The Effects of Yukgunja-tang on the Change of Weight and Serum level in Mice Fed High Fat Diet (육군자탕이 비만마우스의 체중 및 혈청함량변화에 미치는 영향)

  • This experimental study was designed to investigate the effects of Yukgunja-tang(YGJT) on the change of weight and serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level in obese mice induced by high fat diet. I fed normal group fed normal diet and administered DDW 0.2㎖ during 7 weeks, control group fed high fat diet and administered DDW 0.2㎖ during 7 weeks, sample A group fed high diet and administered YGJT 300mg/kg 0.2㎖ during 7 weeks, sample B group fed high diet and administered YGJT 500mg/kg 0.2㎖ during 7 weeks. The results were as follows ; 1. Sample A and Sample B were significantly decreased body weight(4weeks) and serum free fatty acid level in comparison with control group. 2. Sample A was significantly decreased body weight(7weeks), serum total cholesterol level and serum total lipid level, but significantly increased serum HDL-cholesterol level in comparison with control group. 3. Sample A was decreased serum LDL-cholesterol level, serum triglyceride level and serum phospholipid level in comparison with control group. 4. Sample B was increased serum HDL-cholesterol level in comparison with control group. 5. Sample B was decreased body weight(7weeks), serum total cholesterol level, serum LDL-cholesterol level, serum triglyceride level, serum total lipid level and serum phospholipid level in comparison with control group. According to above results, I suggest YGJT is able to be used for the herbal medication of obesity.

Experimental Effects of Ijin-tang add Atractylodis Rhizoma and Atratcylodis Macrocephalae Rhizoma Extract in Obese Mice Induced by High Fat Diet (이진탕가창출백출(二陳湯加蒼朮白朮)이 고지방 식이로 유발된 생쥐의 비만증에 미치는 영향)

  • This experimental study was designed to investigate the effects of Ijin-tang add Atractylodis rhizoma and Atratcylodis macrocephalae rhizoma (IJTAA) on the change of weight and serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid and phospholipid level in obese mice induced by high fat diet. Experimental groups were as follows ; Normal group was fed normal diet and administered distilled water during 7 weeks, Control group was fed high fat diet and administered distilled water during 7 weeks, Sample A group was fed high fat diet and administered IJTAA 500 ㎎/㎏/day/mouse during 7 weeks, Sample B group was fed high fat diet and administered IJTAA 700 ㎎/㎏/day/mouse during 7 weeks. The results were as follows ; 1. In Sample A group and Sample B group, There were significantly decreased in body weight, serum total cholesterol level, serum triglyceride level, serum free fatty acid level, serum total lipid level and serum phospholipid level in comparison with Control group. 2. In Sample A group and Sample B group, There were significantly increased in serum HDL-cholesterol level in comparison with Control group. 3. In Sample A group and Sample B group, There were decreased in serum LDL-cholesterol level in comparison with Control group. According to above results, I suggest IJTAA is able to be used for managing obesity by controllong body weight, serum total cholesterol level, serum triglyceride level, serum free fatty acid level, serum total lipid level and serum phospholipid level.

Changes of Fatty Acid Compositions in Brain Phospholipids of Developing Chicken Embryos (발생중인 닭의 배자에서 뇌 조직내 인지질의 지방산 조성 변화)

  • This study was to investigate the effects of dietary linoleic acid(18:2\omega6, LA) and aipha-linolenic acid(18:3\omega3. \alpha-LNA) levels on brain development and fatty acid compositions of various lipid classes in the chicken embryo brain tissues. Thirty two ISA Brown layers, 52 weeks-old, were divided into four groups. Birds of each group were given corn-soybean meal based diets added with 1) safflower oil 8%, 2) safflower oil 6% + perilla oil 2%, 3) safflower oil 2% + perilla oil 6%, or 4) perilla oil 8%. Mter 15 days fed the diets. the layers were artificially inseminated to obtain fertile eggs. During the incubation. embryonic brains were sampled at 15th and 21st days. Fatty acid contents were quantitated by using heptadecanoic acid (17:0) as an internal standard. No significant differences in brain weight and in contents of various lipids such as phospholipid. triglyceride, cholesterol. cholesterol ester and free fatty acid in the tissues were found among the dietary groups (P<0.05). The ratios of AA/LA in the brain lipid classes were lowered as the dietary levels of perilla oil were increased. Higher LA was found in phosphatidylcholine(PC) than arachidonic acid (20:4\omega6. AA), meanwhile the level of LA was less than AA in phosphatidylethanolamine(PE). Docosahexaenoic acid(22:6\omega3, DHA) was the* major fatty acid in the tissue and its content in PE was 2.5~3 times higher than in PC. DHA level in the phospholipid reached at a peak (1.7~1.8 mg/brain) in dietary groups added with 6% or 8% perilla oil. suggesting that no more increase in that fatty acid level in the brain tissue could be obtained by consuming more \alpha-LNA, the major precursor of DHA.

