본 연구는 비누수 해가림 재배가 인삼 생육 및 품질에 미치는 영향을 구명하기 위하여 인삼의 차광재료를 비누수 해가림과 차광망을 이용하여 실험하였던 바, 얻어진 결과를 요약하면 다음과 같다. 1. 차광재료에 따른 맑은 날의 광량은 비누수해가림이 차광망에 비하여 낮은 경향이었고, 온도는 비누수해가림이 차광망에 비하여 $3{\sim}5^{\circ}C$ 낮게 나타났다. 2. 차광재료에 따른 3, 4, 5년근 인삼의 생존본수는 비누수해가림에 비하여 차광망이 낮게 나타났다. 3. 차광재료에 따른 인삼의 경태는 비누수해가림에서 증가하였고, 생근중도 비누수해가림이 차광망에 비하여 3년근에서는 5.0 g, 4년근에서는 10 g, 5년근에서는 8 g 높았다. 4. 차광재료에 따른 3, 4, 5년근 모두 수삼의 수량은 비누수해가림이 차광망보다 훨씬 높게 나타났다. 5. 차광재료별 3, 4, 5년근 수삼의 크기는 비누수해가림이 차광망에 비하여 대편이 많이 분포하였으며, 진세노사이드 함량도 비누수해가림이 차광망에 비해서 높았다
Background: The quality differences in seedlings of medicinal herbs often affect the quality of medicinal parts. The establishment of the grading standard of Panax notoginseng seedlings is significant for the stable quality of medicinal parts of P. notoginseng. Methods: To establish the grading standard of P. notoginseng seedlings, a total of 36,000 P. notoginseng seedlings were collected from 30 producing areas, of which the fresh weight, root length, root diameter, bud length, bud diameter, and rootlet number were measured. The K-means clustering method was applied to grade seedlings and establish the grading standard. Results: The fresh weight and rootlet number of P. notoginseng seedlings were determined as the final indices of grading. P. notoginseng seedlings from different regions of Yunnan could be preliminarily classified into four grades: the special grade, the premium grade, the standard grade, and culled seedlings. Conclusion: The grading standard was proven to be reasonable according to the agronomic characters, emergence rate, and photosynthetic efficiency of seedlings after transplantation, and the yields and contents of active constituents of the medicinal parts from different grades of seedlings.
Background: Amino acids are one of the major constituents in Panax ginseng, including neutral amino acid, acidic amino acid, and basic amino acid. However, whether these amino acids play a role in ginsenoside conversion during the steaming process has not yet been elucidated. Methods: In the present study, to elucidate the role of amino acids in ginsenoside transformation from fresh ginseng to red ginseng, an amino acids impregnation pretreatment was applied during the steaming process at 120℃. Acidic glutamic acid and basic arginine were used for the acid impregnation treatment during the root steaming. The ginsenosides contents, pH, browning intensity, and free amino acids contents in untreated and amino acid-treated P. ginseng samples were determined. Results: After 2 h of steaming, the concentration of less polar ginsenosides in glutamic acid-treated P. ginseng was significantly higher than that in untreated P. ginseng during the steaming process. However, the less polar ginsenosides in arginine-treated P. ginseng increased slightly. Meanwhile, free amino acids contents in fresh P. ginseng, glutamic acid-treated P. ginseng, and arginine-treated P. ginseng significantly decreased during steaming from 0 to 2h. The pH also decreased in P. ginseng samples at high temperatures. The pH decrease in red ginseng was closely related to the decrease in basic amino acids levels during the steaming process. Conclusion: Amino acids can remarkably affect the acidity of P. ginseng sample by altering the pH value. They were the main influential factors for the ginsenoside transformation. These results are useful in elucidating why and how steaming induces the structural change of ginsenoside inP. ginseng and also provides an effective and green approach to regulate the ginsenoside conversion using amino acids during the steaming process.
There are remarkable differences in growth and morphological characters of roots between mountain and field cultivated Panax ginseng. Growth of root in mountain cultivated ginseng was much slower than that of field cultivated ginseng. However, the factor affecting the retarded growth in mountain ginseng was not known. Soil analysis revealed that phosphorus (P) content of mountain soil was exceptionally low at least ten-fold lower compared to that of field soil. Thus, we suggest that low availability of P in mountain soil may be one of the limiting factors for growth of ginseng in mountain soil environment. We had monitored the growth of ginseng plants after one and three years of phosphate fertilizer application. Three kinds of phosphate fertilizers: fused magnesium phosphate, fused superphosphate, and single superphosphate were applied to mountain soil. Application of phosphate fertilizers increased the fresh-, dry weight, and diameter of ginseng roots and resulted in increased P accumulation in roots. These results demonstrate that slow growth of ginseng in mountain soil environment might be attributed to the low P content in mountain soil. Thus, analysis of P amount in mountain soil will be a good indicator for the selection of suitable site the ginseng cultivation in forest.
