A Study on Korean Green Tea(II) -Analysis of free Amino Acid and Mineral- (한국산(韓國産) 녹다(緣茶)에 대(對)한 연구(硏究) (제 II 보) -Free amino Acid와 무기성분(無機成分)에 대(對)하여-)

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of free amino acids in Korean green tea and the contents of mineral in it was determined by atomic absorption flame emission. The results are summarized as follows: 1) Korean green tea contained Mn 1% or over out of the total ash content and $0.05{\sim}0.20%$ in the water extraction, as the major mineral. 2) Ba, Cr, Ni, Pb and V were analyzed also by small quantities relatively and Co, Tin and Ywere not detected in the water extraction. 3) GLC indicated the presence of 16 components in free ammo acids. 4) The quantities of free amino acids in Korean green tea were determined $2.96{\sim}6.61%$ Alanine, $1.01{\sim}l.89mg%$ Glycine, $2.07{\sim}7.81mg%$ Valine, $1.27{\sim}8.76mg%$ Leucine and Isoleucine, $94.31{\sim}316.27mg%$ Threonine, $9.10{\sim}39.91mg%$ Serine, $2.18{\sim}36.76mg%$ Hydroxyproline, $2.72{\sim}5.90mg%$ Proline, $39.64{\sim}70.02mg%$ Aspartic Acid, $25.93{\sim}101.28mg%$ Glutamic Acid and Lysine, $8.32{\sim}18.30mg%$ Phenylalanine and Tyrosine in trace amount. 5) The total free amino acid contents in Korean green tea ranged from 207.24mg% to 516.06mg% and Moo-Deoung tea contained outstandingly high, 516 mg% or over.

  • The free amino acid contents in several tissues of swine were analyzed qualitatively by means of two dimentional paper chromatography. The tissues used were liver, kidney and spleen that were obtained from normal, immunized and hog cholera infected swines. The results obtained are as follows: 1. Liver: 20 amino acids were detected in normal, 17 in immunized and 15 in infected swines. 2. Kidney: 16 amino acids were detected in normal, 13 in immunized and infected swines. 3. Spleen: 15 amino acids were detected in normal in immunized and 13 in infected swines. 4. Glutamic acid, leucine, serine and threonine were present in high concentration in all of the cases examined. 5. The free amino acids were appeard to be decreased in the infected tissues with hog cholera virus.

Protein and Amino Acid Composition of Water Cress Oenanthe stolonifera DC (미나리의 단백질의 및 아미노산 조성)

  • This study was attempted to compare the nutritive value of leaf with stem of the water cress Oenanthe stolonifera DC. in order to improve the eating habits and as a part of studying on the effective curing nutrients for the damaged liver. The contents of moisture crude proteinon the effective curing nutrients for the damaged liver. The contents of moisture crude protein crude fat and crude ash were 90.40% 2.85%, 0.42% and 0.74% in leaf while the contents of moisture crude protein crude fat and crude ash were 95.15% 0.77% 0.09%, and 0.64% of moisture crude protein crude fat and crude ash were 95.15% 0.77% 0.09% and 0.64% in stem respectively. The quantitative fractionation of proteini of both leaf and stem ranked albumin the highest content followed globulin prolamin and glutelin in order. It has been sh-own by SDS-polyacrylamide gel electrophoresis that water extractable protein of leaf 11 bands but those of stem were not detected. The scope of molecular weight for the main protein of water extractable protein of leaf was between 34.700 and 45,000. The amounts of extractive-nitrogen from leaf and stem of the water cress were 241.02mg% and 271.67mg% respec-tively. The amounts of free amino acid-nitrogen from the leaf and stem were 89.02mg% and 32.02mg% respectively. In free amino acid-nitrogen from the leaf and stem were 89.02mg% and 32.02mg% respectively. In free amino acid composition of both leaf and stem the major components were aspartic acid and glutamic acid. In total amino acid composition of water cress leaf aspartic and glutamic acid were the major components. Whereas alanine and thr-eonine were the major components in stem The assessment of water cress leaf and stem with chemical score. EAAl Rl showed that the values of stem were lower tendancy than those of leaf. Limiting amino acid of leaf was tryptophan while that of stem was lysine.

  • A survey of the free amino acids and free sugars in tomato, watermelon, muskmelon, peach and plum was made by means of amino acid autoanalyzer and thin layer chromatography. The results of the survey are summarized as follows. 1) Fifteen amino acids found in fruit were aspartic acid, glutamic acid, alanine, serine, asparagine, lysine, valine, glycine, methionine, histidine, threonine, leucine, isoleucine, proline and arginine, and an unknown was found. 2) Ten kinds of amino acids were detected in tomato, peach and plum, thirteen amine acids in watermelon and muskmelon (edible part), and eleven amino acids in muskmelon (rind). 3) In general, these fruits contained similar amounts of these thirteen amino acids, and although they were not outstandingly high in any one acid they did contain a nutritionally well-balanced mixture. 4) Amino acids found in the greatest amount in fruit were following: glutamic acid and aspartic acid in tomato, asparagine and lysine in watermelon, alanine, serine and aspartic acid in musk-melon, and aspartic acid and serine in peach and plum. 5) Glucose, fructose, sucrose and maltose were detected in all fruit. The contents of glucose and fructose were high, and those of sucrose and maltose were low.

