Development of a Molecular Marker for Fruiting Body Pattern in Auricularia auricula-judae

  • The fruiting body pattern is an important agronomic trait of the edible fungus Auricularia auricula-judae, and an important breeding target. There are two types of fruiting body pattern: the cluster type and the chrysanthemum type. We identified the fruiting body pattern of 26 test strains, and then constructed two different near-isogenic pools. Then, we developed sequence characterized amplified region (SCAR) molecular markers associated with the fruiting body pattern based on sequence-related amplified polymorphism (SRAP) markers. Ten different bands (189-522 bp) were amplified using 153 pairs of SRAP primers. The SCAR marker "SCL-18" consisted of a single 522-bp band amplified from the cluster-type strains, but not the chrysanthemum strains. This SCAR marker was closely associated with the cluster-type fruiting body trait of A. auricula-judae. These results lay the foundation for further research to locate and clone genes controlling the fruiting body pattern of A. auricula-judae.

Comparison of the growth characteristics of Aurcularia auricula-judae according to gamma ray irradiation dose (감마선 조사량에 따른 Aurcularia auricula-judae의 생육특성 비교)

  • In this study, we examined the effects of gamma irradiation dosage on the mycelial growth of Auricularia auricula-judae and performed analyses of fruiting body yield, growth characteristics, taste, fragrance, and mineral composition. Assessments of mycelial growth in response to gamma irradiation at different intensities revealed an enhancement in the growth of fungi exposed to irradiation at 200 Gy. Fruiting body yield was also highest at 200 Gy, followed by 800 Gy and the control group. On the basis of these observations, we subsequently applied gamma ray doses of 200 and 800 Gy to examine the effects of irradiation on fungal quality characteristics. In terms of the taste of fruiting bodies, we detected no significant differences among the control, 200 Gy, and 800 Gy groups. Contrastingly, with respect to fragrance, we found that fungi treated with 200 Gy were characterized by a pattern that differed from those of the control and other treatment groups. Furthermore, whereas we detected no significant difference among treatments with respect total dietary fiber content, calcium content was found to be higher in the treatment groups compared with the control group, with the highest content being measured in fungi exposed to 800 Gy irradiation. Copper content was confirmed to be higher in the control group, whereas there were no significant differences between the fungi irradiated with 200 and 800 Gy. Contrastingly, the highest levels of zinc were detected in response to 200 Gy irradiation, followed by 800 Gy. Collectively, our findings thus indicate that gamma irradiation can contribute to promoting increases in the fruiting body yield and mineral contents of mushrooms.

  • Fruiting bodies of Phellinus linteus were extracted by hot water and alkali methods. Sugar contents of PL-H (hot water extract) and PL-A (alkali water extract) were 81.1%, 37.4% and protein contents were 6.2%, 21.8%, respectively. Amino acid pattern showed that two extracts contained large amount of aspartic acid and alanine. Two extracts showed characteristic IR absorption pattern for glycosidic bond at $890\;cm^{-1}$. PL-H was divided two fractions by gel filtration chromatography and the molecular weights of each fraction were estimated to be about 10 kD and 225 kD, respectively and also PL-A was estimated 10 kD. Two extracts showed strong antitumor, immunomodulating and antioxidant activities, and were compared with commercialized glycopeptide anticancer drugs.

  • Coprinopsis cinerea was employed to investigate the fungal response to gravity. Mycelium growth revealed a consistent growth pattern, irrespective of the direction of gravity (i.e., horizontal vs. perpendicular). However, the fruiting body grew in the direction opposite to that of gravity once the primordia had formed. For the proteomic analysis, only curved-stem samples were used. Fifty-one proteins were identified and classified into 13 groups according to function. The major functional groups were hydrolases and transferases (16%), signal transduction (15%), oxidoreductases and isomerases (11%), carbohydrate metabolism (9%), and transport (5%). To the best of our knowledge, this is the first report on a proteomic approach to evaluate the molecular response of C. cinerea to gravity.

Effect of sodium chloride on the growth, amino acid content, and fragrance patterns of Pleurotus ostreatus (염화나트륨 농도가 느타리 자실체의 아미노산과 향기성분에 미치는 영향)

  • We investigated the effect of sodium chloride-associated abiotic stress on the development of Pleurotus ostreatus. We examined the growth characteristics of fruiting bodies, constituent amino acids, and fragrance pattern to determine the effect of culturing Pleurotus ostreatus on a sawdust substrate supplemented with sodium chloride in a dose-dependent manner. Pleurotus ostreatus fruiting bodies exhibited an increasing tendency towards augmented yields when grown in the presence of 0.5% sodium chloride as compared with that grown in the control group. However, increasing the supplementation of sodium chloride from 1.0 % to 2.0% resulted in significantly decreased yields of Pleurotus ostreatus fruiting bodies in these groups as compared with that in control groups. Further assessment revealed the presence of 14 types of amino acids in the fruiting bodies, including aspartate, threonine, serine, glycine, alanine, methionine, valine, isoleucine, leucine, phenylalanine, tyrosine, lysine, histidine, and arginine, at lower levels in all the sodium chloride-treated groups than in the control group; except for glutamic acid and proline. Similarly, fragrance pattern analysis of the Pleurotus ostreatus fruiting body by chromatography confirmed that the intensity of the substances presumed to be octane compounds, to which the unique flavor of mushrooms is attributed, was lower in all the sodium chloride-treated groups than in the control group.