Background: The photosynthetic efficiency cool-season, semi-shade ginseng is normal at low morning temperatures, but drops at high afternoon temperatures. Therefore, optimal plant performance would be ensured if it were possible to control daily light transmission rates (LTR). Methods and Results: Plants were grown in a controlled light environment that replicated 11 AM conditions and comparatively analyzed against plant grown under normal conditions. Growth in the controlled light environment resulted in a 2.81 fold increase in photosynthetic efficiency with no change in chlorophyll content, although LTR were high due to low morning temperatures. Increased aerial plant growth was observed in the ginseng plants adapted to the controlled light environment, which in turn influenced root weight. An 81% increase in fresh root weight (33.3 g per plant on average) was observed in 4-year-old ginseng plants grown in controlled light environment compared to the plants grown following conventional practices (18.4 g per plant on average). With regard to the inorganic composition of leaves of 4-year-old ginseng plants grown in controlled light environment, an increased in Fe content was observed, while Mn and Zn content decreased, and total ginsenoside content of roots increased 2.37 fold. Conclusions: Growth of ginseng under a favorable light environment, such as the condition which exist naturally at 11 AM and are suitable for the plant's photosynthetic activity creates the possibility of large scale production, excellent-quality ginseng.
식물조직배양기술을 이용한 Ge 함유 인삼 부정근 생산 시 Ge의 생산성을 향상시키고자, 다양한 킬레이트제가 인삼 부정근의 Ge 축적과 사포닌 형성 및 생육에 미치는 영향을 조사하였다. $GeO_2$ 50ppm과 함께 citric acid, oxalic acid, phosphoric acid, EDTA 및 EGTA와 같은 킬레이트제를 인삼 부정근 배양시 첨가하여 Ge 흡수에 미치는 영향을 조사한 결과, phosphoric acid, EDTA 및 EGTA가 Ge 흡수를 촉진시킨다는 사실을 알 수 있었다. 그러나, EDTA와 EGTA는 인삼부정근의 생육을 억제시킬 뿐만 아니라 Ge 흡수 촉진 효과도 phosphoric acid에 비해 낮으므로 인삼 부정근의 Ge 흡수를 촉진하는데 가장 효과적인 킬레이트제는 phosphoric acid인 것으로 확인되었다. 인삼 부정근의 Ge 흡수 촉진을 위한 phosphoric acid의 최적 농도는 1.0 mM이었고, 이때의 Ge 함량은$22.7{\pm}0.3$ mg%로 대조구$(16.8{\pm}0.7$ mg%)의 1.4배에 해당하는 양이다. 한편, 인삼 부정근의 생장률과 총 사포닌 함량은 phosphoric acid의 농도가 증가할수록 감소하는 경향을 나타내었다.
인삼의 시설생산을 위하여 배지 조성 및 차광 정도를 달리한 환경조건 하에서의 생육특성과 생리활성 변화를 검토하고자 온실 및 실내시험을 수행하였다. 적정한 배지 조합은 차광 정도에 따라 달랐는데 무차광에서는 coco peat 단독, 50% 차광에서는 coco peat와 perlite를 50:50으로 혼합하였을 때, 70% 차광에서는 perlite 단독배지에서 유의적으로 높은 생육을 보였다(P<0.05). 차광은 엽면적, 엽록소 함량, 근장, 지상부 및 지하부 생체중을 유의적으로 감소시켰다. 총 페놀 함량은 지상부(20.7~23.8 $mg{\cdot}kg^{-1}$)보다는 지하부(25.8~28.4 $mg{\cdot}kg^{-1}$)에서, 무차광(20.7~25.8 $mg{\cdot}kg^{-1}$)보다는 차광(23.7~28.4 $mg{\cdot}kg^{-1}$)에서 높게 나타났다. 총 플라보노이드 함량은 지하부(6.9~7.0 $mg{\cdot}kg^{-1}$)보다는 지상부에서(17.4~26.8 $mg{\cdot}kg^{-1}$), 무차광보다는 차광에서 높게 검출되었다. 반면, DPPH 라디컬 소거능 측정을 통한 항산화성은 추출물 농도가 증가할수록 높은 활성을 보였고, 지하부(24.9~28.7%)보다는 지상부에서(31.1~36.5%), 차광보다는 무차광에서 다소 높은 활성을 보였다. 결론적으로 차광은 비닐하우스 조건하에서 재배된 인삼의 생육과 생리활성에 영향을 미치는 것으로 나타났다.

