가용성고형물, 산 함량, 과피색)은 차이가 없었다.

결로 방지 필름으로 포장한 '후지' 사과의 신선도 (Freshness of 'Fuji' Apples Packed Anti-fogging Agents Film)

  • 결로 방지 처리한 필름의 사과의 선도유지효과를 조사하고자 '후지' 사과를 온도 $15^{\circ}C$, 상대습도 67%에 150일 동안 저장하면서 품질변화를 살펴보았다. 대조구용 필름으로 기존 유통중인 LLDPE 필름 L과 여기에 수분응축억제 처리를 한 필름을 포장구 LA라 하였다. 저장 150일 후 대조구에서는 포장구 내부의 과일을 선명하게 볼 수 없을 만큼 수분응축이 심했고, 포장구 LA는 수분응축이 거의 발생되지 않았다. 중량 감소는 두 포장구 모두 1.0~1.1 % 내외의 중량감소가 일어났다. Vitamin C 함량은 두 포장구 모두 감소하였지만 포장구 LA가 대조구보다 Vitamin C 잔존량이 높게 나타났다. 당도는 저장 150일 후 포장구에서 더 높게 유지되었으며 산도는 두 포장구간 유의차가 나타나지 않았다. 에칠렌 가스의 경우 저장 150일 후 대조구 L은 192.2 ppm였고 포장구 LA는 195.7 ppm으로 조사되었다. 이상의 결과에서 수분응축억제 처리한 필름을 처리하지 않은 포장구와 비교해 보았을 때, 여러 분석 항목에서 큰 차이가 발생하지 않았다. 따라서 수분응축억제 처리시 사과의 상품성에는 도움이 될 수도 있으나 신선도 유지에 큰 영향은 없는 것으로 보여진다.