Cultural Characteristics and Fruiting-body Productivity of New Cultivar 'Hwadam' (Lentinula edodes) by Punching Treatments on the Sawdust Medium (톱밥배지 침공처리에 따른 표고 '화담' 의 균사배양 및 자실체 생산성)

  • The effect of punching treatment on bags containing sawdust medium for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was investigated. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached 27.0-28.8℃ on the 23rd day of incubation, and 14.0-15.6% on the 16th day of incubation, respectively, decreasing thereafter. The O2 concentration showed an opposite pattern to the CO2 concentration. As the number of punching treatments on the medium increased during the incubation period, the lightness value decreased, and conversely, the color difference and weight loss rate increased. There was no difference in the cultivation period according to the punching treatment. As the punching treatment increased on the sawdust medium, the fruiting body yield increased; a three-time punching treatment produced the best yield with 605.7 g. Thus, proper punching treatment on a sawdust medium during the incubation period can increase the number of fruiting bodies and yield.

  • The effects of punching treatment on mycelial culture and fruiting body productivity were investigated in a new Lentinula edodes cultivar, "Jadam", in sawdust medium for the stable production of oak mushroom. As the punching volume and number increased, the weight loss rate and color difference increased and the L value decreased. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached their highest values at 33 and 19 days of incubation, respectively. The O2 concentration showed the lowest value on the 14th day of incubation, which was the opposite pattern to the CO2 concentration. As the punching volume and the number increased, the medium temperature and O2 concentration increased, and the CO2 concentration decreased. Higher punching volumes and numbers resulted in higher temperatures and lower CO2 concentrations. The best fruiting body yield was 5 × 70 mm - 30 (punching diameter × depth - number), and the total yield after three cycles was 644.7 g.

  • This study was executed to investigate characterization of physiological genetic in Coriolus versicolor, basidiomycetes. The optimal media for mycelia culture were PDA, CVT-I and MES as solid media, 927 and CVT-III were good as liquid media, respectively. The optimal condition for mycelial culture was pH 5.6 and $25-30^{\circ}C$. Electrophoretic isozymes and protein patterns from mycelia identified very similar in 16001 and KD88001 strain, but the other species were very diffrent in band patterns. Especially, pattern of esterase seemed to be valuable tool as taxonomic techniques for indentifying species of Coriolus versicolor. Fruiting body was cultivated with artificial logs cultivation method; 16001 and KD88001 were very similar to fruiting body shapes and colours but 16001 and CVT-80 were different in their shapes, colours and making primordia. Therefore, 16001 and KD88001 were assumed to the same strain, but 16001, 16002 and CVT-80 had the different genetic background.

Studies on Korean Species of Armillaria (한국산 뽕나무버섯균의 종에 관한 연구)

  • One hundred and ninety two isolates of Armillaria were obtained from mycelial fans on infected hosts, rhizomorphs, and single basidiospores or trauma tissue of fruiting bodies. Mating tests showed that two of these isolates were A. mellea, eight were A. tabescens, 20 were A. ostoyae, and 162 were A. gallica. Armillaria ostoyae was mainly isolated from Pinus koraiensis and Qurecus spp., A. tabescens from fruiting bodies on Pinus densiflora and Qurecus spp., and A. gallica from many tree species but not Pinus koraiensis. Armillaria mellea, A. gallica, A. ostoyae and A. tabescens showed distinct protein banding patterns. Mycelial growth and rhizomorph formation was good on basal medium with ethanol added. A. gallica and A. mellea formed many rhizomorphs, but A. ostoyae did not. A. gallica showed the best rhizomorph formation on media with tannic acid and ethanol, but a. mellea formed the most rhizomorphs on gallic acid. Rhizomorphs showed monopodial branching for A. gallica and dichotomous branching for A. ostoyae. Fruiting bodies. formed in the laboratory on sawdust media most abundantly by A. tabescens. In nature, fruit body formation by A. tabescens was from early to mid August. A. ostoyae and A. gallica fruit bodies were formed from early August to late October. While there are common names in Korea for A. mellea and A. tabescens, such as mulberry mushroom relative, no common names are available for A. gallica and A. ostoyae. Therefore, we refer to a. gallica as the Gastrodia mushroom because it has been used to produce Gastrodia and A. ostoyae as the Korean pine mushroom because it is frequently found as mushrooms on Korean pine.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.