This experiment was carried out using artificial clay and LED in the plastic film house (irradiation time: 08:00~18:00/day). Seedlings (n = 63 per $3.3m^2$) of ginseng was planted on May 17, 2018. LED was combined with red and blue light in a 3:1 ratio and irradiated with different light intensity. The average air temperature from April to September was $12.3^{\circ}C$$-26.0^{\circ}C$ and it was the the highest at $26.0^{\circ}C$ in August. The test area where fluorescent lamp was irradiated tended to be somewhat higher than the LED irradiation area. The chemical properties of the test soil are as follows. pH levels was 5.3~5.5, EC levels 0.45~0.52 dS/m and OM levels 33~37%. The total nitrogen content was 0.35~0.47% and the available $P_2O_5$ contents was 13.7~16.0 mg/kg, which was lower than the suitable level of 70~200 mg/kg. Exchangeable cations K and Mg contents were within acceptable ranges, but the Ca contents was $28{\sim}38cmol^+/kg$ levels higher than the permissible level ($2{\sim}6cmol^+/kg$). Germination of ginseng leaves took 8~9 days and the overall germination rate was 70~75%. The photometric characteristics of LED light intensity are as follows. The greater the light intensity, the higher the PAR (Photosynthetic Action Radiation) value, illuminance and solar irradiation. Photosynthetic rate was also increased with higher light intensity was investigated at $1.7{\sim}3.2{\mu}mol\;CO_2/m^2/s$. Leaf temperature ($23.7{\sim}24.8^{\circ}C$) by light intensity was the same trend. The growth of aerial parts (plant height etc.) were generally excellent when irradiated with 3 times the light intensity, the growth of the ginseng aerial parts were excellent as follows. The plant height was 42.6 cm, stem length was 25.2 cm, leaf length was 9.6 cm and stem diameter was 5.0 mm. The growth of underground part (root length etc.) was the same, and the root length was 24.4 cm, the tap root length was 6.0 cm, diameter of taproot was 18.2 mm and the fresh root weight was 17.2 g. There were no disease incidence such as Alternaria blight, Gray mold and Anthracnose. Disease of Damping off occurred 2.2~3.6% and incidence ratio of rusty root ginseng was 14.6~20.7%. Leaf discoloration rate was 13.7~48.9% and increased with increasing light intensity. Ginsenoside content of ginseng by light intensity is under analysis.
병결점 부근의 저장온도에 따른 수삼의 내적 품질 변화 연구로 수삼을 각각 $-3^{\circ}C$, $-1.5^{\circ}C$ 및 $0^{\circ}C$ 에서 16주간 저장하면서 저장온도에 따른 호흡률, 경도, 가용성 고형물함량, 전분함량, pH, 사포닌함량과 관능적 품질의 변화를 조사하였다. 수삼의 호흡률은 저장온도가 낮을수록 낮았으나 그 차이는 미미하였다. 수삼 표피층의 정도는 저장기간이 경과됨에 따라 증가하였고, 내부 조직의 경도는 감소하는 경향을 보였으나 저장온도에 따라 경도 변화의 차이는 뚜렷하지 않았다. 수삼의 가용성 고형분 함량은 $-3^{\circ}C$에서 저장한 수삼의 경우 $0^{\circ}C$ 및 $-1.5^{\circ}C$에서 저장한 수삼에 비해 낮았다. 요오드 반응에 의한 주근 부위의 발색 정도는 저장 2주후까지 저장온도에 따라 다소 차이가 발생하였으나 이후부터 저장종료시점까지는 큰 차이를 보이지 않았다. 수삼의 pH는 저장기간 종 전반적으로 저장 온도가 낮을수록 낮은 경향을 보였다. 조사포닌 함량은 저장 중 온도에 따라 약간의 차이를 보였지만 저장 16주에도 초기함량과 유사한 수준을 유지하였다. 관능적 품질 평가인자 중 수삼 '고유의향'은 $-3^{\circ}C$에서 저장한 경우 저장 후 8주, $-1.5^{\circ}C$에서 저장한 경우 저장 후 14주까지 양호하였으나 $0^{\circ}C$에서 저장한 수삼은 저장종료시점인 16주 이후까지도 품질이 양호하였다. 수삼의 '고유의 맛'은 저장 12주후까지는 저장온도에 따라 차이가 뚜렷하지 않았으나 저장 14주후부터는 $-3^{\circ}C$에서 저장한 수삼이 $-1.5^{\circ}C$ 및 $0^{\circ}C$에서 저장한 수삼과 유의적인 차이를 보였다.(p < 0.05). 전반적으로 저장온도에 따른 수삼의 내적 품질인자의 변화 조사 결과 저장온도에 따른 차이가 뚜렷하지 않았으며 관능적 품질평가 결과를 기준으로 보면 $-3^{\circ}C$ 보다는 $-1.5^{\circ}C$ 및 $0^{\circ}C$가 품질유지에 효과적인 것으로 판단되었다.
