Effects of Artificial Supercooling Followed by Slow Freezing on the Microstructure and Qualities of Pork Loin

  • This study investigated the effects of artificial supercooling followed by still air freezing (SSF) on the qualities of pork loin. The qualities of pork frozen by SSF were compared with the fresh control (CT, stored at 4℃ for 24 h), slow freezing (SAF, still air freezing) and rapid freezing (EIF, ethanol immersion freezing) treatments. Compared with no supercooling phenomena of SAF and EIF, the extent of supercooling obtained by SSF treatment was 1.4℃. Despite that SSF was conducted with the same method with SAF, application of artificial supercooling accelerated the phase transition (traverse from -0.6℃ to -5℃) from 3.07 h (SAF) to 2.23 h (SSF). The observation of a microstructure indicated that the SSF prevented tissue damage caused by ice crystallization and maintained the structural integrity. The estimated quality parameters reflected that SSF exhibited superior meat quality compared with slow freezing (SAF). SSF showed better water-holding capacity (lower thawing loss, cooking loss and expressible moisture) and tenderness than SAF, and these quality parameters of SSF were not significantly different with ultra-fast freezing treatment (EIF). Consequently, the results demonstrated that the generation of supercooling followed by conventional freezing potentially had the advantage of minimizing the quality deterioration caused by the slow freezing of meat.

냉동과 해동 방법이 오리고기의 특성에 미치는 영향 (Effect of Freezing and Thawing Methods on Duck Meat Characteristics)

  • 본 연구는 냉동과 해동 방법에 따른 토종오리고기의 육질특성을 비교하기 위해 수행하였다. 본 시험에 사용된 공시재료는 8주령 토종오리(평균체중 2.8 kg)를 도압(屠鴨)하여 채취한 가슴육을 이용하였다. 본 시험의 처리구는 냉동 전 신선육을 대조구(CON)로 하고, 시험구는 냉동 2처리(급냉(FF), 완냉(SF)와 해동 2처리(급해동(FT), 완해동(ST))의 $2{\times}2$ 복합요인으로 4처리구로 하여 총 5처리구(CON, FFFT, FFST, SFFT, SFST), 처리구당 3반복, 반복당 3점씩 총 45점을 이용하였다. 급냉(시료를 $-50^{\circ}C$ deep freezer에서 보관)과 완냉(시료를 $-20^{\circ}C$ 냉동고에 보관)의 두 가지 방법으로 냉동된 시료를 1개월 저장 후, 급해동($12^{\circ}C$ 5시간의 유수해동)과 완해동($5^{\circ}C$ 24시간 냉장고 해동)의 두 가지 방법으로 해동하여 분석에 이용하였다. 육색 중 명도는 급냉시킨 경우와 급해동시킨 경우에 대조구에 비해 유의적으로 감소되었다(P<0.05). 황색도는 완해동시킨 경우에는 대조구에 비해 유의적으로 높게 나타났다(P<0.01). 물리적 성상 중 가열감량과 보수력은 대조구가 냉동과 해동 처리구에 비해 낮게 나타난 반면에(P<0.01, P<0.05), 전단력은 냉동과 행동 처리구에 비해 대조구에서 높게 나타났다(P<0.01). 화학적 성상 중 수분의 함량은 완해동한 경우에 급해동에 비해 높게 나타났으며(P<0.05), 단백질 함량은 급해동한 경우 대조구에 비해 유의적으로 높게 나타났다(P<0.05). Myristic acid(C14:0)와 linolenic acid(C18:3n3) 함량은 대조구에서 냉동과 해동 처리구에 비해 높게 나타났다(P<0.05). Stearic acid(C18:0) 함량은 완냉동시킨 경우에 급냉동의 경우보다 높게 나타났으며(P<0.05), 해동의 경우에는 처리구간 유의차를 보이지 않았다. Arachidonic acid(C20:4n6)은 완냉동과 완해동의 경우에 대조구에 비해 유의적으로 높게 나타났다(P<0.01, P<0.05). 필수아미노산 중 대조구의 threonine, glycine, iso-leucine, leucine, tryptophan 함량은 냉동과 해동 처리구에서 대조구에 비해 유의적으로 높은 함량을 나타내었다(P<0.05). Methonine과 valine 함량은 해동에 따른 차이는 보이지 않았으나, 냉동 처리시 대조구에 비해 유의적으로 높게 나타났다(P<0.05). 비필수아미노산 중 alanine, aspartic acid, glutamic acid, serine, tyrosine 함량은 대조구에서 냉동과 해동처리구보다 유의적으로 낮게 나타났다(P<0.01, P<0.05). 결론적으로, 토종오리고기의 냉동 및 해동은 수분과 관련된 육색, 전단력, 가열감량, 보수력에 영향을 미치는 것으로 보여진다.