Hypoglycemic Effect of Fractions of Cassia tora Extract in Streptozotocin-Induced Diabetic Rats (결명자 분획물이 당뇨 유발 흰쥐의 혈당에 미치는 영향)

  • The hypoglycemic effect of fractions of methanol extract of Cassia tora was investigated in diabetic rats. Sprague-Dawley male rats weighing 180∼230 g were divided into 6 groups. Diabetes was induced in the male rats by intravenous injection of streptozotocin at a dose of 45 mg/kg dissolved in citrate buffer (pH 4.5). The diabetic animals then had plasma glucose concentration of above 300 mg/㎗. Fractions of methanol extract of Cassia tora were administered orally into the diabetic rats for 14 days after streptozotocin injection. The food intake and body weight gain were monitored and plasma levels of glucose, triglyceride, free fatty acid, HDL-cholesterol, cholesterol and ALT activity were determined. Levels of glycogen, cholesterol and triglyceride in liver were analysed. Weights of liver and kidney were lighter in all the fraction-administered groups than streptozotocin-control group. Plasma glucose level was significantly decreased by the administration of butanol fraction at 14 days. Plasma triglyceride levels were lower in chloroform, butanol and H$_2$O fraction-administered groups compared to streptozotocin-control group. Plasma cholesterol and HDL-cholesterol levels were not significantly different in all groups. Administrations of each of the four fractions have decreased plasma free fatty acid level, ALT activity and liver triglyceride levels in the diabetic rats. Liver glycogen levels in chloroform, butanol and H$_2$O fraction-administered groups were higher than streptozotocin-control group. It is suggested from the results that butanol fraction of methanol extract of Cassia tora may contain the antihyperglycemic compounds.

Age Related Changes of Microsomal Fatty Acid Composition and Lipid Peroxidation in 2-acetylaminofluorene Treated rats. (2-Acetylaminofluorene의 투여와 나이에 따른 쥐의 세포막 지방산 조성 및 지질과산화물 생성의 변화)

  • For studying the effect of different dietary fats on carcinogenesis, fatty acid composition of membrane and lipid peroxidation were measured. Male Sprague-Dawley rats were fed on diet containing 15%(w/w) beef tallow or soybean oil. A single dose of 50 mg 2-AAF/kg B.W. was injected i.p. in each diet group 10 times. Rats were sacrifled after 1, 5, 10, and 15 weeks from the first injection. By 2-AAF injection, !ipid peroxidation increased slightly compared to control group. The rats fed on different fats had similar MDA production and those fed on soybean oil had slightly higher free radical concentration measured by ESR. In young rats, iipid peroxidation level was high and hydroxy radical production was higher in soybean oil group than in beef tallow group. With age, the lipid peroxidation values were decreased initially then increased. The fatty acid composition in microsomal membrane was reflected by dietary fatty acid composition. In soybean oil group, monoenoic acid was lower and polyunsaturated fatty acid was higher than beef tallow group. Linoleic acid contents showed the most discrepancy among groups. By 2-AAF treatment, iinoleic acid content and unsaturation index increased in soybean oil group. But in beef tallow group, there was no difference in fatty acid contents.

Evaluation of Nutrients during Rack and Bag Culture or Suspended Culture of Pacific Oyster Crassostrea gigas (부유망식과 수하식 양성방법에 따른 참굴(Crassostrea gigas)의 영양상태)

  • The mineral contents, and free amino acid, and fatty acid composition of rack and bag or suspended cultured oysters Crassostrea gigas were compared. The moisture content did not change much from 82.8% to 86.3% in the different culture methods. The glycogen level was low 4.7% during rack and bag culture but high 7.2% during suspended culture. The sodium level was the highest during suspended culture, while copper and zinc level were highest during rack and bag culture. The taurine level was 1,385-1,375 mg/100 g in oyster cultured using the rack and bag method, but that in the market sample was lowest at 1,015.8 mg/100 g. Hydroxyproline, glutamic acid, proline, glycine, and alanine were detected at high levels in the experimental oysters. The polyunsaturated fatty acid content was similar among samples. The docosahexaenoic acid level was 14% with rack and bag culture and 12% with suspended culture, whereas the eicosapentaenoic acid level was 15% with rack and bag culture and 20% with suspended culture. The atherogenic index (AI), thrombogenicity index (TI), and hypocholesterolemic/ hypercholesterolemic (h/H) ratio were important factors in the nutritional evaluation. The AI and TI values were 0.5-0.2, and the h/H ratio were 2.0 and 2.4 with the rack and bag and suspended culture, respectively.