  • For the amino-N -ratio and N- solubity, irradiated samples were higher than non - irradiated them, that is, 15KGy(l9, 72), 7KGy(l9, 55) of amino-N-ratio were more those of 5KGy(17. 91) and l0KGy(l8, 06). during the fermentation of samples. For the Solubility of nitrogen, irradiated samples were decreased sequencely, such as 10KGy, 7KGy, 15KGy and 5KGy. For the amounts of amino acid, Content of the glutamic acid was the highest in the all samples and the main free amino acids in the Steamed Soybeans Were glutamic acid, proline, arginine, and alanine etc. However the amount of amino acid in the irradiated Sample was notably increased in Comparison with the nonirradiated $(10.8017\%)$ and 7KGy amino acid $(15.4166\%)$ was highest.

Content Analyses of Fiber, Protein and Amino Acids of Fully Ripe Fruits of Korea Native Squash, Cucurbita moschata Poir (한국재래종 호박 완숙과의 섬유질, 단백질 및 아미노산 함량 비교분석)

  • We studied active substances like crude cell wall components, crude protein, composing amino acids and free amino acids including orinithine cycle-related amino acids such as asparagine, ornithine and citrullin in fully ripe fruits of Korean native squash, Cucurbita moschata Poir. Crude protein content of 'Jeju 2' was the highest with $2,830\;{\mu}g/g$, while 'Sangju' was the lowest with $1,319\;{\mu}g/g$. Regarding the contents of crude cell wall components, fruit 'Kanghaw' was the highest with 2,961 mg% while 'Namhea' was the lowest with 1,582 mg%. Pectin contents of crude cell wall components were the highest in 'Kanghaw' (2,198 mg%) followed by 'Jeju 2' (2,178 mg%) and 'Jeju l' (1,461 mg%). The main contents of amino acids in squash were glutamic acid, aspartic acid, lysine, leucine and valine, which comprised to be more than 50% of total amino acid contents. Especially, in 'Jeju 2' aspartic acid and threonine were not detected. In fully ripe fruits, a total of 34 kinds of free amino acids were detected including 8 kinds of essential amino acids (histidine, isoleucine, leucine, lysine, phenylalanine, methionine, threonine and valine). More than 50% of the total free amino acids were aspartic acid and asparagine, and also all varieties were detected in ornithine, citrullin, and arginine, which are related to Ornithine cycle. There was a big difference in the contents of arginine in all varieties whereas the contents of ornithine and citrullin were very similar. 'Teaan' 29.34% was 7 times higher than 'Namhea' 4.30% in regards to arginine contents.

Changes in Chemical Composition of Panax ginseng Leaves by Different Harvesting Months (인삼협의 채엽시기별 화학성분에 관한 연구)

  • This study was performed to investigate the changes in chemical composition of Panax ginseng leaf by harvesting at July, August and September. The levels of crude protein of dried ginseng leaf were decreased from 17.12mg% to 14.26% by harvesting month, however, crude fat contents of dried ginseng leaf were increased slightly from 1.90% to 2.49%. Three kinds of free sugar, i.e. glucose, fructose and sucrose were found in dried ginseng leaf and maltose was not found. Free sugar contents were increased by delaying harvest, but free amino acid were decreased. Total free amino acid was decreased in delayed harvesting month, serine was revealed superior in free amino acid composition, and valine was revealed next order. In minerals, contents of Ca were from 1,306.1mg% to 1,923mg%, that of K were higher than others patricualy as 1,266.9∼1,216.0mg%. The contents of minerals were existence in order of Mg, P, Na, Fe, Mn, Zn and Cu, abundantly. Total vitamin C were present of 391.0∼336.1mg%, and the contents were decreased as delayed as harvesting period despite of the plentiful content. In fatty acid composition of ginseng leaf, the palmitic acid content was as 40% higher than other fatty acids, remarkably.

Comparison of Proximate Composition, Mineral Nutrient, Amino Acid and Free Sugar Contents of Several Panax Species (인삼(Panax)속 식물의 일반성분, 무기성분, 아미노산 및 유리당 함량조성)

  • Chemical components of Panax (P) species were compared. p. species used were Korean white ginseng, Korean, Chinese and Japanese red ginseng (P ginseng), American and Canadian ginseng (P. quinquefolium) , and sanchl ginseng (P. notoginseng). No significant difference in the proximate contents was observed among P. species. Ash, crude lipld and total sugar contents in root of P. notoginseng were found to be relatively lower than those of P. ginseng and P. quinquefolium, but the contents of crude protein and crude fiber were similar among those ginsengs. Mineral nutrient con tents showed a little difference among ginseng species. Total nitrogen contents were slightly higher in P. ginseng than P. quinquefolium and P. notoginseng and Fe and Cu were lower in Chinese and Japanese red ginsengs. Kinds and compositions of amino acids were similar but contents of amino acids were different among ginseng species. Total amino acid contents were 76.3∼83.9 mg/g in P. ginseng 53.8∼60.4 mg/g in p. quinquefolium and 54.9 mg/g in P notoginseng. Free sugar contents were lower in P. notoginseng than P. ginseng or P. quinquefolium. Sucrose accounted for 90∼92% of total free sugar contents with relatively high content in white ginsengs, while sucrose and maltose were 32-36% and 55∼60%, respectively, in red ginseng.

  • Cell-free extracts of Streptomyces neyagawaensis SL-387 grown on a chemically defined medium supplemented with DL-3-amino-3-phenylpropionic acid (APP) produced N-acetyl-APP (Ac-APP) in the presence of APP and acetyl coenzyme A. The APP obtained by acid hydrolysis of the Ac-APP was D-configuration: $[\alpha]_D+6.5^{\circ}(H_2O)\;at\;20^{\circ}C$, optical purity 92% enantiomeric excesses (ee). These results suggest that an N-acetyltransferase exists in the cell-free extract as a novel enzyme with specificity for D-APP.