Effects of various freezing containers for vitrification freezing on mouse oogenesis

  • Background: In the present study, various freezing containers were tested for mouse embryos of respective developmental stages; embryos were vitrified and then their survival rate and developmental rate were monitored. Mouse two cell, 8 cell, and blastula stage embryos underwent vitrification freezing-thawing and then their recovery rate, survival rate, development rate, and hatching rate were investigated. Methods: EM-grid, OPS, and cryo-loop were utilized for vitrification freezing-thawing of mouse embryos. Results: It was found that recovery rate and survival rate were higher in the group of cryo-loop compared to those of EM-grid (p < 0.05). Embryonic development rate, two cell embryos to blastocyst, as well as hatching rate were higher in the control group compared to the EM-grid group and OPS group (p < 0.05), yet no difference was noted between the control group and cryo-loop group. Development rate and hatching rate of eight cell morulae and blastocysts were all lower in the treatment groups than the control group whilst hatching rate of blastocysts was higher in the control group compared to the groups of EM-grid and OPS (p < 0.05); although the cryo-loop group was shown to be slightly higher than other groups, it was not statistically significant. Conclusions: In the study, we investigate effects of freezing containers on vitrified embryos of respective developmental stages; it was demonstrated that higher developmental rate was shown in more progressed (or developed) embryos with more blastomeres. There was however, no difference in embryonic development rate was shown amongst containers. Taken together, further additional studies are warranted with regards to 1) manipulation techniques of embryos for various vitrification freezing containers and 2) preventive measures against contamination via liquid nitrogen.

인간의 미성숙난자의 동결보존에 관한 연구 (Cryopreservation of Human Immature Follicular Oocyte)

  • This study was carried out to set up the ovum bank for ovum donation and to determine the best freezing method for human immature oocytes. Human immature follicular oocytes were cryopreserved by slow freezing and rapid thawing method. Immature follicular oocytes were treated by propanediol(PROH) solution by 2 and 4 step method in protocols A & B, respectively. In protocol C, immature oocytes were exposed to sucrose prior to treatment of PROH by 4 step method. We compared survival rate, maturation rate, and fertilization rate of immature oocytes among three protocols. Results were as follows. 1. Oocytes treated by the protocol C showed the highest survival rate( 70.3 %) and maturation rate(34.6%) after thawing. 2. Survival rate of oocytes treated by the protocol C was significantly higher than that of the protocol B after thawing(p<0.05). In conclusion, treatment of oocytes with sucrose prior to expose PROH was the best freezing method. Sucrose may have reduced the toxic effect of cryoprotectant to oocytes. We failed to induce fertilization of oocytes, which were treated by any protocols, by conventional insemination method, but obtained 28.8% fertilization rate by using partial zona dissection(PZD) method. This result suggests that micromanipulation(PZD) of the thawed oocytes before insemination will improve the fertilization rate.