Physicochemical Characteristics of Traditional Doenjang with added Lentinus edodes (표고버섯을 첨가한 전통된장의 이화학적 특성)

  • Traditional doenjang with Lentinus edodes added at a concentration of 0, 3, 5 and 10% (w/w) was prepared and its physicochemical characteristics were analyzed during fermentation for 6 months. The moisture content, salt level and pH for doenjang were decreased with increasing addition of Lentinus edodes powder, but crude protein content was increased. Among the free sugars, glucose, galactose, arabinose, sucrose and maltose were identified and quantified, with the first two being the major free sugar. Minerals were detected in the order of Na> K> P> Ca> Mg> Fe while the content of Cu, Mn and Zn exhibited little change. The contents of each amino acid of traditional doenjang with added Lentinus edodes, glutamic, leucine and aspartic acids, had a much higher level. In fatty acid compositions, palmitic, stearic, oleic and linoleic acids were analyzed, with oleic acid being the most abundant fatty acid at $56.4{\pm}0.8\sim57.5{\pm}0.5%$ in all the tested samples. GMP and IMP contents of nucleotides and their related compounds were extremely high. It was thought that these substances contributed to the sweet taste of traditional doenjang with added Lentinus edodes. These results showed the applicability and high quality of traditional doenjang with added Lentinus edodes.

Quality Characteristics of Minced Ginger During Storage (생강다대기의 저장 중 품질특성)

  • The effects of vinegar, alcohol, and vitamin C on the color, microorganism count, volatile flavor components, free sugar level, free amino acid concentration, and free fatty acid level of minced ginger were investigated during storage for 17 weeks at $10^{\circ}C,\;20^{\circ}C\;or\;30^{\circ}C$. Bacterial levels remained unchanged during storage and mold was not initially detected. Yeast levels in minced ginger were $5{\times}10^1$ CFU/g initially, and yeast was not detected after 2 weeks. The color values increased during storage at high temperature. The volatile flavor component levels decreased during high-temperature storage. The free sugars of minced ginger were glucose, sucrose, and fructose. Sugar levels did not vary greatly with storage temperature. Free amino acid content decreased during high-temperature storage, and glutamine, valine, alanine, asparagine, tyrosine, and leucine were detected. Free fatty acid content increased during storage and the ratio of unsaturated to saturated fatty acids was 3:7.

Features of Waste Water form By-Product Silk Treatment and Composition of Extracted Lipid (부잠폐수의 성상과 유출 유지의 성분)

  • The features of waste water from by-product silk treatment of silk reeling process were investigated and the lipid extracted from waste water was analysed. The COD of waste water from by-product silk treatment was at the level of 605 mgO/$\ell$ Total Dissolved Solid Particles 2,335mg/$\ell$and Total Suspended Solid Particles 2,123mg/$\ell$. The lipid extracted from the waste water from by-product silk treatment was composed of triglyceride 76.8%, free fatty acids 12.5%, diglyceride 5.7% and free sterol 5.0%. In fatty acid composition of lipid, the content of loeic acid, linoleic and linolenic acid was 64.93%, whereas that of palmitic acid was 29.39% and stearic acid 4.93%.

Effect of Vitamin E and Selenium on the Lipid Composition in Serum and Liver of Rats (비타민E와 셀렌이 흰쥐의 혈청(血淸) 및 간장(肝臟)중의 지질성분(脂質成分)에 미치는 영향(影禦))

  • This experiment was undertaken to investigate ana to compare the effect of vitamin E and selenium on the lipid composition in serum and liver of rats. Net weight gain and food efficiency ratio were elevated by supplementation with selenium and/or vitamin E. The weight of kidney, heart and lung were not affected by the diets. However, liver weight was slightly decreased by supplementation with vitamin E and/or selenium and spleen weight was significantly decreased only by vitamin I supplementation. No differences in serum level of total lipid were found in all groups. Serum cholesterol level was significantly decreased, but the levels of triglyceride and free fatty acid in serum were increased by vitamin E supplementation. Contents of crude lipid, cholesterol, free fatty acid, and triglyceride in liver were significantly decreased by supplementation with vitamin E and/or selenium. In fatty aicd composition of liver, the percentage of linoleic acid(18:2) was the lowest in the selenium-Supplemented group and the significantly lower percentage of arachidonic acid(20:4) was found in the vitamin E and/or selenium-supplemented group in comparision to the deficient basal group.