냉동과 해동처리가 계육 가슴살의 natural microflora, 접종된 Listeria monocytogenes와 Campylobacter jejuni에 미치는 영향 (Effects of Freezing and Thawing Treatments on Natural Microflora, Inoculated Listeria monocytogenes and Campylobacter jejuni on Chicken Breast)

  • 본 연구는 다양한 냉동과 해동처리에 따른 계육 가슴살에 natural microflora, 접종된 L. monocytogenes와 C. jejuni 수와 미세구조 변화 구명을 위하여 연구를 수행하였다. $-20^{\circ}C$ 송풍식 냉동처리구의 총 호기성 세균과 C. jejuni 수는 4.06 log CFU/g과 4.09 log CFU/g으로 대조구와 비교하여 각각 약 0.7 log CFU/g과 1.0 log CFU/g의 감소를 보였다. 한편, 계육 가슴살 접종된 L. monocytogenes 수는 냉동방법과 냉동온도에 따라 유의적인 차이를 보이지 않았다. $4^{\circ}C$$25^{\circ}C$ 송풍식 해동처리구의 총 호기성 세균수는 3.70 log CFU/g과 4.02 log CFU/g으로 측정되어 대조구와 비교하여 각각 0.72 log CFU/g과 0.40 log CFU/g 감소한 반면 $25^{\circ}C$ 유수식 해동처리구의 총 호기성 세균 수는 해동과정에서 균수가 급격히 증가하여 5.78 log CFU/g으로 관찰되었다. 해동 중 C. jejuni 수 변화는 해동방법보다 해동온도에 영향을 받는 것으로 나타났다. 냉동-해동 반복 5회 후 총 호기성 세균과 효모 및 곰팡이 수는 감소하여 각각 4.15 log CFU/g과 2.30 log CFU/g을 보였다. 계육 가슴살에 접종된 L. monocytogenes 수는 냉동-해동 반복처리에 유의적인 영향이 없었지만 C. jejuni 수는 냉동-해동 반복 횟수가 증가함에 따라 감소하는 경향을 보였다. 냉동-해동 반복이 증가함에 따라 근섬유 조직을 재 손상시켰으며 특히 냉동-해동 반복 5회 후에는 계육 가슴살 시료의 조직세포 외부뿐만 아니라 내부에서도 공간이 발생하였으며 불균일하게 찢어진 세포가 관찰되었다.

냉해동 처리에 따른 오미자 담금주의 추출 특성 (Extraction Characteristics on Soaked Liquor of Omija (Schisandra chinensis) by Freeze-thawing Process)

  • Fresh Omija (Schisandra chinensis) has good marketability, but its quality is difficult to maintain during storage and distribution. Freezing and freeze-thawing treatments can be utilized for the quality maintenance and processing of cold press juice. In this study, the color, antioxidant properties, and the major components of soaked liquor from Omija with freeze-thawing treatment were analyzed during the extraction periods. Each of the frozen and freeze-thawed Omija samples was soaked in 35% ethanol, extracted for 15 days, and used for analysis. The frozen and freeze-thawed samples showed a tendency toward better color and higher antioxidant activity and major component levels than the controls, and freeze-thawing was the best. The results of this study showed that freeze-thawing treatment improved the color, antioxidant properties, and level of the major components of Omija soaked liquor, and freeze storage is suitable for making soaked liquor.

저온성 표고버섯의 빙점하 저장 잠재력 (Storage potential of low temperature adapted shiitake mushroom under freezing temperature)

  • This study was aimed to find the storage potential of low temperature adapted shiitake mushroom under freezing temperature ($-3^{\circ}C$). Fresh shiitake mushroom was harvested at late Feb. and cooled to target temperature, $-3^{\circ}C$ in the cooling room for 24 hr. Cooled mushroom were then placed in plastic boxes, wrapped with plastic film ($30{\mu}m$), and stored at $-3^{\circ}C$ for 2 months. Weight loss of mushroom was ranged from 2.1 to 3.2%. Dry weight per unit fresh weight, however, was slightly increased because of moisture loss. Firmness of fruit body increased from $0.95kg/cm^{-2}$ (before storage) to $1.13kg/cm^{-2}$ (after 2 month storage). About half amount of starch was lost during 2 month storage. The amount of total and reducing sugars remained relatively constant. After storage, freshness of mushroom was recovered by thawing treatment. When recovered mushroom were packaged with styrofoam tray and PVC wrapping, and exposed to ambient and $10^{\circ}C$, respectively, brown spot on the gill of fruit body was found and slight decay symptom was also found at ambient temperature only but not at $10^{\circ}C$. Results indicated that low temperature adapted shiitake mushroom has a storage potential under freezing temperature ($-3^{\circ}C$). Freezing storage technology of fresh shiitake mushroom will contribute the increase of storability up to 2 months.

Methanol을 이용한 개 정액 동결시 융해후 양호한 활력 및 생존율을 나타내는 정액 처리 조건 (Semen Treatment to Maintain Good Quality of Post-thaw Motility and Viability of Canine Spermatozoa Frozen Using Methanol)

  • This experiment was carried out to investigate the renditions to maintain good post-thaw motility and viability of canine spermatozoa when the semen was frozen using methanol. The semen from two male dogs which had been proven to be fertile in the previous one year was treated with different compositions of semen diluent and was frozen at different freezing temperatures, When canine semen was frozen at-2$0^{\circ}C$, -6$0^{\circ}C$, or -8$0^{\circ}C$, the spermatozoa frozen and stored at -2$0^{\circ}C$ showed very low post-thaw motility and viability from day 2 to 7 and showed no viability since day 15 after freezing. The spermatozoa frozen and stored at -6$0^{\circ}C$ or -8$0^{\circ}C$ showed higher post-thaw motility and viability on day 2, 1, 15 and 30 after freezing than that frozen and stored at-2$0^{\circ}C$(p<0.01), with no difference between two groups. Among different composition groups of the semen diluents of control(tris + egg yolk + glycerol), egg yolk-free, 히ycerol-free, and tris-free, Prior to freezing, the egg yolk-free diluent showed significantly love. motility and viability than the other diluents(p<0.05). On each thawing day (from day 2 to 15 after freezing), control group showed considerably higher motility and viability than the other groups(p<0.01). The canine spermatozoa frozen and stored at -6$0^{\circ}C$ and -8$0^{\circ}C$ showed gradual decrease of motility from day 2 to 30 after freezing and the spermatozoa of these two groups thawed on day 30 showed considerably love. motility than those thawed on day 2 after freezing, respectively(p<0.01). These results indicate that the freezing temperature of either -6$0^{\circ}C$ or -8$0^{\circ}C$ can be applicable to the freezing method using methanol and also all of the components of the semen diluent including cryoprotectant, buffer and cold-shock buffer are very important to maintain motility and viability of canine spermatozoa in the freezing and thawing procedure.

Durability and mechanical performance in activated hwangtoh-based composite for NOx reduction

  • Activated hwangtoh (ACT) is a natural resource abundant in South Korea, approximately 15.0% of soil. It is an efficient mineral admixture that has activated pozzolanic properties through high-temperature heating and rapid cooling. The purpose of this study is to improve a curb mixture that can reduce NOx outside and investigate durability performance. To this end, mortar curb specimens were manufactured by replacing OPC with ACT. The ACT substitution ratios of 0.0, 10.0, and 25.0% were considered, and mechanical and durability tests on the curb specimens were conducted at 28 and 91 days of age. Steam curing was carried out for three days for the production of curbs, which was very effective to strength development at early ages. The reduction in strength at early ages could be compensated through this process, and no significant performance degradation was evaluated in the tests on chloride attack, carbonation, and freezing and thawing. The mortar curb with an ACT of 10.0~25.0% replacement ratio exhibited clear NOx reduction through photocatalytic (TiO2) treatment. This is due to the increase in physical absorption through surface absorption and the photocatalyst-containing TiO2 coating. In this study, the reasonable range of the ACT replacement ratio for NOx reduction was quantitatively evaluated through a comprehensive analysis of each test.

  • This study was performed to evaluate properties of recycled concrete for roadway pavement. Recycled concretes was manufactured for the target compressive strength of 280kg/$\textrm{cm}^2$ and 180kg/$\textrm{cm}^2$ with recycled aggregate ratio of 0%, 20%, 40%, 60%, 80%, respectively. Laboratory experiment was conducted for testing properties of fresh concrete and concrete strength at curing 28days and durability by freezing and thawing treatment. The study result presented a maximum replacement ratio of recycled material.
